Two Cases of IgG-associated Mesangial Glomerulonephritis in Children (소아에서 관찰된 IgG-associated mesangial glomerulonephritis 2례)

  • Rare cases of IgG associated mesangial glomerulonephritis(IgG GN) defined by exclusive or predominant mesangial IgG deposits were reported first by Sato et al.(1993). and subsequently 10 pediatric cases were reported by Yoshikawa et al.(1994). Previous reports suggested that the prognosis of IgG GN is relatively benign course but recent report suggested that prognosis of IgG GN is highly variable. Also the recurrence of IgG GN in a renal transplant was reported by Fakhouri et al. (2002). Such a recurrence highlights the specificity of this type of glomerulonephritis. We experienced two pediatric cases of IgG GN proven by renal biopsy. Case 1. 4-year-old girl with nephrotic syndrome admitted because of general edema. The patient's urinalysis showed proteinuria and microscopic hematuria. Renal biopsy was performed because of relapsed nephritic syndrome. Light microscopic finding was nonspecific with almost normal histology. Immunofluorescent findings showed diffuse segmental IgG(+) and IgM(+) deposits in the capillary walls, and focal segmental spotty C4(trace), C1q(trace) deposits. Electron microscopic findings showed focal portion of mesangial electron dense deposits without mesangial widening. Case 2. 11-year-old girl admitted for evaluation of microsopic hematuria detected through mass school urinary screening program. Renal biopsy was performed for exact diagnosis. Immunofluorescent findings showed focal segmental IgG(+), IgM(+/-) and C3(+/-) deposits. Electron microscopic findings showed focal portion of mesangial electron dense deposits without mesangial widening.

Effect of Si contents on Tensile-Shear Peak Load and Nugget Diameter in the Resistance Spot Welded of Dual Phase Steel for Automotive Body Applications (자동차 차체용 냉연 DP강 저항점용접부의 너깃경과 인장전단강도에 미치는 Si 함유량의 영향)

  • 원가 측면에서 유리한 저항점용접(Resistance Spot Welding)이 차체 용접에 80%이상으로 가장 많이 적용되고 있다. 첨단고강도강(Advanced High Strength Steel)의 저항점용접성 및 용접부 특성에 미치는 공정 변수의 영향에 대한 연구결과는 많으나, 합금원소의 영향에 대해서는 전무하다. 특히, Si는 DP(Dual Phase)강에 첨가 시 균일한 마르텐사이트의 분포를 촉진하는 원소로 저항 점용접성 및 용접부 특성에 영향을 미칠 것으로 예상되며, 이에 대한 연구는 보고된바 없다. 본 연구에서는 냉연 DP강의 저항 점용접시 중요한 인자 중 하나인 너깃경과 전단인장강도에 미치는 Si함유량의 영향을 검토하였다. 사용된 강재 및 용접기는 1.2mm 두께의 Si함유량(0, 0.5, 1.0, 1.5wt%)이 다른 인장강도 780~1000MPa급 냉연 DP강과 단상 AC용접기를 사용하였다. 용접조건은 ISO 18278-2규격에 따라 가압력 4kA, 초기가압시간 40cycle, 유지시간 17cycle로 고정하고, 용접전류만 변화하여 용접을 실시하였다. 너깃경은 용접부 단면을 컷팅 후 폴리싱 하여, 광학현미경과 Image Pro plus를 이용하여 측정했으며, 인장시편규격은 JIS Z 3137를 이용하였다. Si함유량이 증가에 따라 스패터 발생 전류는 감소했고, 너깃경은 직선적으로 증가했다. Si함유량 증가에 따른 너깃경 증가 이유는 저항(R) 측정결과, Si함유량 증가에 따라 모재의 저항이 높아져, 따라서 입열량($Q=I^2Rt$)이 많아지기 때문으로 판단되었다. 인정전단강도는 Si함유량 증가에 따라 직선적으로 증가했다. 이러한 이유는 Si함유량 증가에 따라 너깃경이 증가되기 때문으로 판단되었고, 너깃경과 인장전단강도 사이에 직선적 관계(PL(kN)=$3.2N_{dia.}$-0.81, $R^2$=0.93)를 가지고 있었다. 파단양상은 Si함유량에 상관없이 5.4kA이하에서는 계면파단이 일어났고, 6.0kA이상에서는 풀 아웃 파단이 일어났다. 계면파단주원인은 용접부 가장자리에 지름이 약 $5{\mu}m$이하의 예리한 노치가 존재하여 노치응력집중과 HAZ계면 근처에 미접합부가 존재하기 때문으로 판단되었다. 6.0kA이상에서는 예리한 노치가 없었고, HAZ부가 완전히 접합되어 있기 때문에 풀 아웃 파단이 일어난 것으로 판단되었다. 따라서, Si함유량 증가에 따라 적정용접전류 구간은 감소했고, 너깃경은 직선적으로 증가했다. 또한, Si함유량 증가에 따라 인장전간강도는 증가 했으며, 너깃경과 인장전단강도 사이에 직선적 관계를 가지고 있었다. 파단 양상은 Si함유량에 상관없이 5.2kA이하에서는 계면파단이, 6.0kA이상에서는 풀 아웃 파단이 일어났다.

Expression of Human SOD1 and Mutant SOD1 (G93A) in E. coli and Identification of SOD1 as a Substrate of HtrA2 Serine Protease (대장균에서의 human SOD1과 mutant SOD1 (G93A) 단백질의 발현과 HtrA2의 기질 여부 확인에 관한 연구)

  • Superoxide dismutase (SOD) is physiologically important in regulating cellular homeostasis and apoptotic cell death, and its mutations are the cause of familial amyotrophic lateral sclerosis (FALS). Mitochondrial serine protease HtrA2 has a pro-apoptotic function and has known to be associated with neurodegenerative disorders. To investigate the relationship between genes associated with apoptotic cell death, such as HtrA2 and SOD1, we utilized the pGEX expression system to develop a simple and rapid method for purifying wild-type and ALS-associated mutant SOD1 proteins in a suitable form for biochemical studies. We purified SOD1 and SOD1 (G93A) proteins to approximately 90% purity with relatively high yields (3 mg per liter of culture). Consistent with the result in mammalian cells, SOD1 (G93A) was more insoluble than wild-type SOD1 in E. coli, indicating that research on the aggregate formation of SOD1 may be possible using this pGEX expression system in E. coli. We investigated the HtrA2 serine protease activity on SOD1 to assess the relationship between two proteins. Not only wild-type SOD1 but also ALS-associated mutant SOD1 (G93A) were cleaved by HtrA2, resulting in the production of the 19 kDa and 21 kDa fragments that were specific for anti-SOD1 antibody. Using protein gel electrophoresis and immunoblot assay, we compared the relative molecular masses of thrombin-cleaved GST-SOD1 and HtrA2-cleaved SOD1 fragments and can predict that the HtrA2-cleavage sites within SOD1 are the peptide bonds between leucine 9-lysine 10 (L9-K10) and glutamine 23-lysine 24 (Q23-K24). Our study indicates that SOD1 is one of the substrate for HtrA2, suggesting that both HtrA2 and SOD1 may be important for modulating the HtrA2-SOD1-mediated apopotic cell death that is associated with the pathogenesis of neurodegenerative disorder.

Correlation between Strain and Dielectric properties in Paraelectic $ZrTiO_4$ Thin Films ($ZrTiO_4$상유전 박막의 Strain과 유전 특성 상관성 고찰)

  • 급증하는 무선통신 정보수요는 특히, 고주파대역 (300NHz-300GHz)에서 사용되는 공진기, 필터, 발진기 등과 같은 소자의 품질향상을 요구하고 있다. 고주파용 유전체 중 ZrTiO4 는 $\alpha$-PbO2 계열의 사방정구조를 갖고 있는 유전체로서 높은 유전율 ($\varepsilon$=40)과 높은 품질계수 (Q=1/tan$\delta$=4700 at 7GHz)를 갖고 있고, Sn 첨가시 0ppm/$^{\circ}C$의 공진주파수 온도계수를 얻을 수 있다고 보고되어 있다. 본 연구에서는 약 110$0^{\circ}C$ 이상에서 안정한 상으로 존재하는 ZrTiO4를 저온에서 증착하여 준안정한 상태로 결정화되게 한후, 유전손실 (tan$\delta$)과 유전율($\varepsilon$)을 측정하였다. 또한 증착온도와 열처리과정에 따른 박막의 us형 (Strain) 정도의 변화를 X-선 회절결과로부터 분석하였으며 이를 측정된 유전특성 값과 비교하였다. ZrTiO4 박막은 DC magnetron reactive sputter로 Zr과 Ti 타겟으로부터 high phosphorous doped Si (100) 기판위에 증착하였다. 압력은 4mTorr로 유지하고 박막의 화학양론적 조성비를 맞추기 위해 각 타겟에 가해지는 power는 Zr/Ti=500W/650W로 고정하고, 반응가스의 비율을 Ar/O2=17sccm/3.5sccm으로 유지하여 박막내에 인입되는 산소량을 제어하였다. 증착 직후와 열처리 후의 박막특성을 비교하기 위해 증착온도를 상온에서부터 $600^{\circ}C$까지 변호시키고 증착후 각각의 시편을 80$0^{\circ}C$ 산소분위기에서 2시간동안 열처리하여 시편을 준비하였다. 박막의 상형성 여부와 결정성변화는 $ heta$-2$\theta$X-선 회절법을 사용하여 조사하였고, EPMA를 이용하여 박막의 조성을 확인하였다. 유전특성의 측정을 위해 백금 상부전극을 증착한 후, impedance analyzer를 이용하여 100kHz 영역에서의 유전손실을 측정하고, 측정된 정전용량과 박막의 두께로부터 유전율을 계산하였다. ZrTiO4 박막은 증착온도 20$0^{\circ}C$ 이상에서 결정성을 보이기 시작했으며, 열처리 이후에는 상온에서 비정질이었던 시편이 $650^{\circ}C$ 이상의 온도에서 결정화되기 시작하였다. 증착온도에 따라 유전손실은 0.038에서 0.017 정도로 감소하는 경향을 나타냈으며, 각각 열처리에 의해서 0.034, 0.005 정도로 다시 감소하였다. 박막의 유전율은 약 35 정도의 값을 나타내었으며 X-선 회절 data로부터 분석한 박막의 변형은 증온도에 따라 7.2%에서 0.04%로 감소하였고 이 이경향은 유전손실은 감소경향과 일치하였다.

Geochronology and Petrogenetic processes of the so-called Hongjesa granite in the Seogpo-Deogku Area (석포(石浦)-덕구간(德邱間)에 분포(分布)하는 소위(所謂) 홍제사화강암(洪濟寺花崗岩)의 지질연대(地質年代)와 생성과정(生成過程)에 대(對)한 硏究(연구))

  • Main aspects of this study are to clarify geochronology and petrogenetic processes of the so-called Hongjesa granite, which is a member of various intrusive rocks exposed in the northeastern part of the Ryongnam Massif, one of the Precambrian basements of South Korea. In this study, the Hongjesa grainte is divided into four rock units based on the geologic age, mineralogical and chemical constituents, and texture: the Precambrian Hongjesa granite gneiss (Hongjesa granite Proper) and leucogranite gneiss, the Paleozoic gnessic two mica granite, and the Jurassic muscovite granite. The Hongjesa granite gneiss is identified by its grayish color, slight foliation, and porphyroblastic texture. The leucogranite gneiss is distinct by its light gray color, sand medium to coarse grained texture. The gneissic two mica granite is distinguished from others by its strong foliation, containing gray-colored feldspar phenocrysts with biotite and muscovite in varying amounts. The muscovite granite occurs as a small stock containing feldspar phenocrysts along margin of the stock. These granitic rocks vary widely in composition, reflecting the facts that they partly include highly metamorphosed xenolith and schlierens as relics of magmatic and anatectic processes. In particular, grayish porphyroblasts of microcline perthite is characteristic of the Hongjesa granite gneiss, whereas epidote and garnet occur in both the Hongjesa granite gneiss and leucogranite gneiss. These minerals are considered to be formed by potassic metasomatism and contamination of highly metamorphosed rocks deeply buried under the level of the Hongjesa granite emplacement. The individual synchronous granitic rocks plotted on Harker diagram show mostly similar trends to the Daly's values. The plots of the Hongjesa granite gneiss and gneissic two mica granite concentrate near the end part of the calc-alkalic rock series on the AMF diagrams, whereas those of the leucogranite gneiss and muscovite granite indicate the trend of the Skaergaard pluton. These granitic rocks plotted on a Q-Ab-Or diagram (petrogeny's residua system) fall well outside the trough of the system. This can be attributed to the potassic matasomatism of these rocks. On the ACF diagram, these rocks appear to be dominantly I-type prevailing over S-type. The K-Ar ages, obtained from a total of 7 samples of the leucogranite gneiss, gneissic two mica granite, muscovite granite, porphyritic alkali granite, and rhyolitic rock, in addition to the Rb/Sr ages of the Hongjesa granite gneiss by previous workers, permit the rock units to be arranged in the following chronological order: The middle Proterozoic Hongjesa granite gneiss (1714-1825 m.y.), the upper proterozoic leucogranite gneiss (875-880 m. y.), the middle Paleozoic gneissic two mica granite (384 m. y.) the upper Jurassic muscovite granite (147 m. y.), the Eocene alkali granite (52 m. y.), and the Eocene rhyolitic rock (45 m. y.). From the facts and data mentioned above, it is concluded that the so-called Hongjesa granite is not a single granitic mass but is further subdivided into the four rock units. The Hongjesa granite gneis, leucogranite gneiss, and gneissic two mica granite are postulated to be either magmatic or parautochtonous, intrusive, and the later muscovite granite is to be magmatic in origion.

Clinical Features, Response to Treatment, Prognosis, and Molecular Characterization in Korean Patients with Inherited Urea Cycle Defects

  • The urea cycle, consisting of a series of six enzymatic reactions, plays key roles to prevent the accumulation of toxic nitrogenous compound and synthesize arginine de novo. Five well characterized diseases have been described, resulting from an enzymatic defect in the biosynthesis of one of the normally expressed enzyme. This presentation will focus on two representative diseases; ornithine transcarbamylase(OTC) deficiency and citrullinemia(argininosuccinate synthetase deficiency). OTC deficiency is one of the most common inborn error of urea cycle, which is inherited in X-linked manner. We identified 17 different mutations in 20 unrelated Korean patients with OTC deficiency; L9X, R26P, R26X, T44I, R92X, G100R, R141Q, G195R, M205T, H214Y, D249G, R277W, F281S, 853 del C, R320X, V323M and 10 bp del at nt. 796-805. These mutations occur at well conserved nucleotide sequences across species or CpG hot spot. The L9X and R26X lead to the disruption of leader sequences, required for directing mitochondrial localization of the OTC precursor. Their phenotypes are severe, and neonatal onset. The G100R, R277W and V323M mutations were uniquely identified in patients with late onset OTC deficiency. The other genotypes are associated with neonatal onset. Out of 20 patients with OTC deficiency, only 6 patients are alive; two were liver transplanted, and normal in growth and development at 2, 4 years after transplantation respectively. Citrullinemia is an autosomal recessive disease, caused by the mutations in the argininosuccinate synthetase(ASS) gene. We identified in 3 major mutations in 11 unrelated Korean patients with citrullinemia; G324S, $IVS6^{-2}$ A to G, and 67 bp ins at nt 1125-1126. Among these, the 67 base pair insertion mutation is novel. The allele frequency of each mutation is; G324S(45%), IVS6-2 A to G(32%), and 67 base pair insertion(14%). All patients are diagnosed at neonatal or infantile age. Interestingly, two patients presented with stroke like episode. Out of 11 patients, 5 patients died. Among 6 patients alive, one patient was successfully liver transplanted.

Forest Stand Structure, Site Characteristics and Carbon Budget of the Kwangneung Natural Forest in Korea (광릉 활엽수천연림의 산림식생구조, 입지환경 및 탄소저장량)

  • The study area, Kwangneung Experiment Forest (KEF) is located on the west-central portion of Korean peninsula and belongs to a cool-temperate broadleaved forest Bone. At the old-growth deciduous forest near Soribong-peak (533.1 m) in KEF, we have established a 1 ha permanent plot ($100m{\times}100m$) and a flux tower, and the site was registered as a KLTER(Korean long-term ecological research network) and DK site of KoFlux. In this site, we made a stemmap of trees and analyzed forest stand structure and physical and chemical soil characteristics, and estimated carbon budgets by forest components (tree biomass, soils, litter and so on). Dominant tree species were Quercus serrata and Carpinus laxiflora, and accompanied by Q. aliena, Carpinus cordata, and so on. As a result of a field survey of the plot, density of the trees larger than 2 cm in DBH was 1,473 trees per ha, total biomass 261.2 tons/ha, and basal area $28.0m^2$/ha. Parent rock type is granite gneiss. Soil type is brown forest soil (alfisols in USDA system), and the depth is from 38 to 66 cm. Soil texture is loam or sandy loam, and its pH was f개m 4.2 to 5.0 in the surface layer, and from 4.8 to 5.2 in the subsurface layer. Seasonal changes in LAI were measured by hemispherical photography at the 1.2 m height, and the maximum was 3.65. And the spatial distributions of volumetric soil moisture contents and LAIs of the plot were measured. The carbon pool in living tree biomass including below ground biomass was 136 tons C/ha, and 5.6 tons C/ha is stored in the litter layer, and about 92.0 tons C/ha in the soil to the 30 cm in depth. Totally more than about 233.6 tons C/ha was stored in DK site. These ground survey and monitoring data will give some important parameters and validation data for the forest dynamics models or biogeochemical dynamics models to predict or interpolate spatially the changes in forest ecosystem structure and function.

Genome-wide Survey of Copy Number Variants Associated with Blood Pressure and Body Mass Index in a Korean Population

  • Hypertension is the major factor of most death and high blood pressure (BP) can lead to stroke, myocardial infarction and cardiac failure. Moreover, hypertension is strongly correlated with body mass index (BMI). Although the exact causes of hypertension are still unclear, some of genetic loci were discovered from genome-wide association study (GWAS). Therefore, it is essential to study genetic variation for finding more genetic factor affecting hypertension. The purpose of our study is to conduct a CNV association study for hypertension-related traits, BP and BMI, in Korean individuals. We identified 2,206 CNV regions from 3,274 community-based Korean participants using the Affymetrix Genome-Wide Human SNP Array 6.0 platform and performed a logistic regression analysis of CNVs with two hypertension-related traits, BP and BMI. Moreover, the 4,692 participants in an independent cohort were selected for respective replication analyses. GWAS of CNV identified two loci encompassing previously known hypertension-related genes: LPA (lipoprotein) on 6q26, and JAK2 (Janus kinase 2) on 9p24, with suggestive p-values (0.0334 for LPA and 0.0305 for JAK2 ). These two positive findings, however, were not evaluated in the replication stage. Our result confirmed the conclusion of CNV study from the WTCCC suggesting weak association with common diseases. This is the first study of CNV association study with BP and BMI in Korean population and it provides a state of CNV association study with common human diseases using SNP array.

Wiryeongtang attenuates diabetic renal dysfunction in human renal mesangial cells (위령탕(胃苓湯) 추출물의 사람 유래 신장 메산지움 세포에서의 당뇨병성 신장 손상 개선 효과)

  • Objectives : Diabetic nephropathy is one of the most common chronic complications of diabetes and a leading cause of end-stage renal failure in the world. Mesangial cell proliferation is known as the major pathologic features such as glomerulosclerosis and renal fibrosis. Wiryeongtang (WRT) is a well-known traditional herbal formula as therapeutic agents for chronic edema and dysuresia of renal homeostasis. In the present study, we investigated whether WRT inhibits high glucose (HG)-induced renal dysfunction by TGF-β/Smads signal regulation in cultured mesangial cells.Methods : Inhibitory effect of WRT (10-50 ㎍/ml) on HG-stimulated mesangial cells proliferation and dysfunction were evaluated by [3H]-thymidine incorporation, Western blot, and RT-qPCR.Results : WRT significantly decreased HG-accelerated thymidine incorporation in human renal mesangial cell in a dose-dependent levels. WRT induced down-regulation of cyclins/CDKs and up-regulation of CDK inhibitor, p21waf1/cip1 and p27kip1 expression. In addition, HG enhanced expression of dysfunction biomarker such as collagen IV and CTGF, which was markedly attenuated by WRT. WRT decreased TGF-β1 and Smad-2/Smad-4 expression, whereas increased Smad-7 expression under HG. Furthermore, WRT inhibited HG-induced inflammatory factors level such as ICAM-1 and MCP-1 as well as NF-κB p65 nuclear translocation and intracellular ROS production.Conclusions : These results suggested that WRT may alleviate mesangial proliferation and inflammation possibly involved in renal fibrotic process, further diabetic nephropathy through disturbing TGF-β1/Smad signaling and NF-κB/ROS pathway. Thus, WRT might prove to be effective in the treatment of renal dysfunction leading to diabetic nephropathy.

Purification and Characterization of a Thermostable Xylanase from Paenibacillus sp. NF1 and its Application in Xylooligosaccharides Production

  • High levels of extracellular xylanase activity (211.79 IU/mg) produced by Paenibacillus sp. NF1 were detected when it was submerged-cultured. After three consecutive purification steps using Octyl-Sepharose, Sephadex G75, and Q-Sepharose columns, a thermostable xylanase (XynNF) was purified to homogeneity and showed a molecular mass of 37 kDa according to SDS-PAGE. The specific activity of the purified XynNF was up to 3,081.05 IU/mg with a 14.55-fold purification. The activity of XynNF was stimulated by $Ca^{2+}$, $Ba^{2+}$, DTT, and ${\beta}$-mercaptoethanol, but was inhibited by $Fe^{2+}$, $Zn^{2+}$, $Fe^{2+}$, $Cu^{2+}$, SDS, and EDTA. The purified XynNF displayed a greater affinity for oat spelt xylan with the maximal enzymatic activity at $60^{\circ}C$ and pH 6.0. XynNF, which was shown to be cellulose-free, with high stability at high temperature ($70^{\circ}C-80^{\circ}C$) and low pH range (pH 4.0-7.0), is potentially valuable for various industrial applications. The enzyme hydrolyzed oat spelt xylan to yield mainly xylooligosaccharides (95.8%) of 2-4 degree of polymerization (DP2-4). Moreover, the majority of the xylooligosacharides (DP2-4) products was xylobiose (61.5%). The thermostable xylanase (XynNF) thus seems potentially usefull in the production of xylooligosaccharides.
