Objectives : Hyangsayangyi-tang (香砂養胃湯) is a polyherbal formula, has been used as one of the representative So-eumin (少陰人) prescriptions in Sasang-medicine (四象醫學), a famous Korean medicinal theory. The aqueous extract of Hyangsayangyi-tang (HSYYT) was evaluated for its possible ameliorative effect in the regulation of hypothyroidism related reproductive organ damages in propylthiouracil (PTU)-induced rat model. Methods : HSYYT aqueous extracts were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 500, 250 and 125 mg/kg (bodyweight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28days. Results : PTU induced hypothyroidism and related male reproductive organ (testis, epididymis and prostate) damages were favorably and dose-dependently inhibited by treatment of HSYYT 500 and 250 mg/kg, and they also effectively regulated the PTU-induced abnormal antioxidant defense system changes in the testis. No effective or any harmful changes on the PTU induced hypothyroidism and related male reproductive organ damages were observed in HSYYT 125 mg/kg treated rats as compared with PTU control in this experiment. Conclusions : The results obtained in this study suggest that oral administration of 500 and 250 mg/kg of HSYYT showed favorable effects on the hypothyroidism and related reproductive organ damages through augmentation of antioxidant defense system in the testis, and it is considered that HSYYT may be help to ameliorate the hypothyroidism and related organ damages in clinics.
Objectives: The object of this study was to observe the complex anti-climacterium potentials of Yuklinzu aqueous extracts (YLZ), using bilateral ovariectomy (OVX) female ddY mice similar to women postmenopausal symptoms, as including cardiovascular diseases, obesity, hyperlipidemia, osteoporosis and hepatic steatosis. Methods: In order to evaluate anti-climacterium effects of YLZ, six groups of mice were used; sham control, OVX control, 17β-estradiol, YLZ 500, 250 and 125 mg/kg treated groups. Since 28 days after bilateral OVX surgery, YLZ were administered orally for 84 days, once a day. And then we evaluated anti-climacterium effects divided into five categories; estrogenic effects, anti-obesity, hypolipidemic effects, hepatoprotective effects and anti-osteoporotic effects. The results of YLZ were compared with 17β-estradiol 0.03 ㎍/head/day subcutaneous treated OVX mice. Results: As a result of OVX, obvious changes related to the estrogen-deficient menopausal symptoms - obesity, hyperlipidemia, hepatic steatosis and osteoporosis were displayed in mice. However, these menopausal symptoms induced by OVX were significantly inhibited by 84 days of consecutive treatment of 17β-estradiol, YLZ 500, 250 and 125 mg/kg, respectively. Especially, YLZ showed obvious dose-dependent inhibitory activities on the OVX-induced climacterium changes in mice, and YLZ 500 mg/kg showed comparable inhibitory effects against menopausal symptoms in comparison with those of 17β-estradiol 0.03 ㎍/head/day subcutaneous treatment. Conclusions: The results suggest that oral administration of YLZ 500, 250 and 125 mg/kg has obvious dose-dependent favorable anti-climacterium effects in OVX mice. Especially, YLZ 500 mg/kg showed comparable inhibitory effects against menopausal symptoms in comparison with those of 17β-estradiol 0.03 ㎍/head/day subcutaneous treatment.

Tubers(4-5cm diameter) of Zantedeschia 'Black Magic' were used to study the induction of flowering in GA3 soaking times and concentration. The GA3 soaking were 10 seconds and 30 minutes at the GA3 concentrations of 50, 100, 250, and 500 mg·L1. The time to emergence in GA3 treatment were 1.1~4.0 days shorter than control Growth characteristics were good in GA3 treatment but that was no significance in GA3 soaking time and concentration. When GA3 soaking time and concentration were increased, days to flowering was shorter, but flower stalk length, flower stalk width, flower length, flower width were no difference. The number of flower per tuber was most as 4.0~4.3 in 250~500 mg·L1 GA3 concentration and that was about 2 times compared to control. The normal flower ratio and bulb enlargement were similar compared with GA3 soaking time and concentration. GA3 250~500 mg·L1 treatment is necessary for improvement of number of flowers per bulb.
The major and trace constituents of Mongolian chromite were analyzed by ICP-AES. The dissolution procedures, mixed acid ($HClO_4+H_3PO_4$) digestion and fusion with $Na_2O_2$ flux, have been studied to dissolve the chromite. The optimum dissolution method was found to be a fusion with $Na_2O_2$ flux. The effect of large amount of Na on major and trace constituents was examined when these elements were determined by ICP-AES. There was no effect on major elements at a concentration of Na 250 mg/L solution. The emission intensity of trace constituents containing Na 1,250 mg/L decreased to 1.0-5.2% according to elements and wavelengths. The result of this method was compared with that of neutron activation analysis (NAA) to confirm the accuracy of this procedure. The results between two methods were in a good agreement within less than 5% for $Al_2O_3$, $Cr_2O_3$, MgO and -20 to 8% for Co, Mn, V, Zn, respectively.
The pathogenesis of cholestatic liver injury as well as the modulation of hepatic fibrogenesis is causally associated with involvement of reactive oxygen species and free radical reactions. In this study, we investigated whether flavonoids (fustin, sulfuretin) which were isolated from Rhus verniciflua Stokes (RCS) have antioxidant and antihepatotoxicity effect under the biliary liver fibrosis condition. After surgery (control) and posttreated RCS methanol extract (250mg/kg), ethyl acetate extract (250mg/kg) and flavonoids were administered p.o. 10mg/kg/day in two weeks for control groups. The concentration of clinical parameters and product of hepatic lipid peroxidation and the hydroxyproline content were significantly increased in liver fibrosis developed rats. Among the clinical parameters of serum, value of ALT, AST, SDH, total bilirubin and ${\gamma}$ -GT in posttreated RCS components-group showed significantly lower than in control-group. The content of hydroxyproline in posttreated RCS components-group showed lower than in control group and then the value of MDA in posttreated RCS components-group was also significantly reduced to 40~60% of that in control group. The hepatic xanthine oxidase and aldehyde oxidate activities were posttreated RCS components-group showed significantly lower than in control-group. The hepatic SOD and glutathione peroxidase activities were posttreated RCS components-group showed significantly higher than in control-group. Hence we concluded that active components of fustin and sulfuretin which were isolated from R. verniciflua Stokes were hepatoprotective effect in experimental liver fibrosis.
This study was conducted to investigate the antioxidation and biological activities of 5 kinds of specialized crops (asparagus, aloe, curcumin, backnyuncho and garlic) cultivated in Namhae. All crops were freeze dried and then extracted with 10 folds of $60^{\circ}C$ hot-water, for 8 hours. Collected twice extracted water extract were also freeze dried. Dried extract samples were diluted with deionized water for examination. The total phenolics content was the highest observed in asparagus extract to $23.79{\pm}0.33mg/g$ and total flavonoids of aloe extract was the highest content $17.10{\pm}0.05mg/g$. In all samples, DPPH, ABTs and NO radical scavenging activity were depends on the concentration of the extract tended to increase. ABTs radical scavenging activity was the highest conducted $96.10{\pm}0.06%$ effect examined at $2,000{\mu}g/mL$ in asparagus hot-water extract. NO radical scavenging activity of hot-water extract from curcumin was the highest $250{\mu}g/mL$ concentration observed in $45.04{\pm}1.39%$. In all sample, tyrosinase activity was significantly inhibited with increasing of hot-water extract concentration. As a result, ${\alpha}$-glucosidase inhibition activity was the highest in hot-water extract from backnyuncho on $31.38{\pm}0.94%$ at $250{\mu}g/mL$.
This experiment was designed to improve the in vitro production system of apple rootstock M.26 as being influenced by the growth regulators, TDZ, BA, IAA, IBA, zeatin, and GA$_3$. Different levels of potassium humate (KH), known as cytokinin and auxin-like substance, were also supplemented to the MS basal medium along with IBA 0.6 mg/L to find out it effect on root formation in apple rootstock M.26. ID initiate and establish the in vitro multiplication of shoots byway of meristem culture, MS medium added with zeatin 1.0 mg/L was found to be the most suitable, showing the 100% of survival rate of shoot tips. A combination of thidiazuron (TDZ) 0.2 mg/L and NAA 0.5 mg/L promoted the shoot proliferation when shoot tips were used as explants. MS basal medium plus IBA 0.6 mg/L was very effective for root induction, but an addition of potassium humate (250 mg/L) to the medium containing IBA 0.6 mg/L stimulated the induction and proliferation of the rook by far the better.
Explants of lettuce (Lactuca sativa L.) were cocultured with A. tumefaciens LBA 4404 harboring ${\gamma}$-tocopherol methyltransferase (${\gamma}$-TMT) gene from Arabidopsis thaliana. These explants were transferred to MS medium supplemented with 50 mg/L kanamycin, 500 mg/L carbenicillin, 0.1 mg/L NAA and 0.5 mg/L BA. After 4 weeks, kanamycin resistant shoots were obtained from the explants on the selection medium. The putative transgenic shoots were transferred to rooting MS medium supplemented with 50 mg/L kanamycin and 250 mg/L carbenicillin. Stable incorporation of the Arabidopsis ${\gamma}$-TMT cDNA into lettuce genomic DNA was confirmed by PCR and Southern analysis. HPLC analysis showed that $\alpha$- to ${\gamma}$-tocopherol ratio increased over four fold in a transgenic lettuce line indicating successful expression of the transgenic Arabidopsis ${\gamma}$-TMT in lettuce.
Biologically, MGK-264 (N-octyl bicycloheptene dicarboximide) acts as a synergists for insecticides mainly pyrethrins and pyrethroids. It's used extensively in combination with pyrethrin and piperonyl butoxide and also with personal insect repellent and cockroach repellents. But the toxic effect of MGK-264 in mamalians was a relatively little known therefore in this studies it was initiated to examine the toxic effect of MGK -264 in rats. For 5 weeks it administrated daily in each 250 mg and 500 mg of MGK-264 per kg of body weight in rats. 1) The body weight gain and the LYMPH (%) value in blood were observed a slight tendency to reduce in accordance with amount of dose and number treatment time. 2) The content of cytochrome P-450 and activity of NADPH-cytochrome c reductase were decreased in liver and those were observed some tendency in the kidney as liver but not significant. 3) The liver cholinesterase activity in the both 250 mg/kg and 500 mg/kg per kg of body weight with treated groups and the liver aniline hydroxylase in 500 mg/kg treated group were gradually decreased from 4 weeks after treated groups. In consequence it would sugested that the toxic effect of MGK-264 was low but in could offer hazard effect in liver and nervers system of rats if it was administrated move dose of MGK-264 and agumented in number of treated time.
This study was carried out to investigate the effects of fertilizer and organic resource annual dressing for 30 years of Jeonbug series (silt loam) on soil properties and rice N uptake in paddy field soil. In the study field, treatments including control (NPK), NPK+rice straw, NPK+rice straw compost and nitrogen fertilization levels at 0, 100, 150, 200, 250 kg $ha^{-1}$ have been imposed for 30 years. Soil hardness and bulk density decreased from 15.7 mm and 1.381 Mg $m^{-3}$ in the control to 12.5 mm and 1.244 Mg $m^{-3}$ in NPK+rice straw compost treatment, respectively, indicating improvement of soil physical conditions such as porosity. Co-application of straw compost with NPK also result in a better chemical properties than NPK alone as it increased available phosphate (from 96 to 133 mg $kg^{-1}$), available silicate (from 81 to 116 mg $kg^{-1}$), and cation exchange capacity (from 9.8 to 11.4 $cmol_c\;kg^{-1}$). Soil organic matter concentration of top soil (0 to 7.5 cm in depth) was higher in NPK+rice straw and NPK+rice straw compost than in control. Fertilizer N uptake amount was much higher in NPK+rice straw (nitrogen fertilization level; 250 kg $ha^{-1}$) and NPK+rice straw compost (nitrogen fertilization levels; 200, 250 kg $ha^{-1}$) plots compared to the control (nitrogen fertilization level; 100 kg $ha^{-1}$) plot. Nitrogen use efficiency was showed significantly high in the NPK+rice straw compost (nitrogen fertilization levels; 100, 150 kg $ha^{-1}$) plot compared to the control (nitrogen fertilization level; 100 kg $ha^{-1}$) plot. Therefore, it was suggested that application of organic inputs is helpful in improving soil fertility and physical conditions and thus in N uptake.
