Evaluation of the Microbiological Safety for the Seawater in a Part of the South Coast, Korea (남해안 일부해역 해수의 세균학적 위생안전성 평가)

  • This study was designed to evaluate the sanitary characteristics of sea water in a part of the south coast, Korea and to check the seawater which is in compliance with the recommended bacteriological criteria for shellfish cultivation. The samples of sea water were collected at 14 sampling stations established in the survey area between March 2014 and October 2014. Food poisoning caused by seafood consumption is often associated with pathogenic microorganisms originated from fecal contamination. Therefore, fecal coliform is very important criteria for evaluating the safety of fisheries in coastal areas. The range of geometric mean (GM) and the estimated 90th percentile values of total coliform were 4.1~83.1 MPN/100 mL, and 11.7~834.1 MPN/100 mL, respectively. The GM and the estimated 90th percentile values of fecal coliform were 2.5~22.7 MPN/100 mL and 2.5~170.0 MPN/100 mL, respectively. Therefore, the bacteriological safety of seawater at this shellfish-growing area met with the Korean Shellfish Sanitation Program (KSSP) criteria for a growing area. However, the values are seasonally exceed the KSSP criteria, suggesting that the monitoring and evaluation of seawater quality is very important in shellfish-growing area.

A Study on he Optical and Electrical Properties of $In_2O_3-ZnO$ Thin Films Fabricated by Pulsed Laser Deposition (PLD 법으로 제작한 $In_2O_3-ZnO$ 박막의 광학적 및 전기적 특성)

  • In this study, $In_2O_3-ZnO$ thin films are prepared on quartz substrates by the pulsed laser deposition and their optical and electrical properties are investigated as the function of substrate temperatures ($200{\sim}600^{\circ}C$) at the fixed oxygen pressure of 200 mTorr. The XRD measurement shows that polycrystalline $In_2O_3-ZnO$ thin films are formed. In the XRD measurement, the intensity of the (400) $In_2O_3$ peak at $35.5^{\circ}$ decreases and that of the (222) $In_2O_3$ peak at $30.6^{\circ}$ increases with the increase substrate temperature up to $500^{\circ}C$. From the result of AFM measurement, the morphology of $In_2O_3-ZnO$ thin films are observed as round-type grains. The lowest surface roughness (6.15 nm) is obtained for the $In_2O_3-ZnO$ thin film fabricated at $500^{\circ}C$. The optical transmittance of $In_2O_3-ZnO$ thin films are higher than 82% in the visible region. The maximum carrier concentration of $2.46{\times}10^{20}cm^{-3}$ and the minimum resistivity of $1.36{\times}10^{-3}{\Omega}cm$ are obtained also for the $In_2O_3-ZnO$ thin film fabricated at $500^{\circ}C$.

Changes of Nitrogen-Fixation Activity and Environmental Factors of Growth in Lespedeza bicolor Turcz (싸리(Lespedeza bicolor Turcz.)의 공생 질소고정활성과 생육환경요인의 변화)

  • The nitrogen fixation activity and environmental factors of Lespedeza bicli!oy Turcz, forming annual root nodules by symbiotic Rhizobium sp. were analyzed in the field conditions during the growing period. Seasunal changes of $N_2-fixation$ activity showed the maximum value of $120\;\mu\textrm{M}\;C_2_H4{\cdot}noduie\;g\;fw^{-1}{\cdot}hr^{-1}$ during the active growing period (June) and varied significantly depending on the growth phase and environmental factors. The maximum activities were attained at the conditions of pH 7, $30^{\circ}C$ of temperature, 18 Kpa of oxygen partial pressure and inhibited by water stress and nitrogen sources. The habitat soil was weak acidic and poor in nitrogen, phosphorus and organic matter contents. The leaf area ratios and chlorophyll contents were ranged from 442 to $48;\textrm{cm}^2{\cdot}g\;dw^{-1}$ and from 33 to $38\;\mu\textrm{g}\;chI{\cdot}\textrm{cm}^2$. Nitrogen contents in each organ showed the maximum of 46, 19 and $11\;mg{\cdot}g\;dw^{-1}, respectively for leaf, rool and stem in the early period. The highest phosphorus contents were 4.2, 1.2 and $0.6\;mg{\cdot}g\;dw^{-1}$, respectively for leaf. root and stem in early growing period. The allocation ratios of nitrogen quantity showed 60% for leaves and 73% for roots during the active and late growth period, and 22% [or stems in average. The allocalion ralios o[ phosphorus quantity showed 58% for leaves during the most productive period, 70% for roots in the pre-growth stage and 26% for stems in average.verage.

A Study on Clogging and Water Quality Improvement in Floodplain Filtration with Flood/rest Raw-water Supply (범람/휴지식 홍수터여과에서 폐색현상 및 수질개선도 연구)

  • A pilot-scale experiment of floodplain filtration with a filtration depth of 3.6m was performed employing flood/rest type raw-water supply system in an effort to find ways to improve river water quality by additional treatments of discharged effluent from sewage treatment plant. Soil samples were taken from 3 sites including Gumi, Daegu and Gimhae along the Nakdong river. Reductions of infiltration rates following increases in operating time was investigated in each soil sample, along with the analysis of removal efficiencies of various pollutants according to different depths and infiltration rates. The results show incremental development of clogging on the soil surface with increases in operation time, and illustrate exponential decrease in the infiltration rate. The time required for the removal of the clog from the soil surface was longer than 2 weeks for all soil samples analyzed. The stable infiltration rates for soils were 5 m/day for Gumi and for Daegu and Gimhae was 1 m/day. In unsaturated soils dissolved oxygen levels increased following the increase of filtration depth, suggesting that alternating application of flood and rest for raw-water supply effectively keeps the soil environment aerobic. For all soils, the nature of pollutant removal depending on the depth of filtration remained the same regardless of the infiltration rate. Most of the BOD and turbidity were removed within 1.2 m, about 30% of COD was removed within 3.6m and was expected to be removed further with increases in filtration depth. Nitrification occurred near the surface of all soils; however there was no significant removal of nitrogen in the filtration depths tested in this study. Although removal rate of phosphorus was low for Gumi's soil, it was high enough for other soils, suggesting that the method developed in this study could significantly improve river water quality.

Analysis of the particulate matters in the vertical-flow woodchip wetland treating stormwater from paved road (포장도로 강우유출수 처리목적의 수직흐름형 우드칩 충진 습지에서 입자상 물질분석)

  • In this study, three pilot-scale wetland systems were built for treating stormwater runoff from asphalt road. Each of the system consists of a settling tank and a vertical flow wetland packed with 25%, 50%, and 75% woodchip as treatment media. According to the analysis of the distributions of particle size, it was found that solids ranging in size $0.52-30{\mu}m$ were predominant in the stormwater runoff. After 24-hours settling, those coarser than $20{\mu}m$ were significantly detained. Further retention, especially for the finer-sized fraction, occurred in the wetland through internal recirculation during the dry day periods. As a primary media of the wetland, woodchip showed a high filtration and attachment capacity for the particulates in pre-settled stormwater, whereas overall amount of solids in the wetland effluent increased due to the detachment of woody elements from the media. This was observed mainly during the initial 75 days of operation, and the size and detachment rate were found to be strongly related with the woodchip packing ratio. The mechanism involving woody particle detachment was modeled as a first-order form. In addition, water quality factors and operational parameters affecting the detachment were analyzed and discussed.

  • Oxides of nitrogen (NO$_x$) in exhaust gases was determined by absorbing the gas in alkaline peroxide solution containing 0.03${\%}$ H2O2 and 0.1N NaOH. About 100 ppm of NO$_x$ was rapidly oxidized to NO$_2$ or N$_2$O$_5$ by H$_2$O$_2$ and required a contact time of 2 minutes with the absorbing solution for complete absorption. With vigorous shaking including air or oxygen gas, high concentration of NO$_x$ (>200 ppm) can be absorbed within 30 minutes. The remaining H$_2$O$_2$ affect the absorbance of color solution strongly. However, the excess H$_2$O$_2$ was completely decomposed by zinc powder 0.5g and the sample solution should be adjusted to the pH range 6.1∼6.6 before the reduction so that conversion of nitrate to nitrite ion is possible. The absorbed NO$_x$ is determined colorimetrically by the diazotization-coupling method with sulfonilamide and NEDA as the coupling agent. The sensitivity of the new method was 4.48 ${\times}$ 10$^4$ as molar absorptivity which was high sensitive compared with that obtained for the usual zinc reduction NEDA method with O$_3$. This method was far more rapid, brief and accurate than previously published O$_3$-NEDA method in Korean industrial standard.

Effects of Heat Pretreatment on the Quality under Storage of Fresh Ginger Rhizomes (저장 전 열처리가 생강의 이화학적 및 관능적 특성에 미치는 영향)

  • Fresh ginger rhizomes were heat-treated for 10 min, 30 min, or 60 min using hot air ($40^{\circ}C$, $50^{\circ}C$, or $60^{\circ}C$), and stored in low-density polyethylene (0.04 mm thickness) bags for 2 months at $12{\pm}1^{\circ}C$. We studied the effects of heat pretreatment on changes in gas levels after packaging, and quality characteristics of the rhizomes. Oxygen and carbon dioxide levels progressively fell and rose, respectively, as the temperature of heat treatment rose and the duration of such treatment was extended. The sprouting rate of ginger rhizomes treated at $40^{\circ}C$ was higher than that of other samples. Rotting, softening, and increasing pH of rhizomes were accelerated by treatment at higher temperature for a longer time. Weight loss and soluble solid levels were not affected by heat treatment. Sensory qualities such as appearance, odor, and overall acceptability of rhizomes fell with treatment at a higher temperature for a longer time. These results suggest that heat pretreatment has a detrimental effect on the quality of fresh ginger rhizomes.

Treatment of Liquid Waste Containing Highly Concentrated Ethylenediaminetetraaceticacid by Using Underwater Electrical Discharge (수중 전기방전을 이용한 에틸렌디아민테트라아세트산 폐액의 처리)

  • This study investigated the treatment of liquid waste containing highly concentrated iron(III)-ethylenediaminetetraaceticacid (Fe(III)-EDTA) of 70,000 mg/L by an underwater electrical discharge process using low voltage and high current. When AC voltage is applied to the discharging electrode with the other electrode grounded, the temperature of the liquid waste around the discharging electrode rapidly increases, and at the same time, hydrogen and oxygen gases are formed at the electrode as a result of electrochemical reactions. Ultimately, gases formed by vaporization of water and electrochemical reactions cover the electrode. Since the liquid waste is electrically conductive, it elongates the ground electrode up to the border of the gas layer, where electrical discharge occurs. Without hydrogen peroxide, electrical discharge was able to remove about 50% of Fe(III)-EDTA. As the concentration of hydrogen peroxide added increased, the removal efficiency of Fe(III)-EDTA increased. When the molar ratio of hydrogen peroxide to the initial Fe(III)-EDTA was higher than 24.7, more than 80 g of Fe(III)-EDTA was removed with an energy of 1 kWh. A comparison between tungsten and steel electrodes showed that electrode material did not affect the Fe(III)-EDTA removal. In the present underwater electrical discharge process, the removal of Fe(III)-EDTA was completed within 30 min at molar ratios of hydrogen peroxide to the initial Fe(III)-EDTA higher than 24.7.

Fluid Inclusion and Sulfur Stable Isotope of Buckchang Deposit, Korea (북창광상의 유체포유물 및 황안정동위원소 연구)

  • The Buckchang deposits which is located in the Ockcheon metamorphic zone, are emplaced along $N20-30^{\circ}E$ trending fissure sets. So it is a sort of fissure-filling ore deposits. The results of mineral paragenetic studies suggest two stages of hydrothermal mineralization; stage I: base-metal sulfides stage, stage II: late base-metal sulfides, electrum and silver-bearing sulfosalts stage. The silver-bearing sulfosalts occured as the Buckchang mine are mainly argentite and, minor of canfieldite, tetrahedrite, etc. Au:Ag ratios of the electrums show a highly limited range of nearly 1:1 in atomic %. The temperature, salinity and pressure of the Buckchang deposits estimated from fluid inclusion and sulfur isotope studies are as follows; stage I: $174{\sim}250^{\circ}C$, 0.35~4.01 NaCl eq. wt.%, 0.40~1.00 Kbar, stage II: $138{\sim}222^{\circ}C$, 1.9~8.4 NaCl eq. wt.%, 0.22~0.53 Kbar. The estimated oxygen and sulfur fugacity during stage I mineralization, based on phase relation of associated minerals, range from $10^{-39.7}{\sim}10^{-44.7}$ atm. and $10^{-13.4}{\sim}10^{-18.1}$ atm., respectively. All these evidences suggest that the Buckchang deposits are polymetallic epithermal ore deposits.

Structural and Optical Properties of Sol-gel Derived ZnO:Cu Films

  • 최근 단파장 광전 소자와 고출력 고주파 전자 소자에 대한 수요 때문에 넓은 밴드갭 에너지를 갖는 반도체에 관심이 많다. 이중에서, ZnO는 우수한 화학 및 역학적 안정성, 수소 플라즈마 내구성과 저가 제조의 장점 때문에 광전자 소자 개발 분야에 적합한 산화물 투명 전극으로 관심을 끌고 있다. 불순물이 도핑되지 않은 ZnO는 본질적으로 산소 빈자리 (vacancy)와 아연 격자틈새 (interstitial)와 같은 자체의 결함으로 말미암아 n형의 극성을 갖기 때문에, 반도체 소자로 응용하기 위해서는 도핑 운반자의 농도와 전도성을 제어하는 것이 필요하다. 본 연구에서는 박막 제조시 제어성, 안정성과 용이하게 성장이 가능한 졸겔 (sol-gel) 방법을 사용하여 사파이어와 석영 기판 위에 Cu가 도핑된 ZnO 박막을 성장시켰으며, 그것의 구조, 표면 형상, 평균 투과율, 광학 밴드갭 에너지를 계산하였다. 특히, Cu의 몰 비를 0, 0.01, 0.03, 0.05, 0.07, 0.1 mol로 변화시키면서 ZnO:Cu 박막을 성장시켰다. ZnO:Cu 졸은 zinc acetate dihydrate, 2-methoxyethanol (용매), momoethanolamine (MEA, 안정제)을 사용하여 제조하였다. 상온에서 2-methoxyethanol과 MEA가 혼합된 용액에 zinc acetate dihydrate (Zn)을 용해시켰다. 이때 MEA와 Zn의 몰 비는 1로 유지하였다. 이 용액을 $60^{\circ}C$ 가열판 (hot plate)에서 24 h 동안 자석으로 휘젓으며 혼합하여 맑고 균일한 용액을 얻었다. 이 용액을 3000 rpm 속도로 회전하는 스핀 코터기의 상부에 장착된 사파이어와 석영 기판 위에 주사기 (syringe)를 사용하여 한 방울 떨어뜨려 30 s 동안 스핀한 다음에, 용매를 증발시키고 유기물 찌꺼기를 제거하기 위하여 $300^{\circ}C$에서 10분 동안 건조시킨다. 기판 위에 코팅하는 작업에서 부터 건조 작업까지를 10회 반복한 다음에, 1 h 동안 전기로에 장입하여 석영 기판 위에 증착된 시료는 $550^{\circ}C$에서, 사파이어 기판은 $700^{\circ}C$에서 열처리를 수행하였다. Cu의 몰 비 0, 0.01, 0.03, 0.05, 0.07, 1로 성장된 ZnO:Cu 박막에 대한 x선 회절 분석의 결과에 의하면, 모든 ZnO:Cu 박막의 경우에 관측된 34.3o의 피크는 ZnO (002) 면에서 발생된 회절 패턴을 나타낸다. 이것은 JCPDS #80-0075에 제시된 회절상과 일치하였으며, ZnO:Cu 박막이 기판에 수직인 c-축을 따라 우선 배향됨을 나타낸다. 사파이어 기판 위에 증착된 박막의 경우에, Cu의 몰 비가 점점 증가함에 따라(002)면 회절 피크의 세기는 전반적으로 증가하여 0.07 mol에서 최대를 나타내었으나, 석영 기판 위에 증착된 박막의 경우에는 0.05 mol에서 최대를 보였다. 외선-가시광 분광계를 사용하여 서로 다른 Cu의 몰 비로 성장된 ZnO:Cu 박막에서 광학 흡수율 (absorbance) 스펙트럼을 측정하였으며, 이 데이터를 사용하여 평균 투과율을 계산한 결과, 투과율은 Cu의 몰 비에 따라 현저한 차이를 나타내었다. Cu의 몰 비가 0.07 mol일 때 평균 투과율은 80%로 가장 높았으며, 0.03 mol에서는 30%로 최소이었다. 광학밴드갭 에너지는 Tauc 모델을 사용하여 계산하였고, 결정 입자의 형상과 크기와의 상관 관계를 조사하였다.
