The Genetic Variability and Relationships of Japanese and Foreign Chickens Assessed by Microsatellite DNA Profiling

  • This is the first study in which genetic variability and relationships of a large number of Japanese chicken breeds were revealed along with those of several foreign breeds by using microsatellite DNA polymorphisms. Twenty-eight breeds (34 populations) of native Japanese chickens and seven foreign breeds or varieties were analyzed. The mean number of alleles per locus, the proportion of the polymorphic loci, and the expected average heterozygosity ranged from 1.75 to 4.70, from 0.55 to 1.00, and from 0.21 to 0.67, respectively. Microsatellite alleles being unique to a particular population were detected in some populations. The $D_A$ genetic distance between populations was obtained from allele frequency for every pair of the populations to construct a neighbor-joining tree. According to the phylogenetic tree, excluding a few exceptions, native Japanese chicken breeds and foreign breeds were clearly separated from each other. Furthermore, the tree topology divided native Japanese chickens into four main classes, which was almost in accordance with the classification based on body morphology; that is, (1) Cochin type, (2) Malay type, (3) layer type, and (4) intermediate type between Malay and layer types. This is the first finding for native Japanese chickens.

Preliminary Study on the Effects of Out-of-Plane Deposition Angle on Product Characteristics of a UV Photo-Curing Process (UV 광경화 공정에서 평면 외 적층 경사각에 따른 제품 특성 변화에 관한 기초 연구)

  • The goal of this paper is to investigate the effects of out-of-plane deposition angle on product characteristics of a UV photo-curing process. Specimens are manufactured from a commercialized UV photo-curing machine, the NOBEL V1.0. The influence of the out-of-plane deposition angle of the specimen on surface characteristics, including morphology of the sloped surface, pick-to-pick distance of convex region, and roughness of the sloped surface, is examined via the observation of the sloped surface. In addition, the influence of the radius of curvature of the specimen on the surface roughness of the sloped surface is evaluated. The effects of the out-of-plane deposition angle on impact strength of specimens are investigated via Izod impact experiments. Finally, we discuss the influence of the out-of-plane deposition angle on failure characteristics of specimens for impact loads.

Effects of vitamin A supplementation in the diet of breeding geese on offspring intestinal tissue morphology and immune performance

  • Objective: The effects of maternal and offspring dietary vitamin A (VA) supplementation on early body weight, digestive tract function and immune function in goslings were studied. Methods: Yangzhou geese (180 d old) were randomly divided into 5 experimental groups of 15 females and 3 males (the males were kept until slaughter). Eggs were collected for hatching during the peak laying period. A total of 96 goslings were selected from each treatment group (each fed a basic diet supplemented with 0, 4,000, 8,000, 12,000 or 16,000 IU/kg VA) and randomly divided into 2 groups, with 6 replicates in each group and 8 goslings in each replicate. The gosling diet was supplemented with 0 or 9,000 IU/kg VA. Results: i) Villus length, villus width and the muscle thickness of the duodenum, jejunum and ileum were increased and the crypt depth was reduced after adding 12,000 IU/kg VA to the goslings' diet (p<0.05). Adding 9,000 IU/kg VA to the offspring diet increased the length of the duodenal villi and width of the ileum and decreased the crypt depth of the ileum (p<0.05). ii) Supplementing the maternal diet with 12,000 IU/kg VA increased immune organ weight, the immune organ index and immunoglobulin content in goslings (p<0.05). The bursa weight and immunoglobulin G content of offspring were higher in the 9,000 IU/kg VA supplementation group than in the group with no supplementation (p<0.05). Conclusion: Offspring growth and development were affected by the amount of VA added into maternal diet. The negative effect of maternal VA deficiency on offspring can be compensated by adding VA to the offspring diet. Continued VA supplementation in the offspring diet after excessive VA supplementation in the maternal diet is unfavorable for gosling growth and development.

Sunflower cake with or without enzymatic complex for broiler chickens feeding

  • Objective: This study was to evaluate the sunflower cake and enzymatic complex fed to broilers from 22 to 42 d of age. Methods: In a completely randomized design, a total of 850 birds were allotted in a $2{\times}5$ factorial scheme (with and without enzymatic complex) and five inclusion levels (0%, 5%, 10%, 15%, 20%) of sunflower cake. There were 5 replications and 17 birds in each experimental unit. Data from performance, carcass yield and intestinal morphology were evaluated. Results: Feed intake, weight gain, final weight and feed:gain ratio linearly worsened as sunflower cake increased. For weight gain, final weight and feed:gain ratio, the birds whose diets contained levels of 15% and 20% of sunflower cake showed worse values (p<0.05) than the birds fed the control diet. When fed the enzymatic complex, birds improved (p<0.05) crypt depth and villus:crypt ratio in the jejunum. As inclusion levels of sunflower cake increased, villus depth and villus:crypt ratio in duodenum, jejunum and ileum linearly reduced and the crypt depth linearly increased. Carcass yield linearly reduced as sunflower cake increased. Conclusion: Based on performance, sunflower cake can be used up to 10% in broilers feeding from 21 to 42 days of age.

Evaluation of Corrosion and Cavitation Erosion Resistance of Sealed Aluminum Alloy after Anodizing Treatment in Seawater (양극산화 후 실링처리된 알루미늄 합금의 해수 내 내식성과 캐비테이션 침식 저항성 평가)

  • Various sealing techniques were applied to the anodized 5083 aluminum alloy for marine environment to reduce corrosion and cavitation erosion damage. Electrochemical experiments and cavitation erosion tests were conducted to evaluate the corrosion resistance and cavitation resistance of the anodic oxide film treated with sealing in natural seawater solution. Then, damaged surface morphology was analyzed by scanning electron microscope(SEM) and 3D microscope. As the results of the electrochemical experiments, it was observed that the surface damage of all the experimental conditions in the anodic polarization experiment was locally grown by the combination of crack and corrosion damage. In the Tafel analysis, the corrosion resistance of all sealing treatment conditions was improved compared to the anodizing. On the other hand, cavitation erosion tests showed that the anodizing and all the sealing treatment conditions generated local pit damage by cavitation erosion attack and grew to crater damage in the observation of damaged surface by SEM. Also, the weight loss and the surface damage depth measured with the experiment time presented that most of the sealing treatment conditions showed better cavitation erosion resistance than the anodizing, and they had an incubation period at the beginning of the experiment.

The Study on the Characteristics of Pellets Manufactured with Morphologically Different Domestic Larix Kaemferi Carr Sawdust (국내산 낙엽송의 톱밥 유형에 따른 펠릿특성에 관한 연구)

  • morphologically different Larix Kaemferi Carr sawdust. The pellet characteristics included moisture contents satisfied the first grade (less than 10%) of quality standard of wood pellets announced by Korea Forest Research Institute. Densities were also adequate for the first grade (640kg/$m^3$) on the quality standard and there was no large difference in other characteristics of pellets, however, pellets manufactured with planer shavings had the highest density. Ash contents also passed the first grade (less than 0.7) of quality standard. But bark pellet exceeded the second grade (1.5%) in ash content. Lower absorption ratio of bark pellet showed the effect on hygroscpicity. The heating value of the manufactured wood pellets in this study exceeded the first grade of quality standard(more than 4,300 kcal/kg).

  • Poymer blends of two degradable aliphatic polyesters, relatively expensive material polylactic acid (PLA) and relatively inexpensive material poly(butylene adipate-co-terephthalate) (PBAT), were used in this study. Three different kinds of modifiers were used with various amounts. Diisocyanate type methylenediphenyl 4,4'-diisocyanate (MDI) and hexamethylene diisocyanate (HDI) were used as modifiers and epoxy type coupling agents also used. The melt flow index (MFI) and dynamic viscoelasticity of various compositions of PLA/PBAT blends were studied. The mechanical property and morphology with respect to the fracture surface of PLA/PBAT blends were also investigated using tensile test and field emission scanning electronic microscopy, respectively. These tests were also used to verify the compatibility of PLA/PBAT and the effect of mechanical properties due to the use of modifiers. Tensile properties of PLA/PBAT blends modified with HDI were improved remarkably.

3-D Structure of a Coronal Jet Seen in Hinode, SDO, and STEREO

  • We have investigated a coronal jet near the limb on 2010 June 27 by Hinode/X-Ray Telescope (XRT), EUV Imaging Spectrograph (EIS), Solar Optical Telescope (SOT), SDO/Atmospheric Imaging Assembly (AIA), and STEREO. From EUV (AIA and EIS) and soft X-ray (XRT) images we identify the erupting jet feature in cool and hot temperatures. It is noted that there was a small loop eruption in Ca II images of the SOT before the jet eruption. Using high temporal and multi wavelength AIA images, we found that the hot jet preceded its associated cool jet. The jet also shows helical-like structures during the rising period. According to the spectroscopic analysis, the jet structure changes from blue shift to red one with time, implying the helical structure of the jet. The STEREO observation, which enables us to observe this jet on the disk, shows that there was a dim loop associated with the jet. Comparing the observations from the AIA and STEREO, the dim loop corresponds to the jet structure which implies the heated loop. Considering that the structure of its associated active region seen in STEREO is similar to that in AIA observed 5 days before, we compared the jet morphology on the limb with the magnetic fields extrapolated from a HMI vector magnetogram observed on the disk. Interestingly, the comparison shows that the open field corresponds to the jet which is seen as the dim loop in STEREO. Our observations (XRT, SDO, SOT, and STEREO) are well consistent with the numerical simulation of the emerging flux reconnection model.

Characterization of Electro-deposited Ni-P Layer by Using Dynamic Nano-Indentation Method (동적 나노압침법을 이용한 Ni-P 도막의 특성 연구)

  • Dynamic nano-indentation method was applied to characterize thin electroformed Ni-P layers. The Ni-P layers were produced in a sulphamic acid bath at $50^{\circ}C$ in $0.02A/cm^2$ for 10-60 minutes. The chemical analyses by XRD and EDX showed that the Ni-P layers were very fine grains with mainly $Ni_3P$ with Ni. The surface roughness determined by atomic force microscopy increased with thickness, which was relative to the surface morphology. The nano-hardness and the stiffness of the thin Ni-P layers with thickness of 1.9, 6.2 and $7.5{\mu}m$ were 5.52, 6.52 and 6.77 [GPa] and 56.7, 76.2 and 108.0 [${\mu}N/nm$], respectively. The elastic modulus of the Ni-P layer increased with thickness such as 37.29, 54.50 and 78.76 [GPa], respectively. The surface roughness of the electroplated Ni-P layers with diverse thickness was 8.66, 18.56 and 35.22 [nm], respectively. The enhanced nano-mechanical properties were related to mainly residual stress of the Ni-P layers.

Preparation and Characterization of Gelatin-immobilized Bacterial Cellulose Scaffold for Tissue Engineering Using Gamma-ray Irradiation (감마선을 이용한 조직공학용 젤라틴이 개질된 미생물 셀룰로오스 지지체의 제작 및 특성)

  • Bacterial cellulose (BC) is generated from citrus gel by Gluconacetobacter hansenii TL-2C. BC has good properties such as high-burst pressure, high-water contact and the ultrafine highly nanofibrous structure of mimic natural extracellular matrix (ECM) for tissue engineering. In this study, acrylic acid (AAc) was grafted onto BC surfaces under aqueous conditions using gamma-ray irradiation, and then immobilized gelatin onto AAc-g-BC. The characterization of scaffolds was performed by scanning electron microscopy (SEM), attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR), toluidine blue O (TBO) assay. Morphology of gelatin and AAc incorporation onto BC nanofibers did not changed. Our study suggests that gelatin-immobilized BC nanofibers scaffold has a potentiality to fabricate 3D nanofibrous scaffolds for tissue engineering.
