Naphthoquinone Analog-induced G1 Arrest is Mediated by cdc25A Inhibition and p53-independent p21 Induction in Human Hepatocarcinoma Cells

  • The naphthoquinone analog (2,3-dichloro-6,9-dihydroxy-1,4-naphtoquinone, NA) has an inhibitory effect on cdc25A protein phosphatase in vitro, which is responsible for G1/S transition during cell cycle. However, the exact mechanism inducing the growth inhibition is not understood. In this study, we investigated the regulatory mechanisms of growth arrest induced by NA, as a new potent inhibitor of cdc25A phosphatase, in human hepatocarcinoma SK-hep-1 cells. We found that NA induced the G1 arrest by perturbation of protein tyrosine dephosphorylation of Cdk2, which may be resulting from inhibition of cdc25A phosphatase. In addition, p21 was expressed in a p53-independent manner and participated in the NA-induced G1 arrest by inhibiting Cdk2 activity. Although the exact mechanism is not known, the p21 expression might be related to MAPK activation. From these results, we suggest that NA induces G1 arrest via inhibition of cdc25A and induction of p53-independent p21 expression in SK-Hep-1 cells.

Effect of Water Temperatures and Photoperiods on Gondal Degeneracy in Banded Catfish Pseudobagrus fulvidraco (동자개(Pseudobagrus fulvidraco)의 생식소 퇴화에 미치는 수온과 광주기의 영향)

  • To investigate the role of temperatures and photoperiods as environmental cues regulating reproductive rhythm in Pseudobagrus fulvidraco, rearing experiments were conducted using sveral rearing regimes conbined with photoperiods and water temperatures during gonadal degeneration periods. Gonadosomatic index (GSI) in control was $8.16{\pm}1.50%$, while in other experiment GSI levels in female were lower than that in the control. In case of experimental precinct of 9 light (L) and 15 L, GSI levels were decreased. But GSI level with $20^{\circ}C$ was no diffrence after 40 and 60 days. GSI level in male of control was $0.35{\pm}0.05%$. GSI under 9 L and $25^{\circ}C$ was similar to that in control, whereas its level in other experiments was lower than that in control. Testosterone (T) of female was $3.68{\pm}0.22$ ng/mL at experimental precinct. In case of 9 L and 15 L, concentration of T were lower than experimental precinct in all of water temperature. Estradiol-$17{\beta}$ ($E_2$) and $7{\alpha}$, $20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}OHP$) levels of female were $0.42{\pm}0.02$ and $0.83{\pm}0.01$ ng/mL at experimental precinct. $E_2$ levels of 9 and 15 L were higher than experimental precinct and $17{\alpha}20{\beta}OHP$ levels of 9 and 15 L were higher than experimental precinct. In case of T and 11-ketotestosterone levels were $0.69{\pm}0.11$ and $.62{\pm}0.03$ ng/mL in male. During the period of gonadal degeneration, gonadal maturation did not occur in any of the experimental regimes. However, comparatively high levels of $E_2$ observed at low temperature regimes ($20^{\circ}C$) regardless of photoperiods.

Isoflavanones from the Stem of Cassia siamea and Their Anti-tobacco Mosaic Virus Activities

  • Two new isoflavanones, (3R) 7,2',4'-trihydroxy-3'-methoxy-5-methoxycarbonyl-isoflavanone (1) and (3R) 7,2'-dihydroxy-3',4'-dimethoxy-5-methoxycarbonyl-isoflavanone (2), together with six known isoflavanones (3-8), were isolated from the stems of Cassia siamea. The structure of 1-8 was elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1, 2, 5-8 were evaluated for their anti-tobacco mosaic virus (Anti-TMV) activity. The results showed that compounds 1 and 6 showed potential anti-TMV activity with inhibition rates of 24.6% and 26.9%, respectively. Compounds 2, 5, 7, 8 also showed anti-TMV activity with inhibition rates in the range of 11.8-18.6%.

Sexual Maturity and Reproductive Cycle of Roughscale Sole Clidoderma asperrimum Cultured in Indoor Tank (실내 사육한 줄가자미(Clidoderma asperrimum)의 성 성숙과 생식주기)

  • The gonadosomatic index (GSI), sex steroid hormones and gonadal development of roughscale sole Clidoderma asperrimum cultured in indoor tank were investigated to evaluate its sexual maturation and reproductive cycle. The highest GSI values of female and male were $6.91{\pm}4.03$ (May) and $0.16{\pm}0.08$ (August), respectively. The reproductive cycle would be classified into four successive developmental stages: growing stage (December to February), maturation stage (March to April), ripe and spawning stage (May to June), recovery and resting stage (July to November). The highest plasma testosterone (T) and estradiol-$17{\beta}$ ($E_2$) levels of female were $259.4{\pm}76.8$ and $633.3{\pm}182.5$ pg/mL, respectively in May. Also $17{\alpha}$, $20{\beta}$-dihydroxy-4-pregen-3-one ($17{\alpha}$, $20{\beta}$-OHP) levels of female peaked in April before spawning season ($244.2{\pm}42.5$ pg/mL). The highest plasma testosterone (T) and 11-ketotestosterone levels of male were $231.0{\pm}46.0$ and $273.9{\pm}54.5$ pg/mL, respectively in April. But there was no significant difference in $17{\alpha}$, $20{\beta}$-OHP.

  • Twelve epoxy and hydroxydiosgenin derivatives (DI-1${\sim}$DI-12) were synthesized from diosgenin (25(R)-5-spirosten-3${\beta}$-ol). Diosgenin was epoxidized with m-chloroperoxybenzoic acid (mCPBA) to oxidize 25(R)-4${\alpha}$,5${\alpha}$-epoxyspirostane (DI-1). Diosgenin was reacted with DDQ to form 25(R)-1,4,6-spirostatrien-3-one (DI-2), which was treated with 30% H$_2$O$_2$ to give 25(R)-1${\alpha}$,2${\alpha}$-epoxy-4,6-spirostadien-3-one (DI-3) and treated with mCPBA to form 25(R)-6${\alpha}$,7${\alpha}$-epoxy-1,4-spirostadien-3-one (DI-7), respectively. DI-3 was reduced with NaBH$_4$ to afford 25(R) -1${\alpha}$,2 ${\alpha}$-epoxy-4,6-spirostadien-3${\beta}$-ol(DI-4) and reacted with Li metal in absolute ethanol to form 25(R)-2-ethoxy-1,4,6-spirostatrien-3-one (DI-5). DI-7 was reduced with NaBH$_4$ to produce 25(R)-3${\beta}$,7${\alpha}$-dihydroxy-4-spirostene (DI-8) and treated with Li metal in liquid ammonia to produce 25(R)-7${\alpha}$-hydroxy-4-spirosten-3-one (DI-9). DI-2 was reduced with NaBH$_4$ to form 25(R) -4,6-spirestadien-3${\beta}$-ol(DI-10), which was stirred with 30% H$_2$O$_2$ to synthesize 25(R)-4,6-spirostadien-3-one (DI-11) and reacted with mCPBA to give 25(R)-4${\beta}$,5${\beta}$ -epoxy-6-spirosten-3${\beta}$-ol (DI-12), respectively. The antinociceptive effects of synthesiz ed compounds were measured by hot plate method and compound DI-7 signifcantly exhibited antinociceptive effect. DI-2 decreased the serum triglyceride and total cholesterol levels in poloxamer P-407 injected rat.

Phytochemical Constituents from the Leaves of Soybean [Glycine max (L.) Merr.]

  • Phytochemicals study from the leaves of soybean [Glycine max (L.) Merr.], one of Korean edible plant materials were investigated through various chromatographic procedures. The methanolic leaves extracts of soybean yielded 16 phytochemicals, including 5 isoflavones 1-5, 3 flavones 6-8, 1 flavonol 9, 2 pterocarpans 10 and 11, 2 phenolic compounds 12 and 13, 2 phytosterols 14 and 15, and 1 sugar alcohol 16. The structures were fully characterized by analysis of physical and spectral data and were defined clearly as 4,5,7-trihydroxyisoflavone (1), 4,5,7-trihydroxyisoflavone-7-O-$\beta$-D-glucopyranoside (2), 4,7-dihydroxy-6-methoxyisoflavone (3), 4,7-dihydroxyisoflavone (4), 4,7-dihydroxyisoflavone-7-O-$\beta$-D-glucopyranoside (5), 5,7,4'-trihydroxyflavone (6), 3',4',5,7-tetrahydroxyflavone (7), 3',4',5-trihydroxyflavone-7-O-$\beta$-D-glucopyranoside (8), 3,4',5,7-tetrahydroxyflavonol (9), coumestrol (10), glyceofuran (11), 4-hydroxybenzoic acid (12), methyl-4-hydroxybenzoate (13), soyasapogenol B (14), stigmasterol (15), and D-mannitol (16), respectively. Among them, phytochemicals 7-16 were reported for the first time on the isolation and confirmation from the leaves of this species. These results suggest that the leaves extracts of soybean may possess possible health related benefits to human due to the isolated phytochemicals 1-16 which have been well known potential effects on various chronic diseases.

  • To maximize astaxanthin (3,3'-dihydroxy-$\beta\beta$'carotene-44'-dione) production by high density Haematococcus pluvialis cultures, various, media were examined Among tested media, \`Hong Kong Medium and Modified Bolds Basal Medium showed the best result for cell growth ( $2.0$\times$10^{ 6}$cells /mL) and for astaxanthin content per cell (9.7 mg astaxanthin mg/g cell), respectively, Maximum astasanthin concentration of 6.1mL was obtained at pH 7.5, $20^{\circ}C$~$25^{\circ}C$ Deficiencies of nitrogen source($NaNO_3$ and proteose-peptone) found to simulate astaxanthin formation Relatively low light inten- sity of $60\mu$E ($\m^2$s) was sutiable for vegetative cell growth while higher light intensity was required for higher astaxanthin accumulation.

Anti-Inflammatory and Enzyme Inhibitory Activities of Polyphenols from Peanut (Arachis hypogaea L.) Hull

  • Peanut hull as by-product has been discarded during peanut processing. However, peanut hull contains plenty of polyphenols that shows various physiological activities. The objectives of this study were to investigate anti-inflammatory and enzyme inhibitory activities of polyphenols from 'Sinpalkwang' peanut (Arachis hypogaea L.) hull. Compounds were isolated from methanol extracts of peanut hull by preparative-high performance liquid chromatography after identifying and quantifying polyphenols using Ultra performance liquid chromatography (UPLC) and UPLC-Quadrupole time-of-flight-mass spectrometry profiling. The structures of compounds were elucidated by one-dimensional [1H, 13C] nuclear magnetic resonance (NMR) and two-dimensional NMR (correlated spectroscopy, heteronuclear single quantum coherence and heteronuclear multiple bond correlation). Three compounds were identified as 5,7-dihydroxy-4H-chromen-4-one (peak 2), luteolin (peak 4) and eriodictyol (peak 5). Significant differences in inflammatory mediator such as nitric oxide (NO), interleukin-6 (IL-6) and interleukin-1β (IL-lβ) in lipopolysaccharide stimulated Raw 264.7 macrophages and in enzyme (xanthine oxidase [XO] and α-glucosidase [AG]) inhibitory activities were observed between three compounds (p < 0.05). Peak 5 treated Raw 264.7 macrophages showed lower content of NO (16.4 uM), IL-6 (7.0 ng/mL), and IL-1β (60.6 pg/mL) than peak 2 (NO: 28.3 uM, IL-6: 11.3 ng/mL, IL-1β: 66.9 pg/mL) and peak 4 (NO: 24.7 uM, IL-6: 9.3 ng/mL, IL-1β: 62.6 pg/mL). Peak 5 showed higher XO inhibitory activity (84.7%) and higher AG inhibitory activity (52.4%) than peak 2 (XO inhibitory activity: 45.4%, AG inhibitory activity: 21.6%) and peak 4 (XO inhibitory activity: 37.9%, AG inhibitory activity: 37.5%) at concentration of 0.5mg/mL. This study suggests that peanut hull could be a potential source of anti-inflammatory and physiological materials while creating new use of discarded peanut hull as by-products concomitantly.

Annual Reproductive Cycle and Changes in Plasma Levels of Sex Steroid Hormones of the Female Korean Dark Sleeper, Odontobutis platycephala (Iwata et Jeon) (동사리, Odontobutis platycephala (Iwata et Jeon) 암컷의 생식주기와 혈중 성스테로이드 호르몬의 변화)

  • To clarify annual reproductive cycle of Korean dark sleeper, Odontobutis platycephala (Iwata et Jeon), we examined the seasonal changes of gonadosomatic index (GSI), the proportional frequency of oocyte development stages in the ovary and the changes of sex steroid hormone levels in blood from December 1995 to November 1997. In July and August, GSI was 0.35 to 0.72 and most oocytes in the ovary were chromatin-nucleolus stage and perinucleolar stage (proportional frequency: $87\%\~96\%$). In September, GSI was 1.20 $\pm$ 0.12, some oocytes in the ovary were yolk vesifle stage (proportional frequency: $22.8\%$) and vitellogenic stage which appeared very rarely(proportional frequency: $2.2\%$). GSI increased gradually from October and reached 4.59± 0.61 to December. During this period, oocytes of vitellogenic stage increased slightly (proportional frequency in December: $22.1\%$). In January, GSI was 4.32 $\pm$ 0.72 but the proportional frequency of oocytes in vitellogenic stage increased (proportional frequency: $51.2\%$). from February, GSI was increased sharply and reached to 10.51 $\pm$ 1.04 in March, the highest value throughout the year and the proportional frequency of oocytes in vitellogenic stage also reached the highest levels (proportional frequency: $60\%$). From April, GSI was gradually decreased and fell down to 1.11 $\pm$ 0.35 in June. During this period, the proportional frequency of mature oocytes was the highest in April (proportional frequency of mature oocyte stage: $40\%$ in April, $12\%$ May, $5\%$ June) throughout the year, and atretic ovarian follicles were appeared. The blood level of estradiol-17$\beta$ ($E_2$), which stimulates the hepatic synthesis and secretion of vitellogenin, was $0.84{\pm}0.20\;ng/m{\ell}$ in August, and thereafter was not changed until December. from January, it increased sharply and reached the highest level of $ 2.85{\pm}0.35\;ng/m{\ell}$ in March throughout the year, but fell to $0.14{\pm}0.02\;ng/m{\ell}$ in July(P<0.05), 17$\alpha$-hydroxprogesterone(17$\alpha$-OHP) was the peak $13.37{\pm}0.52ng/m{\ell}$ in March, but no significant changes in other period(below $3ng/m{\ell}$, P<0.05). 17$\alpha$, 20$\beta$-dihydroxy-4-pregnen-3-one(17$\alpha$, 20$\beta$-P), which was known as the final maturation inducing hormone in teleost, was $0.74{\pm}0.09ng/m{\ell}$ in April and $0.54{\pm}0.07ng/m{\ell}$ in May, but no significant changes in other period (below $0.26\;ng/m{\ell}$, p<0.05). Taken together these results, the annual reproductive cycle of O. platycephala divided into 4 periods as follows: 1) ripe and spawning period from April to June, main spawning period was from April to May, 2) Resting period from July to August, 3) Growing period from September to December, 4) Maturing period from January to March. Moreover, It was showed that the changes of sex steroid hormone in blood played a important roles in the annual reproductive cycle of O. platycephala.

Testicular Development and Serum Levels of Gonadal Steroids Hormone during the Annual Reproductive Cycle of the Male Koran Dark Sleeper, Odontobutis platycephala (Iwata et Jeon) (동사리, Odontobutis platycephala (Iwata et jeon) 수컷의 생식주기에 따른 정소 발달과 혈중 생식소 스테로이드의 변화)

  • To clarify annual reproductive cycle of Koran dark sleeper, odontobutis platycephala, we examined the seasonal changes of gonadosomatic index(GSI), testicular development stages and sex steroid hormones in blood from December 1995 to November 1997. Testis was podlike shape from July to October, and tadpole-like shape from November because of its expanded posterior part. GSI was 0.14~0.18 from July to September and increased to $0.43{\pm}0.04$ in October and then was not changed significantly until February. GSI was reincreased to $0.52{\pm}0.09$ from March and then was kept at similer levels until May, but fell down to $0.28{\pm}0.05$ in June. As results of histological observation, testis was divided into 3 parts(anterior, boundary, posterior) in the development progress of germ cells. In July, the testis was composed of only spermatogonia without seminiferous tubules in most fishes. In the anterior part of testis, the ferquency of spermatogenesis stage seminiferous tubules appearing in August was more than 80% from September to December. decreased gradually from January to March and drastically in April, and then disappeared in June. The frequency of spermiogenesis stage seminiferous tubules appearing in December, increased gradually from January to March and drastically to 80% in April, and reached to 90% the highest levels of the year in June. Post-spawning stage seminiferous tubules did not appear throughout the year. The frequency of spermatogonia was 100% and 65% in July and August, and less than 20% in the rest period of the year. In the boundary part, the frequency of spermatogenesis stage seminiferous tubules appearing in August increased from September and reached to 82% in November, decreased from December, adn disappeared in March. The frequency of spermiogenesis stage seminiferous tubules appearing in November was less than 18% until February, and increased to 29%~57% from March to June. The frequency of post-spawning stage seminiferous tubules appeared 12%~25% only from March to June. The frequency of spermatogonia was 100% in July, decreased to 85% in August and 10% in November, and increased gradually from December to 50% in April, and decreased again from May to June. In the posterior part, seminiferous tubules with some seminiferous tubules increased drastically 80%~85% in August and September, decreased drastically from October to November and remained below 10% until February, and disappeared after March. The frequency of spermiogenesis stage seminiferous tubules appearing in August increased sharply from October and reached to 75% in November. decreased to 15% in December and no significant changes until March, and disappeared after April. The frequency of post-spawning stage seminiferous tubules appearing very early in November increased to 82% in December and 85%~95% until June. The frequency of spermatogonia was 100% in July, decreased drastically to 15% in August, disappeared from October to Mrch, but reappeared from April and kept at less than 10% until June. The blood level of testosterone (T) increrased gradually from August was $0.61{\pm}0.09 ng/m\ell$ in November, increrased drastically to $3.99{\pm}1.22 ng/m\ell$ in December and maintained at in similar level until March, and decreased to $0.25{\pm}0.14 ng/m{\ell} ~ 0.17{\pm}0.13ng/m{\ell}$ in April and May and no significant changes until July (P<0.05). The blood level of 17, 20 -dihydroxy-4-pregnen-3-one $ng/m{\ell}$in the rest of year without significant changes(P<0.05). Taken together these results, the germ cell development of testis progressed in the order of posterior, boundary, anterior part during annual reproductive cycle in Korean dark sleeper. The testicular cycle of Korean dark sleeper was as follows. The anterior part of testis : i.e. spermatogonial proliferation period (July), early maturation period (from August to November), mid maturation period (from December to March), late maturation period (from April to May) and functional maturation period (June) were elucidated. The boundary of testis, i.e. spermatogonial proliferation period (July), early maturation period (from August to October), mid maturation period (from November to February) and the coexistence period of late maturation, functional maturation and post-spawn (from March to June) were elucidated. The posterior of testis, i.e. spermatogonial proliferation period (July), mid maturation period (from August ot September), late maturation period (October), functional maturation period (November) and post-spawn period (from December to June) were elucidated. It was showed that the changes of sex steroid hormone in blood played a important roles in the annual reproductive cycle of Korean dark sleeper.
