Sanitization of Commercial Powdered Products Using Gamma Irradiation (감마선 조사를 이용한 시판 분말원료의 위생화)

  • Microbiological populations and the sterility of commercial powdered products treated with gamma irradiation at 0~10 kGy were investigated before using them as ingredients for a non-cooked Saengsik product. We evaluated a total of 14 powdered products: 8 powdered cereals, 3 powdered tubers, and 3 powdered leafy vegetables. The total numbers of bacterial populations in non-irradiated powdered cereals, tubers, and leafy vegetables were 2.7~6.9, 5.6~6.0, and $5.3{\sim}6.8\;log\; CFU{\cdot}g^{-1}$, respectively. Moreover, coliform bacteria were not indicated in adlay, millet, germinated brown rice, soybean, and mulberry leaves powder within detection limit ($2.0\;log\; CFU{\cdot}g^{-1}$). The number of Bacillus cereus exceeded $3.0\;log\; CFU{\cdot}g^{-1}$ (the maximum limit for Saengsik products) in all samples, excluding perilla seeds, buckwheat, barley, oat, potato, and Jerusalem artichoke powder. However, a dose of 6 kGy of gamma irradiation reduced the microbiological populations in all samples, and all the powdered products met the microbial requirements for Saengsik products. Futhermore, it was confirmed that all microorganisms in the 9 powdered products, except fermented brown rice, sweet potatoes, and 3 leafy vegetables, were sterilized by 10 kGy of gamma irradiation.

  • In order to select ${\gamma}-Linolenic$ acid (GLA)-producing fungi, a total of forty-four strains of 4 genera such as Phytophthora, Pythium, Mucor and Rhizopus were obtained from Koran Agricultural Culture Collection (KACC) and then analysed by using GC-FID and GC-MS. GLA was detected on 39 fungal strains, and the highest rate of GLA was found as 24.8% of total fatty acids on Mucor hiemalis f. sp. hiemalis KACC 40264. Total GLA content of Zygomycota was comparatively high - Mucor (14.2%) and Rhizopus (14.3%), whereas that of Oomycetes was low - Phytophthora (3.3%) and Pythium (3.0%). Moreover, total fatty acids of the Zygomycota fungi such as Mucor (15.4 mg/100 ml) and Rhizopus (7.1 mg/100 ml) were higher compared with the Oomycetes such as Phytophthora (2.6 mg/100 ml) and Pythium (4.5 mg/100 ml). Thus, two genera such as Mucor and Rhizopus have higher potential as an useful microbial resource. The total fatty acid content varies even within the strains of the same genus e.g. Mucor. M. blumbeus KACC 40935 showed the highest values on productivity (18.2%) of GLA and total fatty acid contents (50.8 mg/100 ml liquid medium).

Investigation of serum protein electrophoresis fractions analysis in Gyeongju DongGyeong dogs (경주지방의 무미 또는 단미 형태의 개(경주개 동경이)의 혈청 단백질의 분획에 대환 실태조사)

  • Serum proteins of Korean native dogs (Gyeongju DongGyeong dogs) was analyzed by capillary electrophoresis system. Electrophoretic patterns showed six fractions. Total serum protein and ratio of albumin to globulin were $5.99{\pm}0.83$ (g/dl) and $1.41{\pm}0.29$ (g/dl) in DongGyeong dogs. Relative percentages of total serum albumin and $\alpha-1$, $\alpha-2$, $\beta-1$, $\beta-2$, $\gamma$-globulin fraction were $57.94{\pm}5.43$, $3.15{\pm}2.30$, $7.49{\pm}4.09$, $9.43{\pm}3.50$, $7.63{\pm}5.70$, and $14.36{\pm}7.63$, respectively. It was observed that $\beta$-globulin was higher than other fractions. The most striking alternation with age was founded in the $\gamma$-fractions. Also, it was observed that ratios of albumin to globulin in DongGyeong dogs were higher than on other dogs.

Gamma irradiation and subsequent storage reduce patulin content in apple juice

  • Patulin has been reported as a risk factor in various foods, especially apple juice. This study monitored residual patulin and polyphenolic content in apple juice during post-irradiation storage conditions. Response surface methodology (RSM) was applied to monitor the changes in dependent variables (Yn, patulin, and polyphenolics) as affected by independent variables, such as storage temperature (Xi, -20℃ to 20℃), irradiation dose (Xii, 0-2 kGy), and storage period (Xiii, 0-20 days), which were based on a central composite design. The predicted peak point resulted in the lowest residual patulin content of 58.42 ppb with the corresponding independent parameter conditions, such as 18.19℃ of storage temperature, 1.24 kGy of irradiation dose, and 13.42 days of storage period. The residual patulin content of 58.42 ppb is the minimum desirable level, representing a 91% reduction compared to the non-irradiated control (675.00 ppb). A maximum polyphenolics content (11.98 mg/g) was obtained under the predicted maximum conditions of 14.40℃, 0.78 kGy, and 3.4 days. The most influential parameter in reducing residual patulin content while maintaining polyphenolic content in apple juice was irradiation dose (p<0.01), which showed potential to be applied in controlling the patulin levels in apple juice.

Gamma-Radiation Sensitivity of Pathogenic Bacteria in Beef (우육에 오염시킨 병원세균의 방사선 감수성)

  • The gamma-radiation sensitivity of eight kinds of pathogenic bacteria related to beef was investigated in frozen cells $(-18^{\circ}C)$ with 0.1 M phosphate buffer and inoculated cells in beef. In frozen cells, D10 values of pathogenic bacteria related to beef were $0.07{\sim}0.69$ kGy, and inactivation factors were $2.90{\sim}42.86$ at the radiation doses of $2{\sim}3$ kGy. Beef was inoculated with Escherichia coli, Pseudomonas aeruginosa, Salmonella typhimurium, Enterobacter aerogenes, Vibrio parahaemolyticus, Staphylococcus aureus, Listeria monocytogenes, and Aeromonas hydrophila. Inoculated beef samples were packaged in air and irradiated at 0.005 to 3.0 kGy. Ninety percent of the viable pathogenic bacteria in beef was eliminated by doses of $0.1{\sim}0.61$ kGy at room temperature, and the inactivation factors were $3.28{\sim}30.0$ kGy at the radiation doses of $2{\sim}3$ kGy. Therefore, irradiation is considered to be an effective method to control pathogenic bacteria in beef.

Effect of Teminalia chebula Extract on Liver in Rat (가자(Terminalia chebula) 추출물이 흰쥐의 간장 활성에 미치는 영향)

  • In this study, we investigated the effect of Teminalia Chebula (TC) water extract on liver in Rat. Treatment of TC water extract was orally administered 200, 300 mg/kg daily for one week and two weeks. The clinical parameters of serum, values of AST, ALT showed significantly higher than in normal group. Xanthine oxidase and aldehyde oxidase activities were significantly increased comparison with normal group. Microsomal enzymes, aminopyrine N-demethylase and aniline hydroxylase were not affected. Water extract of TC also increased hepatic rnalondialdehyde formation and reduced glutathione content. We also found that water extract of TC decreased activities of glutathione S-transferase and glutathione reductase but was not affected activities of $\gamma$-glutamylcysteine synthetase Thus, it seems that the water extract of TC induced decrease of oxygen free radical scavenger, glutathione content by inhibition of glutathione reductase which may reform oxidized glutathione to reduced glutathione.

  • In order to know the kinds of liver function tests which are applicable to the hepatotoxicity experiment in Korean black goats, 0.3ml of carbon tetrachloride per kilogram body weight was injected intraruminally in 6 Korean black goats and 8 liver function tests were performed sequentially during the 14-day experimental course. Serum total bilirubin concentration, serum alkaline phosphatase activity and BUN concentrotion were suggested not to be applicable to the hepatotoxicity experiment in Korean black goats as they were not altered significantly during the 14-day experimental course. Indocyanine green T 1/2, Serum AST, SDH, gamma-GTP, and OCT activities were suggested to be applicable as sensitive indicators to the hepatotoxicity experiment in Korean black goats.

Effects of ionizing and ultraviolet radiation on microbial mutation and DNA damage (전리방사선 및 자외선의 미생물 돌연변이와 DNA 손상에 대한 영향)

  • Physical sterilization methods using ultraviolet radiation and ionizing radiation such as gamma ray and electron beam are applied in various industry fields due to disinfection effects and economic efficiency but may also cause microbial mutation. In this research, Salmonella enterica and Escherichia coli strains were treated with ionizing and ultraviolet radiation and their survival rate, mutation rate, and DNA damage were studied to evaluate the genetic safety. The survival rate of the strains decreased drastically as the irradiation dose of ultraviolet ray, gamma ray, and electron beam increased, and over 90% of the strain was exterminated at a dosage of $0.40{\sim}25.06mJ/cm^3$, 0.11~0.22 kGy, 0.14~0.53 kGy respectively. In SOS / umu-test, genotoxicity causing DNA damage was identified in all samples. In Ames test, back-mutation rate increased to $3.82{\times}10^{-4}$ and $9.84{\times}10^{-6}$ respectively when exposed to ultraviolet ray and gamma ray. At exposure to ultraviolet ray, gamma ray, and electron beam with dosage of over 99.99% extinction rate of S. enterica TA100, back-mutation rate increased 347 times, 220 times, 0.6 times respectively to the spontaneous back-mutation rate. Rifampicin resistance mutation rate of E. coli CSH100 exposed to ultraviolet ray, gamma ray, and electron beam was $2.46{\times}10^{-6}$, $1.66{\times}10^{-6}$, $4.12{\times}10^{-7}$ respectively. Therefore, gamma radiation is effective in microorganism control from the perspective of disinfection and electron beam has the advantage of sterilizing with little DNA damage and bacterial mutation.

Structure of a Copper(Ⅱ) Hexaazamacrotricyclic Complex : (1,3,6,9,11,14-Hexaazatricyclo[12.2.1.16,9]octadecane)-copper(Ⅱ) Perchlorate

  • The crystal structure of (1,3,6,9,11,14-hexaazatricycol[12.2.1.$1^{6,9}$]octadecane)copper(Ⅱ) perchlorate, Cu($C_{12}H_{26}N_6$)$(ClO_4)_2$, has been determined by the X-ray diffraction methods. The crystal data are as follows: Mr=516.9, triclinic, ${\alpha}=8.572\;(2)$, b=8.499 (3), c=15.204 (3) ${\AA}$, ${\alpha}=80.42\;(5),\;{\beta}=73.57\;(3),\;{\gamma}=69.82\;(4)^{\circ},\;V=994.2\;{\AA}^3,\;D_C=1.726\;gcm^{-3}$, space group $P{\tilde{1}},\;Z=2,\;{\mu}=21.27\;cm^{-1}&, F(000)=534 and T=297 K. The structure was solved by direct methods and refined by full-matrix least-squares methods to and R value of 0.081 for 1608 observed reflections measured with graphite-mono-chromated Mo Ka radiation on a diffractometer. There are two independent complexes in the unit cell. The two copper ions lie at the special positions (1/2, 0, 0) and (0, 1/2, 1/2)and each complex possesses crystallographic center of symmetry. Each Cu ion is coordinated to four nitrogen donors if the hexaazamacrotricyclic ligand and weakly interacts with two oxygen atoms of the perchlorate ions to form a tetragonally distorted octahedral coordination geometry. The Cu_N (sec), Cu_N(tert) and Cu_O coordination distances are 1.985 (14), 2.055 (14) and 2.757 (13) ${\AA}$ for the complex A and 1.996 (10), 2.040 (11) and 2.660 (13) ${\AA}$ for the complex B, respectively. The macrocycles in the two independent cations assume a similar conformation with the average r.m.s. deviation of 0.073 ${\AA}$. Two 1,3-diazacyclopentane ring moieties of the hexaazamacrotricyclic ligand are placed oppositely and almost perpendicularly to the square coordination plane of the ruffled 14-membered macrocycle. The secondary N atoms are hydrogen-bonded to the perchlorate O atoms with distances of 3.017 (23) and 3.025 (19) ${\AA}$ for the complexes A and B, respectively.

Effect of Gamma-Irradiation on the Storage Stability and Quality of Polished Wheat (밀쌀의 저장성(貯藏性)과 품질(品質)에 미치는 감마선(線) 조사(照射)의 영향)

  • In order to study the effect of gamma-ray on the storage stability and quality of polished wheat, the grain was irradiated at $50{\sim}1000$ krad levels and quality assessment was made for three-month storage period. The results are summarized as follows: 1) Growth of molds was apparent at equilibrium moisture content above 14.8% of the grain, but their growth was retarded at 250 krad level. 2) Irradiation at 250 krad level had no adverse effect on fat acidity, reducing sugar content and color of the grain. 3) Irradiation of the grain caused the increase of degree of gelatinization and the decrease of viscosity. 4) Irradiation at 250 krad level brought about higher digestibility of the grain but had no effect on the rate of retrogradation.
