표식유전자를 이용한 담배와 감자의 원형질체 융합 (Protoplast Fusion of Nicotiana glauca and Solanum tuberosum Using Selectable Marker Genes)

  • 표식 유전자를 이용하여 체세포 잡종체를 선발하기 위한 연구의 일환으로 감자조직에는 T-DNA를 도입하여 식물호르몬 무첨가 배지에서도 생장가능한 형질전환체을 획득하고, 담배조직에는 NPT H gene을 도입하여 kanamycin에 대해서 저항성을 나타내는 형질전환체를 획득하여 각각의 특성구명과 원형질체를 유리하여 도입된 유전자 marker를 이용해서 융합을 시도한 바 그 결과는 다음과 같다. 1. 감자 괴경에서 Agrobacterium tumefaciens Ach5와 A. rhizogenes ATCC15834를 접종하여 crown gall tumor 및 hairy root를 유기하였으며 이러한 tumor조직은 식물 호르몬 무첨가 배지에서 생장이 가능하였다. 2. 감자에서 유기된 hairy root로부터 callus 형성은 2.4-D 2mg/1 첨가된 MS 배지에서 가장 양호하였으며 casein hydrolysate 1g/1가 첨가하면 유연 한 callus의 증식이 더욱 왕성하였다. 3. Activated charcoal이 0.5~2.0g/1 첨가된 배지에서는 crown gall tumor callus의 절단면이 갈변되는 것을 방지 할 수 있어 생존률을 높일 수 있었으나 hairy root에서는 갈변되어 고사되었다. 4. 2, 4-D 2mg/1와 casein hydrolysate 1g/1를 첨가한 배지에 hairy root callus를 현탁배의한 결과 양호한 많은 callus 덩어리들을 단시간에 얻을 수 있었다. 5. $Tri^-$parental mating으로 NPTII gene이 coding되어있는 binary vector인 pGA643을 wild type 및 disarmid된 Agrobacterium 내에 도입하여 Agrobacterium tumefaciens Ach5/pGA643, A.tumefaciens $A_4T$/pGA643, A. tumefaciens LBA4404/pGA643를 획득하였다. 이 세 개의 conjugant를 사용하여 0.7% agarose gel 상에서 pGA643을 확인하였다. 6. pGA643이 도입왼 Agrobacterium tumefaciens LBA4404와 담배 조직과 동시배양하여 kanamycin $100\mug$/ml 첨가된 배지에 생존하는 callus를 선발하였으며 동일배지에서 callus의 증식이 가능하였다. 7.형질전환된 담배 callus로부터 식물체 형성은 BA 2mg/1를 첨가한 배지에서 가능하였다. 8. 재분화된 담배의 엽조직은 kanamycin.이 $1000\mug$/ml 첨가된 MS 배지에서도 왕성히 callus가 유기되어, 이는 재분화체에서도 NPTII gene이 그대로 유지되고 있음을 확인할 수 있었다. 9. 담배의 정상 shoot와 형질전환된 shoot를 kanamycin이 $100\mug$/ml이 함유된 MS배지에 기내삽목한 결과, 정상 shoot는 발근이 되지 않고 황화 되었으나 형질전환된 shoot는 발근이되었으며 정상적으로 생장을 하였다. 10. 감자의 T-DNA가 도입된 현탁배양 callus는 cellulase 2%, macerozyme 2%, dricelase 1%에서 양호하게 유리되었다. 11. Osmoticum으로서 mannitol 농도 0.8M에서 담배와 감자의 두 조직 모두 원형질체유리가 가장 효과적이었다. 생존력은 T-DNA가 도입된 감자의 hairy root를 callus로 탈분화 시킨 후 현탁배양한 callus가 mannitol 0.5M에서 97%를 나타냈고, 그리고 NPTII gene이 도입된 담배의 엽조직은 mannitol 0.7M에서 94%로 최고를 타나냈다. 12. 원형질체 융합은 PEG solution 처리 15분 후부터 관찰되기 시작하여 20분에 완전히 융합되었고, 융합된 원형질체는 선발 marker인 호르몬 무첨가 및 kanamycin 첨가배지에서 배양 5일 후에 세포벽이 재생되었으며 4주일 후부터 colony들이 관찰되었다.

  • The boundary conditions for the free-surface including the important meaning for both scientific and engineering purposes are described together with the numerical techniques to implement the conditions. Two kinds of numerical method based on MAC method are introduced. One is applied to the problem of 2D solitary wave propagation and the other is applied to the problem of 3D bow wave breaking.

Whole Mitochondrial Genome Sequence of an Indian Plasmodium falciparum Field Isolate

  • Mitochondrial genome sequence of malaria parasites has served as a potential marker for inferring evolutionary history of the Plasmodium genus. In Plasmodium falciparum, the mitochondrial genome sequences from around the globe have provided important evolutionary understanding, but no Indian sequence has yet been utilized. We have sequenced the whole mitochondrial genome of a single P. falciparum field isolate from India using novel primers and compared with the 3D7 reference sequence and 1 previously reported Indian sequence. While the 2 Indian sequences were highly divergent from each other, the presently sequenced isolate was highly similar to the reference 3D7 strain.

On the Voiced-Voiceless Distinction in Stops of English

  • Phonologically, the difference between the English stops /b, d, g/ and /p, t, k/ is carried by the presence or the absence of the vocal fold vibration throughout their oral closure phase. If phonology has its foundation in phonetics, there must be phonetic evidence for the voiced-voiceless distinction. This study is aimed to determine whether or not the voiced-voiceless distinction is acceptable or proper in English. The determination was based mainly on findings in the existing literature and in informal experiments. In conclusion, there is no phonetic evidence for the voiced-voiceless distinction both in production and perception. The [voice] appears to be one of potential phonetic correlates of the phonologically voiced stop. It is improper to use the [voice] as independent phonological marker, regardless of position (word-initial, intervocalic, word-final). A feature other than the voiced-voiceless feature must distinguish /b, d, g/ from /p, t, k/.

Phenanthrene Derivatives, 3,5-Dimethoxyphenanthrene-2,7-diol and Batatasin-I, as Non-Polar Standard Marker Compounds for Dioscorea Rhizoma

  • Phenathrene derivatives, such as batatasins, are well-known constituents in Dioscorea Rhizoma. Although phenanthrenes have been reported as representative compounds in this plant, standard markers for quality control have been focused on the polar constituents (saponins and purine derivatives). Herein, simple, rapid and reliable HPLC method was developed to determine 3,5-dimethoxyphenanthrene-2,7-diol (DMP) and batatasin-I (BA-I) as non-polar standard maker compounds of Dioscorea Rhizoma. DMP and BA-I were analyzed under optimized HPLC conditions [column: Columbus $5{\mu}$ C18 100A ($30{\times}4.6mm$ i.d., $5{\mu}m$; mobile phase: $H_2O$ with 0.025% $CH_3COOH$ (v/v) for solvent A and $CH_3CN$ with 0.025% $CH_3COOH$ (v/v) for solvent B, gradient elution; flow rate: 2 mL/min; detection: 260 nm), and each experiment was finished within 13 min. Good linearity was achieved in the range from 0.5 to $10.0{\mu}g/mL$ for each compound, and intra- and inter-day precision were in the acceptable levels. The recovery test were performed with three different Dioscorea Rhizoma samples (D. opposita, D. batatas and D. japonica), and showed its accuracy values in the range of 97.2 - 102.8% for three different concentrations of DMP and BA-I. The content levels of DMP and BA-I were ranged under 0.0020%. These results demonstrated that amounts of DMP and BA-I are easily determined with conventional HPLC-UV-DAD method although the content levels were lower than those of saponins and allantoin in Dioscorea Rhizoma. This HPLC method could be used for quality control of various Dioscorea preparations.

Genetical and Pathological Studies on the Mutant Mice as an Animal Model for Deafness Disease

  • A new neurological mutant has been found in the ICR outbred strain mouse. Affected mice display profound deafness and a head-tossing and bidirectional circling behavior, showing an autosomal recessive mode of inheritance. It was, therefore, named cir/Kr with the gene symbol cir. The auditory tests identified clearly the hearing loss of the cir mice when compared to wild type mice. Pathological studies confirmed the developmental defects in the middle ear, cochlea, cochlear nerve, and semicircular canal areas, which were correlated to the abnormal behavior observed in the cir mice. Thus, cir mice may be useful as a model for studying inner ear abnormalities and deafness. We have constructed a genetic linkage map by positioning 14 microsatellite markers across the (cir) region and intraspecific backcross between cir and C57BL/6J mice. The cir mouse harbors an autosomal recessive mutation on mouse chromosome 9. The cir gene was mapped to a region between D9Mit116 and D9Mit38 Estimated distances between cir and D9Mit116, and between cir and D9Mit38 are 0.7 and 0.2 cM, respectively. The gene in order was defines : centromere-D9Mit182-D9Mit51/D9Mit79/D9Mit310-D9Mit212/D9Mit184-D9Mit116-cir-D9Mit38-D9Mit20-D9Mit243-D9Mit16-D9Mit55/D9Mit125-D9Mit281. The mouse map location of the cir locus appears to be in a region homologous to human 3q21. Our present date suggest that the nearest flanking marker D9Mit38 provides a useful anchor for the isolation of the cir gene in a yeast artificial chromosome contig.

통상성 간질성 폐렴 환자 예후인자로서의 섬유모세포병소(fibroblastic foci)의 유용성 (Prognostic Value of Fibroblastic Foci in Patients with Usual Interstitial Pneumonia)

  • 연구배경 :통상성 간질성 폐렴(UIP)은 일반적으로 생존기간 중앙값이 약 3년이하로 보고되는 치명적인 질환이나 환자들간에 예후에 차이가 많다. UIP의 조직학적 소견은 늑막 직하부의 국소적(patchy) 섬유화병변으로 여러 시기의 섬유화 병변이 같이 공존하는 것이 특징이다. 섬유모세포 병소(fibroblastic foci: FBF)는 활발히 증식하는 근-섬유모세포들이 모여있는 급성 폐병소 부위로서 UIP의 활동성 및 예후를 반영할 가능성이 많으나 UIP의 예후인자로서 FBF의 유용성에 관해서는 서로 상반된 결과가 보고되어 논란이 많다. 이에 저자들은 UIP에서 FBF의 예후인자로서의 유용성를 평가하고자 하였다. 대상 및 방법 :1990년 7월부터 2000년 4월까지 서울중앙병원에서 수술적 폐생검검사로 진단되고 일년이상 경과가 관찰된 UIP환자 46명(연령 $59{\pm}12$, 남:여=33:13)으로 모든 조직표본을 재검토하여 ATS/ERS 분류에 의한 IPF-UIP로 확인된 환자들만을 대상으로 하였다. FBF 정도는 폐생검조직에서 0에서 3 (absent:0, mild:1, moderate:2, severe:3)까지 반정량적인 방법으로 측정하였다. 결 과 : 1) 대상 환자들의 46명의 진단 후 생존기간 중앙값은 26개월이었고, 추적관찰 기간중에 사망자는 13명이었다. 2) 사망군과 생존군을 비교 하였을 때 진단시의 FVC와 DLco, 흡연 유무, FBF 정도가 유의한 차이를 보였다. 3) 진단시 FBF 정도가 비중증군 (FBF :2이하)의 103개월로 중증군(FBF :3)의 평균 생존기간이 48 개월에 비해 유의하게 길었다. 또한 FVC가 73.5% 이상인 군에서(평균 생존기간 86개월) 73.5% 미만인 군(65개월)보다, $D_Lco$가 60.5% 이상인 군(평균 생존기간 83개월)에서 60.5% 미만 군(69개월)보다 생존기간이 길었다. 4) 환자의 나이, 성별, 흡연 그리고 Kaplan Meyer 분석을 이용한 단변수 분석 상 생존과 연관이 있는 인자로 나타난 $D_Lco$, FVC, FBF정도에 대하여 Cox regression분석을 시행한 결과, FBF 정도 만이 UIP사망의 독립적인 예측인자로 확인되었다. 결 론 : 수술적 폐생검상 FBF 정도와 환자의 생존기간의 연관성이 관찰되어, FBF가 UIP의 예후의 예측인자로 추측되었다.

외부 하중이 어깨관절의 능동적 재위치 검사 결과에 미치는 영향: 3차원 동작 분석 시스템을 이용한 예비연구 (Effect of External Load on Shoulder Joint Active Relocation Using 3D Motion Capture System: A Pilot Study)

  • Background: There are insufficient objective or quantitative evidence for the better intervention to improve proprioception particularly for the application of external load. There are conflicting opinions whether the external load is effective for proprioception improvement or not. Objects: The purpose of this study was to investigate effects of external load on proprioception of shoulder joint quantitatively using 3D motion capture system. Methods: Nine healthy adults joined for this study. They were asked to perform scapular plane abduction motion with attaching reflective markers on the trunk and upper limb. The 3D positions of finger marker, while they performed the same task with and without external load, were recorded and analyzed. Results: All participants showed decreased variable errors in the vertical direction when the external load was applied (p<.02). Even though other directions (y, z) and absolute errors increased, they did not have statistical significances. Conclusion: Based on this study results, the external load application would be effective for shoulder joint position sense improvement.

측풍영향을 고려한 고속전철 주위의 비압축성 점성 유동 해석 (Incompressible Viscous Flow Analysis around a High-Speed Train Including Cross-Wind Effects)

  • The flow field around a high-speed train including cross-wind effects has been simulated. This study solves 3-D unsteady incompressible Navier-Stokes equations in the inertial frame using the iterative time marching scheme. The governing equations are differenced with 1st-order accurate backward difference scheme for the time derivatives, 3th-order accurate QUICK scheme for the convective terms and 2nd-order accurate central difference scheme for the viscous terms. The Marker-and-Cell concept was applied to efficiently solve continuity equation, which is differenced with 2nd-order accurate central difference scheme. The 4th-order artificial damping is added to the continuity equation for numerical stability. A C-H type of elliptic grid system is generated around a high-speed train including ground. The Baldwin-Lomax turbulent model was implemented to simulate the turbulent flows. To validate the present procedure, the flow around a high speed train at constant yaw angle of $45^{\circ}\;and\;90^{\circ}$ has been simulated. The simulation shows 3-D vortex generation in the lee corner. The flow separation is also observed around the rear of the train. It has concluded that the results of present study properly agree with physical flow phenomena.

코팅 두께에 따른 친수성 무기 필름의 특성 분석 (Properties Characterization of the Hydrophilic Inorganic Film as Function of Coating Thickness)

  • In this paper, we present a novel hydrophilic coating material (Wellture Finetech, Korea) which can be utilized as a coating layer for anti-contamination for electrical and electronic system. The coating material was deposited on 4 inch silicon wafer with several different film thickness. The film thickness was controlled by spin coating speed. After curing of the film, we have scratched by permanent marker to check self-cleaning property of the film. Also we have executed several mechanical tests of the films. As the spin coating speed is increased, the film thickness was thinned from 230 nm to 125 nm. Contact angle of the film was lowered from $30^{\circ}$ to $12^{\circ}$ as the spin coating speed is increased from 700 to 2,500 rpm. On permanent marker scratched film surface coated at 1,000 rpm, we have poured regular city water to investigate self cleaning property of the film. The scratches were gradually separated from the film surface due to super-hydrophilicity of the film. Hardness of spin coated film was 9H measured by ASTM D3363 method. and adhesion of all film was 5B tested by ASTM D3359 method. Also, to get exact hardness value of the film, we have utilized a nano-indenter. As spin speed is increased, the hardness of film was increased from 3 GPa to 5 GPa.
