Detection of Methicillin-Resistant Staphylococcus aureus by In Vitro Enzymatic Amplification of MecA and FemA Gene (메티실린 내성 황색 포도상 구균에서 mecA, femA 유전자의 임상적 의의)

  • Purpose : In the treatment of MRSA infection, rapid detection of MRSA is extremely important. The mecA gene codes the new drug resistant polypeptides called PBP2' which mediates the clinically relevant resistance to all beta-lactam antibiotics. The identical mecA gene has been found in coagulase-negative staphylococcus with the methicillin-resistant phenotype. On the other hand, the femA gene was absent from coagulase negative staphylococcus strains with the methicillin resistant phenotype. This study is aimed at early detection and definite diagnosis of MRSA. Methods : A total of 24 MRSA strains were studied. All strains were tested for antimicrobial susceptibility and purified DNA. We amplified both mecA and femA genes by PCR in 24 strains. Results : In MRSA all the 16 strains (100%) carried femA gene and 11 strains (68.7%) carried mecA gene. In contrast, in methicillin sensitive staphylococcus all the 8 strains (100%) carried femA and only 3 strains (37.5%) were detected mecA. Conclusions : As results, there are difference in the phenotype and genotype of methicillin resistance by PCR of mecA and femA. Such disparities between methicillin resistance and the presence of mecA gene suggest the presence of control gene of the mecA.

  • Disk type single phase switched reluctance motor which is a simple robust construction, simple control circuit and low manufacturing cost, has a characteristics of axial and radial flux machines. However, because this DSPSRM has a complicated magnetic circuit, it is difficult to analyze the design characteristics.. In this study, the calculation of design parameter based on the conventional design theory of electric machine and the characteristics analysis by computer simulation was performed. As the DSPSRM has the characteristics of both AFM and RFM, it is difficult to analyze its characteristics by the 2D FEM. 3D FEM was applied in the analysis of energy distribution and approximated calculation of torque characteristics with rotor positions. With analysis results, prototype of DSPSRM is manufactured.

Analysis of Talus Slope Stability using 2D FEM and 3D Limit Equilibrium Method (2차원 유한요소법과 3차원 한계평형법을 이용한 테일러스 사면안정성 해석)

  • A series of talus slope stability analyses were carried out using 2D FEM and 3D limit equilibrium methods for this study. The FEM analyses on Phase 2 were performed to delineate failure depths based on stress distributions for each slope. The results revealed that the failure surface exist in the colluvium layer of about 3-10 m thickness. Three dimensional models, derived from the FEM analyses and geological field survey, were made for the use in a 3D limit equilibrium analysis. The result shows that all the talus slopes are stable under dry condition, but unstable under saturated condition due to heavy rain.

  • This paper presents the performances of eddy current brake for the vehicle. Because travelling magnetic field problem of eddy current brake cannot be analyzed accurately in Galerkin finite element method(FEM) for large cell Peclet number, the characteristics are analyzed by using 2-D upwind FEM. The magnet stack width are compensated in order to increase the precision of the solution. The validity of the analysis results is obtained by comparing with experimental data.

  • Double-side linear pulse motor(DSLPM) has more advantages than single-side linear pulse motor because noise and vibration can be considerably decreased by countervailing the normal forces, which is generated between two stators and mover. However, DSLPM has more complicated magnetic flux path and layout of stator pole toot/mover tooth rather than single-side linear pulse motor In this paper, DSLPM is designed and fabricated by considering the air gap magnetic density, shape of tooth and slot. In order to verify the characteristics of DSLPM, the air gap magnetic flux density is analyzed by 2D FEM and the magnetic flux path is analyzed by 3D FEM. Also the static thrust forces is obtained with the analyzed results.

  • The two-dimensional (2D) analysis is widely used in geotechnical engineering for slope stability analysis assuming a plane-strain condition. It is implicitly assumed that the slip surface is infinitely wide, and thus three-dimensional (3D) end effects are negligible because of the infinite width of the slide mass. The majority of work on this subject suggests that the 2D factor of safety is conservative (i.e. lower than the 'true' 3D factor of safety). Recently, the 3D finite element method (FEM) became more attractive due to the progress of computational tools including the computer hardware and software. This paper presents the numerical analyses on rotational mode and translational mode slopes using the 2D and 3D FEM as well as 2D limit equilibrium methods (LEM). The results of the parametric study on the slope stability due to mesh size, dilatency angle, boundary conditions, stress history and model dimensions change are analysed. The analysis showed that the factor of safety in 3D analysis is always higher than that in the 2D analysis and the discrepancy of the slope width in W direction on the factor of safety is ignored if the roller type of W direction conditions is applied.

  • Using 2D numerical analysis that covers the largest section of the dam body, a process is generally performed when evaluating its stability against seepage. The quantity of seepage is first obtained by assuming that its bottom topography is in the simple form of a rectangle, it is then calculated by reflecting its sectional shape during this process of analyzing the seepage quantity. Considering that various forms of dams are being constructed on various types of ground, thanks to more recent technological advances, it is judged more appropriate to draw a conclusion by means of the results on reflecting the realistic shape and topographical conditions of the dam body through 3D numerical analysis. Therefore, this study intends to present a method designed to carry out safety management by evaluating the correct quantity of water leakage that passes only through the dam body, having excluded other factors that include the amount of rainfall through the 3D FEM analysis.

  • A new band pass filter (BPF) prepared by dielectric monoblock and its design techniques are presented. The modeling of the three-pole dielectric monoblock BPF has been carried out by CAD. The equivalent circuit of the BPF was established by transmission lines and lumped capacitors. The transmission line characteristic impendances were computed using 2-D FEM. The BPF for PCS has been designed to have a 60 MHz pass-bandwidth with center frequency of 1960 MHz and an attenuation pole at below the passband using a commercial 3-D structure simulator.

  • Electric machines such as motors which have moving parts are desgined for producing mechanical force or torque. The accurate calculation of electromagnetic force and torque is important in the design these machines, Electromagnetic force calculation method using the results of Finite Element Method(FEM) has been presented variously in 2-D problems. Typically the Maxwell's Stress Tensor method and the method of virtual work are used. In the problems including current source, magnetic vector potentials(MVP) have mostly been used as an unknown variables for field analysis by numerical method; e, g. FEM. This paper, thus, introduces both methods using MVP in 3-D case. To verify the usefulness of presented methods, a solenoid model is chosen and analyzed by 3-D and axisymmetrical FEM. In each case, the calculated force are tabulated for several mesh schemes.

Design and Fabrication of Linear-Array Ultrasonic Transducer Using KLM and FEM Simulation for Non-Destructive Testing (KLM 및 FEM 시뮬레이션을 이용한 비파괴검사용 선형배열 초음파 탐촉자의 설계 및 제작)

  • In this paper, a linear-array transducer capable of overcoming the faults of a single element and phased array transducers with convex shape for non-destructive ultrasonic testing was designed and fabricated. A 5.5 MHz linear-array transducer was designed using the PiezoCAD program based on the KLM analysis and the PZFlex program based on the FEM analysis. A 2-2 composite structure was employed to achieve broad-band characteristics. A 128 element linear-array transducer was fabricated and its performance was compared with the simulation results. The center frequency of the fabricated transducer was 5.5 MHz and the -6 dB frequency bandwidth was 70 %. Thus, we expect that the designed transducer can provide an effective inner image of the test material during non-destructive ultrasonic testing.
