Identification of Neuroactive Constituents of the Ethyl Acetate Fraction from Cyperi Rhizoma Using Bioactivity-Guided Fractionation

  • Cyperi Rhizoma (CR), the rhizome of Cyperus rotundus L., exhibits neuroprotective effects in in vitro and in vivo models of neuronal diseases. Nevertheless, no study has aimed at finding the neuroactive constituent(s) of CR. In this study, we identified active compounds in a CR extract (CRE) using bioactivity-guided fractionation. We first compared the anti-oxidative and neuroprotective activities of four fractions and the CRE total extract. Only the ethyl acetate (EA) fraction revealed strong activity, and further isolation from the bioactive EA fraction yielded nine constituents: scirpusin A (1), scirpusin B (2), luteolin (3), 6'-acetyl-3,6-diferuloylsucrose (4), 4',6' diacetyl-3,6-diferuloylsucrose (5), p-coumaric acid (6), ferulic acid (7), pinellic acid (8), and fulgidic acid (9). The activities of constituents 1-9 were assessed in terms of anti-oxidative, neuroprotective, anti-inflammatory, and anti-amyloid-${\beta}$ activities. Constituents 1, 2, and 3 exhibited strong activities; constituents 1 and 2 were characterized for the first time in this study. These results provide evidence for the value of CRE as a source of multi-functional neuroprotectants, and constituents 1 and 2 may represent new candidates for further development in therapeutic use against neurodegenerative diseases.

Phenolic Acids and Antioxidant Activities of Wild Ginseng (Panax ginseng C. A. Meyer) Leaves

  • The compositions and antioxidant activities of tree and hydrolyzed phenolic acids, which are aglycones of esterified phenolic acids, in wild ginseng leaves were investigated. The contents of tree and hydrolyzed phenolic acids in the wild ginseng leaves were $422.4\;{\pm}\;3.5$ and $319.6\;{\pm}\;5.7\;mg/100\;g$, respectively, as gallic acid equivalents. Free phenolic acids were composed of 55.3% benzoic acid derivatives and 44.6% phenylpropanoids. The major constituents of free phenolic acids in the ginseng leaves were syringic (139.4 mg/l00 g) and sinapic (131.2 mg/100 g) acids. On the other hand, hydrolyzed phenolic acids in the ginseng leaves were mainly composed of caffeic (59.4 mg/100 g), ferulic (49.5 mg/100 g), and p-coumaric (33.8 mg/100g) acids. Phenylpropanoid content was higher (82.7%) than benzoic acid derivatives (17.3%). $IC_{50}$ values of DPPH radical scavenging activity were $10.2\;{\mu}g/mL$ for tree phenolic acids and 8.0 mg/mL for hydrolyzed phenolic acids, as gallic acid equivalents. Hydrolyzed phenolic acids also exhibited higher hydroxyl and superoxide radical scavenging activities than free phenolic acids did. These results indicated that the antioxidant activities of the wild ginseng leaves were correlated more closely with phenylpropanoid contents than with total amount of phenolics.

Biosynthesis of Two Flavones, Apigenin and Genkwanin, in Escherichia coli

  • The flavonoid apigenin and its O-methyl derivative, genkwanin, have various biological activities and can be sourced from some vegetables and fruits. Microorganisms are an alternative for the synthesis of flavonoids. Here, to synthesize genkwanin from tyrosine, we first synthesized apigenin from p-coumaric acid using four genes (4CL, CHS, CHI, and FNS) in Escherichia coli. After optimization of different combinations of constructs, the yield of apigenin was increased from 13 mg/l to 30 mg/l. By introducing two additional genes (TAL and POMT7) into an apigenin-producing E. coli strain, we were able to synthesize 7-O-methyl apigenin (genkwanin) from tyrosine. In addition, the tyrosine content in E. coli was modulated by overexpressing aroG and tyrA. The engineered E. coli strain synthesized approximately 41 mg/l genkwanin.

Phenolic Compounds Content and Tyrosinase Inhibitory Effect of Unripe Apple Extracts

  • This study consisted of a quantitative analysis of five phenolic ingredients in differently sized unripe apple extracts, and their tyrosinase inhibitory effects were examined. In the HPLC analysis of phenolic ingredients, small ($4{\pm}1\;g$ per one) unripe apple extracts were observed to have significantly higher quercetin content than larger ($8{\pm}1\;g$ per one) unripe apple and ripe apple extracts. The amount of catechin, epicatechin, p-coumaric acid and chlorogenic acid contents were similar in both the small and large unripe apple extracts. For the results of the tyrosinase assay, small unripe apple extracts provided a potent tyrosinase inhibitory effect, showing 89.2% at 1000 ppm. The tyrosinase inhibitory effects of large unripe apple and ripe apple extracts were weaker than those of the small unripe apple extract. These results suggest that the small unripe apple extract could be useful for de-pigmenting material, while quercetin could be responsible for the potent tyrosinase inhibitory properties of small unripe apple extracts.

Inhibitory Activity against Helicobacter pylori of Isolated Compounds from Pinus koraiensis Siebold et Zucc Leaves

  • A phenol substance was extracted from Pinus koraiensis Siebold et Zucc leaf extracts and its biological efficacy was measured. The highest content of the phenol substance contained in Pinus koraiensis Siebold et Zucc leaves was 13.5 mg/g, which was obtained when it was extracted with 80% ethanol. At a concentration of 200 mg/mL, the phenolic substances extracted with 80% ethanol and water showed antimicrobial activities against Helicobacter pylori, producing clear zones of 10 and 12 mm diameter, respectively. Pinus koraiensis Siebold et Zucc. leaf extracts were separated using a Sephadex LH-20 column and 4 fractions were obtained (fractions A-D). Fractions C and D showed the greatest inhibitory activity against Helicobacter pylori producing 10.1 and 12.3 mm clear zones, respectively. These two fractions were purified using a Sephadex LH-20 and MCI-gel column ($H_2O{\rightarrow}100%$ ethanol). Purified compounds A and B were identified as syringic acid and compound C was identified as p-coumaric acid based on $^1H$-nuclear magnetic resonance (NMR), $^{13}C$-NMR, and fast atom bombardment mass spectrometry spectra. When two or more purified compounds were mixed, a synergistic effect of anti-Helicobacter pylori activity was evident. This result indicates that extracts of Pinus koraiensis Siebold et Zucc leaves could be considered a functional food because of their high antimicrobial properties.

Profiling of the leaves and stems of Curcuma longa using LC-ESI-MS and HPLC analysis

  • Curcuma longa is a plant belonging to the genus Curcuma and is distributed across various Asian regions. This plant is widely known for its rhizomes, which possess a variety of pharmacological properties. However, although the leaves and stems of this plant also contain several health-promoting secondary metabolites, very few studies have characterized these compounds. Therefore, our study sought to quantify the secondary metabolites from the leaves and stems of Curcuma longa L. (LSCL) using liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) and high-performance liquid chromatography (HPLC). Our LC-ESI-MS analyses detected twenty-one phenolic compounds in the LSCL, among which fifteen compounds were detected via HPLC analysis. Four compounds, namely vanillic acid (0.129 mg/g), p-coumaric acid (0.431 mg/g), 4-methylcatechol (0.199 mg/g), and afzelin (0.074 mg/g) were then quantified. These findings suggest that LSCL is rich in secondary metabolites and holds potential as a valuable resource for the development of functional and nutritional supplements in the future.

우리 나라에서 수집한 새포아풀의 분류 및 특성 (Classification and Characteristics of Annual Bluegrass(Poa Annua L.) Collected from Golf Courses in Korea)

  • This study was carried out to get better understandings about morphological, ecological, and genetical characteristics of annual bluegrass collected from different golf courses in Korea and eventually to establish a successful control strategy. Twenty five local lines of annual bluegrass collected from 20 golf courses in Korea were classified into annual or perennial type on the basis of morphological characteristics. Twelve local lines showing obvious morphological differences were selected and then genetically assessed using RAPD analysis. Classification of the 12 local lines through RAPD analysis were considerably similar to that determined by both of morphological differences and phenotype. Responses of the two types of annual blugrass to herbicides were also examined. Shoot growth of annual bluegrass was significantly suppressed by flazasulfuron and the annual type was more susceptible than perennial type, regardless of flazasulfuron concentrations used. By pendimethalin treatment, there was no clear difference in susceptibility between the two types of annual bluegrass. However, by the treatment of dithiopyr, annual type was more sensitive than perennial type in both shoot and root growth. Nine tree species were screened to detect their allelopathic potential on turfgrasses and annual bluegrass. Acacia (Robinia pseudo-acacia) leaves showed selective inhibition in the shoot and root growth as well as their seed germination when treated with 2% and 10%(v/v) of the extract. However, the other leaf extracts except acacia inhibited non-selectively the growth of three turfgrass species such as bentgrass, perennial ryegrass and zoysiagrass and annual bluegrass. The PAL activities of annual bluegrass increased at 24 h after treatment of acacia leaf extract and peaked at 36 h and then decreased till 60h. The highest PAL activity was observed at 36h after treatment of 10%. The highest activity of CA4H in annual bluegrass was observed at 2h after treatment of acacia extract and the level was 4 times greater than that of the control. The phenolic acids such as p-coumaric acid, salicylic acid and ferulic acid were increased with the treatment of acacia leaf extract. The chloroplast membrane and cell wall of annual bluegrass were destroyed by treatment of acacia leaf extract and its inner materials were released. The membranes in annual bluegrass cells might be destroyed by phytotoxic compounds from acacia leaf extract.

토마토 유묘생육 및 양분흡수에 관한 목초액의 영향 (Effect of Wood vinegar on Tomato Seedling Growth and Nutrient Uptake)

  • 유기ㆍ자연농업 등 환경친화적 농업을 수행하는 농가에서 많이 사용하고 있는 목초액의 화학성과 작물생육에 대한 반응을 검토하여 유기ㆍ자연농업자재의 효율적인 이용방안에 대한 기초자료를 얻고자 본 연구를 수행하였다. 목초액의 화학성을 조사한 결과 비료적 가치는 크지 않았다. 특히 농가에서는 일반적으로 사용되는 500∼1000배 목초액은 별도의 비료자원을 사용하지 않고 전적으로 목초액에 의존할 경우 정상적인 작물생육에 요구되는 양분을 충족시키기는 근본적으로 어려울 것으로 사료된다. 목초액 농도를 달리한 조건에서 토마토 유묘의 생육 및 양분 흡수형태를 조사한 결과 500배 희석까지 목초액처리 농도가 높을수록 뿌리 생육량은 증가하였으나 지상부 생육은 억제되었다. 또한, 뿌리와 경엽 중의 양분흡수는 음이온인 질산태 질소와 인산 흡수량이 현저히 감소하는 반면에 양이온인 K, Ca, Mg, Na의 흡수량은 증가하였다. P-coumaric acid, chlorogenic acid, ferulic acid 및 acetic acid를 농도를 달리하여 처리하고 토마토 유묘를 재배한 결과 뿌리의 생육은 chlorogenic acid 또는 ferulic acid 처리로 증가되었고, ferulic acid의 경우 저농도인 10-4M에서는 지상부 및 지하부의 생육이 모두 양호하였다. 따라서 목초액을 시용할 때 도장이 억제되고 건전한 작물생육이 유지된다고 주장하는 것은 목초액 시용에 따른 작물의 지하부 생육향상에 기인한 것으로 판단되었다. 페놀물질간의 상승작용 및 길항작용에 따라 작물생육에 대한 효과는 다를 수 있으므로 다양한 종류의 페놀물질이 포함된 목초액을 작물재배에 사용할 경우 효과발현 정도는 상이할 것으로 사료된다. 한편, 목초액은 사용방법 측면에서 경엽처리 보다는 토양관주 방법으로 영농에 이용하면 작물의 근활력 증진 및 작물생육을 조절할 수 있는 대체농자재로서 활용이 가능할 것으로 사료된다.

그라비올라(Annona muricata) 잎 추출물 및 분획물의 항산화 활성과 성분 분석 (Antioxidative Effects and Component Analysis of Graviola (Annona muricata) Leaf Extract/Fractions)

  • 본 연구에서는 그라비올라(Annona muricata) 잎으로부터 50% 에탄올 추출물, 에틸아세테이트 분획 및 아글리콘 분획을 제조하였고 이들 추출물/분획에 대하여 항산화 활성을 평가하였다. 1,1-Phenyl-2-picrylhydrazyl (DPPH) 라디칼 시험법을 이용한 자유라디칼 소거 활성, 루미놀 발광법을 이용한 총 항산화능 및 $^1O_2$ 소광 효과를 평가하였다. 50% 에탄올 추출물, 에틸아세테이트 분획 및 아글리콘 분획의 라디칼 소거 활성($FSC_{50}$)은 각각 45.6, 29.8 및 $18.0{\mu}g/mL$이었고, 총 항산화능($OSC_{50}$)은 4.4, 1.1 및 $2.8{\mu}g/mL$이었다. 에틸아세테이트 분획의 총 항산화능은 수용성 항산화제로 잘 알려진 L-ascorbic acid ($1.5{\mu}g/mL$)보다 높은 항산화능을 나타내었다. $^1O_2$ 소광 상수 실험 결과, 에틸아세테이트 및 아글리콘 분획은 비교물질로 사용된 L-ascorbic acid와 유사한 활성을 보여주었다. $^1O_2$으로 유도된 적혈구 세포 손상에 있어서, 그라비올라 잎 50% 에탄올 추출물은 농도 의존적($5-50{\mu}g/mL$)으로 세포보호 활성을 나타내었다. 실험에 사용된 그라비올라 잎 추출물의 에틸아세테이트 분획과 아글리콘 분획에 대하여 TLC 및 HPLC를 이용한 성분 분석을 수행하였다. 에틸아세테이트 분획에서는 rutin (quercetin-3-O-rutinoside), kaempferol-3-O-neohesperidoside, nicotiflorin (kaempferol-3-O-rutinoside), p-coumaric acid을 확인하였다. 아글리콘 분획에서는 kaempferol이 존재함을 확인하였다. 이상의 결과들은 그라비올라 잎 추출물이 항산화 화장품 원료로서 응용 가능성이 있음을 시사한다.

칼라꼬마감자와 일반꼬마감자의 영양성분 및 이화학적 특성 (Comparison of Nutritional Components and Physicochemical Properties of Small Colored Potatoes and Small Regular Potatoes)

  • 본 연구에서는 칼라꼬마감자의 영양성분 등의 품질특성을 분석하였고, 항산화활성 효과를 평가하였다. 칼라꼬마감자 100 g(wet weight basis)중에는 수분 79.7%, 탄수화물 17.9%, 조단백질 1.0%, 조지방 0.1%, 조회분 1.3%가 함유되어 있으며, 탄수화물 중 총 식이섬유소 함량은 11.45%이었다. 총 식이섬유의 함량은 4.0%로 나타났다. 또한 칼라꼬마감자 100 g의 총 열량은 337.3 kcal로 분석되었다. 칼라꼬마감자의 구성아미노산 중 asparagine (1,514.3 mg/100g)과 glutamic acid(1,240.3 mg/100g)함량이 가장 높은 함량을 차지하고 있는 것으로 나타났으며, 무기질 함량은 칼륨이 약 1,454.7 mg으로 가장 함량이 높았으며, 인(245.3 mg), 마그네슘(82.8 mg), 칼슘(62.1 mg)순이었다. 미량영양소인 철분(28.1 mg), 망간(28.1 mg), 구리(3.4 mg) 및 아연(2.5 mg)이 함유되어 있었다. 칼라꼬마감자 추출물의 페놀 함량은 chlorogenic acid가 $3,730.60{\pm}0.62mg/100g$로서 가장 많은 양이 함유 되어 있었고 catechin, caffeic acid, gallic acid의 함량이 높은 것으로 나타났으나, p-coumaric acid, ferulic acid는 확인되지 않았다. 칼라꼬마감자의 우수한 영양성분 및 항산화 활성을 나타내는 성분 및 그 활성에 대한 연구가 추가로 필요할 것으로 생각된다. 칼라꼬마감자의 우수한 생리활성을 나타내는 성분에 대해서는 더욱 연구가 필요할 것으로 사료되며, 이러한 결과로 보아 칼라꼬마감자의 영양성분 및 항산화 활성 천연소재로 이용할 수 있을 것으로 사료된다. 따라서 칼라영양성분 및 항산화감자 추출물 제조시 기능성 향상을 위한 추출공정의 개선 및 최적화 등의 추후 연구가 필요한 것으로 사료된다.
