Kinetic Measurement of the Step Size of DNA Unwinding by Bacteriophage T7 DNA Helicase gp4 (T7 박테리오파지 gp4 DNA helicase에 의한 DNA unwinding에서 step size의 반응속도론적 측정)

  • T7 bacteriophage gp4 is the replicative DNA helicase that unwinds double-stranded DNA by utilizing dTTP hydrolysis energy. The quaternary structure of the active form of T7 helicase is a hexameric ring with a central channel. Single-stranded DNA passes through the central channel of the hexameric ring as the helicase translocates $5'\rightarrow3'$ along the single-stranded DNA. The DNA unwinding was measured by rapid kinetic methods and showed a lag before the single-stranded DNA started to accumulate exponentially. This behavior was analyzed by a kinetic stepping model for the unwinding process. The observed lag phase increased as predicted by the model with increasing double-stranded DNA length. Trap DNA added in the reaction had no effect on the amplitudes of double-stranded DNA unwound, indicating that the $\tau7$ helicase is a highly processive helicase. Global fitting of the kinetic data to the stepping model provided a kinetic step size of 10-11 bp/step with a rate of $3.7 s^{-1}$ per step. Both the mechanism of DNA unwinding and dTTP hydrolysis and the coupling between the two are unaffected by temperature from $4∼37^{\circ}C$. Thus, the kinetic stepping for dsDNA unwinding is an inherent property of tile replicative DNA helicase.

Automatic Detection of Stage 1 Sleep (자동 분석을 이용한 1단계 수면탐지)

  • Stage 1 sleep provides important information regarding interpretation of nocturnal polysomnography, particularly sleep onset. It is a short transition period from wakeful consciousness to sleep. Lack of prominent sleep events characterizing stage 1 sleep is a major obstacle in automatic sleep stage scoring. In this study, we attempted to utilize simultaneous EEC and EOG processing and analyses to detect stage 1 sleep automatically. Relative powers of the alpha waves and the theta waves were calculated from spectral estimation. Either the relative power of alpha waves less than 50% or the relative power of theta waves more than 23% was regarded as stage 1 sleep. SEM (slow eye movement) was defined as the duration of both eye movement ranging from 1.5 to 4 seconds and regarded also as stage 1 sleep. If one of these three criteria was met, the epoch was regarded as stage 1 sleep. Results f ere compared to the manual rating results done by two polysomnography experts. Total of 169 epochs was analyzed. Agreement rate for stage 1 sleep between automatic detection and manual scoring was 79.3% and Cohen's Kappa was 0.586 (p<0.01). A significant portion (32%) of automatically detected stage 1 sleep included SEM. Generally, digitally-scored sleep s1aging shows the accuracy up to 70%. Considering potential difficulties in stage 1 sleep scoring, the accuracy of 79.3% in this study seems to be robust enough. Simultaneous analysis of EOG provides differential value to the present study from previous oneswhich mainly depended on EEG analysis. The issue of close relationship between SEM and stage 1 sleep raised by Kinnariet at. remains to be a valid one in this study.

ASTC Block-Size Determination Method based on PSNR Values (PSNR 값 기반의 자동화된 ASTC 블록 크기 결정 방법)

  • ASTC is one of the standard texture formats supported in OpenGL ES 3.2 and Vulkan 1.0 (and later versions), and it has been increasingly used on mobile platforms (Android and iOS). ASTC's most important feature is the block size configuration, thereby providing a trade-off between compression quality and rates. With the higher number of textures, however, it is difficult to manually determine the optimal block sizes of each texture. To solve the problem, we present a new approach based on PSNR values to automatically determine the ASTC block size. A brute-force approach, which compresses a texture on all block sizes and compares the PSNR values of the compressed textures, can increase the compression time by up to 14 times. In contrast, our three-step approach minimizes the compression-time overhead. According to our experiments on a texture set including 64 various textures, our method determined the block sizes from 4×4 to 12×12 and reduced the size of compressed files by 68%.

  • 건설기술과 산업의 발전에 따라 구조물은 대형화되어 가고, 건설공사의 급격한 팽창으로 골재 수용량이 급증함에 따라 천연골재자원은 점차 부족현상을 면치 못할 처지에 있다. 또한, 무리한 천연골재의 채취는 자연환경을 훼손시킬 뿐만 아니라 자연보호 측면에서도 심각한 공해문제로 대두되고 있어 공급량 부족현상은 날로 심화되고 있다. 이에 세계 몇몇 나라에세는 산업부산물을 이용한 골재 생산으로 공해예방과 폐기물 활용방법을 연구하고 있다. 산업부산물중 플라이 애쉬 생산량은 전 세계적으로 매년 약 2억여톤에 달하고 있으나 이중 일부만 활용되고 있는 실정이다. 이와같은 부산물을 활용하기 위한 일환으로 산업부산물인 PFA(Pulverized Fuel Ash)로 만든 인공경량골재의 년생산량이 영국은 600,000$m^3$, 미국은 300,000$m^3$이며, 매년 증가주세에 있다. 고성능 경량골재 콘크리트는 단위중량의 증가없이 내구성과 강도를 향상시켜 실용화 측면에서 경제적인 효과가 있으며, 플라이 애쉬로 만든 경량골재는 시멘트와의 친화력이나 접착면에서 우수한 것으로 알려져 있다. 본 시험에 사용한 골재는 플라이 애쉬로 만든 인공경량 조골재와 강모래이고, 결합제로서 프틀랜드 시멘트를 사용하였다. 부수적인 결합재로서는 플라이 애쉬, 슬래그, 실리카 흄을 사용하였으며, 고성능 경량골재 콘크리트를 개발코자 재령 28일과 180일의 압축강도가 각각 50MPa와 60MPa가 되도록 배합설계를 하였다. 본 연구에서는 플라이 애쉬, 슬래그, 시리카 흄과 같은 산업부산물을 혼입했을때 경량골재 콘크리트의 압축강도, 휨강도, 동탄성계수, 공극체적, 공극률, 단위중량, 공극 크기별 분포등의 변화를 실험적으로 구명하여 재반 구조용 콘크리트에 활용하기 위한 기초자료를 마련코저 한다.있어 특정한 발육단계의 난포 사망기전을 연구하기 어렵다. 또한 난포는 생체 내에서 다양한 호르몬을 동시에 분비하기 때문에 특정한 난소국부호르몬이 사망기전에 미치는 영향을 조사하기 힘든 점이 있다. 최근 들어 난포체외배양이 다양하게 개발되면서, 이러한 어려운 점을 극복할 수 있게 되었다. 본 논문은 각 발육단계의 난포를 절단해 체외배양하면서, apoptosis DNA 절단 현상을 이용하여 각종 난소국부 호르몬들이 난포발육단계별로 사망기전에 미치는 영향을 요약해 보였다. 난포는 발육하면서 점차 복잡한 호르몬 경로를 생존을 위해 필요로 한다.   • Empirical design is still used to avoid a structural damage because impact phenomenon and structural behaviour due to wave impact load can not examined accurately. The damage due to wave impact load is largely affected by impact pressure impulse and impact load area. The objective of this study is, as the second step, to develop an efficient scantling program of bow flare structure, and to predict its impact load area by comparing maximum dented deformations at center of idealized panel structure model of bow flare structure of 300k DWT VLCC using LS/DYNA3D code, which will be used for its verification of dynamic structural analysis, as the next step. Through this study, the impact load area was estimated as $1.5s{\times}1.5s$ stiffener space(s) in the case of panel with stiffeners and as $2.5s{\times}2.5s$, with stringers, under impact pressure curve with peak height 6.5MPa, tail height 1.0MPa, and duration time 5.0msec.

  • The non-steroidal ecdysone agonist RH 5849 showed almost similar LC.o values( 18.1-26.5 ppm) at all stages of larval development of the tobacco cutworm, Spodoptera litura, when treated by a leaf-disk dipping method. The feeding-inhibition rate for the 4th instar larvae was dose-dependent, and simultaneously the weight gain of 3rd instar larvae ceased within 48 hour after feeding of the cabbage leafdisk dipped into RH 5849 4.2 ppm solution. The systemic larvicidal effect of RH 5849 was compared at cabbage and tobacco whole plant test. The $LC_{50}$ values below 20 ppm(mg/kg soil) was lasted for 15 days in cabbage, 30 days in tobacco respectively.

Clinical Outcomes of Ultrasound-Guided Transmeniscal Injection in Medial Compartment Knee Osteoarthritis (무릎 내측 구획 골관절염에서 초음파 유도하 경 반월상 연골 주사의 임상적 효과)

  • Purpose: The purpose of this study was to introduce the ultrasound-guided transmeniscal injection in medial compartment knee osteoarthritis and analyze the clinical outcomes. Materials and Methods: The electronic medical records of 36 patients with medial compartment knee osteoarthritis who were treated with an ultrasound-guided transmeniscal injection from March 2019 to July 2019 were accessed for this retrospective review. Using an ultrasound guided spinal needle, the patients received an intra-articular steroid injection at the medial compartment of the knee. A physical examination was conducted at the initial visit (pre-injection), and at one week, four weeks, and eight weeks after the injection. The numeric pain rating scale (NRS), Lequesne index, and Western Ontario and McMaster Universities Osteoarthritis (WOMAC) score were measured at each visit and analyzed over time. The percentage change of the patients who revealed substantial improvement was analyzed. The NRS, Lequesne index, and percentage of patients, who revealed substantial improvement over time classified by osteoarthritis grade, were analyzed. Results: The NRS and Lequesne index decreased at one week, four weeks, and eight weeks after the injection compared to the initial baseline, and the pain-relief effect continued without change until eight weeks. The percentage of patients who showed substantial improvement at one, four, and eight weeks was 50.0%, 47.2%, and 52.8%, respectively. The WOMAC scores decreased at one, four, and eight weeks compared to the initial baseline, and the decrease was continued without any difference until eight weeks. The percentage of patients with osteoarthritis stage 1 or 2 who revealed more than substantial improvement was significantly higher at one, four, and eight weeks than those with osteoarthritis stages 3 or 4 (p<0.05). Conclusion: In patients with medial compartment knee osteoarthritis, the pain reduction and functional improvement persisted for at least eight weeks after the ultrasound-guided transmeniscal injection at the medial compartment. In particular, patients with medial compartment osteoarthritis stage 1 or 2 showed more effective pain reduction.

Pharmacokinetics and tissue levels of a sustained-release recombinant porcine somatotropin in pigs (돼지에서 서방형 성장호르몬의 약물동태 및 조직잔류성)

  • 서방형 돼지성장호르몬(sustained-release formulation of porcine somatotropin, PST-SR)을 1주 간격으로 6차례 피하 및 근육주사하고 혈액과 조직중의 돼지 성장호르몬(PST)과 insulin-like growth factor 1(IGF-1)의 농도를 측정하여 다음과 같은 결과를 얻었다. 대조군의 혈중 PST와 IGF-1의 농도는 각각 2.41과 95.2 ng/ml 이었다. 1. PST-SR을 투여한 후 PST의 혈중농도는 8시간만에 최대에 도달하여(30 ng/ml) 곧 감소하였다. 혈중농도 반감기(decay half life)는 91~227시간이었다. IGF-1의 혈중농도는 투여후 12시간에 최대에 도달하였으며(165 ng/ml), 이후 서서히 감소되었고 반감기는 77~99시간이었다. 2. 혈중 PST농도-시간의 자료는 제재에서 PST가 유리되는 과정에는 두단계 즉, 투여후 24시간까지의 유리속도가 빠른 단계와 그 이후의 유리속도가 느린 단계가 있음을 보여주었다. 3. 여섯번의 반복투여기간에는 PST의 혈중농도는 투여직후 증가하여 24시간 이후 다음 투여전까지 지속적으로 감소되는 패턴이 반복되었고, 최종투여후 1주일경에는 정상수준으로 회복되었다. 반면에 투여가 반복됨에 따라 매 투여직후의 PST의 혈중 최고치는 다소 증가되는 경향을 보였다(20~40 ng/ml). IGF-1의 혈중농도는 투여가 반복됨에 따라 누적적인 증가현상이 뚜렷하였으며, 이후 2주일후 까지도 정상농도보다 높게 유지되었다(200ng/ml). 임상용량 투여군에서 PST 및 IGF-1의 혈중농도는 투여경로에 따른 차이는 나타나지 않았다. 4. 최종(6번째) 투여후 6, 8, 10, 14일에 조사한 간장, 신장, 소장, 근육, 지방 및 주사부위의 조직중의 PST 농도는 6일째에 이미 대조군 수준으로 회복되었다. IGF-1의 경우 최종투여후 6일에는 간장, 신장, 소장, 지방조직에서 정상보다 높은 농도로 잔류하나 이후 14일까지 모두 대조군 수준으로 감소되었다. 5. 이상의 결과는 본 실험에서 사용된 서방형 PST제제는 최소 1주간 유효성이 유지되며, 동시에 PST는 투여 6일째에, IGF-1은 투여후 14일에 정상수준으로 회복됨을 보여주고 있다.

Development of disaster severity classification model using machine learning technique (머신러닝 기법을 이용한 재해강도 분류모형 개발)

  • In recent years, natural disasters such as heavy rainfall and typhoons have occurred more frequently, and their severity has increased due to climate change. The Korea Meteorological Administration (KMA) currently uses the same criteria for all regions in Korea for watch and warning based on the maximum cumulative rainfall with durations of 3-hour and 12-hour to reduce damage. However, KMA's criteria do not consider the regional characteristics of damages caused by heavy rainfall and typhoon events. In this regard, it is necessary to develop new criteria considering regional characteristics of damage and cumulative rainfalls in durations, establishing four stages: blue, yellow, orange, and red. A classification model, called DSCM (Disaster Severity Classification Model), for the four-stage disaster severity was developed using four machine learning models (Decision Tree, Support Vector Machine, Random Forest, and XGBoost). This study applied DSCM to local governments of Seoul, Incheon, and Gyeonggi Province province. To develop DSCM, we used data on rainfall, cumulative rainfall, maximum rainfalls for durations of 3-hour and 12-hour, and antecedent rainfall as independent variables, and a 4-class damage scale for heavy rain damage and typhoon damage for each local government as dependent variables. As a result, the Decision Tree model had the highest accuracy with an F1-Score of 0.56. We believe that this developed DSCM can help identify disaster risk at each stage and contribute to reducing damage through efficient disaster management for local governments based on specific events.
