Cyclophosphamide가 마우스의 면역기억에 미치는 영향 (Effects of Cyclophosphamide on Immunological Memory in Mice)

  • The use of alkylating agent cyclophosphamide(CY), a widely used antitumor drug is well known as a potent immunosuppressant and has been used as a probe for investigating the functional capabilities of lymphocyte subsets of both T and B cells that play an important role in the regulation of the immune response. The present study was undertaken in an effort to assess the effects of CY on immunological memory in murine model. CY, given as a single dose of CY(250mg/kg) before sensitization with sheep red blood cells(SRBC) enhanced the primary response of Arthus and delayed-type hypersensitivity(DTH), as measured by footpad swelling reaction, but suppressed their tertiary DTH response. The similar CY pretreatment enhanced both the primary and tertiary hemagglutinin(HA) responses to SRBC, and the tertiary antibody response against polyvinylpyrroridone(PVP), a thymus-independent antigen but not the primary response against PVP. CY, given as a single dose of 250mg/kg 2 days before the primary immunization and two doses of 100mg/kg 2 days before the secondary and tertiary immunization, markedly suppressed the tertiary DTH and HA responses to SRBC. However, CY, given as small multiple daily doses(10mg/kg) over 4 days before sensitization but not after sensitization, enhanced the secondary HA response to SRBC. Contact sensitivity to dinitrofluorobenzene(DNFB) was suppressed by the drug, given either as a single large dose(300mg/kg) or as multiple dose(10mg/kg) administered 2 days before, together with or after DNFB sensitization. This suppression was more pronounced and more significant when CY was given as multiple dose. However, the enhancement of the secondary contact sensitivity to DNFB by CY was not clear-cut. The splenectomy appears to increase the enhancing effect of CY on contact sensitivity. These results suggest that CY selectively influences the immune response depending on the time of the drug administration relative to immunization and that the secondary or tertiary immune response involve memory cells with different susceptibilities to CY. Moreover, these results suggest that multiple low doses may sesectivley inhibit suppressor T cell proliferation involving DTH, HA or contact sensitivity without effecting helper T cells, but high doses presumably inhibit helper T cells and suppressor T cells with effecting B cells.

압출성형과 효소처리가 신령버섯 β-Glucan의 추출에 미치는 영향 (Effects of Extrusion and Enzyme Treatment on Extraction of β-Glucan from Agaricus blazei Murill)

  • 압출성형공정과 효소처리가 신령버섯 추출물의 ${\beta}$-glucan 함량에 미치는 영향을 조사하였다. 스크루 회전속도 250 rpm, 원료 사입량 100 g/min, 사출구 3 mm 원형으로 고정하였으며, 수분 함량 20, 30%, 배럴 온도 130, $140^{\circ}C$로 조절하여 압출성형 신령버섯을 제조하였다. 수분 함량 20%, 배럴 온도 $130^{\circ}C$의 압출성형공정을 통한 신령버섯 추출혼합물은 ${\beta}$-glucan 함량이 16.91 mg/g으로 다른 공정들보다 증가하였다. 수분 함량 20%, 배럴 온도 $130^{\circ}C$의 압출성형 공정과 Rohament CL로 처리한 추출물의 ${\beta}$-glucan 함량은 같은 압출성형공정의 Viscozyme L 처리 추출물 17.35 mg/g과 Plantase TL 처리 추출물 17.51 mg/g보다 18.32 mg/g으로 가장 많이 증가하였다. 결론적으로 압출성형공정과 Rohament CL 처리한 추출물의 ${\beta}$-glucan 함량이 18.32 mg/g으로 압출성형공정과 효소처리를 하지 않은 대조구의 14.45 mg/g보다 약 26.7% 정도 증가함을 나타내었다. 이는 신령버섯 추출 시에 압출성형공정과 효소처리가 ${\beta}$-glucan 함량을 대조구에 비해 증가시킬 수 있다고 판단되었다.

Acute and Repeated 28-Day Oral Dose Toxicity Studies of Thymus vulgaris L. Essential Oil in Rats

  • Thymus vulgaris L. is widely used as an ingredient in cooking and in herbal medicine. However, there is little information about its toxicity. The present study was performed to evaluate the acute and repeated 28-day oral dose toxicity of thyme essential oil in rats. For the acute toxicity test, two groups of three rats were used. The rats received a single dose of essential oil: 300 or 2,000 mg/kg of body weight (bw). The rats were observed individually during the first four hours, and then daily until day 14. For the toxicity test with repeated doses, four groups of 10 rats were used. Doses of 100, 250, and 500 mg/kg/day were tested for 28 days. At the end of the experiment, blood was collected and the animals were sacrificed. Histopathological examination showed that in the lungs of rats given the 2,000 mg/kg bw dose, polymorph nuclear infiltrates, hemosiderin macrophages, and interstitial space thickening were present. In the repeated dose study, all rats survived the 28-day treatment period and apparently showed no signs of toxicity. The hematological and biochemical parameters were not altered. The histopathological study of the organs showed severe changes in the lung, with the dose of 500 mg/kg/day; in the other organs, no alterations were observed or the changes were slight. The body weight was only altered in male rats given the 500 mg/kg dose. The relative weight of the organs did not show any significant changes. Our studies revealed that the essential oil of Thymus vulgaris has moderate oral toxicity according to the results of the acute test, whereas the results of the 28-day oral toxicity test suggest that the no-observed-adverse effect level (NOAEL) is greater than 250 mg/kg/day.

맥류 종자의 페룰산(ferulic acid) 함량과 붉은곰팡이 저항성 (Ferulic Acid Content of Barley and Wheat Grains and Head Blight Resistance)

  • 페룰산 함량이 붉은곰팡이병과 곰팡이독소 저항성 곡류 육종의 선발지표가 될 수 있는지 알기 위해 밀, 보리, 벼 육성 품종 80점의 페룰산 함량을 조사하였다. 페룰산 함량은 보리 20품종이 1.66-2.77 mg/g, 밀 40품종이 0.56-1.53 mg/g이었으며 벼 20품종은 0.91-2.13 mg/g이었다. 이 중 페룰산 함량이 다른 밀과 보리 각 7품종을 선정하여 F. graminearum과 F. asiaticum 각 2균주의 붉은곰팡이병과 독소 생성량을 분석하였다. 밀 품종의 평균 병원성은 페룰산 함량이 높은 보리에 비해 유의하게 낮았고 밀 품종 간에는 상관관계가 없었다. 곰팡이독소도 병원성과 같이 페룰산 함량이 낮은 밀이 보리보다 생성량이 낮았다. 그러나 보리에서는 F. asiaticum 균주의 병원성과 독소 생성량이 페룰산 함량과 음의 상관관계를 나타내었다. 이 결과는 페룰산 함량이 밀과 보리에서 F. graminearum과 F. asiaticum에 의한 붉은곰팡이병과 독소 생성에 저항성 요인이 아님을 보여준다.

심장이식 환자에서 Cyclosporine에 의한 중추신경독성 -1례 보고- (Cyclosporine-Assoc iated Central Neurotox ic its after Hearat Transplantat ion 1 Case Report)

  • 승모판막 폐쇄부전증으로 승모판막 치환술을 받은 45세 남자가 확장성 심근증으로 심장이식수술을 받았다. 환자는 수술전 면역억제를 위하여 cyclosporine 400 mg, Immuran 250 mg과 Solumedrol 500 mg을 투약하였 다 술후 cyclosporine을 2 mg/kg/day로 정주했는데 술후 8시간이 지나도 혼수상태가 지속되어 cyclosporine을 1 mg/kg/day로 감량하였다. 당시 혈장내 cyclosporine농도는 345$\mu\textrm{g}$/L, 크레아티닌 수치는 1.8 mg/dl였으며 마그 네슘 수치는 정상수준이 였다. 환자의 의식은 술후 31시간째 완전히 회복되었으나 술후 36시간경부터 전신근 력약화, 초조감, 환청 및 환각을 호소하였다. 신경증상들은 술후 4일째 정상으로 회복되었으며, 환자는 술후 28일째 퇴원하였고 12개월째 후유증은 보이지 않고 있다.

Paenibacillus kribbensis AM49로 발효시킨 택사의 항산화 활성 및 NO 생성 효과 (Antioxidant activity and NO production of the Alisma orientale Juzep fermented by Paenibacillus kribbensis AM49)

  • Objectives : The purpose of this study was to investigate the antioxidant activities and nitric oxide (NO) production in RAW 264.7 macrophages in extract of Alisma orientale Juzep (EAOJ) and fermented extract (FAOJ) by Paenibacillus kribbensis AM49 (P. kribbensis AM49). Methods : The Alisma orientale Juzep was fermented with P. kribbensis AM49 at 37℃ for 72 hours. We measured total polyphenol and total flavonoid, DPPH radical scavenging activity, FRAP activity and reducing power by spectrometric assay in EAOJ and FAOJ at concentrations at 0.5, 1, 5, 10 mg/㎖. Positive control was used ascorbic acid. Furthermore, we examined effect of EAOJ and FAOJ on the cell viability and NO production in RAW 264.7 macrophages. Results : The total polyphenol and total flavonoids content of FAOJ were increased 9.16 mg/g, 2.59 mg/g to 12.58 mg/g, 3.45 mg/g. DPPH radical scavenging activity, FRAP activity and reducing power were dose dependently increased according to the treatment concentration (0.5, 1, 5, 10 mg/㎖) of EAOJ and FAOJ. In particular, DPPH radical scavenging activity, FRAP activity of FAOJ was significantly increased at 5, 10 mg/㎖. Reducing power of FAOJ at 10 mg/㎖ was similar to ascorbic acid at 0.1 mg/㎖. In addition, the cell viability and NO production in RAW 264.7 macrophages were significantly increased at the concentrations of 250, 500, 1000 ㎍/㎖. Conclusions : These results suggest that FAOJ by P. kribbensis AM49 has effects to antioxidant activity. In addition, the cell viability and NO production in RAW 264.7 macrophages were significantly increased.

품종에 따른 국내산 및 중국산 메밀의 품질특성 (Evaluation of quality characteristics of Korean and Chinese buckwheats)

  • 본 연구는 국내산 및 중국산 단메밀과 쓴메밀의 일반성분, 아미노산 조성, 지방산 조성과 루틴 함량을 분석하였다. 다양한 메밀의 수분, 조단백질, 조지방, 조회분, 탄수화물의 함량은 각각 8.78~13.37, 11.00~12.11, 2.87~3.18, 1.80~2.58, 70.2~73.8%로 분석되었다. 국내산 및 중국산 메밀의 주요 아미노산은 aspartic acid(1,105.1~1,403.5mg%), glutamine (2,250.9~2,996.1 mg%), arginine(932.5~1,388.6 mg%)로 분석되었으며, K(423.7~569.4 mg%), Mg(181.8~255.9 mg%), P(328.6~555.0 mg%)가 주요 미네랄로 분석되었다. 한편, 지방산 조성은 포화지방산인 palmitic acid(14.2~16.1%)와 불포화 지방산인 oleic acid(37.0~40.8%), linoleic acid(31.7~38.6%)가 주요 지방산으로 분석되었다. 또한, 국내산 및 중국산 메밀의 주요 phenolic compound는 루틴으로 분석 되었으며, 쓴메밀(261.0~265.0 mg/g)이 단메밀(4.39~5.68 mg/g)보다 높은 함량을 보였다. 본 연구를 종합하면, 국내산 단메밀은 중국산 단메밀과 비교하여 영양성분은 크게 차이를 보이지 않았으나, 메밀의 주요 활성성분인 루틴은 약간 높게 함유되어 있었다. 또한 국내산 쓴메밀은 중국산 쓴메밀에 비하여 Ca, Fe 함량이 높았으며, 루틴 역시 약간 높은 함유량을 보였다. 따라서, 메밀에서 주요 활성성분으로 제시되고 있는 루틴의 함량이 중국산 메밀보다 국내산 메밀에 높게 함유되어 있기 때문에 국내산 메밀에서 더 높은 생리활성이 나타날 것으로 기대된다.

Toxicity Study of Red Ginseng Acidic Polysaccharide (RGAP) : Single and 2-week Repeated Oral Dose Toxicity Study in Rats

  • The present study was conducted to investigate the single and 2-week repeated dose toxicity of red ginseng acidic polysaccharide (RGAP) in Sprague-Dawley rats. The test article was administered orally to rats at dose levels of 0, and 2000 mg/kg/day for single dose toxicity study and at dose levels of 0, 250, 500, and 1000 mg/kg/day for repeated dose toxicity study. In both studies, there were no treatment-related effects on mortality, clinical signs, food and water consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, necropsy findings and organ weights of all animals treated RGAP. Based on these results, it was concluded that the 2-week repeated oral dose of RGAP may have no toxic effect in rats at a dose level of 1000 mg/kg/day. In the condition of this study, the no-observed-adverse-effect level (NOAEL) was considered to be 1000 mg/kg/day for both sexes.

우엉 에탄올 추출물의 항산화활성과 항돌연변이 효과 (Antioxidative and Antimutagenic Effects of $Arctium$ $lappa$ Ethanol Extract)

  • The antioxidant activities of the ethanol extract of Arctium lappa were assessed by measuring the 1,1-diphenyl-2-picrylhydrazyl( DPPH) radical scavenging effect, inhibition of $Fe^{2+}$-induced lipid peroxidation, inhibition of malondialdehyde(MDA)-bovine serum albumin(BSA) conjugation reaction and antimutagenic capacities using the Ames test. The DPPH radical scavenging activity and inhibition of $Fe^{2+}$-induced lipid peroxidation of the $Arctium$ $lappa$ ethanol extract significantly increased in a dose-dependent manner. In the radical scavenging assay using DPPH, the $IC_{50}$ of the Arctium lappa extract was 296 ${\mu}g$/assay(1.29 mg of dry sample). In addition, the $IC_{50}$ in the inhibition of $Fe^{2+}$-induced lipid peroxidation was 1,759 ${\mu}g$/assay(7.65 mg of dry sample). This extract also significantly inhibited the MDA-BSA conjugation reaction with an $IC_{50}$ of 57.58 mg/assay(250 mg of dry sample). However, no inhibitory effects against the direct and indirect mutagenicities in $Salmonella$ Typhimurium TA98 and TA100 were observed. Based on these results, the ethanol extract of $Arctium$ $lappa$ was shown to display considerable antioxidative activities.

Influence of Annealing Temperatures on Corrosion Resistance of Magnesium Thin Film-Coated Electrogalvanized Steel

  • To improve the corrosion resistance of an electrogalvanized steel sheet, we deposited magnesium film on it using a vacuum evaporation method and annealed the films at $250-330^{\circ}C$. The zinc-magnesium alloy is consequently formed by diffusion of magnesium into the zinc coating. From the anodic polarization test in 3% NaCl solution, the films annealed at $270-310^{\circ}C$ showed better corrosion resistance than others. In X-ray diffraction analysis, $ZnMg_2$ was detected through out the temperature range, whereas $Mg_2Zn_{11}$ and $FeZn_{13}$ were detected only in the film annealed at $310^{\circ}C$. The depth composition profile showed that the compositions of Mg at $270-290^{\circ}C$ are evenly and deeply distributed in the film surface layer. These results demonstrate that $270-290^{\circ}C$ is a proper temperature range to produce a layer of $MgZn_2$ intermetallic compound to act as a homogeneous passive layer.
