Effect of Culture Media on Embryonic Cell Growth in Zebrafish, Danio rerio (배지에 따른 제브라피쉬(Danio rerio) 배아 유래세포의 성장 효과에 관한 연구)

  • To optimize the cell culture conditions of zebrafish embryonic cells, we compared the efficiency of three types of medium, DMEM, K-NAC and D-NAC. In this study, we showed that the cells grown in K-NAC have better plating efficiency than DMEM, especially in the case of low cell seeding density. However, cells grew slower in K-NAC than those in DMEM in confluent cultures. The effect of 0.1% zebrafish embryo extracts was minimal. The presence of 1% trout serum in culture medium significantly increased the growth rate of cells(p<0.05). No difference was found at $2{\sim}3{\times}10^5$ cell seeding density(p<0.05). At $4{\sim}5{\times}10^5$ cell seeding density, cells grew better in DMEM than K-NAC (p<0.1). The results suggest that supplementation of NAC and A2P in Keratinocyte SFM may improves plating efficiency when cells are plated at low population. No difference was found for cell growth in either medium with 5%, 10% or 15% FBS supplemented (p<0.05). Cells culture in D-NAC grew significantly better than those in DMEM(p<0.05). Our results clearly showed that the use of NAC and A2P in the culture medium has a positive effect on cell growth regardless of the amount of FBS added.

Ecology and Growth of Weeds and Weedy Rice in Direct-seeded Rice Fields (직파재배답(直播栽培沓)에서의 잡초(雜草) 및 적미(赤米) 발생(發生) 생태(生態))

  • This experiment was carried out at Yeongnam Crop Experiment Station since 1992 to obtain basic information on ecology of weeds and weedy rice in direct-seeded rice fields. Annual grass weeds of Echinochloa sp., Leptochloa sp., Digitaria sp. and Setaria sp.(C4 plant) and weedy rice(C3 plant) were important species in direct seeded rice compared to transplanted rice field. Period of seedling emergence of barnyard grass was varied from 8 days to 20 days depending on seeding date while it was shorter 4 to 6 days than rice. Weed occurrence and the degree of yield loss were varied by cultivation method. In direct seeding method weeds increased 2 to 3 times compared to manual transplanting. The greatest yield loss was recorded in direct seeding(40 to 100%) followed by mechanical transplanting(25 to 35%) and hand transplanting(10 to 20%), in order. Double cropping of rice-barley was reduced weeds about 30% than rice single crop due to alleopathic effect of barley residue or stubble. Weedy rice(red rice) occurrence was closely related with seeding date and tillage methods. Early seeding and reduced tillage enhanced the growth of weedy rice.

Effect of Spatial Soil Salinity Variation on the Emergence of Soiling and Forage Crops Seeded at the Newly Reclaimed Tidal Lands in Korea (신간척지토양의 공간적 염농도 변이가 녹비·사료작물의 출현에 미치는 영향)

  • Relation between spatial variation of soil salinity and emergence of five upland crops such as sudan grass, sesbania, barnyard grass, corn and soybean was studied in the three reclaimed lands of Korea during two years from 2007 to 2008. Although soil salinity is vary high at seeding season, desalting treatment by three days-flooding before seeding, reach at favorable level lower than $6dS\;m^{-1}$ of soil salinity for emergence of soiling and forage crops and then plant number emerged(No. $m^{-2}$) was 55~149 for sudan grass, 118~266 for barnyard grass, 46~115 for sesbania, 3~11 for corn and 6~19 for soybean in 2007. However plant number emerged under no desalting treatment varies place by place because of soil salinity difference in 2008. Plant number emerged after seeding according to soil salinity was well expressed as logarithmic function, and sharply decrease with increase of soil salinity. It is accordingly concluded that desalting treatment of flooding before seeding of upland crops is essential for good emergence in the newly reclaimed land from tidal flat.

Changes in Methane Emissions from Paddy under Different Tillage and Cultivation Methods (벼 재배 시 경운 및 재배방법에 의한 메탄발생 양상)

  • The increase in carbon stock and sustainability of crop production are the main challenges in agricultural fields relevant to climate change. Methane is the most important greenhouse gas emitted from paddy fields. This study was conducted to investigate the effects of tillage and cultivation methods on methane emissions in rice production in 2014 and 2015. Different combinations of tillage and cultivation were implemented, including conventional tillage-transplanting (T-T), tillage-wet hill seeding (T-W), minimum tillage-dry seeding (MT-D), and no-tillage-dry seeding (NT-D). The amount of methane emitted was the highest in T-T treatment. In MT-D and NT-D treatments, methane emissions were significantly decreased by 77%, compared with that in T-T treatment. Conversely, the soil total carbon (STC) content was higher in MT-D and NT-D plots than in tillage plots. In both years, methane emissions were highly correlated with the dry weight of rice ($R^2=0.62{\sim}0.96$), although the cumulative emissions during the rice growing period was higher in 2014 than in 2015. T-T treatment showed the highest $R^2$ (0.93) among the four treatments. Rice grain yields did not significantly differ with the tillage and cultivation methods used. These results suggest that NT-D practice in rice production could reduce the methane emissions and increase the STC content without loss in grain yield.

The Effects of Water Level and Temperature on Seed Germination and Early Seedling Development of Rice (Oryza sativa I.)

  • The application of direct seeding cultivation reduces time, labor, and cost. However, this application often has poor seedling establishment and leads to lower yield as compared to transplanting system. The tolerance to anaerobic and low temperature germination is important to improve seedling establishment and the wide-spread application of direct seeding method. This study was carried out to evaluate the responses of three japonica cultivars to different temperatures (15℃, 18℃, 21℃, 24℃, and 27℃) and different flooding levels (1 cm, 4 cm, 7 cm, 10 cm) during germination and seedling development. The mean survival percentage significantly increased (P<0.05) when the flooding level decreased and when temperature increased. There were significant effects of the interaction between temperature and water depth on survival percentage and seedling height. When temperature decreased from 27℃ to 15℃, the germination duration significantly increased from 6.4 days to 16.3 days while the germination speed, survival percentage, and seedling height decreased from 5.3 seeds day-1, 61.9% and 190.6 cm to 2.2 seeds day-1, 33.2%, and 47 cm, respectively. The increase in temperature under submergence condition was associated with the increased expression of Amy3D and ALDH2a but the decreased expression of ADH1 and PDC1. The results of this research would be used for further studies and breeding programs to improve rice seedling establishment and the application of direct seeding cultivation.

Yield and Quality of Forage Mixture as Affected by Maturity of Rye Cultivar and Oat-Rye Seeding Rate (호밀품종의 조만성과 연맥-호밀의 파종량이 혼파사초의 수량과 사료가치에 미치는 영향)

  • Evaluation works of oat in mixture with rye on forage yield have not been reported. The objective of this study was to determine yield and quality of forage mixture as affected by maturity of rye cultivar and oat-rye seeding rate at Suweon from 1999 to 2000. The experiment was arranged in a split plot design with three replications. Main plots consisted of maturity of rye. such as early(cv. Koolgrazer) and late(cv. Kodiak)maturing cultivars. Sub-plots consisted of seeding rate (T1: Oat 2000 and rye 0kg/ha, Ts: Oat 150 and rye 40kg/ha, T3: Oat100 and rye 80kg/ha, T4: Oat50 and rye 120kg/ha. and T5: Oat0 and rye 160kg/ha). Crude protein(CP) content of oat-rye mixture harvested in the fall was not influenced by maturity of rye cultivar, but that of oat-rye mixture was increased from 13.6 to 19.3% as the seeding rate of rye increased(P<0.05), however, maturity of rye cultivar significantly affected CP content of oat-rye mixture in the spring(P<0.01). Acid detergent fiber(ADF) content of oat-rye mixture harvested in the spring was not significantly affected by rye cultivar, but the ADF was decreased from 27.8 to 20.7% as the seeding rate of rye increased(P$<$0.01). When rye was harveste in the spring, ADF content of late maturing cultivar 'Kodiak' was shown as 28.0%. This was lower than that of early maturing cultivar 'Koolgrazer' which was shown as 35.8%(P$<$0.01). Among treatments, neutral detergent fiber(NDF) and in vitro dry matter digestibility(IVDMD) of oat-rye mixture showed a similar trend made on ADF. In this experiment, the highest forage yield (12.356kg/ha) was obtained from early maturing rye cultivar and seeding rates of 100kg/ha of oat and 80kg/ha of rye mixture. A significant interaction between maturity of rye cultivar and seeding rate was found(P$<$0.01). The above results indicate that an early maturing rye cultivar at the seeding rate of 100kd/ha in mixtures with 80kg/ha of oat could be recommended as a succeeding cropping system after corn for silage.

  • This experiment was carried out to select adaptable winter wheat varieties to deep seeding for developing early maturing, cold tolerant, and high yielding varieties. Varieties adaptable to deep seeding had deep crown and cold tolerant or early maturing characteristics. Varieties which had deep crown at 6cm seeding depth were Namkwang, Wonkwang, Suweon #202, Milyang #5, Milyang #7, Kitagamigomugi, Norin # 4, J ukoku # 81, Sage, Blueboy, Expection, Oasis, C.I. 14034, Rossalka, Benhur, Biserka, Martonvasar-1, and Martonvasar-2.

Dry Matter Digestion Kinetics of Two Varieties of Barley Grain Sown with Different Seeding and Nitrogen Fertilization Rates in Four Different Sites Across Canada

  • Our objective was to determine the differences in the rate and extent of dry matter digestion between barley subjected to differing agronomic variables. Two malting barley varieties, Copeland and Metcalfe were seeded at rates of 200 and 400 plants/$m^2$. Each of these varieties received nitrogen fertilizer at rates of 0, 30, 60 and 120 kg/ha, resulting in a total of 20 different barley grain samples. Samples were ground through a 6mm screen and approximately 3 g of each weighed into 50 ${\mu}m$ Dacron bags and sealed. The bags were incubated in three ruminally cannulated Holstein cattle for periods of 0, 3, 6 and 24 h. Using the data obtained from these incubations, rates of digestion were able to be predicted. The soluble fraction ranged from 0.229-0.327, the slowly degradable fraction ranged from 0.461-0.656, and the undegradable fraction ranged from 0.038-0.299. The rates of digestion ranged from 0.127-0.165 $h^{-1}$ and the effective degradability ranged from 0.527-0.757. At the Canora location, the Copeland samples which received 120 kg/ha of nitrogen fertilizer had a significantly lower (p = 0.013) soluble fraction than the rest of the samples at that location. A significant interaction (p = 0.009) was seen between the seeding rate and nitrogen fertilizer application with samples from the Canora location, as well as significant differences (p = 0.029) between nitrogen application rates in samples from the Indian head location. The rate of digestion of samples from the Indian head location differed (p = 0.020) between the two seeding rates, with samples seeded at 200 seed/$m^2$ having a slightly higher rate of degradation. Differences in the effective degradability were seen between the different nitrogen application rates with samples from both the Canora and Indian head locations, as well as an (p = 0.004) interaction between the seeding rate and nitrogen fertilizer application rate. Although there was not a clear correlation between the different variables, both nitrogen application and seeding rate did have a significant effect on the rates and extent of digestion across each of the four locations.

  • The research of 3-dimensional (3-D) scaffold for tissue engineering has been widely investigated as the importance of the 3-D scaffold increased. 3-D scaffold is needed to support for cells to proliferate and maintain their biological functions. Furthermore, its architecture defines the shape of the new bone and cartilage growth. Polycaprolactone (PCL) has been one of the most promising materials for fabricating 3-D scaffold owing to its excellent mechanical property and biocompatibility. However, there are practical problems for using it, in vitro and in vivo; extracellular matrix components and nutrients cannot penetrate into the inner space of scaffold, due to its hydrophobic property, and thus cell seeding and attachment onto the inner surface remain as a challenge. Thus, the surface modification strategy of 3-D PCL scaffold is prerequisite for successful tissue engineering. Herein, we utilized a mussel-inspired approach for surface modification of 3-D PCL scaffold. Modification of 3-D PCL scaffolds was carried out by simple immersion of scaffolds into the dopamine solution and stimulated body fluid, and as a result, hydroxyapatite-immobilized 3-D PCL scaffolds were obtained. After surface modification, the wettability of 3-D PCL scaffold was considerably changed, and infiltration of the pre-osteoblastic cells into the 3-D scaffold followed by the attachment onto the surface was successfully achieved.
