The graphene oxides (GOs) were tested as a fluorescent quencher in the field of DNA-diagnostics. The various suspensions of GO nanoplates were prepared by changing the synthesis conditions. The suspensions were stable for at least 6 weeks by differing degrees of functionalization of various oxygen-containing groups of atoms. Depending on the properties of GO nanoplates, their fluorescent quenching abilities, which were determined by the amount of the tagged immobilized oligonucleotide, were also changed. GO suspension synthesized at $75^{\circ}C$ of reaction mixture showed the fluorescent quenching of 16.39 nmol/mg, which would be a potential substitution of molecular fluorescent quencher in test-systems for DNA-diagnostics.
수분 후 담배의 배에 중이온 ($^{14}$ N) beam을 조사하여 유도한 variegated담배의 잎 조직을 기내배양하여 재생된 식물체의 특성을 조사하였다. NAA 0.1mg/L 와 BAP 1/0mg/L를 첨가한 MS배지에서 variegated담배의 잎을 배양하면 백색부위에서는 백색 식물체만이 유도되었으나 녹색부위에서는 녹색 식물체가 47.2%, 백색 식물체가 37.4%, variegated식물체가 15.4% 유도되었다. 재생된 녹색 식물체는 자가수분한 후대 (F$_1$)에서 녹색 식물체 1,651 개:백색 식물체 54개로 분리되었다. 또한 variegated 잎에서 재생된 녹색 식물체와 정상주를 정역교배 한 결과, 전부 녹색체가 분리되었다. 이 결과는 variegated잎의 표현형 유전은 최소한 모성 유전이 아님을 보여준다. Variegated잎의 녹색부위를 배양하여 재생된 variegated식물체의 잎을 이용하여 DNA gel blot한 결과, 엽록체 유전자인 psbA, rbcL, 165 rDNA, 23S rDNA의 양은 흰색부위 잎에서 정상주인 녹색 잎과 variegated식물체 잎의 녹색부위 보다 많았다.
국내 야생화들로부터 효모를 분리하여 이들을 고부가가치의 의약이나 건강 식품산업에 응용하기 위한 연구의 일환으로 경기도와 제주도 일부 지역의 야생화들을 채집하여 효모들을 분리, 동정하고 이들 가운데 항통풍성 xanthine oxidase 저해물질 생산 균주를 선발한 후 생산 조건을 검토하였다. 서울 구로동과 일부 경기도 지역 야생화 31점에서 26종, 54균주의 효모들을 분리, 동정하였고 이들 중 Pseudozyma sp.가 13종으로 가장 많이 분리되었다. 제주도 송악산과 성산 일출봉 지역의 야생화 21점에서 모두 14종, 32균주의 효모들을 분리, 동정하였고, 이들 균 중에서 Sporobolomyces ruberrimus가 6종으로 가장 많이 분리되었다. 86효모 균주들을 YEPD 배지에서 2일간 배양하여 각각의 배양 상등액과 무세포 추출물을 제조한 후 이들의 항통풍성 Xanthine oxidase 저해활성을 측정한 결과, Pseudozyma hubeiensis 228-S-1의 무세포 추출물이 19.5%의 가장 높은 Xanthine oxidase 저해활성을 보였다. 또한 Pseudozyma hubeiensis 228-S-1을 YEPD 배지에 접종하여 $30^{\circ}C$에서 36시간 배양하였을 때, 세포내 항통풍성 xanthine oxidase 저해물질을 가장 많이 생산하였다.
Sixteen microsatellite markers (simple sequence repeat (SSR) markers) were employed to examine the genetic stability of 27 randomly chosen date palm (Phoenix dactylifera L.) plants produced through somatic embryogenesis with upto forty two in vitro subcultures. No microsatellite DNA variation was observed among all micropropagated plants. Our results indicate that the micropropagation protocol used for rapid in vitro multiplication is appropriate and suitable for clonal propagation of date palm and corroborated that somatic embryogenesis can also be used as one of the safe modes for production of true-to-type plants of date palm. This is the first report on the use of microsatellite DNA markers to establish the genetic stability in micropropagated date palm plants.
We isolated and identified culturable Arctic bacteria that had inhabited soils around the Korean Arctic Research Station Dasan located at Ny-Alsund, Svalbard, Norway $(79^{\circ}N,\;12^{\circ}E)$. The collected soils were diluted in distilled water; the diluted soil-water was spread on 3M petri-films at Dasan Station. The petri-films were transported to the laboratory at KORDI, and cultured at $4^{\circ}C$. Colonies grown on the petri-films were subsequently cultured on nutrient agar plates at $4^{\circ}C$ every 7 days. The pure colonies were inoculated into nutrient liquid media, genomic DNA was extracted, and phylogenetic analysis was performed on the basis of 165 rDNA sequences. A total of 227 strains of bacteria were isolated. Among them, 16S rDNA sequences of 185 strains were identical with those of known strains isolated in this study, and 42 strains were finally identified. Phylogenetic analysis using 16S rDNA indicated that the 30 strains belonged to Pseudomonas, 7 strains to Arthrobacter, two strains to Flavobacterium, and the remaining to Achromobacter, Pedobacter, and Psychrobacter. Among the 42 strains, 14 bacteria produced protease: they were 6 strains of Pseudomonax, 4 strains of Arthrobater, an Achromobacter strain, 2 strains of Flavobacterium, and a Pedohacter strain. We expect these Arctic bacteria can be used for screening to develop new industrial enzymes that are active at low temperatures.
India possesses a total buffalo population of 105 million out of which 26.1% inhabit Uttar Pradesh. The buffalo of Uttar Pradesh are described as nondescript or local buffaloes. Currently, there is no report about the genetic diversity, phylogenetic relationship and matrilineal genetic structure of these buffaloes. To determine the origin and genetic diversity of UP buffaloes, we sequenced and analysed the mitochondrial DNA D-loop sequences in 259 samples from entire Uttar Pradesh. One hundred nine haplotypes were identified in UP buffaloes that were defined by 96 polymorphic sites. We implemented neutrality tests to assess signatures of recent historical demographic events like Tajima's D test and Fu's Fs test. The phylogenetic studies revealed that there was no geographic differentiation and UP buffaloes had a single maternal lineage while buffaloes of Eastern UP were distinctive from rest of the UP buffaloes.
The genes for ribosomal RNA in Rhizobium meliloti and Bradyrhizobium japonicum were analyzed by southern hybridization of BamHI, EcoRI, HindIII digested chromosomal DNA with purified 5' $^{32}P$-labeled 16S and 23S rRNA. The big differences in the hybridization pattern of both rhizobia were found. The comparative results were discussed in relation to the copy number and conservativity of restriction sites in the rRNA genes of both rhizobia.
This paper dealt with the distribution of Shigella spp. on 5 piggeries in Taegu and Kyungpook during the period from August to October 1987. Isolated Shigella were examined for serogrouping, antimicrobial drug resistance and detection of R plasmid. Genetic properties of R plasmid in Shigella have examined to fertility inhibition (Fi) and gel electrophoresis was performed for the isolation of plasmid DNA. The results obtained were summarised as followings; 1. Of total 2,978 samples from 5 piggeries, 82 strains (2.8%) of Shigella spp. were isolated from 82 samples. The isolated strains were identified as S dysenteriae (60 strains), S flexneri (20 strains) and S sonnei (2 strains). 2. Of the 82 strains examined 67 (95.1%) were resistant to one or more antibiotics, such as ampicillin (Am), chloramphenicol (Cm), kanamycin (Km), nalidixic acid (Na), rifampicin (Rf), streptomycin (Sm), sulfademethoxine (Su), and tetracycline (Tc) and higher resistant to Su (90.2%), Sm (63.4%) and Tc (63.4%). 3. Of the 78 resistant Shigella strains 26 (33.3%) harbored conjugative R plasmids and the transfer frequency of Sm (50.0%), Cm(33.3%) resistance was much higher than that of the other drug resistance. 4. The most common resistant patterns were SmSuTc, Su and AmSmSuTc. 5. Out of the 26 Shigella R plasm ids examined for Fi, 14(53.8%) were $Fi^+$ and the remainder were $Fi^-$. 6. The plasmid DNA profiles in Shigella spp. (9 strains) isolated from pigs were confirmed as being 2 to 9 fragments by the gel electrophoresis. Their molecular size ranged 2.17 to 87.62 kilobase (Kb). All strains of Shigella spp. consisted in 15.4 Kb plasmids.
임상 검체에서 분리 동정한 E.faecalis 균주에 대하여 gentamicin및 타 항균제에 대한 감수성을 조사했고, R-플라스미드 DNA패턴 및 제한효소에 의한 DNA단편분석,filter-mating에 의한 gentamicin 플라스미드 DNA 전이, R-플라스미드 DNA 제거, tetracycline 내성 유전자확인, 항원분석을 실시하였다. 분리된 40주 E. faecalis 모두가 gentamicin에는 내성이었고, 25주는 요 검체에서 분리되었으며, 이 40균주들 중 95%가 입원환자에서 분리되어 중요한 원내감염균임을 알 수 있었다. Gentamicin 고도내성 E. faecalis는 24주(60%)이었고, 최소억제농도의 범위는 ampicillin이 1~64$\mu$g/ml, Chroramphenicol 이 8~128$\mu$g/ml 과 erythromycin 128 $\mu$g/ml, vancomycinol 1~2 $\mu$g이었다. 분리한 Gentamicin 고도내성 E. faecalis HL-1은 4가지 항균제에 모두 내성이었고, 7종류의 플라스미드가 있었다. HL-2와 HL-3은 모두 6종류, HL-4는 7종류, HL-5는 4종류 그리고 HL-6은 5종류의 플라스미드가 있었다. E. faecalis HL-1 과 HL-6의 내성전달 빈도는 6.3$\times10^{-4}$ 과 3.7$\times10^{-5}$이 었으며, E faecalis HL-1의 내성전달 빈도가 높았다 접합에 의해 E.faecalir HL-1과 HL-6의 플라스미드 중 51.7 Kb크기의 전이된 DNA가 확인되어 gentamicin 내성이 플라스미드 전이에 의한 것임을 알 수 있었다. Tetracycline 유전자는 E.faecalis transconjugants R-1의 2.15 Kb 플라스미드에 있었다. 공여균주 및 transconjugants균주의 항원성을 Immunoblotting으로 분석한 결과 E. faecalis HL-1과 E. faecalis transconjugants R-1균주는 97.8, 46.8 Kd의 분자량을 갖는 단백질과 공통적으로 반응하였고, E. faecalis HL-6와 E. faecalis transconjugants R-6균주는 46.8 Kd의 분자량을 갖는 단백질과 공통적으로 반응하였다. 그 반응의 강도는 85.8, 97.8, 46.8, 33.7, 63.5 와 74.8 Kd 순이었다. 공여균주 E. faecalis HL-6에서는 반응하지 않았던 97.8, 95.8, 74.8와 63.5 Kd의 단백질이 E. faecalis transconjugants R-6균 주에서 반응하였다. 이들 종 특이성 단백질을 Invitro에서는 배양되지 않는 E. faecalis에 대한 혈청학적 진단의 항원으로 이용한다면 유용할 것이다.
