Physicochemical Properties and Biological Activities of Collagens with Different Molecular Weights from Alaska Pollack (Theragra chalcogramma) Skin (명태 껍질 유래 콜라겐의 분자량에 따른 이화학적 특성 및 생리활성)

  • This study was conducted to investigate the physicochemical properties and biological activities of collagens with different molecular weights from Alaska pollack (Theragra chalcogramma) skin as well as their efficacies as functional materials. The molecular weights of collagens were between 1~10 kDa (below 1 kDa (AP1), 1~3 kDa (AP2), 3~10 kDa (AP3), and above 10 kDa (AP4). The protein content of AP4 (40.19 g/100 g) was the highest. Collagen contents of AP1, AP2, AP3, and AP4 were 36.43, 32.23, 19.23, and 14.89%, respectively. The free amino acid and essential amino acid contents of AP1 were higher than those of AP2, AP3, and AP4. Fourier transform infrared spectroscopy spectra of collagens with different molecular weights showed wavenumbers representing the regions of amide I, amide II, amide III, and amide A, respectively. The electron-donating ability (29.51%) and SOD-like activity (38.45%) of AP1 were higher than those of AP2, AP3, and AP4. Tyrosinase inhibition activity of AP1 improved with higher treatment concentration. The rate of inhibition of MMP-1 production in HS68 cells exposed to UVB was suppressed by treatment with AP1 (29.78%) and AP2 (26.49%) at 1 mg/mL. Furthermore, there was a strong correlation between DPPH, superoxide dismutase, tyrosinase activity, and MMP-1 inhibition rate of collagens with different molecular weights.

  • The hot-water extract of fruiting bodies in Fomitella fraxinea had potent anti-complementary activities. After fractionation of water-soluble polysaccharides by DEAE-Sephadex A-25 column chromatography, major anti-complementary activity was concentrated into the FF-AP1 among three polysaccharides (FF-NP, FF-AP1, FF-AP2). FF-AP1 was fractionated into $FF-AP1{\alpha}$ and $FF-AP1{\beta}$ obtained from the adsorbed fraction and unadsorbed fraction by affinity chromatography using a ConA-sepharose 4B column, respectively. $FF-AP1{\beta}$, which exihibited the highest anti-complementary activities had an IR absorption peak of $890cm^{-1}$, and a M.W. of about 15,000 (gel filtration). Anti-complementary activity of FF-AP1 decreased greatly by pronase treatment and periodate oxidation. $FF-AP1{\beta}$ responsible for potent anti-complemenary activities of Fomitella fraxinea was an acidic protein-containing heteroglycan consisted of 48% glucose, 13% mannose, and 12% galactose as major component sugars, 9.6% protein, 6% uronic acids.

  • IETF (Internet Engineering Task Force) CoAP (Constrained Application Protocol) protocol is supported communication between sensor or actuator nodes by in a constrained environment, such as small amount of memory, and low power. CoAP and HTTP protocol can convert easily, and can use to monitor or controll the infrastructure utility through low-power sensor and actuator networks in IoT (Internet of Thing) and M2M (Machine-to-Machine) environment. IETF CoRE(Constrained RESTful environments) Working Group proposed CoAP protocol in 2010, and began to standardize it. Recently, CoAP protocol is published RFC (Request for Comments) 7252. In this paper, we design and implement of CoAP protocol for testing the interoperability in heterogeneous operating environments. For this experiment, we developed the CoAP client program based on Windows environment and CoAP server program in Linux environment to test the interoperability.

Effects of Rotarod Exercise and Electroacupuncture on Muscle Activity and Serum BDNF Level in the Ataxic Rats by the 3-Acetylpyridine (3-Acetylpyridine에 의한 운동실조 동물모델에서 로타로드 운동과 전침이 근활성도와 혈청 BDNF에 미치는 영향)

  • The 3-Acetylpyridine(3-AP) induces cerebellar injury which is chemoablation of the inferior olive nucleus. The purpose of this study was to investigate the effects of exercise(Ex) and electroacupuncture(EA) on muscle activity of hindlimb and serum brain derived neurotrophic factor(BDNF) in the ataxia rats by the 3-AP. 12-week-aged male Sprague-Dawley rats were randomly divided into the 5 groups: Control, 3-AP, 3-AP+Ex, 3-AP+EA and 3-AP+Ex+EA groups. Maximal Height Vertical Jump(MHVJ) was significantly decreased in the 3-AP compared with control group, and it was increased in Ex, EA and Ex+EA groups than 3-AP group. The muscle activity of hindlimb was significantly increased in the 3-AP groups than control group, also it was most decreased in Ex+EA group. The concentration of BDNF of the serum was significantly decreased in the Ex, EA and Ex+EA groups than 3-AP group. The results of this study showed that dynamic exercise and electroacupuncture have a positive effect on functional recovery and in the ataxia rats by the 3-AP.

  • In this paper, it is analyzed propagation distribution characteristics of radio wave, which is radiated on all sides from Access Point device for estimating scheme of wireless AP without GPS receiver environment. To tracking AP device is traced from wireless terminals and wireless terminal is traced from fixed AP devices, above all it is important to analysis the characteristics of receiving power in according to distance. Therefore in this paper, radio propagation characteristics of 2.4GHz wave is analyzed effect extent and quality in according to distance in doors and out doors, and utilized to location estimation.

  • ISO 10303-AP241 is an Application Protocol of ISO 10303, which specifies the representation of AEC facility's life cycle information. The Working Draft of AP241 was submitted to the TC184/SC4 of ISO in July, 2007 by Korean team of ISO TC184/SC4/WG3. This paper introduces the currently developed draft Ap241 core model, and describes the life cycle support modules which are required to represent the life cycle information of AEC facilities. The draft core model is developed as a generic data model which is useful as a basis for implementation in a data warehouse. This core model could be used in combination with reference data such as PLCS RDL, Gellish table and RDL of ISO 15926. These concepts already exists in ISO 10303-AP221, AP239 and ISO 15926. In order to support maintenance and feedback of operational information concerning the AEC facilities, the AP241 team re-used many modules from AP239, and developed various STEP Application Modules which are adapted from the ISO 15926.

Structural characteristics of a red ginseng acidic polysaccharide rhamnogalacturonan I with immunostimulating activity from red ginseng

  • Background: Many researchers reported that the various immune activities of red ginseng are due to acid polysaccharides. But, the exact structural characteristics of the acidic polysaccharide in red ginseng have not been fully elucidated. Therefore, we isolated the acidic polysaccharide from red ginseng and characterized the structural property of the active moiety of this polysaccharide, which contributes to the immunostimulatory activity of red ginseng. Methods: A polysaccharide (RGP-AP-I) was purified from red ginseng via size-exclusion chromatography using Sephadex G-100. Immunostimulatary activity of RGP-AP-I was investigated via anti-complementory and macrophage stimulatory activity. The structure of RGP-AP-I was characterized by HPLC, sugar composition, β-glucosyl Yariv reagent and methylation analysis. Results: Peritoneal macrophages stimulated using RGP-AP-I significantly augmented the production of various cytokines such as interleukin (IL)-6, IL-12, and tumor necrosis factor (TNF)-α. The primary structure of RGP-AP-I was elucidated by assessing its sugar composition and methylation analysis. RGP-AP-I is a 96 kDa acidic polysaccharide, and comprises nine different monosaccharides, which mainly include sugars such as rhamnose (Rha, 9.5%), galacturonic acid (GalA, 18.4%), galactose (Gal, 30.4%), and arabinose (Ara, 35.0%). RGP-AP-I exhibited an considerable reaction with the β-glucosyl Yariv reagent, revealing the presence of arabino-β-3,6-galactan. Methylation analysis indicated that RGP-AP-I comprises 21 different glycosyl linkages, such as 3-, 4-, 6- and 3,6-linked Galp; 5-linked Araf; 2,4-linked Rhap; and 4-linked GalAp, which are characteristics of rhamnogalacturonan I (RG-I). Conclusion: we assumed that the immunostimulatory activity of RGP-AP-I may be due to the RG-I structure, which comprises a main chain with a repeating linkage unit, [→2)-Rhap-(1→4)-GalAp-(1→] and three groups of side chains such as (1→5)-linked arabinan, (1→4)-linked galactan, and arabino-β-3,6-galactan, which branch at the C(O)4 positions of Rha residues in the main chain of RGP-AP-I.

  • Total of 11 strains of Ldctobacillus isolated from lactobacillus-fermented milk and-beverage in March 1980 were examined for susceptibility to 8 drugs, and transferability and transfer frequency of R plasmids by conjugation. Of 11 isolates each 2 strains were classified as L. cellobiosus and L. helveticus, each 1 strain as L. plantarum, L. lactis, L. acidophilus, L. delbrueckii, L. casei subsp. casei, L. casei subsp, tolerans and L. salivarius subsp, salivarius by Bergey's manual. Resistance was the most active to na lidixic acid(NA), followed in decreasing order by chloramphenicol(CP), ampicillin(AP), kanamycin(KM) and streptomycin(SM). All of isolates were resistant to NA, each 10 strains to CP and AP, 7strains to KM and 6 strains to SM, indicating all of the isolates were resistant to two or more drugs in combination. No strain was resistant to erythromycin(EM), penicillin(PC) and tetracycline(TC). The most frequently encountered resistant patterns were CP NA AP SM KM, followed by CP NA AP KM, NA AP, CP NA, CP NA AP and CP NA AP SM in order. Transfer experiment of drug resistance showed that of 11 resistant strains, 9 strains transferred parts of their resistance to AP or AP CP or SM AP, indicating 9 strains carried R plasmids determining R(AP), R(AP CP) and R(SM AP). The conjugal frequency of R(AP) from Lactobacillus to E. coli ranged from 2.5{\times}10^{-1} to $5.6{\times}10^{-4}%$, that of R(CP) ranged from 5.0{\times}10^{-1} to 5.0{\times}10^{-3}% and that of R(SM) ranged from 6.0{\times}10^{-5} to 1.4{\times}10^{-5}%, at $37^{\circ}C$ for 18 hours of incubation.

Computed Tomography Assessment of Severity of Acute Pancreatitis in Bangladeshi Children

  • Purpose: Acute pancreatitis (AP) is common among children in Bangladesh. Its management depends mainly on risk stratification. This study aimed to assess the severity of pediatric AP using computed tomography (CT). Methods: This cross-sectional, descriptive study was conducted in pediatric patients with AP at the Department of Pediatric Gastroenterology and Nutrition, BSMMU, Dhaka, Bangladesh. Results: Altogether, 25 patients with AP were included, of whom 18 (mean age, 10.27±4.0 years) were diagnosed with mild AP, and 7 (mean age, 10.54±4.0 years) with severe AP. Abdominal pain was present in all the patients, and vomiting was present in 88% of the patients. Etiology was not determined. No significant differences in serum lipase, serum amylase, BUN, and CRP levels were observed between the mild and severe AP groups. Total and platelet counts as well as hemoglobin, hematocrit, serum creatinine, random blood sugar, and serum alanine aminotransferase levels (p>0.05) were significantly higher in the mild AP group than in the severe AP group (p=0.001). The sensitivity, specificity, positive predictive value, and negative predictive value of CT severity index (CTSI) were 71.4%, 72.2%, 50%, and 86.7%, respectively. In addition, significant differences in pancreatic appearance and necrosis were observed between the two groups on CT. Conclusion: CT can be used to assess the severity of AP. In the present study, the CTSI effectively assessed the severity of AP in pediatric patients.

SIMULATED AP1000 RESPONSE TO DESIGN BASIS SMALL-BREAK LOCA EVENTS IN APEX-1000 TEST FACILITY

  • As part of the $AP1000^{TM}$ pressurized water reactor design certification program, a series of integral systems tests of the nuclear steam supply system was performed at the APEX-1000 test facility at Oregon State University. These tests provided data necessary to validate Westinghouse safety analysis computer codes for AP1000 applications. In addition, the tests provided the opportunity to investigate the thermal-hydraulic phenomena expected to be important in AP1000 small-break loss of coolant accidents (SBLOCAs). The APEX-1000 facility is a 1/4-scale pressure and 1/4-scale height simulation of the AP1000 nuclear steam supply system and passive safety features. A series of eleven tests was performed in the APEX-1000 facility as part of a U.S. Department of Energy contract. In all, four SBLOCA tests representing a spectrum of break sizes and locations were simulated along with tests to study specific phenomena of interest. The focus of this paper is the SBLOCA tests. The key thermal-hydraulic phenomena simulated in the APEX-1000 tests, and the performance and interactions of the passive safety-related systems that can be investigated through the APEX-1000 facility, are emphasized. The APEX-1000 tests demonstrate that the AP1000 passive safety-related systems successfully combine to provide a continuous removal of core decay heat and the reactor core remains covered with considerable margin for all small-break LOCA events.
