Pseudomonas tolaasii 박테리오파지에 특이적인 다클론항체 형성 및 이를 이용한 파지 교차 반응성 (Pseudomonas tolaasii bacteriophage-specific polyclonal antibody formation and its cross reactivity to various phages)

  • Pseudomonas tolaasii는 느타리버섯에 갈반병을 일으키는 병원균주로, 다양한 변이균주들을 분리하여 $P1{\alpha}$$P1{\beta}$, $P1{\gamma}$ 세 가지 소그룹으로 분류하였다. 각 그룹별 균주들에 특이적인 박테리오파지를 이용한 파지테라피는 갈반병 방제에 매우 성공적이었다. 본 연구에서는, 박테리오파지들의 특성을 구명하기 위하여 소그룹별 대표균주을 이용하여 파지를 분리하였고, 이들의 다클론항체를 제작하여 파지들 사이에 유연관계를 조사하였다. 파지 준비물은 $10^{10}pfu/mL$ 이상으로 토끼의 다리 근육에 주사하였고, 3회의 반복주사에 의해 다클론항체가 얻어졌다. 파지 ${\phi}6264$에 대한 항체의 역가는 $2{\times}10^7Ab/mL$ 이상, 파지 ${\phi}HK2$에 대해서는 $1{\times}10^6Ab/mL$, 파지 ${\phi}HK19$${\phi}HK23$에 대해서는 $1{\times}10^7Ab/mL$ 이상이었다. 항체와 이에 특이적인 파지 사이에는 매우 높은 반응특이성이 있었고, 소그룹이 다른 파지의 항체와 파지 사이에도 일부 교차반응성을 확인하였다. 파지 ${\phi}6264$에서 생성된 $Ab{\phi}6264$는 모든 $P1{\alpha}$ 소그룹의 파지들과 반응성을 보였으나, 파지 ${\phi}HK16$을 제외한 다른 소그룹의 파지들과는 반응하지 않았다. $P1{\gamma}$ 소그룹에서 생성된 $Ab{\phi}HK23$$P1{\beta}$ 소그룹의 모든 파지들을 불활성화시켜 가장 넓은 항체범위를 보였다. 항체와 파지를 이용한 숙주균과의 관계를 분석하였을 때, 16S rRNA 유전자 분석에 의한 숙주균의 근연관계와 항체를 이용한 숙주균의 파지들 사이의 구조적 근연관계는 상당히 차이가 있음을 확인하였다. 결론적으로, 박테리오파지의 숙주균 특이성과 항체를 이용해 측정한 파지의 껍질단백질 구조유사성 사이에는 약한 상관성을 보였다.

제주연안 갯녹음(백화) 지역의 해수에 분포하는 세균군의 분자생물학적 분석 (Molecular Analysis of Bacterial Communities Distributed in Sea Water of Whitening Areas of Jeju Coast)

  • 본 연구에서는 갯녹음(백화)현상이 세균생태계와 어떤 관련 이 있는 지에 대한 기초자료 및 정보를 얻기 위하여 갯녹음 현상이 일어난 제주도 성산과 강정지역 연안의 해수에 존재하는 세균군을 16S rRNA중폭기법을 이용하여 조사하였다. 강정지 역에서는 Alcanivorax, Paracoccus, Damselae, Pseudomonas, Rhodovulum, Silicibacter, Sulfitobacter, Roseobacter등 다양한 종류의 세균이 분포되어 있었으며, Alcanivorax가 20%의 빈도로 가장 많이 나타났다. 반면, 성산 지역에서는 Pseudomonas속 균주가 우점종으로서 존재하였으며, Pseudomonas tolaasii (혹은 Pseudomonas corrugata)와 유연관계가 가까운 세균은 44%, Pseudamonas mandeli와 가까운 세균이 24%, Verrucomicrobiales와 가까운 세균은 4%,기타 동정되지 알은 세균은 세 group으로 구분되었으며, 각각 8%,8%, 12%를 차지하여, 두곳에 분포되어 있는 세균군상이 상당한 차이점 이 있음을 확인하였다. 갯녹음 지역인 강정과 성산 해수의 8월(표품 채집시기)의 수온은 $27^{\circ}C$~$27.4^{\circ}C$, 염분의 농도는 30.24-30.60%, pH는 8.23~8.36,용존산소량(DO)은 각각 7.20~7.28로, 두지역에서 매우 비슷한 것으로 조사된 바 있는 데, 이는 수온이나 염분의 농도, 또는 pH보다는 다른 원인에 의하여 두 갯녹음 지역에 분포하는 세균군의 차이를 가져왔음을 제시해 주었다.

토양으로부터 단리한 Bacillus subtilis NSC 유래 Cellulase의 특성 규명 (Characterization of Cellulase from Bacillus subtilis NSC Isolated from Soil)

  • 삼림 토양 시료를 이용하여 cellulase 생산균주를 단리한 결과, Carboxymethylcellulose (CM-cellulose)를 기질로 첨가한 고체배지상에서 명확한 활성환을 형성하는 총 6종류의 cellulase 생산균주를 단리하여, 단리한 균주 유래의 16S rDNA유전자 염기서열 분석을 통하여 균주 동정을 진행하였다. 그 결과 본 연구에서 단리된 균주는 Bacillus subtilis 4종류, Bacillus amyloliquefaciens 1종류, Bacillus cereus 1종류로 동정되었다. 이 중에서 CM-cellulose에 대한 가장 높은 cellulase 활성을 보이는 Bacillus subtilis를 선정하여 Bacillus subtilis NSC로 명명하였다. Bacillus subtilis NSC 유래 cellulase는 균주 배양 36~48시간에서 가장 높은 cellulase 생산성을 보였으며, 최적 pH 및 온도를 검토한 결과, 본 균주 유래 cellulase는 pH 5.0과 $40^{\circ}C$에서 가장 높은 효소 활성을 나타내었다. 그리고, pH 4.0~5.0 조건에서 30분간 효소처리를 하여도 효소활성의 감소가 없었으며, $40^{\circ}C$까지는 30분간의 열처리에도 효소활성의 저하 없이 안정한 특성을 보였다. CM-cellulose, Alkali swollen cellulose, Sigmacell-cellulose, Alpha-cellulose, 그리고 Avicel을 기질로 기질 특이성을 한 결과 CM-cellulose에 대하여 가장 높은 효소 활성을 나타내었으며, cellulose 결정구조를 보유하고 있지않는 CM-cellulose와 Alkali swollen cellulose에 대해서는 명확한 효소활성을 보였다. 하지만, cellulose 결정구조를 보유하고 있는 Sigmacell Cellulose, Alpha-cellulose, 그리고 Avicel 기질에 대해서는 CM-cellulose 활성의 각각 8%, 8%, 그리고 4%의 매우 낮은 효소활성을 나타내었다. Bacillus subtilis NSC 유래 cellulase의 0.26 U/ml 조효소액과 0.52 U/ml 조효소액을 이용하여 CM-cellulose의 분해 특성을 검토하였을 때 두 조효소액 모두 반응 CM-cellulose에 대해 반응 120분 후에 0.43 U/ml와 0.76 U/ml의 효소 활성을 나타내었다.

Comparison of cecal microbiota composition in hybrid pigs from two separate three-way crosses

  • Objective: The intestinal microbiota plays an important role in host physiology, metabolism, immunity, and behavior. And host genetics could influence the gut microbiota of hybrid animals. The three-way cross model is commonly utilized in commercial pig production; however, the use of this model to analyse the gut microbial composition is rarely reported. Methods: Two three-way hybrid pigs were selected, with Saba pigs as the starting maternal pig: Duroc× (Berkshire×Saba) (DBS) pig, Berkshire×(Duroc×Saba) (BDS) pig. One hundred pigs of each model were reared from 35 days (d) to 210 d. The body weight or feed consumption of all pigs were recorded and their feed/gain (F/G) ratio was calculated. On day 210, 10 pigs from each three-way cross were selected for slaughter, and cecal chyme samples were collected for 16S rRNA gene sequencing. Results: The final body weight (FBW) and average daily gain (ADG) of DBS pigs were significantly higher than those of BDS pigs (p<0.05), while the F/G ratios of DBS pigs were significantly lower than those of BDS pigs (p<0.05). The dominant phyla in DBS and BDS pigs were Bacteroidetes (55.23% vs 59%, respectively) and Firmicutes (36.65% vs 34.86%, respectively) (p>0.05). At the genus level, the abundance of Prevotella, Roseburia, and Anaerovibrio in DBS pigs was significantly lower than in BDS pigs (p<0.01). The abundance of Eubacterium, Clostridium XI, Bacteroides, Methanomassiliicoccus, and Parabacteroides in DBS pigs was significantly higher than in BDS pigs (p<0.05). The FBWs and ADGs were positively correlated with Bacteroides, ClostridiumXI, and Parabacteroides but negatively correlated with the Prevotella, Prevotella/Bacteroides (P/B) ratio, Roseburia, and Anaerovibrio. Conclusion: These results indicated that host genetics affect the cecal microbiota composition and the porcine gut microbiota is associated with growth performance, thereby suggesting that gut microbiota composition may be a useful biomarker in porcine genetics and breeding.

Report of seven unrecorded bacterial species in Korea belonging to the family Acetobacteraceae

  • Several genera belonging to the family Acetobacteraceae were generally considered to be acetic acid bacteria (AAB) which produce acetic acid from alcohols using an oxidation pathway. Some species of the family Acetobacteraceae have been of interest to the industry due to their capability to produce vinegar. In 2018-2020, several bacterial strains were isolated from plants, fruits and vinegar in various regions of the Republic of Korea. Based on the 16S rRNA gene sequences, seven species classified into the family Acetobacteraceae were found to be ones unrecorded in the Republic of Korea, including two Asaia species, one Gluconacetobacter species, three Gluconobacter species and one Komagataeibacter species. As a result, we report Asaia lannensis AF11C3 (=KACC 22050) isolated from plant Chrysanthemum zawadskii, Asaia platycodi AF15C2 (=KACC 22051) isolated from plant Isodon inflexus, Gluconacetobacter liquefaciens C23-3 (=KACC 22064) isolated from fruit of Pyrus pyrifolia, Gluconobacter cerinus BGF2-R2 (=KACC 22053) isolated from fruit of Prunus armeniaca, Gluconobacter kondonii FR39A4 (=KACC 22388) isolated from persimmon fruit, Gluconobacter thailandicus FR36C4 (=KACC 22057) isolated from fruit of Pseudocydonia sinensis and Komagataeibacter melaceti SPV-2 (=KACC 22058) isolated from potato vinegar.

Properties of a Fibrinolytic Enzyme Secreted by Bacillus amyloliquefaciens RSB34, Isolated from Doenjang

  • Nine bacilli with fibrinolytic activities were isolated from doenjang, a traditional Korean fermented soy food. Among them, RSB34 showed the strongest activity and was identified as Bacillus amyloliquefaciens by 16S rRNA and recA gene sequencing. During growth on LB up to 96 h, RSB34 showed the highest fibrinolytic activity ($83.23mU/{\mu}l$) at 48 h. Three bands of 23, 27, and 42 kDa in size were observed when the culture supernatant was analyzed by SDS-PAGE and 27 and 42 kDa bands by fibrin zymography. The gene encoding the 27 kDa fibrinolytic enzyme AprE34 was cloned by PCR. BLAST analyses confirmed that the gene was a homolog to genes encoding AprE-type proteases. aprE34 was overexpressed in Escherichia coli BL21(DE3) using pET26b(+). Recombinant AprE34 was purified and examined for its properties. The $K_m$ and $V_{max}$ values of recombinant AprE34 were $0.131{\pm}0.026mM$ and $16.551{\pm}0.316{\mu}M/l/min$, respectively, when measured using an artificial substrate, N-succinyl-ala-ala-pro-phe-p-nitroanilide. aprE34 was overexpressed in B. subtilis WB600 using pHY300PLK. B. subtilis transformants harboring pHYRSB34 (pHY300PLK with aprE34) showed higher fibrinolytic activity than B. amyloliquefaciens RSB34.

Coproduction of Thermostable Amylase and ${\beta}$-Galactosidase Enzymes by Geobacillus stearothermophilus SAB-40: Application of Plackett-Burman Design to Evaluate Culture Requirements Affecting Enzyme Production

  • A locally isolated thermophile, Geobacillus sp. SAB-40, producing thermostable extracellular amylase constitutively and an induced intracellular ${\beta}$-galactosidase was characterized and identified based on 16S rRNA sequencing. A phylogenetic analysis then revealed its closeness to Geobacillus stearothermophilus. To evaluate the effect of the culture conditions on the coproduction of both enzymes by G stearothermophilus SAB-40, a Plackett-Burman fractional factorial design was applied to determine the impact of twenty variables. Among the tested variables, $CaCI_2$, the incubation time, $MgSO_4{\cdot}7H_2O$, and tryptone were found to be the most significant for encouraging amylase production. Lactose was found to promote ${\beta}$-galactosidase production, whereas starch had a significantly negative effect on lactase production. Based on a statistical analysis, a preoptimized medium attained the maximum production of amylase and ${\beta}$-galactosidase at 23.29 U/ml/ min and 12,958 U/mg biomass, respectively, which was 3-and 2-fold higher than the yield of amylase and lactase obtained with the basal medium, respectively.

Biosurfactant Production from Novel Air Isolate NITT6L: Screening, Characterization and Optimization of Media

  • In this paper, an air isolate (NITT6L) has been screened based on hemolytic activity, emulsification activity, drop collapsing test, and oil displacement test, as well as lipase activity. It was found that strain NITT6L was able to reduce the surface tension of the medium from 61.5 to 39.83 mN/m and could form stable emulsions with tested vegetable oils. Morphological, biochemical, 16S rRNA sequencing analyses, and fatty acid methyl ester analysis using gas chromatography confirmed that the air isolate under study was Pseudomonas aeruginosa. Characterization of the biosurfactant using agar double diffusion assay revealed that the biosurfactant was anionic in nature, and CTAB-methylene blue assay and Molisch test revealed its glycolipid nature. The FT-IR spectrum confirmed that the crude biosurfactant was a rhamnolipid. Using unoptimized medium containing sucrose as the carbon source, the isolate was found to produce 0.3 mg/ml of rhamnolipid in batch cultivation (shake flask) at $37^{\circ}C$ and pH 7. Optimization of the medium components was carried out using design of experiments and the yield of rhamnolipid has been enhanced to 4.6 mg/ml in 72 h of fermentation.

In-Depth Characterization of Wastewater Bacterial Community in Response to Algal Growth Using Pyrosequencing

  • Microalgae have been regarded as a natural resource for sustainable materials and fuels, as well as for removal of nutrients and micropollutants from wastewater, and their interaction with bacteria in wastewater is a critical factor to consider because of the microbial diversity and complexity in a variety of wastewater conditions. Despite their importance, very little is known about the ecological interactions between algae and bacteria in a wastewater environment. In this study, we characterized the wastewater bacterial community in response to the growth of a Selenastrum gracile UTEX 325 population in a real municipal wastewater environment. The Roche 454 GS-FLX Titanium pyrosequencing technique was used for indepth analysis of amplicons of 16S rRNA genes from different conditions in each reactor, with and without the algal population. The algal growth reduced the bacterial diversity and affected the bacterial community structure in the wastewater. The following in-depth analysis of the deep-sequenced amplicons showed that the algal growth selectively stimulated Sphingobacteria class members, especially the Sediminibacterium genus population, in the municipal wastewater environment.

Isolation of bacteria capable of removing 2-methylisoborneol and effect of cometabolism carbon on biodegradation

  • 2-Methylisoborneol (2-MIB) is one of typical odorants in potable water sources, which is hardly removed by conventional water treatment process. In this study, three strains capable of removing 2-MIB singly from drinking water were isolated from activated carbon of sand filter. They were identified to be Shinella zoogloeoides, Bacillus idriensis and Chitinophagaceae bacterium based on 16S rRNA gene sequence analysis. In mineral salts medium without external carbon source, removal efficiencies of $20{\mu}g/L$ 2-MIB in three days were 23.3%, 32.9% and 17.0% for Shinella zoogloeoides, Bacillus idriensis and Chitinophagaceae bacterium, respectively. The biodegradation of 2-MIB was significantly improved with the presence of cometabolism carbon(glycerol, glucose, etc.). In the period of 20 days, Bacillus idriensis can remove 2 mg/L MIB to $368.2{\mu}g/L$ and $315.4{\mu}g/L$ in mineral salts medium without and with glycerol respectively. The removal of 2-MIB by Bacillus idriensis was from 2 mg/L to $958.4{\mu}g/L$ in Xiba river samples on 15 days.
