Effects of particle size and adaptation duration on the digestible and metabolizable energy contents and digestibility of various chemical constituents in wheat for finishing pigs determined by the direct or indirect method

  • Objective: This experiment was conducted as a $3{\times}2{\times}2$ factorial design to examine the effects of particle size (mean particle size of 331, 640, or $862{\mu}m$), evaluation method (direct vs indirect method) and adaptation duration (7 or 26 days) on the energy content and the apparent total tract digestibility (ATTD) of various chemical components in wheat when fed to finishing pigs. Methods: Forty-two barrows ($Duroc{\times}Landrace{\times}Yorkshire$) with an initial body weight of $63.0{\pm}0.8kg$ were individually placed in metabolic cages and randomly allotted to 1 of 7 diets with 6 pigs fed each diet. For the indirect method, the pigs were fed either a corn-soybean meal based basal diet or diets in which 38.94% of the basal diet was substituted by wheat of the different particle sizes. In the direct method, the diets contained 97.34% wheat with the different particle sizes. For both the direct and indirect methods, the pigs were adapted to their diets for either 7 or 26 days. Results: A reduction in particle size linearly increased the digestible energy (DE) and metabolizable energy (ME) contents as well as the ATTD of gross energy, crude protein, organic matter, ether extract (EE) and acid detergent fiber (ADF) (p<0.05), and had a trend to increase the ATTD of dry matter of wheat (p = 0.084). The DE, ME contents, and ATTD of gross energy, crude protein, dry matter and organic matter were higher (p<0.05) when determined by the direct method, but the ATTD of ADF, EE, and neutral detergent fiber were higher when determined by the indirect method (p<0.05). Prolongation of the adaption duration decreased the ATTD of neutral detergent fiber (p<0.05) and had a trend to increase the ATTD of EE (p = 0.061). There were no interactions between particle size and the duration of the adaptation duration. The ATTD of EE in wheat was influenced by a trend of interaction between method and adaptation duration (p = 0.074). The ATTD of ADF and EE in wheat was influenced by an interaction between evaluation method and wheat particle size such that there were linear equations (p<0.01) about ATTD of ADF and EE when determined by the direct method but quadratic equations (p = 0.073 and p = 0.088, respectively) about ATTD of ADF and EE when determined by the indirect method. Conclusion: Decreasing particle size can improve the DE and ME contents of wheat; both of the direct and indirect methods of evaluation are suitable for evaluating the DE and ME contents of wheat with different particle sizes; and an adaptation duration of 7 d is sufficient to evaluate DE and ME contents of wheat in finishing pigs.

고추향미유의 저장 중 화학적 특성 변화 (Chemical Changes of Red Pepper Seasoning Oil during Storage)

  • 라면 수프의 품질 안정화 기술을 개발하기 위한 기초자료로 활용하고자, 라면 수프의 원료 중의 하나인 고추향미유를 $65^{\circ}\C$에서 6주간 저장하면서 화학적 변화를 조사하였다. 시료의 산가는 저장기간이 길어짐에 따라 약간 증가하였으며 과산화물가는 저장 4주에 크게 증가하다가 저장 6주에서는 감소하였다. ASTA value와 $CIE\;L^*$ 값, $CIE\;a^*$값, 그리고 $CIE\;b^*$ 값 모두 4주까지는 저장기간이 길어짐에 따라 증가하다가 6주후에는 감소하는 경향을 나타내었다. 시료를 구성하는 주요 지방산은 linoleic(51.02%), Oleic(30.17%), 그리고 palmitic(11.81%) acid이었다. 저장기간이 증가할수록 myristic acid(14:0)와 palmitic acid(16:0)는 약간 증가하였으나 linoleic acid(18:2)는 감소하였고, 불포화지방산과 포화지방산의 비율은 저장기간이 증가할수록 감소하였다. 시료에 가장 많이 존재하는 유리지방산은 linoleic acid(18:2)이었는데 6주간 저장 후에는 linoleic acid(18:2)의 양이 85.5에서 65.2(mg/kg oil)로 크게 감소하였고, 총유리지방산의 양은 저장기간이 증가할수록 증가하였으며, 유리 불포화지방산과 유리 포화지방산의 비율은 저장기간이 증가할수록 감소하였다.

흑미 첨가량을 달리한 떡볶이 떡의 품질에 관한 연구 (Quality of Tteokbokki tteok prepared by adding various concentration of brown rice)

  • 쌀 소비확대를 위하여 가공용으로 재배 육성된 흑미의 특성과 떡복이용 떡에 가공적성이 가능한지를 파악하고자 일반성분, 수분 결합력, 수분 흡수력, 호화특성을 조사하였으며, 삼광을 대조구로 하여 흑설, 흑진주 떡볶이 떡을 제조하여 저장기간에 따른 색도, 물성 및 관능적 특성을 조사하였다. 일반성분을 분석한 결과 수분이 14.89~7.15%, 조단백질이 4.79~6.81%, 조지방이 0.48~2.66%, 조회분이 0.39~1.60%로 나타났다. 특히, 흑진주는 조단백질과 조지방 함량이 가장 높았고 삼광이 조단백질함량이 가장 낮게 나타났다. 수분결합력은 흑진주가 가장 높았고 그 다음으로 흑설이 높은 것으로 나타났다. 수침시간에 따른 수분흡수율은 수침 초기에 흡수속도가 높았으며 대조구인 삼광은 수침 3시간, 현미로 도정한 흑미는 12시간 까지는 완만하게 증가하다가 그 이후에는 거의 변화가 없어 평형에 도달하였다. 호화특성에서 호화개시온도는 흑설이 가장 높았으며 최고점도(peak viscosity peak), 최저점도(hot viscosity), 최종점도(final viscosity), 강하점도(breakdown) 등의 온도에 따른 점도 변화는 흑설이 가장 낮은 값을 보여 주었다. 차반점도(setback)는 백미인 삼광이 가장 높았으며 현미 흑미인 흑설과 흑진주는 비슷한 수준의 값을 나타내었다. 흑미 떡볶이 떡의 색도는 흑미 첨가량이 증가할수록 L값 및 b값은 감소하고 a값은 증가하였으며 저장기간이 경과함에 따라 L값은 증가한 반면 a값 및 b값은 감소하는 경향을 보였다. 흑미 떡볶이 떡의 물성변화는 흑미의 첨가량이 증가함에 따라 경도, 응집성, 씹힘성은 감소하는 반면 부착성은 증가하였으며 저장기간이 경과함에 따라 경도의 경우 증가하는 경향을 보였다. 또한, 관능적 특성의 전반적인 기호도 조사에서 흑미 10% 첨가군을 가장 선호하는 것으로 나타났지만 20% 첨가군과의 유의적인 차이는 없었다(p<0.05). 따라서, 떡볶이 떡 제조 시 적절한 흑미 첨가량은 10~20%가 적당할 것으로 보이며 흑미 첨가가 관능적 기호도에 좋은 영향을 줄 것으로 기대된다.

Effects of Different Levels of Vitamin-Mineral Premixes on Growth Performance, Nutrient Digestibility, Carcass Characteristics and Meat Quality of Growing-Finishing Pigs

  • Two experiments using growing and finishing pigs (Landrace$\times$Duroc$\times$Yorkshire) were conducted to determine the effects of different levels of vitamins and trace minerals (VTM) on growth performance, nutrient digestibility, carcass characteristics and meat quality of growing-finishing pigs, and to evaluate the suitability of vitamin-mineral levels commonly used in the swine industry in Korea. A total of 120 three crossbred (Landrace$\times$Duroc$\times$Yorkshire) growing (Experiment I: 20.90 0.44 kg average initial body weight) and finishing (Experiment II: 53.55 0.97 kg average initial body weight) were used in 6 and 9 weeks feeding trials, respectively. Pigs were allotted on the basis of sex and weight to 5 treatments with 6 replications per treatment with 4 pigs per pen in a completely randomized block design. Treatments were: 1) control, 2) fat soluble vitamin 200% vs. control, 3) water soluble vitamin 200% vs. control, 4) trace mineral 50% vs. control, 5) trace mineral 200% vs. control. In experiment I, during the overall experimental period (0 to 6 weeks, 21 to 54 kg body weight), ADG of the 200% trace mineral supplemented group was significantly higher than that of the 50% trace mineral supplemented group. There were no significant differences among other the treatments. Digestibility of crude protein was lowest in mineral 50% supplemented group (p<0.05). Calcium digestibility was significantly higher in the 200% fat soluble vitamin supplemented group than in the other treatments (p<0.05). In experiment II, during the overall experimental period (0 to 9 weeks, 54 to 106 kg body weight), growth performance was not significantly affected by dietary vitamin and trace mineral levels. However, increasing level of water soluble vitamins at the level of 200% compared to control had a tendency to improve the overall growth performance. Overall carcass characteristics except for carcass length did not differ among pigs fed the dietary treatments. Loin eye area, pH, drip loss and shear force of meat were not affected by dietary vitamin and trace mineral levels. There was a trend for less fat content (%) in pork when the level of vitamin and trace mineral was increased, but the difference was not significant. Flavor score was the lowest in control and highest in the 200% fat soluble vitamin supplemented group (p<0.05). Juiciness of muscle was lower in the 200% fat soluble vitamin supplemented group than other dietary treatments, except for trace mineral 50% supplemented group (p<0.05). Based on these results, it is suggested that "typical" commercial levels of vitamin and trace minerals used by feed companies in Korea are sufficient to meet requirement for the maximum growth of growing-finishing pigs. Our results suggests that a reduction in trace mineral levels in commercial diets could be considered to reduce feed cost and nutrient excretion with economic and environmental benefits.

CAD SYSTEM을 이용한 한복의 기성복 설계에 관한 연구(I) -여아 색\ulcorner 저고리 및 치마를 중심으로- (A Study on the Korean Costume Pattern Design Using CAD System(I) -With Concentration on the Girl's Color-Strip Blouse & Skirt-)

  • The research, aiming an automatic pattern design of Korean Costume by utilizing CAD system, tried Grading, Seaming and Marking with a theme of girl's color-strip blouse & skirt. The content & conclusion of the research can be outlined as follows; 1) As we reviewed the current situation of Korean costume which becomes more likely celebrational or festival costume, while the demand of it is getting increase, it's being dealt mostly as ready-made dresses with different qualities & designs. Especially childrens ready-made ones were highly demanded to be picked up as the theme. 2) For the original drawing of children's color-strip blouse & skirt, Kyung-Ja Park's drawing method was used here, and for the substitute of particular body parts absolute size, Joo-Won Lee's standard size chart from a size-study by ages. 3) To work with CAD system we had input master pattern, drawn for age 5 as basic size, then graded six step-sizes for 1-11 years old. For add-subtract of particular body parts size, we graded through computing the standard variation among items to get the pattern developed into ready-made standard size, we can make precise plotting by grading wanted size very rapidly if we correct the rule of changed items different from standard size to make utilization possible enough with easy order method of ready-made Korean Dress. 4) We produced Marker after attaching a margin to seam accordingly by parts for each pattern using P/D/S to mark, In mass gament-cutting, the loss of time and material can minimized. In this research the apparel CAD system which has been utilized and only be western fashion industry was introduced for the design of ready-made Korean costume and utilized it in Grading, Marking which are critical steps to improve productivity and have reported the result in the research. Thus we expect that less cost, improved productivity and better quality with minimized loss of material from marking as well as from prompt and precise size-drawing. Furthermore the utilization of CAD system is considered as an effective one in terms of the research & the development to remove effective one in terms of the research & the development to remove irrational elements in the design and production process of Korean costume as well as in terms of the study of Korean Costume development through creative works of Korean Costume.

백서장기(白鼠臟器)에서의 Chromatin의 분리(分離)와 그 RNA 합성능(合成能)에 미치는 X-선전신조사(線全身照射)의 영향(影響)에 관(關)한 연구(硏究) (Studies on the Chromatin Isolated from the Organs of Animals Received Whole-body X-ray Irradiation)

  • 근년(近年) 고등동물세포(高等動物細胞)에 있어서 유전자(遺傳子)의 본체(本體)인 DNA에서 RNA를 경과(經過)해서 특이적(特異的)인 단백질(蛋白質)의 생합성(生合成)에 도달(到達)하는 경로(經路)에 대(對)해서는 많은 연구(硏究)에 의해서 확립(確立)되어졌으나 그 조절기구(調節機構)에 대(對)해서는 불명(不明)한 점(點)이 많다. 개체(個體), 기관(器管), 세포내구조(細胞內構造) 급(及) DNA의 준위(準位)에서의 방사선(放射線)의 장해(障害)에 대(對)해서도 연구(硏究)되고 있으나 소위(所謂) 방사선감수성(放射線感受性) 급(及) 비감수성(非感受性)의 각장기(各臟器)에서 분리(分離)한 Chromatin (DNA-Histone-잔여단백(殘餘蛋白)의 고차구조결합체(高次構造結合體)에 대(對)한 DNA, RNA, 전단백질(全蛋白質)과 유전수식체(遺傳修飾體)라고 생각되는 Histon-단백(蛋白)의 화학조성(化學組成)을 검출(檢出)했으며 겸(兼)해서 chromatin의 생물활성(生物活性)인 RNA 합성능(合成能)(priming activity)에 대(對)한 방사선(放射線)의 영향(影響)을 조사(調査)하는데 의의(意義)가 있다. 전리방사선(電離放射線) 조사(照射)에 의해서 생체(生體)의 DNA의 합성조해(合成阻害)가 잘 알려진 사실(事實)이나 분화(分化)한 생체조직(生體組織)에서의 DNA의 합성(合成)보다도 일반대사(一般代謝)에 중요(重要)한 역할(役割)을 한다는 것도 생각된다. 세포(細胞)의 대사(代謝)는 내분비계등(內分泌系等)의 "Effector-DNA-RNA-단백합성(蛋白合成)이라는 정보유전기구(情報遺傳機構)에 의해서 제어(制禦)되어 있다. 이 연구(硏究)는 방사선생물학상(放射線生物學上) 중요(重要)한 것은 논할(論) 필요(必要)도 없으며 방사선동위원소표지화합물(放射線同位元素標識化合物)을 사용(使用)하여 생화학적(生化學的)으로 추구(推究)하였다.

아밀라아제 처리에 의한 메밀의 당화가 페놀화합물 함량과 항산화 활성의 증가에 미치는 영향 (Saccharification of Fagopyrum esculentum by Amylase Treatments increases Phenolic Compound Content and Antioxidant Activity)

  • 본 연구에는 메밀을 ${\alpha}$-amylase, ${\beta}$-amylase, glucoamylase 세 종류의 효소로 당화한 결과, 당도와 rutin, quercetin, 총 폴리페놀 함량이 증가하였고, 항산화 활성이 개선됨을 확인하였다. 당화 전 약 $1^{\circ}Brix$를 나타내던 메밀은 ${\alpha}$-amylase, ${\beta}$-amylase, glucoamylase로 당화하였을 때 최고 $10.27^{\circ}Brix$, $5.13^{\circ}Brix$, $11.13^{\circ}Brix$를 각각 나타내었다. Rutin과 quercetin의 함량도 당화 전보다 당화를 거치면서 증가하는 경향을 나타내었으며, ${\alpha}$-amylase로 당화하였을 때 rutin은 1.76배, quercetin은 2.09배 증가하였으며, ${\beta}$-amylase로 당화하였을 때 rutin은 1.58배 증가하였으며, glucoamylase로 당화하였을 때 rutin은 3.36배, quercetin은 6.58배 증가하였다. 총 폴리페놀과 DPPH radical 소거능을 측정한 결과, 당화를 하기 전과 비교하여 당화 후 총 폴리페놀 함량과 DPPH radical 소거능이 유의하게 증가하였다. 이러한 결과를 바탕으로 메밀의 당화는 메밀에 함유되어 있는 기능성 성분을 증가시켜 기능성 식품으로의 개발가능성을 높이는 것으로 판단된다.

클로로프렌 고무의 해수에 의한 물성 변화 및 노화 수명 예측 (Life Time Prediction and Physical Properties of Chloroprene Rubber Aged by Seawater)

  • 본 연구에서는 클로로프렌 고무의 열 및 해수에 의한 노화 현상에 따른 물성 변화 및 사용 수명을 예측하였다. 가속 스트레스 수준은 열노화 $80^{\circ}C$, $100^{\circ}C$, $120^{\circ}C$, 해수침지노화 $40^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$에서 2년 3개월(20,000Hr)동안 가속노화를 통해 인장시험을 수행하여 신장률, 인장강도, 경도를 통한 물성변화 측정을 하였다. 분석 결과신장율 저하가 가장 주된 고장지배모드임을 알 수 있었다. 아레누이스 식을 이용한 노화수명 예측 결과 열노화된 CR의 예측 수명은 $23^{\circ}C$에서 125년이며, 해수침지노화된 CR의 예측수명은 $23^{\circ}C$에서 9년이었다. SEM과 SEMEDS 분석 결과, 해수노화시편이 균열과 SEM-EDS에서는 산소의 농도가 증가하였으며, 이를 바탕으로 FT-IR에서는 해수와의 반응에 의하여 C-O와 C=O 결합이 생성되었음을 관찰하였다. 또한 DSC와 TGA 분석에서는 해수침지노화에 의하여 유리전이온도가 상승하며 분해온도가 낮아짐을 관찰하였다.

Changes in plant hydraulic conductivity in response to water deficit

  • How do plants take up water from soils especially when water is scarce in soils? Plants have a strategy to respond to water deficit to manage water necessary for their survival and growth. Plants regulate water transport inside them. Water flows inside the plant via (i) apoplastic pathway including xylem vessel and cell wall and (ii) cell-to-cell pathway including water channels sitting in cell membrane (aquaporins). Water transport across the root and leaf is explained by a composite transport model including those pathways. Modification of the components in those pathways to change their hydraulic conductivity can regulate water uptake and management. Apoplastic barrier is modified by producing Casparian band and suberin lamellae. These structures contain suberin known to be hydrophobic. Barley roots with more suberin content from the apoplast showed lower root hydraulic conductivity. Root hydraulic conductivity was measured by a root pressure probe. Plant root builds apoplastic barrier to prevent water loss into dry soil. Water transport in plant is also regulated in the cell-to-cell pathway via aquaporin, which has received a great attention after its discovery in early 1990s. Aquaporins in plants are known to open or close to regulate water transport in response to biotic and/or abiotic stresses including water deficit. Aquaporins in a corn leaf were opened by illumination in the beginning, however, closed in response to the following leaf water potential decrease. The evidence was provided by cell hydraulic conductivity measurement using a cell pressure probe. Changing the hydraulic conductivity of plant organ such as root and leaf has an impact not only on the speed of water transport across the plant but also on the water potential inside the plant, which means plant water uptake pattern from soil could be differentiated. This was demonstrated by a computer simulation with 3-D root structure having root hydraulic conductivity information and soil. The model study indicated that the root hydraulic conductivity plays an important role to determine the water uptake from soil with suboptimal water, although soil hydraulic conductivity also interplayed.

효소분리 및 용매정제법으로 제조한 고농도 Sialic Acid(23%)가 함유된 GMP 가수분해분말의 마우스 골수세포의 소핵시험을 이용한 안전성 평가연구 (In Vivo Evaluation of the Safety of Hydrolyzed GMP Powder containing Highly Concentrated Sialic Acid (23%) produced by Enzyme Separation and Solvent Enrichment Method using Micronucleus Test in Mice)

  • 본 시험물질인 GMP으로부터 sialic acid를 분리 및 이의 농도를 23%로 높게 정제한 GMP 가수분해물(23%-GNANA) 분말에 대해 발암성 유발 유 무 판단의 기초자료를 얻기 위하여 마우스 골수세포를 이용한 소핵시험을 실시하였다. 결과로서, 다염성 적혈구의 소핵출현 적혈구의 출현비율은 음성 대조군과 시험물질 투여군간 평균 차이의 유의성이 없었다(p<0.05). 또한 동물개체별로 관찰한 결과에서도 음성대조군과 시험물질 투여군간 유의성이 없었으며(p<0.05), 시험물질의 용량의존성 반응은 전체적으로 나타나지 않았다. 이러한 결과로 보아, GMP로부터 제조한 23%-GNANA는 고농도로 sialic acid가 농축되었을 뿐, 일반 식품인 GMP 수준에서 마우스 골수세포에 대해 소핵의 유발성이 없는 것으로 판단되었다.
