Pseudomonas sp. strain DJ77에서 Plant-Type의 Ferredoxin을 암호화하는 phnM 유전자의 구조 (Genetic Structure of the phnM Gene Encoding Plant-Type Ferredoxin from Pseudomonas sp. strain DJ77)

  • Pseudomonas sp. DJ77로부터 전보에서 클로닝한 pHENX7의 하류방향으로 약 3kb 정도를 포함하는 pYCS500을 클로닝하였다. PYCS500의 제한효소지도를 작성하고 부분적으로 염기서열을 분석한 결과 465 bp의 HindIII-ClaI절편에서 282 bp로 이루어진 하나의 open reading frame(ORF)을 발견하였다. phnM으로 명명된 이 ORF는 93개의 아미노산으로 구성된 polypeptide를 암호화하고 있었으며 계산된 분자량은 10,008 Da이었다. PhnM은 NahT, XylT, DmpQ, AtdS, PhlG, PhhQ, TbuW 등 plant-type ferredoxin 형태의 단백질과 37.7%-53.9%의 상동성을 나타내었으며 이들이 공통적으로 가지고 있는 motif가 일치하였다.

임계열유속 향상을 위한 나노물질의 산화처리에 대한 연구 (Study on the Oxidation Treatment of Nanoparticles for the Critical Heat Flux)

  • Pool boiling, one of the key thermal-hydraulics phenomena, has been widely studied for improving heat transfer efficiencies and safety of nuclear power plants, refrigerating systems, solar-collector heat pipes, and other facilities and equipments. In the present study, the critical heat flux (CHF) and heat-transfer coefficients were tested under the pool-boiling state using graphene M-5 and M-15 nanofluids as well as oxidized graphene M-5 nanofluid. The results showed that the highest CHF increase for both graphene M-5 and M-15 was at the 0.01% volume fraction and, moreover, that the CHF-increase ratio for small-diameter graphene M-5 was higher than that for large-diameter graphene M-15. Also at the 0.01% volume fraction, the oxidized graphene M-5 nanofluid showed a 41.82%-higher CHF-increase ratio and a 26.7%-higher heat-transfer coefficient relative to the same nanofluid without oxidation treatment at the excess temperature where the CHF of distilled water occurs.

시티딘 정량을 위한 식물조직 센서 (Plant Tissue Sensor for Determination of Cytidine)

  • 시티딘은 장미조직이 가지고 있는 시티딘 디아미나제에 의해 우리딘과 암모니아 기체를 생성한다. 이 장미조직 50mg을 암모니아 기체감응센서에 고정시켜 장미조직센서를 제조하고, 수용액 중의 시티딘을 정량하는데 최적 실험조건을 조사하였다. pH8.4, 온도 37$^{\circ}C$, 0.2M 인산 완충용액을 사용하였을 때 시티딘의 농도가 $7.0 {\times}10^{-4}$에서 $1.0{\times}10^{-2}$M까지 직선이었고, 53mV/decade의 감응기울기를 나타내었다. 검출한계는 $3.0 {\times} 10^{-4}$ M 상대표준편차는 3.4%였다. 또한 이 조직센서는 선택성이 탁월하였다.

  • 다중 교반형 생물반응기 내에서 배추 PLD에 의한 인 지방질의 염기 가수분해 반응 및 염기전이 반응에 대한 특성 실험에서 다음과 같은 결론을 얻었다. 반응 최적 조건으로서 pH 5.6, 온도 37$^{\circ}C$였고 반응 촉진제로서 $Ca^2^+$의 농도는 가수분해 반응시 20mM, 염기전이 반응에서는 40mM이었다. Diethylether와 butylacetate도 강력한 반응촉진제로서 반응성을 높였고 이들의 농도는 20%가 효과적이었으며, 반면에 EDTA 및 $Ba^2^+,Mn^2^+,Zn^2^+$등은 PLD의 활성을 강력하게 저해하였다, 염기 수용체로서 glycerol을 비롯한 알콜 화합물은 대부분이 반응성을 양성으로 나타내었으며 15종의 알콜 화합물 가운데 glycerol, ethyleneglycol, propyleneglycol등이 반응성이 높게 나타났으며 이들의 최적농도는 20%가 효과적이었다.

Increased DNA Polymerase Fidelity of the Lamivudine Resistant Variants of Human Hepatitis B Virus DNA Polymerase

  • Although efficient antiviral lamivudine is used for HBV-infected patients, a prolonged treatment with nucleoside analogs often results in lamivudine-resistant variants. In this study, we evaluated the fidelity of the lamivudine-resistant variants. The FLAG-tagged wild-type (FPolE) and Met550 variants (FPolE/M550A, M550V, and M550I) of HBV DNA polymerases were expressed in insect cells then purified. Like many other reverse transcriptases, no $3'{\rightarrow}5'$ exonuclease activity was detected in the HBV DNA polymerase. Since there is no proofreading activity, then the use of the site-specific nucleotide misincorporation method is beneficial. From the $f_{ins}$ value analysis, it is evident that M550I and M550V exhibit higher fidelity values than the wild-type HBV DNA polymerase, while M550A exhibits similar fidelity values. It is therefore suggested that lamivudine resistance comes from the stringency to dNTP binding and the discrimination of dCTP and lamivudine in M550V and M550I.

공업도시의 금속원소 조사와 특성 연구 (Study on Investigation and Characteristics of Metallic Elements in Industrial Complex)

  • Objectives: In this study, the size distribution of airborne particulates ($PM_{10}$) was measured by using Cascade Impactors. The purpose of this study was to assess the size distribution of metal and ionic materials of $PM_{10}$. Methods: Samples were collected in the Kunsan industrial complex from April 2006 to January 2007. Results: The mass fraction of $PM_{10}$ had a bimodal distribution between 2.1-3.1 ${\mu}M$, and the average mass fraction of particles less than 2.1-3.1 ${\mu}M$ was 47%. Average concentrations of PM10 were 68.05 ${\mu}g/m^3$ and seasonal concentration 95.44 ${\mu}g/m^3$ for spring, 49.03 ${\mu}g/m^3$ for summer, 81.99 ${\mu}g/m^3$ for fall, 52.66 ${\mu}g/m^3$ for winter, respectively. Conclusions: Seasonal variations of $PM_{10}$ were significant for showing peak values in spring. The average concentrations of Cd, Cr, Pb, and Fe were 1.54, 4.51, 14.11, and 254.3 $ng/m^3$, respectively. The ratios of fine particles to total mass were 0.47 for $PM_{10}$, 0.45 for Cr, and 0.16 for Fe, 0.91 for Cd and 0.49 for Pb, respectively.

Lipoxygenase Inhibitory and Antioxidant Activities of Isolated Compounds from Moutan Cortex

  • Phytochemical investigation on the ethyl acetate and n-butanol fractions of Moutan Cortex resulted in the isolation and characterization of a new monoterpene glycoside (3) and twenty known monoterpene glycosides (1, 2, 4-21). The structure of 3 was determined by spectroscopic data interpretation and physico-chemical properties. Compounds 1 and 8 presented a remarkable inhibitory activity against lipoxygenase-1 (LOX-1) with $IC_{50}$ values of 45.2 and $37.5\;{\mu}M$, respectively. Compounds 9, 10, 13, 18, 19, and 21 showed significant 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect with $IC_{50}$ values of 9.8, 25.5, 6.4, 15.2, 18.7, and $23.7\;{\mu}M$, respectively. Benzoylpaeoniflorin (8), which exhibited the highest inhibitory effect with an $IC_{50}$ value of $37.5{\pm}0.7{\mu}M$, was further analyzed the inhibition kinetics by Lineweaver-Burk plots. Results indicated that 8 is a non-competitive inhibitor, and the kinetic parameter values were estimated to be ($31.04\;{\mu}M$, Ki), ($0.29\;{\mu}M/min$, $V_m$), and ($48.50\;{\mu}M$, $K_m$).

  • Solar ultraviolet-A (UV-A) irradiances indicent on a horizontal surface at Taegu of Korea during 1995-1998 were calculated with 5 min averages of measurements taken every 30 seconds by a broadband UV- A sensor. The maximum and minimum of monthly averages of daily UV-A dose were 499.37 KJ m-2 day-1 inJuly and 171.09 KJ m--2 day-1 in December for 4 years of the observation period. The maxima of daily UV-A dose and instandaneous UV-A were observed as 846.46KJ m-2 day-1 June 7, 1998 and 37.22 W m-2 at 12 : 15, July 16, 1998, respectively. Increasing trends in annual maxima of daily UV-A dose and instantaneous UV-A were averaged to 6.4% and 6.7% , respectively, per year during 1995-1998 at Taegu, Korea.

  • The effects of the moments of the retrial time to the system performance measures such as blocking probability, mean and standard deviation of the number of customers in service facility and orbit are numerically investigated. The results reveal some performance measures related with the number of customers in orbit can be severely affected by the fourth or higher moments of retrial time.

A Spirulina maxima-derived peptide inhibits HIV-1 infection in a human T cell line MT4

  • Human immunodeficiency virus (HIV) is the causative agent of acquired immune deficiency syndrome (AIDS). Anti-HIV agents targeting various steps in HIV life cycle have been developed; however, so far, no effective drugs have been found. We show here that a peptide isolated from Spirulina maxima (SM-peptide) inhibits HIV-1 infection in a human T cell line MT4. SM-peptide inhibited $HIV-1_{IIIB}$-induced cell lysis with a half-maximal inhibitory concentration ($IC_{50}$) of 0.691 mM, while its 50 % cytotoxic concentration ($CC_{50}$) was greater than 1.457 mM. Furthermore, the SM-peptide inhibited the HIV-1 reverse transcriptase activity and p24 antigen production. This suggests that SM-peptide is a novel candidate peptide, which may be developed as a therapeutic agent for acquired immunodeficiency syndrome patients.
