THERMO-SENSITIVITY OF N-VINYL PYRROLODONE-CO-2- HYDROXYETHYLMETHACRYLATE HYDROGELS

  • The copolymerization of HEMA with different hydrophilic and hydrophobic co-monomers allows for the manipulation of their intrinsic properties. 2-Hydroxyethylmethacrylate (HEMA)-based hydrogels thus are of great interest due to their outstanding physico-mechanical properties and chemical stability. The idea to use HEMA in order to create thermo-sensitive polymers was based on our assumption that thermal-sensitivity comes from a suitable hydrophilic-hydrophobic balance of macromolecules. In this work we have chosen N-vinyl pyrrolidone as a hydrophilic co-monomer with the relatively hydrophobic HEMA due to its good polymerizing properties as well as its non-toxicity in a polymer state and deserved recognition as a biocompatible material. As a result, copolymerization of NVP and HEMA was successful in obtaining new types of thermo-sensitive polymers composed of hydrophilic and hydrophobic monomers.

Hydrogel Chamber를 이용한 수정 및 배양 (Utilization of Hydrogel Chamber for Fertilization and In Vivo Culture)

  • The in-vitro fertilization in human clinic and animal reproduction is a very important technique but the rate of success is still low. When the in-vitro fertilization and culture of gametes or embryos were done under the condition which Hema hydrogel chamber were implanted into the peritoneal cavity of mouse, the in-vitro fertilization and development of embryos could be significantly improved and the cell-block under in-vitro culture could be overcome. Also, the Rema hydrogel chamber was very useful for the protection of isolated blastomeres. It is concluded that the polymerized Hema (pHema) hydrogel chamber may be effectively used in the fields of embryo transfer and in vitro fertilization.

콘택트렌즈의 재질별 각막부종의 비교 (Comparison of Corneal Edema Induced by Material of Contact Lens)

  • 목적: 콘택트렌즈를 권장착용시간 동안 착용한 후에 렌즈의 재질에 따라 발생하는 각막부종의 정도를 비교해 보고자 한다. 방법: 안질환이나 약물사용이 없는 성인남녀 43명 77안을 대상으로 하였다. Narafilcon A, HEMA, 실리콘 하이드로젤 재질의 정기교체형 렌즈 3종의 렌즈를 사용하였다. 중심각막두께는 Pentacam을 사용하여 렌즈 착용 전과 8시간 착용 후를 측정하였다. 착용 전과 후의 차이 값으로 각막부종의 정도를 판단하였다. 결과: Narafilcon A, HEMA 재질의 렌즈, 실리콘 하이드로젤 재질의 정기교체형 렌즈의 평균 각막부종은 각각 $2.36{\pm}6.15{\mu}m(Mean{\pm}SD)$, $23.61{\pm}10.71{\mu}m$, $18.25{\pm}8.64{\mu}m$였다. Narafilcon A를 착용한 뒤 측정된 중심각막두께는 착용 전 측정한 중심각막두께와 통계적으로 유의한 차이가 없었다. HEMA와 실리콘 하이드로젤 재질의 렌즈에서 착용 후에 유발된 각막부종은 그 정도에 있어 narafilcon A와 통계적으로 유의한 차이가 있었다. 결론: 정기교체형 실리콘 하이드로젤 재질 렌즈에서 유발된 각막부종의 양은 HEMA 재질의 렌즈에서 유발된 각막부종의 양과 통계적으로 유의한 차이가 없었지만 narafilcon A와는 유의한 차이가 있었다. 따라서 콘택트렌즈 재료의 조성에 따라 권장착용시간을 초과하지 않도록 주의해야 할 것으로 판단된다.

Hydroxyethyl methacrylate (HEMA)와 Triethylene glycol dimethacrylate (TEGDMA)가 실험적 복합레진의 특성에 미치는 영향 (EFFECT OF HEMA AND TEGDMA ON THE PROPERTIES OF EXPERIMENTAL COMPOSITE RESINS)

  • Purpose : The purpose of this study was to compare HEMA and TEGDMA as diluents for the composite resin. Material and methods : Eight kinds of experimental light curable composite resins were prepared and used. Concentrations of monomer and filler were same for all experimental composites, except, the diluent's ratios to the monomer. The ratio of diluents to Bis-GMA were 15%, 20%, 25%, and 30%, and two kinds of diluents were used, so total experimental groups were eight including one control group of 25% TEGDMA. Results : Depth of cure, flexural strength, shear bond strength to bovine enamel, shear bond strength to bovine dentin, water absorption and solubility of composites in water were measured. Sample size for each groups were 10. Arithmetic means were used as each groups representative values, and regression test for two diluents and low concentrations, Duncan's multiple range test, and Two-way ANOVA test were done for kinds of diluents and its concentrations at level of 0.05. Conclusion : Following results were obtained ; 1. There were not significant differences in effects of HEMA and TEGDMA to depth of cure, flexural strength of composites and shear bond strength to bovine enamel (p>0.05). 2. Increase of the concentrations of the diluents made the depth of cure (p<0.001) and flexural strength (p<0.05) a little higher. 3. Shear bond strength to dentin was higher on HEMA containing composites than TEGDMA containing composites (p<0.001). 4. Water absorption was higher on HEMA containing composites than TEGDMA containing composites (p<0.01).

Preparation and Physical Properties of Silicone Hydrogel Ophthalmic Lens Containing Hydrophilic Monomer

  • The major physical characteristics of macromolecules used in silicone hydrogel ophthalmic lenses include optical transmittance, oxygen permeability, water content, and refractive index. For the preparation of highly functional silicone hydrogel lens materials, two silicone monomers were used in the presence of 2-hydroxyethyl methacrylate (HEMA) and hydroxypropyl methacrylate (HPMA). The samples containing HEMA and HPMA had oxygen transmissibility (Dk) values in the range of 73.38–50.98 × 10-11 (cm2/s) (mLO2/mL×mmHg) and 71.94–42.80 × 10-11 (cm2/s) (mLO2/mL×mmHg), respectively. Furthermore, the water contents of samples containing HEMA and HPMA were in the range of 32.73–34.67% and 31.94–33.74%, respectively, and the refractive indices were in the range of 1.4348–1.4364 and 1.4385–1.4407, respectively. Thus, silicone monomers containing HEMA and HPMA are expected to be useful for fabricating high-oxygen-permeability silicon hydrogel ophthalmic lenses.

표면개질된 폴리설폰 막에 대한 오일에멀젼의 수투와 플럭스 (Water Permeation Flux of Oil-Emulsion through Surface-Modified Polysulfone Membrane)

  • A hollow-fiber type polysulfone UF membrane was surface-coated with hydroxyethylmethacrylate (HEMA). The effect of various coating parameters on permeation flux, such as concentration ratio of AIBN/HEMA, solvent(water or methanol), and UV irradiation time, was investigated. The water permeation flux of oil-emulsion increased with concentration ratio of AIBN/HEMA, and increased with UV irradiation time. The flux of the membrane coated in solution using methanol as a solvent was greater than that of the membrane coated in solution using water as a solvent. The flux of both the coated and the uncoated membrane declined with the operation. The flux decline means that the membrane fouling by oil-emulsion occurs. However, the fouling of the coated membrane was much less than that of the uncoated membrane.

Benzalkonium Chloride가 BPDM/HEMA계 접착제의 상아질 접착에 미치는 영향 (EFFECT OF BENZALKONIUM CHLORIDE ON DENTIN BONDING WITH BPDM/HEMA SYSTEM)

  • The purpose of this study was to elucidate the effect of benzalkonium chloride on tensile bond strength of BPDM/HEMA dentin bonding. One hundred sixty dentin specimens from freshly extracted bovine mandibular incisors were used, and 0, 0.02, 0.1 or 0.5% benzalkonium chloride solution was applied to the dentin specimen with/after phosphoric acid. 32% phosphoric acid was used when the specimens were bonded with One-Step$^{TM}$, a BPDM/HEMA system and 10% was used when bonded with All-Bond$^{(R)}$ 2, a NTG-GMA/BPDM system. Aelitefil$^{TM}$ composite resin was bonded to the pretreated dentin specimen with the use of All-Bond$^{(R)}$ 2 or One-Step$^{TM}$ dentin bonding agent. After the bonded specimens were stored in $37^{\circ}C$ distilled water for 24 hours, tensile bond strength was measured. The fractured dentin specimens were examined under the scanning electron microscope. The results were as follows : Benzalkonium chloride application after acid-treatment resulted in decrease of dentin bond strength of One-Step$^{TM}$, a BPDM/HEMA system (p>0.05). Benzalkonium chloride application did not exert any influence on dentin bond strength of All-Bond$^{(R)}$ 2, a NTG-GMA/BPDM system (p>0.05). There was no relationship between the concentration or application method of benzalkonium chloride and the dentin bond strength of One-Step$^{TM}$ or All-Bon$^{(R)}$ 2 (p<0.05). On SEM examination of the fractured dentin-resin interface, while mixed failure was prominent in dentin bonding with One-Step$^{TM}$, adhesive and mixed failures were seen together in dentin bonding with All-Bond$^{(R)}$ 2 regardless of the concentration and application method of benzalkonium chloride.

상아질을 통한 HEMA의 확산에 인산농도가 미치는 영향 (EFFECT OF PHOSPHORIC ACID CONCENTRATION ON THE DIFFUSION OF HEMA THROUGH DENTIN)

  • The purpose of this study was to investigate the effect of phosphoric acid concentration on the movement of 2-hydroxyethylmethacrylate(HEMA) from bonding resin - resin composite combination through dentin in vitro. Freshly extracted human third molar teeth were divided into four groups each of 10 teeth. A closed chamber with 1 ml distilled water was attached to the CEJ of each tooth. An occlusal cavity of 4mm diameter & remaining dentin thickness of 1.0-1.5mm was prepared in each tooth. Dentin was treated with 10% phosphoric acid gel for 15 seconds. 32% phosphoric acid gel for 15 seconds, or with 35% phosphoric acid gel for 15 seconds. A control group not treated with acid gel was also prepared. The cavities were rinsed, dried and then treated with the HEMA-containing All-Bond 2 primer & bonding resin which was light-cured for 10 seconds. The cavities were then restored with Z100 composite resin(shade:A3.5:3M Dent. Prod. USA) & light cured for 30 seconds. Water samples were retrieved from the chambers over a time course (4.32, 14.4, 43.2, 144 & 432 minutes ; 1, 3 & 10 days) and analyzed by high performance liquid chromatography. The results were as follows. 1. HEMA was detected in the pulp chambers of all teeth from 4.32 minutes after resin placement The highest rate of release was in the first sample period (0-4.32 min) & rate of release declined exponentially thereafter. 2. No significant differences were found for mean release rate for HEMA over a time course among the four groups (p>0.05). 3. The diffusion rate was significantly (p<0.05) less for 10% phosphoric acid gel than 32% phosphoric acid gel at the second sample period(4.32-14.4 min). 4. No significant differences were found for cumulative HEMA diffusion among the four groups at 10 days(p>0.05) and mean total(cumulative) release at 10 days for all groups was in the 9 - 16 nmol range. 5. The cumulative release was significantly (p<0.05) less for 10% phosphoric acid gel than 32% phophoric acid gel at the third(14.4-43.2 min) & fourth(43.2-144 min) sample period.

Poly(Dimethylaminoethyl Methacrylate)-Based pH-Responsive Hydrogels Regulate Doxorubicin Release at Acidic Condition

  • Stimuli-responsive biomaterials that alter their function through sensing local molecular cues may enable technological advances in the fields of drug delivery, gene delivery, actuators, biosensors, and tissue engineering. In this research, pH-responsive hydrogel which is comprised of dimethylaminoethyl methacylate (DMAEMA) and 2-hydroxyethyl methacrylate (HEMA) was synthesized for the effective delivery of doxorubicin (Dox) to breast cancer cells. Cancer and tumor tissues show a lower extracellular pH than normal tissues. DMAEMA/HEMA hydrogels showed significant sensitivity by small pH changes and each formulation of hydrogels was examined by scanning electron microscopy, mechanical test, equilibrium mass swelling, controlled Dox release, and cytotoxicity. High swelling ratios and Dox release were obtained at low pH buffer condition, low cross-linker concentration, and high content of DMAEMA. Dox release was accelerated to 67.3% at pH 5.5 for 6-h incubation at $37^{\circ}C$, while it was limited to 13.8% at pH7.4 at the same time and temperature. Cell toxicity results to breast cancer cells indicate that pH-responsive DMAEMA/HEMA hydrogels may be used as an efficient matrix for anti-cancer drug delivery with various transporting manners. Also, pH-responsive DMAEMA/HEMA hydrogels may be useful in therapeutic treatment which is required a triggered release at low pH range such as gene delivery, ischemia, and diabetic ketoacidosis.
