가잠의 연화병 바이러스에 관한 연구 II. 연화병 바이러스 Polypeptide의 성상 (Studies on Flacherie and Ina-flacherie Viruses of the Silkworm, Bambyx mori II. Some Properties of Polypeptide of Flacherie Virus)

  • 1. 연화병 바이러스의 단백질을 SDS-polyacrylamide gel 전기영동으로 분석한 결과 분자량 37,500(FP I), 30,500(FP II) 및 26,500(FP III)에 상당하는 3종류의 polypeptide가 얻어졌으며, 이들의 비율은 FP I이 6.6%, FPII가 25.0% 및 FP III가 68.4%였다. 2. 연화병 바이러스의 단백질을 amino산 분석을 행한 결과, 17종의 amino산이 분리되었으며, 이 amino산 조성은 타곤충 바이러스의 amino산 조성과 유사하였다.

사람 골모세포 FOB에서 아미노산 수송계 L의 발현 및 역할 (Expression and Role of the System L Amino Acid Transporter in FOB Human Osteoblast Cells)

  • 사람의 정상 골모세포 FOB에서 아미노산 수송계 L의 발현 및 이들 수송계 L을 통한 아미노산 수송특성을 밝히기 위하여, FOB 세포에서 RT-PCR, western blot 분석 및 아미노산 uptake 실험 등을 수행하여 다음과 같은 결과들을 얻었다. FOB 세포에서 아미노산 수송계 L의 두 아형인 LAT1, LAT2및 그들의 보조인자 4F2hc의 발현을 확인할 수 있었다. FOB 세포에서 $_{L}-leucine$의 수송은 $Na^+$-비의존적이었다. FOB 세포에서 $_{L}-leucine$의 수송은 아미노산 수송계 L의 선택적 억제제인 BCH에 의해 완전히 차단되었다. FOB 세포에서 여러 아미노산들에 의한 L-leucine수송억제 실험결과는 Xenopus oocyte에서 시행되어 보고되어진 LAT1과 LAT2 수송억제 실험 결과의 특성을 모두 포함하였다. 본 연구의 결과로 사람의 정상 골모세포주인 FOB에서 는 세포성장 및 증식을 위한 중성 아미노산의 수송에 아미노산 수송계 L의 두 아형인 LAT1과 LAT2가 중요한 역할을 하고 있음을 확인할 수 있었다.

Influence of the Structural Characteristics of Amino Acids on Direct Methylation Behaviors by TMAH in Pyrolysis

  • Direct methylation behaviors of 20 amino acids with tetramethylammonium hydroxide (TMAH) were studied under diluted conditions with silica. Amino acid concentration was controlled by dilution with silica ($SiO_2$) and the molar ratios of amino acid/silica were 0.20, 0.50, and 2.0. The molar ratios of amino acid/TMAH (0.51 - 4.64) also varied. It was found that arginine, asparagine, aspartic acid, cysteine, glutamic acid, and glutamine did not generate any directly methylated pyrolysis products, whereas alanine, glycine, isoleucine, leucine, methionine, phenylanaline, valine, and proline generated all the directly methylated pyrolysis products. Tri- and tetra methylated products of lysine consisted of two types. Histidine and threonine hardly generated the partly methylated products. Mono- and dimethylated products of serine, tryptophan, and tyrosine were not observed. Relative intensities of the methylated products varied with the amino acid concentration, TMAH concentration, and pyrolysis temperature. Direct methylation behaviors of amino acids were explained by the structural characteristics of amino acids.

Regulation of 3-Deoxy-D-arabinoheptulosonate-7-phosphate (DAHP) Synthase of Bacillus sp. B-6 Producing Phenazine-1-carboxylic acid

  • The 3-Deoxy-D-arabinoheptulosonate 7-phosphate (DAHP) synthase is the first enzyme of aromatic amino acid-, folic acid-, and phenazine-1-carboxylic acid biosynthetic pathways. DAHP synthase of Bacillus sp. B-6 that produces phenazine-1-carboxylic acid was feedback inhibited by two intermediary metabolites of aromatic amino acid biosynthetic pathways, prephenate and chorismate, but not by other metabolites, such as anthranilic acid, shikimic acid, p-aminobenzoic acid, and 3-hydroxyanthranilic acid. DAHP synthase of Bacillus sp. B-6 was not inhibited by end products, such as aromatic amino acids, folic acid, and phenazine-1-carboxylic acid. The inhibition of DAHP synthase by prephenate and chorismate was non-competitive with respect to erythrose 4-phosphate and phosphoenolpyruvate. Prephenate and chorismate inhibited 50% of the DAHP synthase activity at concentrations of $2{\times}10^{-5}\;M$ and $1.2{\times}10^{-4}\;M$, respectively The synthesis of DAHP synthase of Bacillus sp. B-6 was not repressed by exogenous aromatic amino acids, folic acid, and phenazine 1-carboxylic acid, single or in combinations.

  • Dyeing properties of a series of 4-amino-4'-fluorosulfonylazobenzene disperse dyes on poly(ethylene terephthalate) (PET) were investigated. Build-up properties and color properties on PET were examined. In particular, the 4-aminoazobenzene dyes containing a nitro group instead of a fluorosulfonyl group at 4'-position were also synthesized in order to compare their dyeing properties on PET with that of 4'-fluorosulfonyl analogues.

  • Timmis반응을 이용하여 1,3-dimthyllumazine유도체를 합성하였다. 4-Amino-1,3-dimethyl-5-nitrosouracil(1)을 2,4-pentanedione, ethyl cyanoacetate, 그리고 ethyl acetoacetat와 반응시켜 6-acetyl-1,3,7-trimethyllumazine (2), ethyl 7-amino-1,3-dimethyllumazine-6-carboxylate (4), ethyl 1,3,7-trimethyl-lumazine-6-carboxylate (5)를 좋은 수율로써 합성하였다. 화합물 2,4,5의 6-아세틸 및 에스테르기에 대한 곁사슬 반응으로 다양한 1,3-dimethyllumazine 유도체들이 합성되었다. 합성된 화합물의 구조 및 물리적 특성은 NMR, UV, IR스펙트럼, 그리고 원소분석에 의하여 확인하였다.

Synthesis and Characterization of Fluorinated Polyimides for Optical Waveguide Application

  • Fluorinated polyimides with high thermal stability, low optical absorption loss in the optical communication wavelengths of 1.3 and 1.55㎛, and low birefringence and water absorption have been investigated for optical waveguide applications. These polyimides were prepared from 2,2-bis(3,4-dicar-boxyphenyl) hexafluoropropane dianhydride (6FDA) with 1,4-bis-(4-amino-2-trifluoromethyl-phenoxy) tetrafluorobenzene (ATPT) , 1,4-bis-(4-amino-2-trifluoromethyl-phenoxy) benzene (ATPB) , and 1,3-bis-(4-amino-2-trifluoromethyl-phenoxy ) 4,6-dichlorobenzene (ATPD ), respectively. The polyimides obtained had glass transition temperatures that ranged from 260 to 280$\^{C}$, and low water absorption less than 0.4% and low optical loss (> 0.2 dB/cm) at 1.3 and 1.55㎛. Birefringency values of polymer films ranged from 0.0041 to 0.0066.

미생물을 이용한 환경오염원의 분해에 관한 연구 II (Studies on the Decomposition of Environmental Pollutants by Utilizing Microorganisms)

  • 1. When Chong Ju and Chung Ju soils possessing different physicochemical properties were treated with 500 ppm of TOK and incubated in flooded anaerobic condition for 2, 4, and 6 months, respectively, they produced 4-Chloro-4'-amino diphenyl ether, 2,4-Dichloro-4'-amino diphenyl ether(amin-TOK), N-[4'-(4-Chlorophenoxy)] phenyl acetamide, and N-[4'-(4-Chlorophenoxy)] phenyl formamide as the metabolities. This result indicates that TOK undergose the reduction of its $NO_2\;to\;NH_2$ group, dechlorination, acetylation, and formylation under this condition. The cleavage of ether linkage does not occur. In addition, TOK degrades more readily in Chung Ju soil which is characterized by pH 6.43 and higher contents of $Ca^{++}$ and C.E.C. than in Chong Ju soil which is lower in pH, $Ca^{++}$, and C.E.C. 2. In the aerobic incubation of TOK of 25ppm in Chung Ju soil suspension for 21 days, the ratio of the resulting metabolites, TOK : amino-TOK : 4-Chloro-4'-amino diphenyl ether was 100 : 130 : 76. Meanwhile, in the 42 day incubation, the ratio was 100 : 19 : 5, which indicates that TOK in aerobic condition dose not necessrily degrade as a function of the incubation period. 3. The citrate buffer extract of Chung Ju soil has the capability of degrading TOK, which was verified to be due to the action of the microorganisms involved. 4. Twelye strains of soil bacteria were isolated from the TOK-treated soils. In the incubation of TOK in pure cultures of the respective isolates, the strain T-1-1 isolated from Chong Ju soil had almost no degradability whereas the strain T-2-3 was the most potent. The degradation of TOK by the isolates constituted mostly the reduction of the nitro group to amino group. 5. In a test for the degradability of TOK by some selected microorganisms, Pseudomonas species were more potent than fungi. Yet, Isolate B which had been isolated from Chung Ju soil suspension was the most potent.

Recent Advances in Amino Acid Nutrition for Efficient Poultry Production - Review -

  • The nutritional value of protein varies between feedstuffs. It is possible to feed animals using crystalline amino acids as a sole nitrogen source, but in practice only some limiting amino acids are added to the diet. In order to use feedstuffs efficiently, it is important to determine exact amino acid requirements. Reported values differ widely because the requirements are affected by various factors. In this report, therefore, the factors affecting amino acid requirements are reviewed as follows: 1) availability of dietary amino acids, conversion factors of nitrogen to protein, interaction of amino acids, and strain, sex and age of animals; 2) amino acid requirements for maximum performance and maintenance, usefulness of non-essential amino acids; 3) plasma amino acid concentration as a parameter to determine amino acid requirements; and 4) nitrogen excretion to reduce environmental pollution. These factors should be considered, it is to improve the dietary efficiency, which is to reduce excess nitrogen excretion for environmental pollution.

가잠견사 Fibroin의 품종별 Amino산 분석 (Amino Acids Analysis of Silk Fibroins among Varieties)

  • 본 보고는 가잠사 fibroin의 amino산 성분을 품종별로 Paper chromatography에 의해 분석하여 품종간의 amino산 성분차이를 관찰하고 동 amino산이 잠체내에서의 유전성과 생합성에 어느정도의 관련성이 있나 고찰하기 위한 것으로 다음과 같은 결론을 얻었다. 1. Paper chromatography에 의한 각종 amino산의 전개상황은 품종간에 대체로 유의하였다. 2. 그러나 숫자적 분석치는 타인의 것과 유의하면서도 품종간에 다소의 차이를 보였다. 3. 각 교잡종 견사 fibroin의 amino산 성분비율은 모체의 유전인자와 어느 정도 관련성이 있는 것으로 보인다. 4. 영양성 amino산은 견사 fibroin의 성분으로서보다 잠아성장에 더욱 관련하는 것 같다. 5. 지금까지 문한상에 발견되지 않은 미지의 amino산이 특수영양종견사 fibroin에 나타났으며 Hydroxyprolin이 아닌가 추측된다. 6. 견사 fibroin의 견사선내에서의 생합성기구로서 Stepwise process나 Template 학설 어느측이나 일방적으로 지대할 수 없는 실험결과이었다.
