참당귀의 무기성분, 유리당, 아미노산 및 지방산 함량 특성 (Characterization of Inorganic Components, Free Sugars, Amino Acids, and Fatty Acids in Angelica gigas Nakai)

  • Background : The major compounds of Angelica species are decursin, decursinol angelate, nodakenin, umbelliferone and ${\beta}$-sitosterol, which act anti-inflammatories, reduce pains, protect the liver and enhance the immune system. This study investigated the chemical compositions, minerals, metals, sugars and overall amino acid composition in Angelica gigas Nakai. Methods and Results : Powder of Angelica roots smaller than 30 mesh were used. Physico-chemical analysis revealed the presence of carbohydrates (62.0%), crude proteins (13.9%), moisture (11.4%), crude fats (7.3%) and ash (5.4%). Results showed that potassium was present in the highest amount (1,859 ppm), followed by magnesium (214.5 ppm), calcium (147.3 ppm) and sodium (6.0 ppm). Free sugar profiles showed the presence of sucrose (29.3 g/100 g). The total amino acids concentrations was 9,752 mg/100 g, the most common and dominant amino acids were arginine (2,181 mg/100 g), glutamic acid (1,212 mg/100 g) and aspartic acid (834 mg/100 g). The total free amino acids contents was 1,476 mg/100 g, in which the most common amino acid were arginine (932 mg/100 g), glutamic acid (127 mg/100 g), and ${\gamma}$-aminobutyric acid (80.4 mg/100 g). The fatty acid composition of A. gigas showed a higher concentration of unsaturated fatty acids such as linoleic acid (443.9 mg/100 g) and palmitic acid (181.3 mg/100 g) according to gas chromatography. Conclusions : These results showed that Angelica roots can be used in various fields of foods and medicines, and in the preparation of cosmetics.

양송이(洋松茸)의 단일담포자(單一擔胞子) 분리(分離)에 의(依)한 신계통(新系統) 선발(選拔)에 관한 연구(硏究) (Improvement of Cultivated Mushroom Agaricus bisporus by Means of Single Basidiospore Selections)

  • 앙송이 단일담포자(單一擔胞予)의 발아(發芽)에 가장 알맞은 배지(培地)는 water agar였고 이때 발아율(發芽率)은 26%였다. 단일담포자(單一擔鐵子) 6,372개(個)를 분리(分離)하여 1,622개(個)를 발아(發芽)시켰으며 이들은 균총형태(菌叢形態)에 따라 strandy type과 fluffy type으로 구분(區分) 할 수 있었으며 이중 전자(前者)는 균사생장(菌絲生長) 속도(速度)가 빨랐고 수량(收量)도 높았다. 발아(發芽)된 단일담포자중(單一擔胞子中) 퇴비추출배지상(堆肥抽出培地上)에서 strandy type 778개(個)를 선발(選拔)하여 생산력(生産力)을 검정(檢定)한 결과(結果) 1차(次)에서 공시(供試) 균주(菌株)의 89개(個), 2차(次)에서 19개(個), 3차(次)에서 9개(個) 균주(菌株)를 우수(優秀) 계통(系統)으로 선아(選芽)할 수 있었다. 이들중 No. 645 외(外) 8균주(菌株)는 수량(收量)이 5%이상(以上) 증수(增收)되었으며 그중 No. 1567, 1708 두 균주(菌株)는 수량(收量)이 10% 이상(以上) 증수(增收)되었고 품질(品質)도 양호(良好)하여 우량계통(優良系統)으로 선발(選拔)할 수 있었다. 따라서 단일담포자(單一擔胞子)의 계통분리(純系分離)에 의(依)해 양송이 신계통(新系統) 육성(育成)이 가능(可能)할 것으로 보인다.

White Sport Syndrome Virus Disease of Shirmp and Diagnostic Methods

  • Since 1993, the White Spot Syndrome Virus (WSSV) disease occurred in China among cultured shrimps resulting in mass mortality. Epizootiological surveys undertaken during the outbreak period of 1993-1994 indicated that all stages of Penaeus chinensis, P. japonicus and P. monodon were infected. Consequent to the transport of contaminated shrimp seedlings and seawater, the disease spread all over the farms of China. The disease was more rapidly transmitted at temperatures above $25^{\circ}C$. Challenge experiments showed the causative agent was highly virulent. White spots appeared on the carapace of both span-taneous and experimentally infected shrimps. Moribund shrimps contained turbid hemolymph, hypertrophied Iymphoid organ and a necrotic mid-gut gland. Electron microscopy showed the presence of viral particles in the gills, stomach, lymphoid organ, and epidermal tissue of the infected shrimp. The visions were slightly ovoid with an envelope and averaged 350 $\times$ 150 nm; nucleocapsids measured 375 $\times$ 157 nm. With discontinuous sucrose gradient of 35, 50 and 60% (w/v), the virus was separated from hemolymph of the infected shrimp. The estimated molecular weight of genomic DNA was 237 Kb with EcoR I, 247 Kb with Hind III and 241kb with Pst I. A total of 9 hybridoma colones secreting monoclonal antibodies (MAbs) were produced from mouse myeloma and spleen cells immunized with WSSV. The immunofluorescence assay of gill tissue showed that the MAbs reacted with diseased but not with healthy shrimp. The MAbs belonged to IgGl, IgG2b subclass and IgM class, all with kappa light Immune-electron-microscopy with colloidal gold marker showed the presence of 5 MAbs epitopes on the envelope and one on the capsid of the virus. Baculoviral mid-gut gland necrosis showed the specificity of the MAbs produced. For diagnosis 5 different methods were selected. Using Kimura primers for PCR, or MAbs for immunoblot, ELISA or FAT method, in situ hybridization was carried out to show the gene. All these methods detected WSSV in the organ samples of the diseased shrimp but not in healthy one.

  • 식물생장 촉진 호르몬인 auxin을 생산하는 균주를 선발하기 위해 경산지역 경작지 토양에서 29종의 세균을 분리하였으며 Salkowsky test를 통해 auxin을 생산하는 14종을 선발하였다. 이 중 CAS(chrome azurol S) blue agar에서 siderophore의 생산을 확인 후 auxin과 siderohroe를 동시에 생산하는 AH18을 최종 선발하였다. AH18의 mung-bean adventitious root induction test를 통해 식물생장 촉진 능이 대조구에 비해 1.5배나 뛰어남을 알 수 있었고, 토마토 pot test에서 토마토 시들음병의 윈인균인 F. oxysporum에 대해서 길항력도 나타내었다. 선발된 AHl8 균주는 16S rDNA와 Biolog system을 통해 Bacillus subtilis 동정되었다. B. subtilis AH18은 Sucrose-asparagine-MgsO$_4$(SAM) medium (pH 6.0)에 접종하여 30$^{\circ}C$에서 3일간 배양 시 siderophore를 가장 많이 생산하는 것을 알 수 있었다.

닭 뉴캐슬병 바이러스의 특이 검출을 위한 polymerase chain reaction 법 (Polymerase chain reaction for the detection of Newcastle disease virus)

  • To study the specific tools for the diagnosis of Newcastle disease virus (NDV) in chicken, polymerase chain reaction (PCR) and its presumable conditions were evaluated for the detection of hemagglutinin-neuraminidase (HN) gene of NDV RNA. For these purposes, Kyojeongwon strain of the NDV was propagated in allantoic cavity of SPF embryonating chicken eggs, and viral RNA was extracted from fractionated virus after the allantoic fluids were ultracentrifuged with sucrose gradient. The first-strand cDNA was then made for the HN gene of NDV RNA by reverse transcription at $42^{\circ}C$ for 1 hour using specific primer complementary to the HN gene. The single-stranded cDNA was used as template in the PCR of the HN-DNA, and various conditions of the PCR were evaluated to set up method for the specific detection of the HN-DNA. The PCR conditions promising for the detection of HN gene consist of preheating at $94^{\circ}C$, 5 min, 30 cycles of denaturation at $94^{\circ}C$, 1 min, annealing at $55^{\circ}C$, 1 min and polymerization at $72^{\circ}C$, 2 min, and a cycle of extension at $72^{\circ}C$, 5 min. when NDVs of allantoic fluids without fractionation were applied to the above PCR condition, the HN genes were detected effectively not only from Kyojeongwon but from other velogenic strains such as Herts and a field isolate.

건강음료로서의 자작나무 수액의 유출량에 미치는 영향인자 (Factors Affecting on Sap Flows of Birch Trees, Betula platyphylla as a Healthy Beverages)

  • Lately public interest in tree saps of maple and birch trees stimulated to increase demands for sap drink as a natural medical beverage. In order to understand factors affecting tree sap flows, birch species, particularly Betula platyphylla in Mt.Sobaek area, were monitored for daily sap flows according to factors, such as DBH, tapping hole sizes and direction of hole drilling on the trunks. The chemical constituents of saps were also analyzed. The sap flow initiated from the end of March and finished at the end of April. The flow maxima appeared from April 14th to April 26th. Total amounts of the sap flow obtained from birch tree were over 1,800 ml per day per one tree. Sap flows were increased with increasing DBH and tree age. Six milimeter drilled hole resulted in the highest sap flows. Sap flows increased with increases of diameter and height of the trees. Hole drilling to downward side(south-facing) of tree produced almost doubled sap flows than that of upward one(north-facing), while rightside drilling produced same amounts of saps to that of leftside one. Six mm drilling gave the best results not only in maximum flows but also effective hole occluding rates. The saps have in the range of 4.5 to 5.6 pH and l.0 - 2.0% of saccharinity. Sucrose, glucose, fructose and high mineral contents were found in the saps of birch tree. The tree saps could be utilized not only medicinal drink, but also one of the healthy beverages.

한국아동(韓國兒童)의 치아우식경험과 치면상(齒面上) Streptococcus mutans 분포(分布)에 대(對)한 연구(硏究) (Relationship between the Caries Experience of Korean School Children and the Distribution of Streptococcus mutans in Dental Plaque)

  • Various investigations have been carried out to elucidate the causative relationship between specific oral bacterial species and dental caries since it was first demonstrated that selected streptococoal species produced dental caries in germfree rats when fed a high-sucrose diet. Now, S. mutans is considered to play an important role in the development of dental caries in animals and humans, and only a limited number of species of bacteria other than S. mutans are occasionally found to be cariogenic in experimental animals. In this regard, association of the number of S. mutans in approximal plaque with caries experience (DMFT) was studied from 137 Korean school children($10{\sim}11$ year old). Biotypes of the collected strains of S. mutans were determined, and their relationship with caries status was also examined. The following results were emerged from the study. 1. S. mutans was detected in the plaques of all children. 2. Statistically significant positive correlation(r=0.445, p<0.001) was found between the caries experience(DMFT) and the number of S. mutans in approximal plaques. 3. The number of S. mutans were significantly higher(p<0.001) in plaques removed from carious surface than from sound surface. 4. The most frequent biotype was biotype I(78.8%), followed by IV(33.1%) and V(09.5%). Biotype II was isolated in the plaque of two children(1.7%) only. 5. There was no apparent relation of specific biotypes to carious status.

Variations in Physicochemical Properties of Brown Rice (Oryza sativa L.) During Storage

  • Present study deals with variations of physicochemicals including $\gamma$-aminobutyric acid (GABA), $\gamma$-oryzanol, free sugar, lipoxygenase activity, fat acidity, and germination rate from Korean brown rice cultivars. With increase of storage time and temperature, GABA, $\gamma$-oryzanol, lipoxygenase activity, and fat acidity increased, whereas free sugar and germination rate was reduced. Among cultivars, 'Gopumbyeo' exhibited the highest contents in GABA and $\gamma$-oryzanol during 12 weeks storage at $25^{\circ}C$ (GABA: $28.6{\pm}5.6{\rightarrow}170.4{\pm}4.6\;mg/100\;g$, $\gamma$-oryzanol: $6.1{\pm}0.7{\rightarrow}6.7{\pm}0.4\;mg/g$) and 'Ilpumbyeo' significantly decreased in free sugar during 12 weeks storage at $10^{\circ}C$ ($1,423.7{\rightarrow}1,058.4\;mg/100\;g$). Moreover, 'Taebongbyeo' exhibited the highest quality owing to low lipoxygenase activity, low fat acidity, and high germination rate. In free sugar compositions, sucrose exhibited the highest content (>70%), followed by fructose (>7%), raffinose (>5%), glucose (>3%), and maltose (>2%) during storage. Based on our results, changes of physicochemicals in stored brown rice may be important information in processing food and functional properties.

한국산 곤약의 성분 조성에 관한 연구 (Studies on the Chemical Components of Elephant - foot Produced in Korea)

  • 한국산 구약 감자(elephant-foot=Amorphoph-alus Konjak K.Koch)의 이화학적 성분 조성을 조사하기 위하여 국산 생감자와 분말을 시료로 하여 외국산 정분과 같이 분석한 결과 생감자는 수분이 80.64%였고 고형분의 대부분은 당류, 단백질, 희분이고 섬유나 지방은 미량 존재하였다. 건조 분말 정제품인 정분은 당류 74.42-80.41%, 수분은 10% 전후였고 만난 함량은 한국산이 일본, 중국산보다 상당히 적었고 역시 가용성 유리당도 일본산이 높고 한국산이 낮았는데 구성당은 한국산은 Sucrose가 많은 대신 외국산은 Fructose, Glu-cose 함량이 높았다. 한국산 정분의 단백질 함량은 외국산보다 2배 많았고 이의 amino acid 조성은 Glutamic acid, Aspartic acid, Arginine 등이 총아미노산의 45%를 차지하고 Valine, Serine, Leucines, Glycine의 순서로 많았다. 정분의 무기물은 K가 많고 P>Na>Ca순으로 많게 함유하였고 chip의 정분 수율은 한국산이 61%, 중국산은 57.6%였으며 정분의 백도는 색도 측정치는 중국산이 약간 높았으나 육안으로는 별 차이가 없이 비슷하였다.

첨가물질에 따른 옥수수와 녹두전분겔의 관능적 기계적 특성 (Sensory and Instrumental Characteristics of Corn and Mung bean Starch Gels with Additives)

  • 첨가물질이 무처리 그리고 탈지옥수수와 녹두전분겔의 텍스쳐 특성에 미치는 영향을 알아 보기 위하여 실온에서 24시간과 72시간 저장하면서 관능적 기계적 특성을 조사하였다. 슈크로오스 지방산 에스테르, 셀룰로오스 유도체와 식용유를 전분량에 대해 0.5%첨가한 전분겔의 종합적인 맛은 24시간 저장한 전분겔의 경우에 응집성, 휘어짐성에서 높은 정의 상관관계를 보였고 72시간 저장한 전분겔에서는 탄성 , 응집성 ,색깔, 부드러움성 , 휘어짐성 , 견고성 , 투명도에서 정의상관관계를 보였다. 첨가물질의 유무에 관계없이 탈지에 의한 옥수수전분 겔의 물성적 특성이 크게 증가되었으며 첨가물질의 물성적 특성에 대한 영향은 모든 전분겔에서 낮게 나타났다. 종합적인 맛은 첨가물질을 넣은 전분겔이 다소 낮았고 24시간 저장한 전분겔은 응집성에서 72시간 저장한 전분겔은 탄성에서 가장 높은 상관관계를 보였다. 기계적 검사에 의한 텍스쳐 특성치도 관능검사 결과와 비슷한 경향을 보여 첨가물질의 종류에 따른 유의적인 차이는 보이지 않았다.
