Construction and analysis of painting probe for homogeneously staining regions in human neuroblastoma cell line IMR-32

  • Neuroblastoma, a pediatric malignant neoplasm of neural crest origin, has a wide range of clinical virulence. The mechanisms contributing to the development of neuroblastomas are largely unclear, but non-random chromosomal changes identified over the past years suggest the involvement of genetic alterations. Amplification of the human N-myc proto-oncogene is frequently seen either in extrachromosomal double minutes or in homogeneously staining regions (HSRs) of aggressively growing neuroblastomas. N-myc maps to chromosome 2 band 24, but HSR have never been observed at this band, suggesting transposition of N-myc during amplification. We have constructed and analyzed the region-specific painting probe for HSR in neuroblastoma IMR-32 to determine the derivative chromosomes. Microdissection was performed on HSR using an inverted microscope with the help of microglass needles and an micromanipulator. We pretreated the microdissected fragments with Topoisomerase I which catalyzes the relaxation of supercolled DNA, and performed two initial rounds of DNA synthesis with T7 DNA polymerase followed by conventional PCR to enable the reliable preparation of Fluorescent in situ hybridization probe from a single microdissected chromosome. With this method, it was possible to construct the region-specific painting probe for HSR. The probe hybridized specifically to the HSRs of IMR-32, and to 2p24, 2p13 of normal chromosome. Our results suggest there was coamplification of N-myc together with DNA of the chromosome 2p24 and 2p13. Moreover, the fluorescent signals for the amplified chromosomal regions in IMR-32 cells were also easily recognized at a Thus this painting probe can be applied to detect the similar amplification of N-myc in neuroblastoma tissue, and the probe pool for HSR may be used to identify the cancer-relevant genes.

Expression of Matrix Metalloproteinase-2 and -13 and Tissue Inhibitor of Metalloproteinase-4 in Varicose Veins

  • Background: The relationship between the degree of expression of matrix metalloproteinases or tissue inhibitor of metalloproteinases and venous reflux remains to be investigated. Materials and Methods: Primary varicose vein tissues were obtained from 23 patients, 18 females and 5 males, aged from 19 to 73. Cephalic or basilic veins were obtained for the control group from 10 patients who underwent vascular access for maintenance hemodialysis. Two operative techniques (high ligation with stripping or endovenous laser coagulation) were used. The expression of matrix metalloproteinase-2 and 13 and tissue inhibitor of metalloproteinase-4 in the varicose vein group and control group was assessed semi-quantitatively by immunohistochemical slides stained with primary antibodies. Results: Twenty (87%) of the varicose vein group patients had greater or lesser saphenous vein diseases with reflux. The focal weak (+) stain for matrix metalloproteinases-2, and 13, and tissue inhibitor of matrix metalloproteinase-4 was dominant in the varicose vein group; the focal or diffuse strong stain (++ or +++) was prevalent in the control group. The differences were statistically significant (p<0.01). The degree of reflux and the duration of symptoms were not significantly related to the expression of MMP-13 (p=0.317 and p=0.654, respectively). Conclusion: Further study should be performed to investigate the relationship between the clinical characteristics related to venous hypertension or reflux and expression of MMPs and TIMP in varicose veins.

The Effects of the Combined Biofeedback and Brief Emotion Regulation Nursing Intervention Based on the Gross Model for Sexually Abused Adolescents (성폭력 피해 청소년을 위한 바이오피드백 병행 Gross 모델기반 단기 정서조절 간호중재 효과)

  • Purpose: This study aimed to evaluate the effects of a combined biofeedback and brief emotion regulation (C-BABER) program for sexually abused adolescents. Methods: This study employed a non-equivalent control group pretest-posttest design. The participants included 26 sexually abused adolescents from eight Sunflower Centers of South Korea-with 13 in the experimental group and 13 in the control group. The experimental group received four sessions of the individual C-BABER program, each lasting 60 minutes. Results: Compared with the control group, sexually abused adolescents in the experimental group exhibited significant score differences in traumatic symptoms, including depression (Z = - 2.24, p = .025), dissociation (Z = - 2.21, p = .027), anxiety (Z = - 2.02, p = .044), and posttraumatic stress (Z = - 2.01 p = .045); and impulsivity, including positive urgency (Z = - 3.35, p = .001) and negative urgency (Z = - 2.28, p = .023). Additionally, the experimental group exhibited significant score differences in meta-mood, including emotional attention (Z = - 2.45, p = .014), emotional clarity (Z = - 2.30, p = .021), and emotional repair (Z = - 2.28, p = .022); and emotional regulation modes, including emotional suppression (Z = - 2.65, p = .008) and cognitive reappraisal (Z = - 1.98, p = .047). Regarding bio-attention, significant changes were identified in the experimental group for the bio-attention rate and attention maintenance time in the posttest compared to the pretest (p = .001). Conclusion: The C-BABER program for sexually abused adolescents is effective in decreasing traumatic symptoms and impulsivity, and in improving meta-mood, emotional regulation mode, and bio-attention. Therefore, we recommend providing sexually abused adolescents the C-BABER program to help them regulate their emotions and effectively adapt to their lives.

Association of Human Epidermal Growth Factor Receptor-2 Expression and Clinicopathological Findings in Patients with Colorectal Cancer

  • Background: To determine the frequency of HER-2 overexpression in colorectal cancer (CRC) patients, and to explore the relationship between clinicopathological prognostic factors and their effects on survival, based on immunohistochemistry (IHC) and fluorescent in situ hybridization (FISH) analysis. Materials and Methods: The study included 80 patients with a histologically proven diagnosis of CRC that received adjuvant FOLFOX-4 chemotherapy at our department between March 2006 and September 2010. Patient data were analyzed retrospectively. Results: The median follow-up period and age of the patients were 24 months and 59 years, respectively. In immunohistochemical staining, 3+ staining was found in 2 patients (2.5%) while 2+ was in 13 (16%). FISH for HER-2 was performed for all of these 15 patients; samples which were 3+ showed positivity but the ones with 2+ were negative. There was no significant correlation between HER-2 expression and age, gender, tumor localization, histological subtype, grade, lymphovascular and perineural invasion, or pTN stage (P>0.05), even when the patients with HER-2 overexpression were analyzed separately. There was also no significant relationship between progression-free survival (PFS) and overall survival (OS), and HER-2 expression, gender, tumor localization, obstruction-perforation, bleeding, histological type, grade, lymphovascular and perineural invasion, or pT staging (P>0.05); however, there was a significant relationship between lymph node involvement, and PFS and OS (P<0.05). Conclusions: Evaluation of HER-2 overexpression in a more comprehensive, multi-center, prospective trial with standardized methods will be an appropriate approach.

  • In order to evaluate the correlation of age with development stage on permanent lower posterior teeth. the author exmined the roentgeregrams in standard films taken by intraoral technic and analysed the development phases of 1358 teeth of 500 males ranging from 9 to 15 years. The development was divided into 7 phases : Crown complete (Cr. C.). Root length 1/4(R. 1/4) Root length /2 (R. 1/2) Apical closure complete (A.C) The obtained results were as follows : 1. The formation of roots in full length on posterior teeth was complete as follow : a. Roots of 1st premolar : 12.72 years b. Roots of 2nd premolar : 12.94 years c. Meral Roots of 2nd molar : 13.38 years d. Distal Roots of 2nd molar : 13.46 years 2. The formation of apical forman of premolar was closured as follows : a. Apical foramen of root of 1st premolar : 13.64 years b. Apical foramen of root of 2nd premolar : 13.93 years 3. As a general rule. the mesial roots of second molar were developed earlier than distal roots of second molar. 4. In the correlation of age with the development stage, the regression equations. the correlation coefficents. and the sample numbers were “Y = 0.8370x + 10.2160, r = 0.71(p<0.01), n = 318”on lower first premolar, “Y = 0.6984x + 10.2148, r = 0.71(p<0.01), n = 385”on lower second premolar, “Y = 0.8810x + 10.2040, r = 0.65(p<0.01), n = 344”on mesial Root of lower second molar, and “Y = 0.7310x + 10.7940, r = 0.66(p<0.01), n = 311”on Distal Root of lower second molar respectively.

Induction Chemotherapy and Radiotherapy in Locally Advanced Non-Small Cell Lung Cancer (NSCLC) (국소 진행된 비소세포성 폐암에서 유도 화학요법 및 방사선치료)

  • Purpose : We peformed this study to evaluate the prognostic factors and the effect of induction chemotherapy in locally advanced non-small cell lung cancer (NSCLC). Materials and Methods : A retrospective analysis was done for 130 patients with locally advanced NSCLC treated with curative radiotherapy alone or induction chemo-radiotherapy from January 1986 to October 1996. Eighty-five patients were treated with radiotherapy alone, forty-five with induction chemotherapy and radiotherapy. Age, sex, performance status, histopathologic type, and stage were evenly distributed in both groups. The patients were treated with 6 MV or 10 MV X-ray. Conventional fractionation with daily fraction size 1$.8\~2.0$ Gy was done. Of the patients, 129 patients received total dose above 59.6 Gy ($56\~66$ Gy, median 60 Gy). Induction chemotherapy regimen were CAP (Cyclo-phosphamide, Adriamycin, Cisplatin) in 6 patients, MVP (Mitomycin, Vinblastine, Cisplatin) in 9 patients, MIC (Mitomycin, Ifosfamide Cisplatin) in 13 patients, and EP (Etoposide, Cisplatin) in 17 patients. Chemotherapy was done in $2\~5$ cycles (median 2). Results : Overall 1-, 2-, and 3-year survival rate (YSR) for all patients were $41.5\%,{\;}13.7\%,{\;}and{\;}7\%$, respectively (median survival time 11 months). According to treatment modality, median survival time, overall 1-, 2-, and 3-YSR were 9 months, $32.9\%,{\;}10.\5%,{\;}6\%$ for radiotherapy alone group, and 14 months, $57.8\%,{\;}20\%,{\;}7.6\%$ for induction chemotherapy group, respectively (f=0.0005). Complete response (CR) to overall treatments was $25\%$ (21/84) in radiotherapy alone and $40.5\%$ (17/42) in induction chemotherapy group (p=0.09). The Prognostic factors affecting overall survival were hemoglobin level (p=0.04), NSE (neuron-specific enolase) level (p=0.004), and respense to overall treatment(p=0.004). According to treatment modalities, NSE (neuron-specific enolase) (p=0.006) and response to overall treatment (p=0.003) were associated with overall survival in radiotherapy alone group, and response to overall treatment (p=0.007) in induction chemotherapy group. The failure Pattern analysis revealed no significant difference between treatment modalities. But, in patients with CR to overall treatment, distant metastasis were found in 11/19 patients with radiotherapy alone, and 3/13 patients with induction chemotherapy and radiotherapy (p=0.07). Locoregional failure patterns were not different between two groups (10/19 vs 6/13). Conclusion : Induction chemotherapy and radiotherapy achieved increased 2YSR compared to radiotherapy alone, At least in CR patients, there was decreased tendency in distant metastasis with induction chemotherapy. But, locoregional failures and long-term survival were not improved. Thus, there is need of more effort to increasing local control and further decreasing distant metastasis.

Factors Associated with Cognitive Impairment of the Elderly in Nursing Homes (노인요양시설 입소노인의 인지장애 관련요인)

  • The purpose of this study was to identify cognitive impairment and it's related factors of the elderly in long term care facilities. The participants of this study were 229 residents of five nursing homes. Data were analyzed using two sample t-test, ${\chi}^2$-test, and logistic regression. The results revealed eight factors related to cognitive impairment: gender(OR=2.41, p=.035), age(75-84: OR=3.26, p=.002, Over 85: OR=4.46, p<.001), living with family before admission(OR=2.17, p=.015), hearing(OR=8.88, p=.004), family history of dementia(OR=4.39, p=.009), ADL(OR=0.82, p<.001), hypertension(OR=4.07, p<.001), and diabetes(OR=3.42, p=.001). Based on the findings of this study, continuous nursing intervention focused on adjustable factors would be helpful in order to improve nursing home elderly people's quality of life.

Body Weight Control Behavior and Obesity Stress of College Women (여대생의 체중조절행위와 비만스트레스)

  • This study was conducted to investigate weight control behavior and obesity stress of college women. The data were collected from September 7 to September 13 of 2013 and the subjects were 213 college women in C city of Gyoung-nam Province. The data were analyzed by SPSS 20.0 program using descriptive statistics, t-test, ANOVA, Scheffe's test. The score of obesity stress was moderate($18.26{\pm}5.66$). The level of obesity stress was affected by various general characteristics and weight control behavior characteristics including economic status(F=3.99, p=0.020), figure recognition(F=14.41, p<.001), satisfaction of body figure(F=15.88, p<.001), interest of weight control(F=4.82, p=.001), control of food amount(F=2.41, p=.050), body figure compulsion(F=24.06, p<.001), experience of dieting (F=6.04, p<.001), sleeping hours(t=2.10, p=.036), desired losing weight (F=10.47, p<.001). Therefore, it is necessary to be considered these variables during development of body weight control programs for college women.

  • The crystal structure of fenothiocarb(S-4-phenoxybutyl dimethylthiocarbamate), C13H19NO2S is monoclinic, space group P21/c, a=9.045(1)Å, b=14.577(2)Å, c=10.727(2)Å, β=103.56(1)°, Z=4, V=1375.20(6)Å3, Dc=1.23g/cm3, λ(Mo-Kα)=0.71069Å, μ=2.3cm-1, F(000)=544, temperature : 293±3K, R=0.049 for 1543 unique observed reflections. The structure was determined by direct method and refined by full-matrix least squares method. The molecules are contacted to the c axis with two fold screw and van der Waales force.

  • The P2P technology is becoming a technology for receiving a service, such as the latest YouTube and Facebook. P2P technology can be easily downloaded to a large amount of multimedia content over a distributed environment. However, pictures, music, a large amount of multimedia content such as videos each user is unable to manage the content management operating smoothly because, Song communication processing overhead caused when the reception is likely to remain the problem. In this paper, we propose a convergence content management technique that provides a low communication overhead of the frequency of use of the convergence content when trying to provide a large amount of convergence content to users in a distributed environment. Our method can be used to select the appropriate frequency is high among the shared content on the P2P multimedia environments that are distributed across a frequency of use of the convergence content and provides the service. Experimental evaluation proposed method, the convergence content browsing speed than conventional techniques improved communication about 13.3% and reduced overhead to about 9.5%.
