The use of JIP test to evaluate drought-tolerance of transgenic rice overexpressing OsNAC10

  • In this study, the JIP test was exploited to assess drought-tolerance of transgenic rice overexpressing OsNAC10. Two types of promoters, RCc3 (root-specific) and GOS2 (constitutive), were used to drive the transcription factor OsNAC10, a gene involved in diverse functions including stress responses. Three-month-old plants were exposed to drought for 1 week and their fluorescence kinetics was evaluated. Our results showed that drought-treated non-transgenic plants (NT) have higher fluorescence intensity at the J phase (2 ms) compared to transgenic plants, indicating a decline in electron transport beyond the reduced plastoquinone ($Q_A^-$). As manifested by negative L bands, transgenic plants also showed higher energetic connectivity and stability over NT plants under drought conditions. Also, the pool size of the end electron acceptor at the photosystem I was reduced more in NT than in transgenic plants under drought conditions. Furthermore, the transgenic plants had higher $PI_{total}$, a combined parameter that reflects all the driving forces considered in JIP test, than NT plants under drought conditions. In particular, the $PI_{total}$ of the RCc3:OsNAC10 plants was higher than that of NT plants, which was in good agreement with their differences in grain yield. Thus, the JIP test proved to be practical for evaluating drought-tolerance of transgenic plants.

The Analgesic Effect and Its Opioidergic Mechanism of Electroacupuncture on Inflammatory Pain in the Type II Collagen-induced Arthritis Rats (전침(電鍼)의 collagen 유발(誘發) 관절염(關節炎)에 대한 진통(鎭痛) 효과(效果) 및 그 기전(機轉)에 관한 연구(硏究) - opioidergic mechanism을 중심으로 -)

  • Objectives : The aim of this study is to evaluate the analgesic effect of electroacupuncture on Jogsamni (ST36) in the collagen-induced arthritis rats and investigate the role played by opioid receptor subtypes $({\mu},\;{\delta},\;{\kappa})$ in the antinociceptive effect of electroacupuncture (EA) In the thermal hyper algesia test. Methods : Immunization of male Sprague-Dawley rats with bovine type H collagen emulsified in incomplete Freund's adjuvant, followed by booster injection 2 weeks later induced collagen-induced arthritis (CIA). The thermal hyperalgesia was evaluated weekly with tail flick latency (TFL). In the fourth week after first immunization, EA stimulation (2 Hz, 0.07 mA, 0.3 ms) was delivered into Jogsamni (5736) for 20 minutes. Analgesic effect was evaluated by using the tail flick latency (TFL) after intraperitoneal injection of normal saline, naloxone, naltrindole and nor-binaltorphimine respectively to CIA rats. Results : The results were as follows; 1. The TFL were gradually decreased in CIA as time elapsed after e immunization of arthrogenic collagen and the maximum value was reached between the third to fifth week. 2. EA stimulation on 5736 inhibited chronic inflammatory pain induced by CIA. 3. The analgesic effect of EA was inhibited by pretreatment of ${\mu}-receptor$ antagonist (naloxone),${\delta}-receptor$ antagonist (naltrindole) and ${\kappa}-receptor$ antagonist (nor-binaltorphimine) respectively. Conclusion : Electroacupuncture has an analgesic effect on the CIA rat and has an antinociception mediated by 8, 5, H receptors.

Chemical Changes and Volatile Carbonyl Formation Occurred in Rice during Cooking Process (취반(炊飯)중 쌀성분(成分)의 화학적(化學的) 변화(變化) 및 휘발성(揮發性) Carbonyl 성분(成分)의 생성(生成))

  • Cooked rice was prepared by the conventional cooking method being used among Korean families. The bottom layer (very slightly roasted and charred) and upper layer of the cooked rice in a cooking vessel were collected separately in order to determine the chemical changes and volatile carbonyl formation occurring in rice during the cooking process. Amino acids, especially free amino acid contents were reduced with the decrease of reducing sugar in rice after cooking. A little change was shown in the fatty acid composition of lipid fractions, however, decreased iodine values and increased fatty acid values were observed after cooking process. Volatile carbonyl compounds from cooked rice were isolated and 8 carbonyls of them were identified. A significant difference of relative composition of the identified carbonyls was found between the upper layer and bottom layer of cooked rice. Generally, the cooked rice obtained from the bottom layer had shown a little more changes in chemical composition and a stronger browning flavour than those of the upper layer of cooked rice.

Development of Ring Light for Shadowless Shooting for Medical Purpose (의료용 무영 촬영을 위한 링라이트 개발)

  • In this research a ring light was developed so that a partial shadowless shooting for the patient's affected area at the medical treatment room and surgical operation room using high luminance light emitting diode (LED) for which attention is being paid as new lighting parts for medical purpose. LED which was applied to the development used high luminance three color LED for full color for which various color materialization and the adjustment of radiation intensity are possible and we can get white light in order to emphasize the delicate expression for generic tone of shooting object, strong highlight, simple shadow and three dimensional effect at the time of close-up shadowless shooting of the affected area. And at the time of design of ring light, the characteristics of LED and the loss of light at the time of penetrating light diffusion PC were considered so that intensity of illumination for over 150 lx can be obtained. The result of measurement of the intensity of illumination of the ring light that was developed revealed that maximum intensity of illumination of 225.7 lx was obtained, while smoke index was measured to be maximum 78 Ra in the case of Red(50%) Green(100%) and Blue LED(60%). We could confirm that response speed was also very fast as 1.72 ms.

  • Purpose : This purpose of this study is to make a better public hospital patient room to reflect the views of the patient. Methods: Survey to examine patient experience on healing environment in public hospital have been conducted for the data collection. 229 open-ended answers have been analysed statistically by MS Excel program. Results : 1) If there is no toilet space in the six-patient room, installation of toilet in the patient room is necessary. 2) The most Frequently used patient furnitures are necessary to accomodate the patients needs. Depending on the patient condition, installation of a portable or fixed the room furniture for the convenience to the using furniture. 3) According to the flow of modern times, wireless internet access is required to install of expand the space. Also be used to establish a regulations of internet space. 4) To create healing environment infection free environment should be provided. 5) Need for improvements of patient room for relatives of patient. It is necessary to secure a comfortable space to stay. 6) Separation is needed among the patients through therapeutic classification. Implication : In order to provide a better patient room environment, it is necessary to reflect the patient's opinion.

Proteomic Analysis of Fructophilic Properties of Osmotolerant Candida magnoliae

  • Candida magnoliae, an osmotolerant and erythritol producing yeast, prefers D-fructose to D-glucose as carbon sources. For the investigation of the fructophilic characteristics with respect to sugar transportation, a sequential extraction method using various detergents and ultracentrifugation was developed to isolate cellular membrane proteins in C. magnoliae. Immunoblot analysis with the Pma1 antibody and two-dimensional electrophoresis analysis coupled with MS showed that the fraction II was enriched with membrane proteins. Eighteen proteins out of 36 spots were identified as membrane or membrane-associated proteins involved in sugar uptake, stress response, carbon metabolism, and so on. Among them, three proteins were significantly upregulated under the fructose supplying conditions. The hexose transporter was highly homologous to Ght6p in Schizosaccharomyces pombe, which was known as a predominant transporter for the fructose uptake of S. pombe because it exhibited higher affinity to D-fructose than D-glucose. The physicochemical properties of the ATP-binding cassette transporter and inorganic transporter explained their direct or indirect associations with the fructophilic behavior of C. magnoliae. The identification and characterization of membrane proteins involved in sugar uptake might contribute to the elucidation of the selective utilization of fructose to glucose by C. magnoliae at a molecular level.

  • In the recent IT field, the biggest concerns may be Mobile environment. This means that it becomes part of our life: the Mobile is applied to the areas of teaching, studying, commercial transaction, entertainment etc. In this paper, we implemented Mobile E-BOOK bookstore using WEB, DB, and a Mobile technology. This system takes duplex from to be available in Mobile WEB environment by PDA and actuality WEB site under desk top environment. This paper have presented big possibility in terms of embodied electronic commerce based on the Mobile-based E-BOOKstore system that has not attempted yet commercially.

Bioactive Components from Red Tide Plankton, Cochlodinium polykrikoides (Cochlodinium polykrikoides 적조 조체의 생리활성 성분)

  • Large amounts (300 grams) of natural red tide plankton, Cochlodinium polykrikoides, was collected at the Yokji island coastal waters, Kyeongnam, Korea, on October, 1993. Using the bioassay systems, bioactive materials were screened from methanol extracts of C. polykrikoides. Live C. polykrykoides was toxic to fishes, however, the water soluble and chloroform soluble fraction of their methanol extracts did not shown ichthyotoxicity (5 mg/ml), and toxicity to mice (50 mg, i.p,). These fractions did not show any peaks corresponding to paralytic shellfish toxins or diarryhetic shellfish toxins on the fluorometric HPIC chromatograms. Neither fractions did not show antibiotic activities by paper disk (10 mg/disc) test and chloroform soluble fraction showed only $20\%$ growth inhibition activity on the Lymphoid P-388 at the concentration of $180\;{\mu}g/ml$. Hemolytic activity was detected by both fractions. Fatty acid analysis by GC, GC/MS and proton NMR showed that the chloroform soluble fraction composed of $25.3\%$ of DHA (dorosahexaenoic acid) and $15.3\%$ of IPA (eicosapentaenoic acid) as the hemolytic components.

Major gene identification for SREBPs and FABP4 gene which are associated with fatty acid composition of Korean cattle (한우의 지방산 조성에 영향을 미치는 SREBPs와 FABP4의 유전자 조합 규명)

  • Disease of human and economic traits of livestocks are affected a lot by gene combination effect rather than a single gene effect. In this study, we used SNPHarvester method that supplement existing method in order to investigate the interaction of these genes. The used genes are SREBPs (g.3270+10274 C>T, g.13544 T>C) and FABP4 (g.2634+1018 A>T, g.2988 A>G, g.3690 G>A, g.3710 G>C, g.3977-325 T>C, g.4221 A>G) that are closely related to the fatty acid composition affecting the meatiness of Korean cattle. The economic traits which are used are oleic acid (C18:1), monounsaturated fatty acid (MUFA), marbling score (MS). First, we have utilized the SNPHarvester method in order to find excellent gene combination, and then used the multifactor dimensionality reduction method in order to identify excellent genotype in gene combination.

Self Charging Sulfanilic Acid Azocromotrop/Reduced Graphene Oxide Decorated Nickel Oxide/Iron Oxide Solar Supercapacitor for Energy Storage Application

  • A self-charging supercapacitor is constructed through simple integration of the energy storage and photo exited materials at the photo electrode. The large band gap of $NiO/Fe_3O_4$ heterostructure generates photo electron at the photo electrode and store the charges through redox mechanism at the counter electrode. Sulfanilic acid azocromotrop/reduced graphene oxide layer at the photo electrode trapped the photo generated hole and store the charge by forming double layer. The solar supercapacitor device is charged within 400 s up to 0.5 V and exhibited a high specific capacitance of ~908 F/g against 1.5 A/g load. The solar illuminated supercapacitor shows a high energy and power density of 33.4 Wh/kg and 385 W/kg along with a very low relaxation time of ~15 ms ensuring the utility of the self charging device in the various field of energy storage and optoelectronic application.
