우엉차의 껍질 제거와 로스팅에 따른 추출온도별 항산화 효과 (Effects of Roasting and Peeling Process and Extraction Temperature on the Antioxidant Activity of Burdock Tea)

  • Background: We investigated the optimal aqueous extraction conditions for recovery of high yields of total phenolic compounds from roots of Arctium lappa L. (burdock, Asteraceae), and we compared their antioxidant capacity. Methods and Results: The antioxidant activity of the extracts was tested using 2,2-diphenyl-1-picrylhydrazyl, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt, and oxygen radical absorbance capacity assays. In addition, the major phenolic compounds present in the extracts were determined by high performance liquid chromatography analysis. Our results suggest that the roasted burdock $100^{\circ}C$, 15 min extract exhibited the strongest radical scavenging activity and possessed the highest concentration of phenolic compounds. The polyphenol content of both dried burdock and roasted burdock significantly increased with increase in the extraction temperature and time. Conclusions: These results indicated a relationship between phenolic compound levels in burdock and their free radical scavenging activities. This suggests that phenolic compounds significantly increase the antioxidant potential of burdock extracts.

  • 유리화탄소전극 위에 탄소나노튜브(CNT), 전하전달체(CTC), 글루코스 산화효소(GOx), 폴리이온복합체(PIC, poly-L-lysine hydrobromiderhk과 poly(sodium 4-styrenesulfonate) 혼합물)를 순차적으로 도포하여 글루코스/산소 바이오전지용 효소전극을 제조하였다. 또한, CNT, bilirubin oxidase (BOD), 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS), 그리고 PIC 등의 층으로 제조한 전극을 바이오캐쏘드로 사용하여 바이오전지를 제조하였다. CNT와 CTC가 전극의 성능에 미치는 영향을 조사하였으며, 글루코스농도 5, 20, 200 mM에서 각각 3.6, 10.1, $46.5{\mu}W/cm^2$의 최대전력밀도를 나타내었으며, 본 연구에서 제시한 전극이 바이오전지 및 바이오센서의 개발에 활용될 수 있다는 것을 보여주었다.

Effects of Extraction Methods on In Vitro Biological Capacities and Rheological Properties of Polysaccharides from Red Pepper Stems

  • The purposes of this study were to produce polysaccharides from red pepper stems using different extraction methods and evaluate their chemical composition, in vitro biological capacities, and rheological properties. Two polysaccharides were extracted from red pepper stems using an autoclave and alkali treatments, and the extracts were named PAU and PAL, respectively. The contents of total phenolics and flavonoids were significantly higher in PAU than those in PAL. PAU exhibited greater scavenging activities on 2,2-diphenyl-1-picrylhydrazyl radicals, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radicals, superoxide radicals, and nitrite compared to PAL, suggesting that PAU served as better antioxidants. Similarly, in vitro inhibitory abilities against carbohydrate hydrolyzing enzymes of PAU were higher than those of PAL. Steady shear rheological analysis demonstrated that PAU showed higher psuedoplastic shear-thinning behavior compared to PAL. Based on the results from dynamic shear rheological properties, it was found that both samples had predominantly viscous behavior rather than elastic behavior.

n-Hexane 및 Ethyl Acetate를 이용한 Willamette 품종 라즈베리추출물의 페놀 화합물 함량 및 항산화 효능 연구 (Studies on the Phenolic Compound Content and Antioxidant Efficacy of Rubus idaeus (Raspberry) Cultivar 'Willamette' Fruit Extract Using n-Hexane and Ethyl Acetate)

  • 본 연구는 willamette 품종의 동결건조라즈베리분말, n-hexane 및 ethyl acetate를 이용하여 라즈베리 추출물(Rubus idaeus (Raspberry) fruit extract, RIFE)을 제조한 다음 페놀 화합물 함량, 철 이온 환원능력, 그리고 라디칼 소거 능력의 측정을 수행하였다. Willamette 품종 라즈베리추출물은 10% 농도까지 세포독성을 보이지 않았다. 해당 농도로 실험을 수행한 결과 총 페놀 화합물 함량은 375.3 ppm, 총 플라보노이드 함량은 43.46 ppm이 포함된 것을 확인하였고, ferric reducing antioxidant power (FRAP) reagent를 이용한 철 이온 환원능력은 FeSO4 0.532 mM에 해당하는 환원능력을 가졌음을 확인하였다. 이를 바탕으로 항산화 소재의 대표적 원료인 L-ascorbic acid와 비교하는 라디칼 소거능 실험을 실시하였다. 그 결과 1,1-diphenyl- 2-picrylhydrazyl (DPPH) 라디칼 소거능력은 94.5 ± 0.7%를 나타냈고, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) 라디칼 소거능은 99.4 ± 2.82%를 나타냈으며, nitric oxide (NO) 라디칼소거능은 88.5 ± 0.4%를 나타냈다. 표준인 L-ascorbic acid 용액과 비교할 때 25 - 50 ppm 사이의 효능을 지닌 DPPH 라디칼 소거능 / 100 ppm에 근접한 효능을 지닌 ABTS 라디칼 소거능 / 1,000 ppm 이상의 효능을 지닌 NO 라디칼 소거능을 가졌음을 확인하였다. 이러한 결과들은 willamette 품종 라즈베리추출물이 항산화 활성을 가지는 효과적인 화장품 소재로 적용 가능하다는 것을 시사한다.

수수 수집종의 농업적 형질 및 생리활성 분석을 통한 우수계통 선발 (Selection of Superior Sorghum Accession by Assessing Agronomic Characters and Biological Activity)

  • Background: Sorghum is a major cereal food crop used in many parts of the world. It has been grown on a subsistence level by farmers, under various conditions of environmental stresses in the semi-arid tropics of Africa and Asia. This plant has received significant attention because of its ability to reduce cholesterol in the blood, and its anti-dementia, antioxidant, and antimicrobial properties. It is possible to develop a functional and commercially viable sorghum variety by using superior cultivars of sorghum. The objective of this study was to build a database of superior sorghum accession. Methods and Results: We used 250 sorghum accessions collected from different geographical bioregions in Korea. We determined various agronomic characters including germination rate and ear length of these accessions. To determine the antioxidant capacity, we measured the 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt radical scavenging activity, total phenolic contents, and total flavonoid contents. Accession 189 showed higher germination (> 80%) than the other accessions. Higher 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was observed in 11-SB-078 ($RC_{50}$; $1.89{\pm}2.88{\mu}g/m{\ell}$), and higher 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt radical scavenging activity was recorded in 11-SB-116 ($RC_{50}$; $35.48{\pm}2.42{\mu}g/m{\ell}$) than in the other accessions. The ear length ranged from 15 cm to 48 cm, the total phenolic contents ranged from $3mg{\cdot}GAE/g$ to $77mg{\cdot}GAE/g$, and total flavonoid contents ranged from $0.09mg{\cdot}QE/g$ to $1.07mg{\cdot}QE/g$. Conclusions: Among 250 sorghum accessions, we selected 10 with both superior agronomic characters and highly functional food quality.

편백나무(Chamaecyparis obtusa) 추출물의 항산화, 미백효과에 관한 연구 (Studies on the Antioxidant and Whitening Effects of Chamaecyparis obtusa Extract)

  • 본 연구는 화장품 소재로서 편백나무(Chamaecyparis obtusa)의 부위별 이용 가능성을 알아보기 위하여 잎·가지(Leaf), 수피(Bark), 목재(Wood), 뿌리(Root)로 분리하여 99.9 % 에탄올에 추출한 시료로 항산화, 미백효능을 확인하였다. 이를 위해 다음과 같은 연구를 진행하였다. 항산화 평가는 11-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능, 2 2′-azino-bis (3-ethylbenzothiazoline-sulfonic acid) diammonium salt (ABTS+) 라디칼 소거능으로 확인하였다. 미백 효과를 평가하기 위해서 mushroom tyrosinase 저해 활성 평가, MTT assay를 통한 편백나무 추출물의 세포독성평가를 진행하였으며 cellular tyrosinase 저해율 측정과 melanin 생합성량을 확인하였다. 항산화 활성 결과 편백나무 부위 중 수피(Bark) 추출물이 가장 효능이 우수하였으며 B16F10 melanoma cell내 편백나무 수피(Bark)는 tyrosinase 활성 억제 및 멜라닌 생합성 억제 효과를 나타내었다. 따라서 편백나무 수피 에탄올 추출물의 안전하고 우수한 미백 기능성 화장품의 천연물질 소재로서의 개발 가능성을 확인하였다.

귤홍의 함유성분 분석과 항산화 활성 (Component Analysis and Antioxidant Activity of Citrus Peel)

  • This study examined the components and physicochemical properties of citrus peel as a natural health food source by measuring the general and antioxidative contents of citrus peel. Total contents of carbohydrates, crude protein, crude lipid, and ash were 72.1%, 6.6%, 1.3%, and 3.7% respectively. Caloric content of citrus peel was 334.5 kcal, while total dietary fiber was 32.1%. Regarding mineral contents, Ca was the most abundant mineral, followed by K, Mg, and P. Total phenol contents of the 70% ethanolic extracts of citrus peel was $66.4{\pm}8.74mg\;GAE/g$. Total flavonoid contents of the 70% ethanolic extracts were $14.4{\pm}3.28mg\;RE/g$. The antioxidative activities of citrus peel were significantly increasing in a dose dependent manner on DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging, FRAP(ferric reducing antioxidant power) activity and reducing power. Therefore, the general nutrients and other antioxidant bioactive materials in citrus peel proved that citrus peel possesses a high potential materials as a nutritious food.

Effects of Various Extracts from Turmeric (Curcuma longa L.) on Antioxidant Activity

  • The antioxidant activity of 70% methanol, 70% ethanol and chloroform-methanol (CM, 2:1, v/v) extracts from turmeric (Curcuma longa L.) was investigated in this study. The antioxidant potential of various extracts of turmeric was evaluated by using different antioxidant tests, namely DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, ABTS [2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ferric reducing antioxidant power and ${\beta}$-carotene bleaching assays as well as total phenol content and total flavonoid content. All extracts exhibited abundant total phenol content, total flavonoid content and strong antioxidant activity. Particularly, higher activity was exhibited by CM extract with $46.32{\pm}0.35mgCAE/g$ and $15.58{\pm}0.39 mgQE/g$ of total phenol content and total flavonoid content. Generally, antioxidant activity of various extracts from turmeric increased in the following order: 70% methanol extract, 70% ethanol extract and CM extract, respectively. Results suggested that turmeric could be widely used as a source of antioxidant and a kind of functional material in food.

마카(Lepidium meyenii)의 부위별 함유성분 및 항산화 활성 비교 (Component Analysis and Antioxidant Activity of Maca)

  • This study examined the components and physicochemical properties of Lepidium meyenii as a natural health food source. To accomplish this objective, the general and antioxidative contents of Lepidium meyenii were measured. Total contents of carbohydrates, crude protein, crude lipid, and ash were 56.04%, 33.77%, 1.49%, and 3.7%, respectively(dry wright basis). Total phenol contents of the 70% ethanolic extracts of Lepidium meyenii were 189.9 mg/g, 152.5 mg/g. Total flavonoid contents of the 70% ethanolic extracts were 166.0 mg/g, 78.2 mg/g. The antioxidative activities of Lepidium meyenii were significantly increased in a dose dependent manner on DPPH(1,1-diphenyl-2-picrylhydrazyl) radical scavenging, ABTS (2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging, FRAP(ferric reducing antioxidant power) activity, reducing power. The general nutrients and other antioxidant bioactive materials in Lepidium meyenii were also potential materials for good health food.

Study on Antioxidant Activity of Thermal Treatment Yam (Dioscorea batatas DECNE.) by n-Butanol and Ethyl Acetate Extracts

  • In Korea, yam with thermal treatment is widely used to produce tea and beverage, which appeals to all age groups. To better understand the antioxidative activity of thermal treatment yam (Dioscorea batatas $D_{ECNE.}$), n-butanol and ethyl acetate extracts were evaluated with different antioxidant testing system. Their antioxidant activities, including ABTS [2,2' -azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power as well as total phenol contents were assessed in vitro. Results showed ethyl acetate extract ($51.63{\pm}2.51mg\;CAE/g$) possessed similar total phenol content compared with n-butanol extract ($53.93{\pm}1.00mg\;CAE/g$). Ethyl acetate extract also exhibited similarly stronger antioxidant activity compared with n-butanol extract apart from ABTS radical scavenging activity assay.
