Lignans from the Root of Acanthopanax chiisanensis Nakai

  • Five lignans were isolated from the chloroform fraction of Acanthopanax chiisanensis Nakai root by open column chromatography. Their structures were elucidated as (-)-sesamin (1), helioxanthin (2), savinin (3), taiwanin C (4), and 3-(3",4"-dimethoxybenzyl)-2-(3',4'-methylenedioxybenzyl) butyrolactone (5) by spectral analysis. Among them, compounds 2, 3, 4, and 5 were isolated for the first time from this plant.

A Study on the Establishment of Design Criteria for Anchorage According to Port Characteristics

  • This study suggests design criteria to evaluate the availability of anchorage in Korea to contribute to ship safety by presenting necessary design criteria for anchorage volume according to port development. Accordingly, the concept of "necessary volume of anchorage" is introduced to evaluate the volume of anchorage available in Korea's major ports, and classify these ports into three types according to the characteristics of incoming ship. Numerical simulations designed using MATLAB-SIMULINK have been carried out to track the irregularly of arrival and, waiting times along with the environmental conditions that affect anchorage and necessary volume of anchorage have been suggested based on these tests. Finally, in order to complete a function equation analysis, the necessary volume of anchorage with reference to cargo volume is addressed using regression analysis as follows. Group $A-Y_{NA}=0.0002X_{HA}-3.67$, Group $B-Y_{NB}=0.0002X_{HB}-6.82$, Group $C-Y_{NC}=0.0001X_{HC}+9.02$. This study contributes to a review of anchorage volume from the perspective of harbor development.

Ferroelectric Properties of Bi3.25La0.75Ti3O12 Thin Films Prepared by MOD (MOD 법으로 제작된 Bi3.25La0.75Ti3O12 박막의 강유전 특성)

  • We have fabricated $Bi_{3.25}La_{0.75}Ti_3O_12$ (BLT) thin films on the Pt/Ti/$SiO_2$/Si substrates using a metalorganic decomposition (MOD) method with annealing temperature from $550^{\circ}C$ to $750^{\circ}C$. The structural properties of BLT films examined by x-ray diffraction (XRD). From XRD analysis. BLT thin films show polycrystalline structure. The layered-perovskite phase was obtained by spin-on films at above $600^{\circ}C$ for 1h. Scanning electron microscopy (SEM) showed uniform surface composed of rodlike grains. The grain size of BLT films increased with increasing annealing temperature. The BLT film annealed at $650^{\circ}C$ was measured to have a dielectric constant of 279, dielectric loss of 1.85(%), remanent polarization of $25.66\mu C/\textrm{cm}^2$, and coercive field of 84.75 kV/cm. The BLT thin films showed little polarization fatigue test up to $3.5{\times}10^9$ bipolar cycling at 5 V and 100 kHz.

A Study on Microorganisms Decontamination Using a Vapor-Phase Hydrogen Peroxide System (과산화수소 증기 시스템을 이용한 미생물 제독에 관한 연구)

  • Objectives: Effectiveness and conditions of vapor-phase hydrogen peroxide (VPHP) system on decontamination of Geobacillus stearothermophilus(GS) spores, Escherichia coli (E.coli) and Enterobacteria phage felix01 (felix01) were determined. Methods: The VPHP system was designed to vaporize 35% (w/w) solution of hydrogen peroxide, continuously to inject and withdraw VPHP. The system and VHP 1000ED (Steris) were operated such that dehumidification and conditioning were initiated without samples in the chamber. Then the samples were loaded into and removed. Coupons (glass, anodizing, silicon, viton) with GS spores ($1{\times}10^6$ colony forming unit/mL [CFU/mL]), E.coli ($1{\times}10^7$ CFU/mL) and felix01 ($1{\times}10^7$ plaque forming unit/mL[PFU/mL]), and Biological Indicator (BI) with GS spores ($1{\times}10^6$ CFU/mL) on stainless steel coupons were used. The tested samples were sonicated and vortexed, and then were plated for enumeration, followed by incubation at $55^{\circ}C$, 24 hr for GS spores, and at $37^{\circ}C$, 24 hr for E.coli and felix01. BI analysis in broth culture was only qualitative. Results: The efficacy of the VPHP system on decontamination was almost equivalent to that of VHP 1000ED. The conditions for complete decontamination with the VPHP system was as follows: concentration; 700~450 ppm, relative humidity; approximately 55%, and temperature; $34{\sim}32^{\circ}C$. When comparing the decontamination efficiency among different kinds of coupons, glass was the most effective, however, all kinds of coupons were decontaminated completely after 60 min exposure in both systems. Conclusion: The VPHP system can be recommended as an alternative system for traditional system using ethylene oxide, formaldehyde or chlorine dioxide.

Alterations of Protein Expression in Macrophages in Response to Candida albicans Infection

  • Although macrophages are an important first line of cellular defense, they are unable to effectively kill phagocytosed C. albicans. To determine the physiological basis of this inability, we investigated the alterations of macrophage proteins caused by C. albicans infection. Since the formation of C. albicans hyphae caused cell death, proteins were prepared 3 h after infection and examined by two-dimensional gel electrophoresis (2-DE). The most prominent changes were in glycolytic enzymes, which could have caused energy depletion of the infected cells. Also changed were proteins involved in maintenance of cellular integrity and NO production. Treatment of the macrophages with either cytochalasin D or taxol did not alter their inability to kill C. albicans. Our results indicate that multiple factors contribute to cell death as the pathogenic form of C. albicans becomes fully active inside macrophage cells.

Pythium subutonaiense, A New Aquatic Oomycete from Southern China Based on Morphological and Molecular Characters

  • A new species, Pythium subutonaiense, isolated from aquatic environments (lake) in China is being described based on morphological characters and molecular evidence. The isolates grew at temperatures between $5^{\circ}C$ and $38^{\circ}C$, and the optimum temperature was $30^{\circ}C$, with a radial growth rate of 17.6 mm at $25^{\circ}C$ per day. It is homothallic and characterized by globose to sub-globose shaped and mostly terminal or sometimes catenulate hyphal swellings, filamentous non-inflated sporangia, and smooth oogonia with hypogynous and monoclinous antheridia that contained one plerotic oospore. In phylogenetic analysis, inferred based on the internal transcribed spacer region of the ribosomal RNA gene and mitochondrial cytochrome c oxidase subunit 1 gene, the new species formed a distinct lineage in Pythium clade B. Differences between the new species and phylogenetically related and morphologically similar species are discussed.

Physicochemical Characterization and Dissolution Properties of CS-891 with Different Crystallinity

  • Ground CS-891 (N-[1-(4-methoxyphenyl)-1-methylethyl]-3-oxo-4-aza-5a-androst-1-ene-$17{\beta}$-carboxamide) of poorly water soluble drug was obtained using a Heiko Seisakusho model TI-100 vibration mill, and samples with different crystallinity were prepared at mixture ratios of 10:0, 7:3, 5:5, 3:7 and 0:10 (intact;ground CS-891). Physicochemical characterizations were obtained using qualitative and quantitative X-ray diffractometry, different scanning calorimetry (DSC), scanning electron microscopy (SEM), Quantasorb surface area analyzer, and controlled atmosphere microbalance. With increase of amorphous CS-891 in mixture ratios, the intensities of X-ray diffraction peaks of crystalline CS-891 were decreased, whereas surface area, water absorption, and exothermic peaks in DSC were increased. The apparent solubility of ground CS-891 was $4.4\;{\mu}g/ml$ and the solubility of intact CS-891 was $3.1\;{\mu}g/ml$ at $37{\pm}1^{\circ}C$. The apparent precipitation rates of CS-891 in a supersaturated solution during the solubility test were increased with an increase of amorphous CS-891, and a crystalline form of CS-891 transformed from amorphous CS-891 after the solubility test was found by X-ray diffraction analysis, DSC and SEM. The dissolution profiles of CS-891 with different crystallinity at $37{\pm}1^{\circ}C$ by the USP paddle method were investigated, and the apparent dissolution rate constant of ground CS-891 was about 5.9-fold higher than that of intact CS-891. A linear relationships between the crystallinity of CS-891 and the apparent dissolution rate constant (r>0.96) were obtained.

Organic Matter Degradation and Nitrification Characteristics in Aerobic Digestion of Sewage Sludge (슬러지 호기성 소화공정에서의 유기물 분해 및 질산화 특성)

  • Continuous operation of aerobic sludge digestion reactor was attempted for 279 days. Anaerobic digester sludge, the target material of the experiment, was pretreated by sodium hydroxide at $40^{\circ}C$ for 120 minutes, and the pretreated sludge was fed to 5 L CSTR (continuous stirred tank reactor). Reactor performance was affected by properties of input sludge and HRT (hydraulic retention time). 6 days of HRT showed best and stable performance, and under this condition, removal rates of $NH_3$-N, SCOD, TKN, TCOD, SS, and VSS were 97.4%, 81.7%, 68.7%, 61.4%, 50.6%, and 47.0%, respectively. 73.9% of SS in anaerobic digester sludge was reduced by pretreatment and aerobic digestion. Effluent sludge had low soluble COD of 350 mg/L. This implied the sludge was stabilized and suitable for use as liquid fertilizer. Nitrification took place when HRT was higher than 4 days. $NO_3$-N concentration was as high as 658 mg/L while $NH_3$-N was as low as 20 mg/L.

Unified approach to predict the dynamic performance of transportation system considering wind effects

  • Natural hazards, including the wind hazard and others, threaten the integrity of the modern society. A transportation system usually consists of roadways, bridges and related vehicles. Harsh environmental conditions, caused by such as wind, exist in the real world frequently and affect the dynamic performance of the transportation system through their interactions. Long-span bridges are usually the backbones of transportation lines. In windy conditions, the information about the dynamic performance of bridges and vehicles considering full interactions of environmental factors is very essential for people to assess the overall operational conditions and safety risks of the transportation lines. Most of existent approaches target specifically at several isolated tasks considering partial interaction effects. In order to improve the understanding of these related-in-nature problems integrally as well as the consistency of different approaches, a unified approach to integrally predict the dynamic performance of long-span bridges and vehicles under wind is introduced. Such an approach can be used as a general platform to predict the dynamic responses of vehicles and bridges under various situations through adopting both commercial and in-house software. Dynamic interaction effects can be fully considered automatically for each situation. An example of a prototype bridge in US is given for the purpose of demonstration.

Draft genome sequence of a marine Flavobacteria Flagellimonas eckloniae DOKDO 007T (해양 Flavobacteria Flagellimonas eckloniae DOKDO 007T 의 유전체 염기서열 해독)

  • Flagellimonas eckloniae DOKDO $007^T$, isolated from the rhizosphere of the marine algae Ecklonia kurome collected from Dokdo Island, South Korea, is a marine Flavobacteria belonging to the family Flavobacteriaceae. The genome consists of 4,132,279 bp, 3,527 coding sequences with 37.85% G + C contents and two contigs in one scaffold chromosome. This strain contains a gene encoding proteorhodopsin, as well as other retinal biosynthesis genes, allowing it to utilize sunlight as an energy source. The strain contained only few segment of flagellar constructing gene cluster and this is not consistent with genus name Flagellimonas, therefore, revision of the genus name is required.
