Effects of Salviae Miltiorrhizae Radix Hot Aqueous Extract on Nitric Oxide and Prostaglandin E2 Production and on 1,1-diphenyl-2-picryl hydrazyl Radical Scavenging in Macrophages

  • Objectives: The objective of this study is to investigate the effects of Salviae Miltiorrhizae Radix hot aqueous extract on nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production and on 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical scavenging in macrophages. Methods: Salviae Miltiorrhizae Radix (300 g) was heated at $100^{\circ}C$ with distilled water (2 L) for 4 hours. The extract was filtered and concentrated to 100 mL by using a rotary evaporator, was frozen at $-80^{\circ}C$, and was then freeze-dried by using a freezing-drying system. The RAW 264.7 macrophage was subcultured by using $10-{\mu}g/mL$ lipopolysaccharide (LPS). In order to evaluate cytotoxicity, we performed 3-(4,5-dimrthylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays and measured the cell viability. The NO production was measured by using Griess assays, and the $PGE_2$ production was measured by using enzyme immunoassays. The antioxidant activity, the 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical scavenging capability, was measured by using the DPPH method. Results: Cell viability with the 1-, 5-, 25-, 125- and $625-{\mu}g/mL$ Salviae Miltiorrhizae Radix hot aqueous extract was not significantly decreased compared to the cell viability without the extract. When 125 and $625{\mu}g/mL$ of Salviae Miltiorrhizae Radix hot aqueous extract were used, nitric oxide (NO) production in LPS-stimulated RAW 264.7 macrophages was significantly inhibited compared to that in the control group. When 25, 125, and $625{\mu}g/mL$ of Salviae Miltiorrhizae Radix hot aqueous extract were used, $PGE_2$ production in LPS-stimulated RAW 264.7 macrophages was significantly inhibited compared to that in the control group. The 125- and $625-{\mu}g/mL$ Salviae Miltiorrhizae Radix hot aqueous extracts had high DPPH free-radical scavenging capabilities in RAW 264.7 macrophages. Conclusion: This study indicates that Salviae Miltiorrhizae Radix hot aqueous extract suppresses NO and $PGE_2$ production and improves DPPH free-radical scavenging capability. Thus, it seems that Salviae Miltiorrhizae Radix hot aqueous extract may have an anti-inflammation effect and antioxidant activity.

  • 유기발광소자는 다른 디스플레이에 비해 높은 명암비와 색재현성의 장점을 갖는 차세대 디스플레이로서, 얇은 박막 특성을 가지고 있기때문에 모바일용 디스플레이 기술로 많이 사용되고 있다. 하지만 낮은 발광효율, 높은 구동전압 및 전압에 따른 색좌표 변화의 문제점을 가지고 있어 이를 극복하기 위한 많은 연구가 진행되고 있다. 유기 발광 소자의 발광효율을 높이며 구동 전압을 낮추기 위해 호스트물질에 다양한 도펀트를 도핑하고 있다. 높은 발광효율을 가지는 도펀트인 5,6,11,12-tetraphenylnaphthacene (rubrene)을 사용한 유기발광소자는 rubrene의 안정된 분자 에너지 레벨로 인해 전자들이 포획되는 현상이 나타나 효율이 감소되는 원인이 규명되지 않았다. 본 연구에서는 rubrene을 발광층으로 사용하여, 전공수송층인 N,N_-bis-(1-naphthyl)-N,N_-diphenyl-1,1-biphenyl-4,4-diamine (NPB)의 두께에 따른 I-V 변화와 전계발광 스펙트럼를 분석하여 두께에 따른 rubrene의 전자 포획를 관찰하였다. rubrene보다 큰 lowest unoccupied molecular orbital 에너지를 갖는 NPB와 에너지장벽으로 낮은 highest occupied molecular orbital 에너지를 갖는 4,7-diphenyl-1,10-phenanthroline을 각각 교차되게 적층한 유기발광소자의 I-V 변화와 전자 전공 재결합층의 위치변화에 따른 전계발광 스펙트럼을 비교 분석하였다. 이 결과는 발광층 내부의 rubrene의 상대적인 위치와 에너지장벽과의 상관관계에 따른 전자 포획 메커니즘을 이해하는데 도움 줄 것이다.

Diarylheptanoid and Flavonoid with Antioxidant Activity from Alnus japonica Steud on DPPH Free Radical Scavenging Assay

  • Alnus japonica Steud (Betulaceae) has long been used as a Korean traditional medicine for gastric disorders, hepatitis, and fatty liver. As a part of our study on the identification of secondary metabolites of naturally occurring bioactive compounds, we isolated 1,7-bis(4-hydroxyphenyl)-3,5-heptanediol(1), 5-hydroxy-1,7bis(4-hydroxyphenyl)-3-heptanone(2), 5,3'-dihydroxy-7,4'-dimethoxyflavone(3) and 3,5,7,3',4'-pentahydroxyflavone(4) from the dichloromethane and ethylacetate-soluble fractions of Alnus japonica Steud. These compounds showed significant antioxidant activity in a concentration-dependent manner. The $IC_{50}$ values of compounds 1, 2, 3 and 4 were 30.1, 37.4, 20.2 and 13.7 ${\mu}g/mL$, respectively, through the scavenging capacity of 1,1-diphenyl-2-picrylhydrazyl radical assay.

Photochemical Generation of Phenylsilylene and Its Chemistry (광분해 반응에 의한 Phenylsilylene의 생성과 그 반응성에 관한 연구)

  • The photochemical precursors, 1,1,1,3,3,3-hexamethyl-2-phenyltrisilane(2) and 2,3-dicarbomethoxy-1,4,5,6,7-pentaphenyl-7-silanorbornadiene(5) were synthesized in the yield of 10% and 73%, respectively. Irradiation of 2 at 254 nm in the presence of triethylsilane gave 1,1,1-triethyl-2-phenyldisilane (6) in 44% yield which was the product of phenylsilylene insertion into the Si-H bond. Irradiation of 2 in the presence of diphenylacetylene gave 1-phenyl-1-silacyclopenta-2,4-diene(4) in 68% yield together with 1,2-diphenyl-1,2-disilacyclohexa-2,5-diene(26%) which were formed from [2+2] addition of the silacyclopropene to diphenylacethylene and formed from dimerization of silacyclopropene, respectively. From the neat photolysis of precursor 2,1,5-dihydrosilanthrene(11), intermolecular C-H insertion product of phenylsilylene and 1,2-diphenyltrisilane(12), Si-H insertion product of phenylsilylene to the precursor were obtained in the yield of 5% and 7%, respectively. In the same experimental condition, both photolyses of 5 in the presence of triethylsilane and methanol showed that the intramolecular 1,5-sigmatropic rearrangement of precursor 5 to give the formation of silylenolether was more favorable process than the generation of phenylsilylene.

  • The photochemical reactions and spectral properties of 1,1-diphenyl-2-vinylcyclopropane were studied. Upon direct irradiation of the compound at 253.7nm, 1,1-diphenylethylene was formed as a major product along with a small amount of 4,4-diphenylcyclopentene. Sensitized photoreactions showed quite a different phenomena from those of conjugated dienes supporting the spectral results that the cyclopropane ring does not transmit the conjugation in the compound.

A Study on the Effectiveness of a Mixed Fermented Extract of Vitamin Tree Fruit, Acai Palm Fruit, Mango, Lemon, Apricot and Blueberry as an Active Ingredient (비타민나무열매, 아사이팜열매, 망고, 레몬, 살구 및 블루베리의 혼합 발효추출물의 효능 연구)

  • In this study, the cell viability, radical scavenging ability, elastase activity, moisturizing activity, and antimicrobial activity of mixed fermented extract of vitamin tree fruit, acai palm fruit, mango, lemon, apricot and blueberry as an active ingredient were measured. The experiment was performed at three concentrations(0.5 / 1.5 / 2%), since cytotoxicity was not shown at these three concentrations. Based on this, a radical scavenging experiment was conducted to compare with L-ascorbic acid, a representative raw material of an antioxidant material. The results showed that the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was 95.1 ± 0.6%, 94.3 ± 0.7%, 95.3 ± 0.6%, 95.1 ± 0.7%, 95.1 ± 0.3%, 95.5 ± 0.3%, and 95.4 ± 0.4% respectively, when the concentration of the mixed fermentation extract was 2%. As the concentration increased at the three concentrations (0.5/1.5/2%) and the content of blueberry extracts increased, sample 7 showed excellent elastase activity at a concentration of 2% to 0.9 cm, such as retinol crystal, and moisturizing activity was also found to be 71.5%, 75.6%, and 81.6%, respectively. Particularly, at the 2% concentration of the mixed fermented extract, the antibacterial activity effect on Propionibacterium was very excellent in sample 7. Therefore, it is considered that it is worth developing the mixed fermented extracts of 2% concentration as a cosmetic component with antioxidant, moisturizing, and antibacterial properties.

  • Recently, the phosphorescent organic light-emitting devices (OLEDs) have been extensively studied for their high internal quantum efficiency. In this study, we synthesised several phosphorescent metal complexes, and certified their composition using NMR. We also investigated the characteristics of the phosphorescent OLEDs with the green emitting phosphor, $Ir(ppy)_{3}$. The devices with a structure of indium-tin-oxide(ITO)/N,N'-diphenyl-N,N'-(3-methylphenyI}-1,1'-biphenyl-4,4'-diamine (TPD)/metal complex doped in host materials/2,9-dimethyl-4,7-diphenyl-l,10-phenanthroline(BCP)/tris (8-hydroxyquinolinato) Aluminum($Alq_{3}$)/Li:Al/Al was fabricated, and its electrical and optical characteristics were studied. By changing the doping concentration of tris(2-phenylpyridine)iridium ($Ir(ppy)_{3}$), we fabricated several devices and investigated their characteristics.

In Vitro Antioxidant Activity of Some Selected Prunus Species in Korea

  • In the course of the investigations of natural antioxidants, we examined the antioxidant activities of the methanol (MeOH) extracts of some selected Prunus species, including P. buergeriana, P. davidiana, P padus, P. pendula for. ascendens, P. sargentii, P. serrulata var. spontanea and P. yedoensis by three methods as represented by the 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, total ROS (reactive oxygen species) and the peroxynitrite ($ONOO^-$) scavenging activity tests. We also evaluated the activities of the organic solvent-soluble fractions, including the dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), n-butanol (n-BuOH) fractions and the water ($H_2O$) layer of P. serrulata var. spontanea leaves. By means of bioassay-directed fractionation, we isolated eleven known flavonoids (1-11) from the EtOAc soluble fraction of the MeOH extract of the Prunus serrulata var. spontanea leaves, exhibiting strong antioxidant activity and characterized as prunetin (1), genistein (2), quercetin (3), prunetin $4'-O-{\beta}-glucopyranoside$ (4), kaempferol $3-O-{\alpha}-arabinofuranoside$ (5), prunetin $5-O-{\beta}-glucopyranoside$ (6), kaempferol $3-O-{\beta}-xylopyranoside$ (7), genistin (8), kaempferol $3-O-{\beta}-glucopyranoside$ (9), quercetin $3-O-{\beta}-glucopyranoside$ (10) and kaempferol $3-O-{\beta}-xylopyranosyl-(1{\rightarrow}2)-{\beta}-glucopyranoside$ (11). Compounds 3 and 10 showed good activities in all tested model systems. Compounds 2 and 8 showed scavenging activities in the DPPH and $ONOO^-$ tests, while compounds 5, 7, 9 and 11 were active in the $ONOO^-$ and ROS tests. On the other hand, compounds 1, 4 and 6 did not show any activities in the tested model systems.

  • The syntheses of novel heterocyclic base modified pyrimidine nucleosides are described. 5, 6-dimethyl-4-hydroxy-3-methoxy-1-$(\beta$-D-ribofuranosyl)2(1H)-pyridinone 7 was synthesized by condensation of silylated 5, 6-dimethyl4-hydroxy-3-methoxy-2(1H)-pyridione 7 was synthesized by condensation of silylated 5, 6-dimethyl-4-hydroxy-3-methoxy-2(1H)-pyridinone with $\beta$-D-ribofuranose-1-acetate-2, 3, 5-tribenzoate in dichloroethane in the presence of Lewis acid followed by debenzoylation. The 2, 2'-anhydro-5, 6-dimethyl-2-hydroxy-3-methoxy-1-$\beta$-D-arabinofuranosyl-4-pyridinone 8 was obtained from the reaction of the free ribonucleoside 7 and diphenyl carbonate in DMF. None of these compounds showed any significant antiviral ad antitumor activities in vitro tests.

Analysis of the Antioxidant Properties of 2,2-diphenyl-1 Picrylhydroazyl, Hydroxyl Radicals, and Nitric Oxide in Alaska Pollock Roe, with or without Natural Fermented Seasoned (알래스카 명란의 DPPH, OH, NO의 항산화 특성 분석)

  • Alaska pollock (Gadus chalcogrammus) is a marine fish species belonging to the family Gadidae. In this study, whether the Alaska Polloack Roe have antioxidant properties, 2,2-diphenyl-1 picrylhydroazyl (DPPH), hydroxyl radical (OH) reducing activity, and nitric oxide radical (NO) scavenging activity were evaluated in distilled water extract (DWE) and ethanol extract (ETE) of raw Alaska pollock roe, Gochujang Pollock roe, and fermented seasoned Pollock roe. The DPPH scavenging activity of the DWE with Gochujang Pollock roe was 71.9% at a concentration of 1.0 mg/ml and that of the ETE was 73.7% at the same concentration. The DPPH scavenging activity of the DWE with fermented seasoned Pollock roe was 78.0% at 1.0 mg/ml, whereas that of ETE was 78.4% at the same concentration. The $IC_{50}$ values of the DWE and ETE of raw Pollock roe for DPPH were $11.65{\mu}g/ml$ and $11.47{\mu}g/ml$, respectively. The OH scavenging activities of raw Pollock roe, Gochujang Pollock roe, and fermented seasoned Pollock roe ethanolic extracts at a concentration of 1.0 mg/ml were 70.9%, 79.0, and 80.6%, respectively. The $IC_{50}$ values of the DWE and EWE of raw Pollock roe for NO were $11.45{\mu}g/ml$ and $11.41{\mu}g/ml$, respectively. The DPPH, OH, and NO scavenging abilities in DWEs and ETEs of Gochujang and fermented seasoned Pollock roe were higher than those of instant (no Gochujang or season treatment) treatment Pollock roe. Both the Gochujang and fermented seasoned Pollock roes have natural radical scavenging ability and may be useful potential antioxidant food supplements.
