To isolate mutanse-producing bacteria, soil samples were collected from several areas in chonnam Province, South Korea. A total of 70 strains of actinomycetes were isolated from the soil samples. All isolated actinomycetes were inoculated on mutanase screening media to identify new bacterial strains producing mutanase activity. One strain in particular exhibited a strong mutanase-producing activity, and was identified as Microbispora rosea based on its morphological, cultural, and physiological characteristics, and also by 16S rDNA sequences.
Two specimens of Chromis albicauda were collected from Jeju Island, Korea. The species is most similar to C. analis, but is distinguishable by the presence of a white caudal fin when alive, 4-5 scale rows on the anterior portion of the preorbital, and a blackish blotch around anal opening. The results of a molecular analysis based on mitochondrial DNA 16S rRNA sequences confirm that the two individuals are not C. analis but C. albicauda as the genetic distance between the two species is 0.044. A comparison of the two individuals of C. albicauda with yellow chromis shows that none of the previously reported yellow chromis is C. analis, but rather they are C. albicauda. Herein, we suggest the new Korean name "Huin-ggo-ri-no-rang-ja-ri-dom" for C. albicauda.
We isolated and identified three protease-producing bacteria that had inhabited the region around the Korean Arctic Research Station Dasan located at Ny-Alesund, Svalbard, Norway $(79^{\circ}N,\;12^{\circ}E)$. Biofilms were collected from the surface of a floating pier and from dead brown algae in a tide pool near the seashore. The biofilm samples were transported to the Korea Polar Research Institute (KOPRI) under frozen conditions, diluted in sterilized seawater, and cultured on Zobell agar plates with 1% skim milk at $10^{\circ}C$. Three clear zone forming colonies were selected as protease-producing bacteria. Phylogenetic analysis based on 16S rDNA sequences showed that these three stains shared high sequence similarities with Pseudoalteromonas elyakovii, Exiguobacterium oxidotofewm Pseudomonas jessenii, respectively. We expect these Arctic bacteria may be used to develop new varieties of protease that are active at low temperatures.
The present study aimed to investigate the microbial diversity in Gouda cheese within the four months of ripening, via next-generation sequencing (NGS). Lactococcus (96.03%), and Leuconostoc (3.83%), used as starter cultures, constituted the majority of bacteria upon 454 pyrosequencing based on 16S rDNA sequences. However, no drastic differences were observed among other populations between the center and the surface portions of Gouda cheese during ripening. Although the proportion of subdominant species was <1%, slight differences in bacterial populations were observed in both the center and the surface portions. Taken together, our results suggest that environmental and processing variables of cheese manufacturing including pasteurization, starter, ripening conditions are important factors influencing the bacterial diversity in cheese and they can be used to alter nutrient profiles and metabolism and the flavor during ripening.
Two specimens of Chlorophthalmus nigromarginatus Kamohara, 1953 (51.02 mm, 66.04 mm in standard length) belonging to the family Chlorophthalmidae were collected on February 8, 2022 in the eastern Jeju-do Island, Korea using a bottom trawl. The species has the following morphological characteristics: snout length almost the same as orbit diameter, bump absent on predorsal, caudal fin with posterior black margin. C. nigromarginatus is morphologically very similar to C. acutifrons but can be distinguished by caudal fin with black margin (C. acutifrons has no black margin). Molecular analyses showed that our specimens almost matched with C. nigromarginatus according to mtDNA 16S rRNA sequences (99.8%). Therefore, we propose a new Korean name for C. nigromarginatus, "Geom-eun-kko-ri-pa-rang-nun-mae-tung-i".
A commensal thermophile, Symbiobacterium toebii, isolated from hay compost (toebii) in Korea commensally interacted with a thermophilic Geobacillus toebii sp. nov., which was a new species within the genus Geobacillus on the basis of the phenotypic traits and molecular systematic data. S. toebii required the crude extracts and/or culture supernatant of the Geobacillus toebii for axenic growth and could grow on the temperature between 45 and $70^{\circ}C$ (optimum: $60^{\circ}C$; 2.4 h doubling time) and pH 6.0 and 9.0 (optimum: pH 7.5). The G+C content of the genomic DNA was $65 mol\%$, and the major quinones were MK-6 and MK-7. A phylogenetic analysis of its 16S rDNA sequence indicated that Symbiobacterium toebii was closely related with solely reported Symbiobacterium thermophilum. The presence of the commensal thermophile 16S rDNA and accumulation of indole in all the enriched cultures indicate that Symbiobacterium toebii is widely distributed in the various soils. The genome of S. toebii constituted a circular chromosome of 3,280,275 base pairs and there was not an extra-chromosomal element (ECE). It contained about 4,107 predicted coding sequences. Of these protein coding genes, about $45.6\%$ was encoded well-known proteins and annotated the functional assignment of 1,874 open reading frames (ORFs), and the rest predicted to have unknown functions. The genes encoding thermostable tyrosine phenol-lyase and tryptophan indole-lyase were cloned from the genomic DNA of S. toebii and the enzymatic production of L-tyrosine and L-tryptophan was carried out with two thermostable enzymes overexpressed in recombinant E. coli.
우리나라의 전통 발효 된장으로부터 균체외 효소로 mannanase를 생산하는 세균 2주가 분리되었다. 분리균 WL-6과 WL-11은 형태적 특성, 생화학적 성질 및 16S rDNA의 염기서열에 따라 Bacillus subtilis로 확인되었다. 이들 두 균주로부터 각각 mannanase 유전자를 대장균에 클로닝하여 염기서열을 결정한 결과 mannanase 유전자는 362 아미노산으로 구성된 단백질을 코드하며 1,086 뉴클레오티드로 동일하게 이루어졌다. WL-6과 WL-11 mannanase (Man6, Man11)의 아미노산 잔기 배열은 서로 8개 잔기가 다르며 GH family 26에 속하는 B. subtilis의 mannanases와 매우 상동성이 높았다. Man6과 Man11의 아미노 말단의 26개 아미노 잔기가 signal peptide로 예측되었다. 재조합 대장균로부터 각각 생산된 Man6과 Man11은 94~95% 정도가 균체내에 존재하였고, mannotriose, mannotetraose, mannopentaose, mannohexaose와 같은 만노올리고당과 locust bean gum을 유사하게 분해하여 주된 반응산물로 mannobiose와 mannotriose를 생성하였다. Man6는 55℃와 pH 6.0, Man11은 60℃와 pH 5.5에서 각각 최대 반응활성을 보였으며, Man11이 Man6에 비해 열안정성이 높았다.

We isolated and identifed culturable Arctic bacteria that have inhabited around Korean Arctic Research Station Dasan located at Ny-Alsund, Svalbard, Norway $(79^{\circ}N,\;12^{\circ}E)$. The pure colonies were inoculated into nutrient liquid media, genomic DNA was extracted, and phylogenetic analysis was performed on the basis of 16S rDNA sequences. Out of total 227 strains, 198 strains were overlapped or unidentified, and 43 bacteria were finally identified: 31 strains belonged to Pseudomonas, 7 strains Arthrobacter, two Flavobacterium sp., an Achromobacter sp., a Pedobacter sp., and a Psychrobacter sp. For isolation of diverse bacteria, we need more effective transport method than 3M petri-films, which were used for convenience of transportation that was restricted by volume. We also need to use other culture media than nutrient media. We expect these Arctic bacteria can be used for screening to develop new antibiotics or industrial enzymes that are active at low temperature.
A membrane-based oligonucleotide array was used to detect predominant bacterial species in human fecal samples. Digoxygenin-labeled 16S rDNA probes were generated by PCR from DNA that had been extracted from fecal samples or slurries. These probes were hybridized to an array of 120 oligonucleotides with sequences specific for 40 different bacterial species commonly found in human feces, followed by color development using an alkaline phosphatase-conjugated antibody and NBT /BCIP. Twenty of the species were detected by this method, but E. coli, which was present at $\~$1 $\times 10$^5$ CFU per gram feces, was not detected. To improve the sensitivity of this assay, reverse transcriptase-PCR was used to generate probes from RNA extracted from fecal cultures. Coupled with a chemiluminescence detection method, this approach lowered the detection limit for E. coli from $\~1$$\times 10$^6$ to ${\leq}$ 1 $\times 10$^5$ These results indicate that the membrane-array method with reverse transcriptase-PCR and chemiluminescence detection can simultaneously identify bacterial species present in fecal samples at cell concentrations as low as${\leq}$ 1 $\times 10$^5$ CFU per gram.

Cyanobium is a genus of picoprokaryotic cyanophytes, which includes species worldwide. The present study investigated the morphology, ultrastructure, and molecular phylogeny of the unrecorded genus Cyanobium Rippka & Cohen-Bazire 1983 and species Cyanobium gracile Rippka & Cohen-Bazire 1983. A C. gracile culture from a freshwater sample collected from the Adongji pond was established by single-cell isolation. Morphological data were analyzed using light and transmission electron microscopy. C. gracile lives as solitary cells without gelatinous envelopes and is ovate, oval, or shortly rod-shaped. Thylakoids are laid along the cell walls, with three thylakoid membranes parallel to each other. Nucleoplasm was observed in the center of the cell. Molecular phylogeny performed with data from 16S small subunit ribosomal DNA gene (SSU rDNA) sequences showed that the three strains of C. gracile, including the type strain (PCC6307) and a newly recorded strain (Adong101619), formed a distinct clade with a high supporting value (maximum-likelihood=100, pp=1.00). Based on morphology and molecular data, we report the newly recorded C. gracile in Korea.
