Performance Analysis of Location-Aware System based on Active Tags (능동태그 기반 위치인식 시스템의 성능 분석)

  • Location awareness is one of the key functionalities to build an U-city. Recently, many of works of the location-aware systems are emerging to commercially apply to on-going large-scale apartment complex based on U-city. As dwellers or cars being attached with active tags are moving in the U-city complex, the active tags periodically broadcast their own identifiers and receivers fixed along the street or in building use those information to calculate location of them. There are several issues to be considered for such an environment. The first is that the number of active tags operating in the same region are large as much as tens of thousands, and the second is that the active tags should be alive without change of batteries more than a year, hence low power consumption is very important. In this paper we propose i) a new architecture for location-aware system considering such issues, ii) technical issues to implement it using active tags, and iii) a mathematical analytic model to investigate overall performance and verify it by comparing with actual experimental results. Through the analysis we can show the theoretical boundary of the lowest packet loss rate and the maximum number of tags with acceptable performance for the systems based on active tags. The results can be applied to practical design of location-based systems of U-City projects.

Efficient Transformation Method of Soybean Using Meristematic Tissues of Germinating Seeds (발아종자의 분열조직을 이용한 효율적인 콩 형질전환 방법)

  • An efficient transformation method for soybean [Glycine max (L.) Merr.] using meristematic tissues of germinating seeds has been established. The embryonic axes were excised from germinating seeds of Korean soybean cultivar, Iksannamulkong and 0.5-2 cm long segment containing meristematic tissues were prepared by cutting hypocotyl region. The explants were inoculated with Agrobacterium tumefaciens strain LBA4404 harboring a binary vector with the bar gene as a selectable marker gene and a ${\beta}-glucuronidase$ (GUSINT) reporter gene, and then co-cultured for 7 days on co-cultivation medium (CCM). The meristematic tissues were cultured on shoot induction medium (SIMP6) supplemented with 0.4 mg/l $N_6-benzylaminopurine$ (BAP) and 0.1 mg/l indolebutyric acid (IBA) in the presence of 6 mg/l L-phosphinotricin (PPT) for 2 weeks and the surviving explants were transferred to shoot elongation medium (SEMP6). Transformation was confirmed by Southern blot analysis and the transformation efficiencies ranged from 1.48 to 2.07%. The new modified transformation method was successfully implemented for obtaining several transgenic lines with SMV-CP gene. It is expected that this method could efficiently be used for the transformation of recalcitrant soybean cultivars.

A New Pale Yellow Large Gerbera hybrida Cultivar, 'Honey Cream' with High Yield for Cut Flower (크림색 대륜 절화용 다수성 거베라 신품종 '하니크림' 육성)

  • A new pale yellow gerbera(Gerbera hybrida Hort.) cultivar 'Honey Cream' was developed from a cross between 'Princessa' and 'Picasso', followed by seedling and line selections at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services(ARES) in 2006. Three times evaluations were conducted from 2003 for the detailed characteristics of the new cultivar. 'Honey Cream' has semi-double type large flowers in good harmony with pale yellow(RHS, 11-D) ray floret and green center. It has good, stable flower shape and strong peduncle, and its vase life was 12.3 days. The average flower yield of 'Honey Cream' was about 48.5 stems per plant/year in greenhouse trails during 2003 and 2006. This cultivar was registered to the Korea Seed and Variety Service(KSVS) for commercialization in 2007. Year-round production of this cultivar is possible in the greenhouse condition in Korean climate.

A New Orange Mini Gerbera hybrida 'Yetty' with High Yield for Cut Flower (오렌지색 소륜 다수성 절화용 거베라 신품종 '예티' 육성)

  • A new orange mini gerbera (Gerbera hybrida Hort.) 'Yetty' was developed from a cross 'Haiti' and 'Nova Zembra' followed by seedling and line selections at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) from 2001 to 2006. Three times evaluations were conducted from 2004 for the detailed characteristics of the new cultivar, 'Yetty' has single type mini flower in good harmony with orange (RHS 25-A) ray floret and browen center. It has good stable flower shape and strong peduncle, and its vase life was 14.8 days. The average yield of 'Yerty' was 98.5 flowers per plant a year in greenhouse yield trials carried out from 2004 to 2006. This cultivar registered for commercialization in 2007. 'Yerty' would be cultured under greenhouse condition in Korea.

Perceptions and Attitudes of Americans in Korea toward Edible Insect-based Pet Food (식용곤충 함유 반려동물 식품에 대한 국내거주 미국인 소비자 인식 및 태도 연구)

  • We examined American consumers' perceptions and attitudes toward edible insect-based pet food. In this study, 16 Americans in Korea who owned dogs were categorized into two groups, and focus-group discussions were conducted under three conditions. First, we observed the free association perception of edible insect-based pet food, and attitudes were analyzed after providing a newspaper article related to it. Finally, consumer attitude was examined in the context of purchasing. The study found that the participants had high awareness of the eco-friendliness of edible insects and showed a positive attitude toward news articles related to it. However, when it came to purchasing, they considered nutritional and health functional values compared to environmental values. Meanwhile, the rejection of insects was nevertheless the most important negative factor in pet food, as in general food. The results are significant in confirming the possibility of using insect-based pet food by examining the perceptions and attitudes toward the environmental, nutritional, and health functional values of edible insects under three conditions for American consumers.

Application of mass-spectrometry compatible photocleavable surfactant for next-generation proteomics using rice leaves (벼의 차세대 단백질체 분석을 위한 질량분석기 호환의 광분해성 계면활성제의 적용)

  • The solubilization of isolated proteins into the adequate buffer containing of surfactants is primary step for proteomic analysis. Particularly, sodium dodecyl sulfate (SDS) is the most widely used surfactant, however, it is not compatible with mass spectrometry (MS). Therefore, it must be removed prior to MS analysis through rigorous washing, which eventually results in inevitable protein loss. Recently, photocleavable surfactant, 4-hexylphenylazosulfonate (Azo), was reported which can be easily degraded by UV irradiation and is compatible with MS during proteomic approach using animal tissues. In this study, we employed comparative label-free proteomic analysis for evaluating the solubilization efficacies of the Azo and SDS surfactants using rice leave proteins. This approach led to identification of 3,365 proteins of which 682 proteins were determined as significantly modulated. Further, according to the subcellular localization prediction in SDS and Azo, proteins localized in the chloroplast were the major organelle accounting for 64% of the total organelle in the SDS sample, while only 37.5% of organelle proteins solubilized in the Azo were predicted to be localized in chloroplast. Taken together, this study validates the efficient solubilization of total protein isolated from plant material for bottom-up proteomics. Azo surfactant is suitable as substitute of SDS and promising for bottom-up proteomics as it facilitates robust protein extraction, rapid washing step during enzymatic digestion, and MS analysis.

U-Net Cloud Detection for the SPARCS Cloud Dataset from Landsat 8 Images (Landsat 8 기반 SPARCS 데이터셋을 이용한 U-Net 구름탐지)

  • With a trend of the utilization of computer vision for satellite images, cloud detection using deep learning also attracts attention recently. In this study, we conducted a U-Net cloud detection modeling using SPARCS (Spatial Procedures for Automated Removal of Cloud and Shadow) Cloud Dataset with the image data augmentation and carried out 10-fold cross-validation for an objective assessment of the model. Asthe result of the blind test for 1800 datasets with 512 by 512 pixels, relatively high performance with the accuracy of 0.821, the precision of 0.847, the recall of 0.821, the F1-score of 0.831, and the IoU (Intersection over Union) of 0.723. Although 14.5% of actual cloud shadows were misclassified as land, and 19.7% of actual clouds were misidentified as land, this can be overcome by increasing the quality and quantity of label datasets. Moreover, a state-of-the-art DeepLab V3+ model and the NAS (Neural Architecture Search) optimization technique can help the cloud detection for CAS500 (Compact Advanced Satellite 500) in South Korea.

Development of Salt-Tolerant Transgenic Rice Using Soybean PR10 Gene (콩의 Pathogenesis-Related 10 유전자를 이용한 내염성 벼 형질전환 계통 개발)

  • This study was conducted to understand the role of soybean pathogenesis-related 10 (GmPR10) gene in salt tolerance and to develop salt-tolerant rice using GmPR10 cDNA. GmPR10 transgene was expressed constitutively in the shoot and root of the $T_1$ transgenic rice plants. Interestingly, however, the levels of the transgene expression were increased temporally up to over four- to five-fold in the shoot and root by 125 mM NaCl treatment, peaking at six hours after the treatment and decreasing thereafter. Electrolyte leakage of leaf cells under 125 mM NaCl treatment was lower in all the transgenic lines than in the control variety, Dongjin-byeo. Ability of seedlings to recover from 125 mM NaCl treatment for two weeks was higher in the transgenic plants than in the control plants. These results demonstrated that GmPR10 had function to increase cell integrity and promote growth under the saline stress imposed by NaCl. The transgenic line GmPR10-3 which showed highest ability to recover from the saline stress could be used as a potential source for salt tolerance in rice breeding programs.

Trait Variation and Molecular Characterization of Ds insertional rice lines (벼 Ds 삽입변이 계통의 특성변이 및 분자생물학적 특성)

  • The 1,874 rice lines were selected from 3,000 Ds insertional mutant pool by Basta herbicide treatment and were surveyed for trait variation and molecular characteristics of genes knocked out by Ds insertion. Compared with "Donjin", an original japonica cultivar used for transformation, Ds insertion mutant pool showed large variation in major agronomic traits including tiller, panicle, and heading etc. Southern blot analysis demonstrated that these lines on the average had two Ds copies in Donjin genome, resulting in 38.4% of one copy, 32.5% of two copies, 16.7% of three copies, and 11.3% of over four copies. GUS analysis showed that 3.9% of lines (73/1,860) had tissue-specific expression in leaves, nodal parts, floral organs such as stigma and pollen, and roots. Data set obtained from agricultural trait variation and molecular characteristics for individual Ds insertional lines would provide researchers with more information for understanding the function of unknown rice genes controlling economically important traits.

  • A new pink gerbera (Gerbera hybrida Hort.) cultivar 'Summer Ring' was developed from a cross between 'Grandeur' and 'Nova Zembla' followed by seedling and line selections at the Flower Breeding Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) in 2005. Characteristics of the new cultivar were evaluated three times from 2003 to 2004. 'Summer Ring' has semi-double type mini flower in good harmony with pink (RHS, 56-D) ray floret and green center. It has good stable flower shape and strong peduncle, and its vase life was 14.7 days. The average yield of 'Summer Ring' flower was 69 per plant/year in greenhouse yield trails during 2003 and 2004. This cultivar was registered for commercialization in 2005. Commercial production of 'Summer Ring' could be possible throughout the country under greenhouse condition.
