RP-HPLC법에 의한 무우의 매운 성분인 4-methylthio-3-butenyl isothiocyanate의 분리 및 정제 (Purification of 4-Methylthio-3-butenyl isothiocyanate the Pungent Principle in Radish Roots by RP-HPLC)

  • 무우 속의 매운 맛 성분을 용매추출법 ($CCl_4$,사용)으로 분리하여 RP-HPLC법에 의해 정제한 후에 관능검사에 의해 pungency test를 하고, UV spectrum분석, GC/MC 분석을 수행한 결과 4-methylthio-3-butenyl isothiocyanate 임을 확인하였다. RP-HPLC에서 mobile Phase로 70% acetonitrile을 사용하였는데, 정제된 4-methylthio-3-butenyl isothiocfanate는 단일 peak로 분리되어 나왔고 이때의 retention time은 5.2분이였다. 아울러 RP-TLC에서 Rf치는 0.9이었다.

한국산 무우의 휘발성 함유황화합물에 관하여 (Volatile Sulfur Components from Fresh Radishes of Korean Origin)

  • 생무우(태백무우, 알타리무우)에 존재하는 휘발성 함유황화합물을 수증기 증류 및 용매추출($CCl_4$)방법에 의해 추출하여 GC/MS 분석법에 의해 분리 확인하였다. 수증기 증류로 얻은 2종 무우의 휘발성 함유황 성분들의 GC profile은 유사하였으며, 4-methyl-pentyl, 3-methylthiopropyl, 4-methylthio-3-butenyl, 4-rnethylthiobutyl, 5-methylthiopentyl isothiocyanate 와 5-methylthio-4-pentenenitrile, 1-methylthio-3-pentanone, dimethyl sulfides 등이 분리 확인되었다. 한편 $CCl_4$추출물에는 4-methylsulfinyl-3-butenyl isothiocyanate도 존재하였다. 수증기 증류에서 는 4-methylthiobutyl isothiocyanate가, 그리고 $CCl_4$추출에서 는 4-methylthio-3-butenyl isothiocyanate가 각각 주된 휘발성 함유황 화합물이었다.

배추 종자 및 어린 싹의 성장 시기에 따른 3-butenyl isothiocyanate 및 total glucosinolates 함량 변화 (Changes in 3-Butenyl Isothiocyanate and Total Glucosinolates of Seeds and Young Seedlings during Growth of Korean Chinese Cabbages)

  • 배추의 품종별 종자 및 성장 시기에 따른 3-butenyl isothiocyanate 및 total glucosinolates를 분석하기 위하여 동풍배추(Winter Pride) 및 55일 배추(55 Days) 품종의 종자, 발아 1일, 3일, 5일, 7일 된 어린 배추를 떡잎(cotyledon), 배축(hypocotyl), 뿌리(root) 부분으로 나누고 anion-exchange column(Dowex 1${\times}$2-100)을 통과시키고 아세톤 침전법을 사용하여 제조한 crude myrosinase로 반응시켜 함량 변화를 조사하였다. 종자에서는 55일 배추의 3-butenyl isothiocyanate의 함량이 동풍 배추 종자보다 다소 높았다. 55일 배추의 3-butenyl isothiocyanate 함량은 3-day-old seedlings의 떡잎과 배축에서 가장 높았으며 동풍 배추의 떡잎에서는 3-day-old seedlings까지 함량이 증가한 후 감소하였으며 배축은 3-day-old seedlings까지 함량이 증가하였다. Total glucosinolates를 분석한 결과 종자에서는 동풍 배추보다 35일 배추의 함량이 다소 높게 조사되었으며 55일 배추의 떡잎은 3-day-old seedlings에서 가장 높은 함량을 보였다. 배축 부분은 7-day-old seedlings까지 증가하였으며 뿌리 부분의 glucosinolates는 성장 기간동안 점차 감소하는 경향을 나타내었다. 동풍 배추의 떡잎과 배축 부분은 -day-old seedlings까지 증가하다가 감소하였다.

브로콜리 용매추출물의 Bioactive Organosulfur 화합물의 분리 및 동정 (Isolation and Identification of Bioactive Organosulfur Phytochemicals from Solvent Extract of Broccoli)

  • 브로콜리 중의 bioactive organosulfur phytochemicals를 분석하기 위해 브로콜리를 용매추출하여 GC/MSD에 의해 분리동정하였다. 브로콜리에 함유된 phytochemicals 중에서 isothiocyanate류가 40.42%로 대부분을 차지하였으며, glucosinolate분해산물인 nitrile류는 5.12%로 isothiocyanate류에 비해 적었고, sulfide류는 0.84%로 매우 적었다. 분리 동정된 isothiocyanate류의 종류로는 3-butenyl 4-methtlthiobutyl, 4-methrlthio -3- butenyl, 5- methylthiopentyl, 2- phenylethyl, 3-methylsulfinyl propyl 및 4-methylsulfinylbutyl isothiocyanate의 7종이었다. Isothiocyanate류 중에서 3-butenyl isothiocynate는 22.05%로 가장 많았으며, 그 다음으로 sulforaphane 16.50%로 2종의 isothiocyanate가 대부분을 차지하였다. Nitrile의 종류는 4-methylthiobutyl, 5-methylthiopentyl, 2-phenylethyl 및 4-methylsulfinylbutyl nitrile의 6종류가 확인되었다. 한편, 동정된 sulfide종류로는 dimethyl disulfide, dimethyl trisulfide 및 dimethyl tetrasulfide이었다.

Determination of 3-Butenyl Isothiocyanate in Different Parts and Cultivars of Chinese Cabbages

  • Chinese cabbage (Brassica campestris L. Pekinensis) is Brassica vegetable that contains high amounts of glucosinolates. Glucosinolates and their breakdown products are thought to contribute to health promotion by preventing some cancers. Chinese cabbage is the most commonly consumed vegetable in Asian countries including Korea. In this study, qualitative and quantitative analyses of 3-butenyl glucosinolate (Gluconapin) from different cultivars and different parts of the cabbage were performed. Gluconapin of Chinese cabbage was extracted by hot ethanol ($80^{\circ}C$), isolated by an anion exchange column and identified by GC/MS and LC/MS. The levels of glucosinolates in Chinese cabbage varied according to the different parts, cultivars, and blanching time. In general, the concentrations of 3-butenyl isothiocyanate (ITC) were higher in the leaf than in the midribs parts. The cultivar 'Bulam no. 3' had a much greater content of 3-butenyl ITC than the cultivar 'Garak no. 1,' and the levels of butenyl ITC were highest after two weeks of storage. Blanching treatment decreased the concentration of 3-butenyl ITC. The ITC concentration varied extensively among different crops of the same species, and according to the different parts on the cabbage, the storage duration and the boiling time.

돌산갓의 Isothiocyanate류의 분석 (Analysis of Isothiocyanates in Dolsan Leaf Mustard(Brassica juncea))

  • The composition of isothiocyanates in Dolsan leaf mustard was investigated. Five major volatile isothiocyanates detected in leaf mustard were sec-butyl isothiocyanate, allyl isothiocyanate, 3-butenyl isothiocyanate, n-hexyl isothiocyanate and ${\beta}-phenylethyl$ isothiocyanate. In both leaf and leaf stalk allyl isothiocyanate and 3-butenyl isothiocyanate were the most abundant. The compositional difference of isothiocyanates between leaf and leaf stalk was that n-hexyl and ${\beta}-phenylethyl$ isothiocyanates were present more in leaf than leaf stalk.

Glucosinolates (3-Butenyl Isothiocyanate, Total Glucosinolates) Change in Seeds and Young Seedlings on Chinese Cabbages

  • The main objective of this study was to investigate the patterns of glucosinolates (3-butenyl isothiocyanate, total glucosinolates) in different parts of young seedling and seeds on Korean Chinese cabbages 55 days and Winter pride cultivars. For determination of glucosinolates, two cultivars of Chinese cabbages seeds and different parts of 1-day-old, 3-day-old, 5-day-old, and 7-day-old seedlings were used for analytical sample preparation provided with an anion exchanges column, and measured by GC and UV-Visible Spectrophotometer. 3-Butenyl ITC concentration was the highest in the cotyledon and the hypocotyl parts of 55 days cultivar seedling. In the cotyledon part of Winter pride cultivar seedling, 3-butenyl ITC amount was increase to 3-day-old seedling and then reduced. The cotyledon of 55 days cultivar contained the highest concentration of total glucosinolates while those were increased in the hypocotyl and decreased by degrees in the root. Total glucosinolates amounts were increased to 5-day-old seedling and then decreased in the cotyledon and the hypocotyl parts of seedling on Winter pride cultivar. There was no significant difference in the root part. In the seeds, both of 3-butenyl ITC and total glucosinolates, 55 days cultivar concentration was higher than Winter pride cultivar. The study has shown the variability in 3-butenyl ITC and total glucosinolates concentrations among cultivars, growth stages, and parts.

RP-HPLC법에 의한 무우의 4-methylthio-3-butenyl isothiocyanate의 정량 (Quantitative Determination of 4-methylthio-3-butenyl isothiocyanate in Radish Root by RP-HPLC)

  • RP-HPLC방법에 의해 무우 속에 존재하는 MTBI의 함량을 정량적으로 측정 해 본 결과 10n moles/ml와 120 nmoles/ml사이에서 좋은 직선적인 상관관계(r=0.9997)를 나타내었으며, 편리하고 정확(오차 5%내외)하며 재현성(recovery 99%)이 높았다. 또, MTBI는 알카리용액 중에서는 서서히 분해되었으나 산성용액 중에서는 매우 빠르게 분해되었으며, 이 결과는 231nm에서 UV흡광도가 서서히 감소하는 결과와도 일치하였다. 한편, MTBI는 유기용매(65% acetenitrile)에서 비교적 안정하였다. 또한. 한국산 생무우 중에 함유된 MTBI는 최적추출조건을 PH8.5에서 1분내에 분석하였을때 $210\sim420{\mu}moles/100g$ 정도이었다.

Mitochondria-mediated Apoptosis in Human Lung Cancer A549 Cells by 4-Methylsulfinyl-3-butenyl Isothiocyanate from Radish Seeds

  • 4-Methylsulfinyl-3-butenyl isothiocyanate (MTBITC) found in the radish (Raphanus sativus L.), is a wellknown anticancer agent. In this study, the mechanisms of the MTBITC induction of cell apoptosis in human A549 lung cancer cells were investigated. Our PI staining results showed that MTBITC treatment significantly increased the apoptotic sub-G1 fraction in a dose-dependent manner. The mechanism of apoptosis induced by MTBITC was investigated by testing the change of mitochondrial membrane potential (${\Delta}{\Psi}m$), the expression of mRNAs of apoptosis-related genes by RT-PCR, and the activities of caspase-3 and -9 by caspase colorimetric assay. MTBITC treatment decreased mitochondrial membrane potential by down-regulating the rate of Bcl-2/Bax and Bcl-xL/Bax, and activation of caspase-3 and -9. Therefore, mitochondrial pathway and Bcl-2 gene family could be involved in the mechanisms of A549 cell apoptosis induced by MTBITC.

깍두기 숙성중 매운맛 감소에 관련된 인자들의 변화 (Changes in the Factors Associated with Decrease of Pungency in ‘Kagdugi’ during Fermentation)

  • 깍두기 숙성중 일반적 특성변화로는, 숙성이 진행됨에 따라 pH는 낮아져 3일 후에는 pH 4 이하로 감소되었으며, 산도는 증가하였다. 아스코르브산의 함량은 숙성적기에 약간 증가된 후 다시 감소되었으나 숙성기간동안 최저 1 mM 정도를 유지하였다. 깍두기 숙성중 매운맛이 감소되는 정도를 RP-HPLC에 의해 정량적으로 평가한 결과, 마쇄시 생성되는 MTB-NCS는 숙성이 진행됨에 따라 감소하여 숙성 1일에 81%, 2일에 18%, 2일에 8%, 4일에 3%로 되어 관능검사 결과와 일치하였다. 따라서, 깍두기 숙성중 매운맛의 감소는 MTB-NCS의 생성 감소에 기인된 것이었으며, MTB-NCS의 감소원인은 숙성에 따른 pH 저하로 인한 myrosinase 활성도 감소와 기질인 MTB-glucosinolate의 분해에 의한 것으로 나타났으며, 특히 숙성 3일을 중심으로 이러한 변화가 현저히 관찰되었다.
