Effect of Salts on the Extraction Characteristics of Succinic Acid by Predispersed Solvent Extraction

  • Predispersed solvent extraction (PDSE) of succinic acid with Tri-n-octylamine (TOA) dissolved in 1-octanol from aqueous solutions of 50 g/L succinic acid was examined. It was found that the equilibrium data in PDSE was equal to that in conventional solvent extraction in spite of the lack of mechanical mixing in PDSE. The influence of salts on succinic acid extraction and the stability of colloidal liquid aphrons (CLAs) were also investigated. Results indicated that in the presence of sodium chloride, less succinic acid was extracted by CLAs and the stability of CLAs decreased. However, the stability of CLAs was sufficient to make PDSE practically applicable to real fermentation broth, considering the concentration range of salts in the fermentation process for succinic acid.

Synthesis and Biopharmaceutical Studies of Cefazolin Butyrolactone Ester, a Novel Prodrug of Cefazolin (Cefazolin Butyrolactone Ester의 합성 및 생물약제학적 연구)

  • A butyrolactone ester of cefazolin (CFZ-BTL) was synthesized by the esterification of cefazolin (CFZ) with $\alpha$-bromo-${\gamma}$-butyrolactone. The synthesis was confirmed by the spectroscopic analysis. The CFZ-BTL was more lipophilic than the CFZ when assessed by n-octanol/water partition coefficients at various pH. The CFZ-BTL itself did not show any antimicrobial activity in vitro, but after oral administration of CFZ-BTL to rabbits, exerted significant anti-microbial activity in serum samples when measured by the inhibion zone method in nutrient agar plates, due to conversion of CFZ-BTL to an active metabolite, probably CFZ, in the body. The CFZ-BTL was also converted into CFZ as confirmed by in vitro incubation study, with tissue homogenates (liver, blood and intestine) of rabbits. The liver showed the fastest conversion rate, probably via the hydrolysis mechanism. In vivo metabolism of CFZ-BTL to CFZ was also confirmed in vivo serum samples by HPLC. The oral bioavailability of CFZ-BTL in rabbits was 1.6-fold increased when compared to CFZ, resulting from followed by enhanced lipophilicity increased passive absorption in the intestine.

  • Copper nano particles have been considered as the materials for conductive ink due to its good thermal, electrical conductivity and low cost. However, copper nanoparticles oxidize easily, decreasing dispersion stability and electrical conductivity. Therefore, it is important to develop a method to minimize oxidation of copper nano particles to improve its dispersion stability property in copper nano ink. In this study, copper nano particles were coated with 1-Octanethiol VSAM(Vaporized Self Assembled Multilayers) to prevent oxidation and coated copper powders were dispersed in conductive ink successfully by studying its relationship of different chain length of solvents to 1-Octanethiol coating layer to fabricate nano ink. Various alcohol solvents, such as 1-Hexanol, 1-Octanol, and 1-Decanol were used. The coating layer was observed using FESEM and TEM. Furthermore, dispersion of copper nano particles in nano inks, was characterized using Turbiscan analyzer, viscometer, and contact angle measurement tool.

  • Several physicochemical properties such as water solubility, vapor pressure, hydrolysis and octanol/water partition coefficient(Kow) of flupyrazofos, the first organophosphorus insecticide developed in Korea, were measured based on EPA and OECD methods. Water solubility was low showing 0.80 ppm at $25^{\circ}C$ and in hydrolysis study, half-life at $25^{\circ}C$ was 266.5 hr(pH 4.0), 180.0 hr(pH 7.0) and 120.9 hr(pH 9.0) demonstrating instability in alkaline solution. At $40^{\circ}C$ hydrolysis rate was $2{\sim}4$ times higher than that at $25^{\circ}C$. The equation log P=0.673-(1565.4/T) was obtained from vapor pressure experiments at three different temperatures (25, 35, $45^{\circ}C$) and $2.81{\times}10^{-5}$ torr was obtained at $25^{\circ}C$. This value is similar to that of diazinon and 1,000 times lower than that of DDVP suggesting it would not give environmental contamination by volatilization. High log Kow(5.24) was observed and this might result in bioconcentration through food chain. However, its possibility is not likely to be high due to its relatively rapid hydrolysis.

  • Methanotrophs are microorganisms that possess the unique ability to utilize methane as their sole source of carbon and energy. A novel culture system, known as the compulsory circulation diffusion system, was developed for rapid growth of methanotrophic bacteria. Methanol dehydrogenase (MDH, EC 1.1.99.8) from Methylomicrobium sp. HG-1, which belongs to the type I group of methanotrophic bacteria, can catalyze the oxidation of methanol directly into formaldehyde. This enzyme was purified 8-fold to electrophoretic homogeneity by means of a 4 step procedure and was found in the soluble fraction. The relative molecular weight of the native enzyme was estimated by gel filtration to be 120 kDa. The enzyme consisted of two identical dimers which, in turn, consisted of large and small subunits in an ${\alpha}_2{\beta}_2$ conformation. The isoelectric point was 5.4. The enzymatic activity of purified MDH was optimum at pH 9.0 and $60^{\circ}C$, and remained stable at that temperature for 20 min. MDH was able to oxidize primary alcohols from methanol to octanol and formaldehyde.

Adsorption Characteristics for Nitrosamines in Granular Activated Carbon Process (입상활성탄 공정에서의 nitrosamine류 흡착 특성)

  • This study accessed the adsorption characteristics of the nine nitrosamine species on coal-based granular activated carbon (GAC). The breakthrough appeared first for NDMA and sequentially for NMOR, NPYR, NMEA, NDPA, NDEA, and NPIP. On the other hand, NDBA and NDPHA were not detected in the treated effluent for the operation period. The maximum adsorption capacity (X/M) for the seven nitrosamine species with apparent breakthrough points ranged from $27.5{\mu}g/g$ (for NDMA) to $671.0{\mu}g/g$ (for NPIP). Carbon usage rate (CUR) for NDMA was 1.07 g/day, 13.4 times higher than that for NPIP (0.08 g/day). The X/M values for the seven nitrosamine species were fitted well with a linear regression ($r^2$ = 0.94) by their octanol-water partitioning coefficient ($K_{ow}$).

  • A novel sample pretreatment technique, headspace hanging drop liquid phase microextraction (HS-LPME) was studied and applied to the determination of flavors from solid clove buds by gas chromatography-mass spectrometry (GC-MS). Several parameters affecting on HS-LPME such as organic solvent drop volume, extraction time, extraction temperature and phase ratio were investigated. 1-Octanol was selected as the extracting solvent, drop size was fixed to 0.6 $\mu$L. 60 min extraction time at 25 ${^{\circ}C}$ was chosen. HS-LPME has the good efficiency demonstrated by the higher partition equilibrium constant ($K_{lh}$) values and concentration factor (CF) values. The limits of detection (LOD) were 1.5-3.2 ng. The amounts of eugenol, $\beta$-caryophyllene and eugenol acetate from the clove bud sample were 1.90 mg/g, 1.47 mg/g and 7.0 mg/g, respectively. This hanging drop based method is a simple, fast and easy sample enrichment technique using minimal solvent. HSLPME is an alternative sample preparation method for the analysis of volatile aroma compounds by GC-MS.

  • Dielectric studies of methyl acrylate with 1-propanol, 1-butanol, 1-heptanol and 1-octanol binary mixtures have been carried out over the frequency range from 10 MHz to 10 GHz at temperatures of 283, 293, 303 and 313 K using Time Domain Reflectometry (TDR) for various concentrations. The Kirkwood correlation factor and excess inverse relaxation time were determined and discussed to yield information on the molecular structure and dynamics of the mixture. The values of the static dielectric constant, relaxation time and the Kirkwood correlation factor decrease with increased concentration of methyl acrylate in alcohol. The Bruggman plot shows a non-linearity of the curves for all the systems studied indicates the heterointeraction which may be due to hydrogen bonding of the OH group of alcohol with C=O of the methyl acrylate. The excess inverse relaxation time values are negative for all the systems at all the temperatures indicates that the solute-solvent interaction hinders the rotation of the dipoles of the system.

Distribution between Air-Soil Concentration of Organochlorine Pesticides (유기염소계 살충제의 대기-토양간 분배)

  • This paper was studied to investigate the equilibrium state between organochlorine pesticides (OCPs) concentration of air and soil, and to know whether soil is a secondary pollution source of OCPs in air or not. The OCPs concentration of air ($C_{air}$) and soil ($C_{soil}$) is not related to molecular weight, vapor pressure ($P_L$) and octanol-air partition coefficient ($K_{oa}$). The slope of the regression line between soil-air partition coefficient ($K_{sa}$) and scavenging coefficient ($C_{soil}/C_{air}$) was 0.2952, which the OCPs concentration between air and soil did not reached to the equilibrium state. The soil/air fugacity quotients ($f_{soil}/f_{air}$) of p, p'-DDT/DDD/DDE and ${\beta}-HCH\;(0.30{\sim}0.67$), which is smaller than 1, means the deposit of OCPs from air to soil. However, $f_{soil}/f_{air}$ of heptachlor, heptachlor epoxide, ${\alpha}/{\gamma}-chlordane$, trans-nonachlor, endosulfan sulfate and ${\alpha}/{\gamma}-HCH\;(1.90{\sim}73.25)$, which is greater than 1, means that soil is secondary pollution source of OCPs in air.

대웅세파(DWC-751)의 체내동태 시험

  • 결과 및 고찰: 이 약의 흰쥐에서의 체내동태는 혈중농도로 볼 때 2-exponential pharmacokinetics에 따르고, HPLC법으로 정량한 경우의 $T_1$/$_2$$\alpha$, $T_1$/$_2$$\beta$, AUC, C $L_{T}$, C $L_{R}$, V $D_{SS}$ 는 각각 1.90min, 21.89min, 1899.36$\mu\textrm{g}$ㆍmin/ml, 10.66ml/min/kg, 7.48ml/min/kg, 0.28l/kg으로 bioassay법과는 약간의 차이를 보였다. 분포특성은 간장과 신장에 많이 이행하였으며, 폐로의 이행도 적지만 관찰되었다. 이 약의 단백결합률은 그 농도가 31.3$\mu$M일 때 42.3%였고 water/n-octanol계에서의 유상으로의 분배는 거의 일어나지 않았다. 이 약의 분포용적이 작은 것은 단백결합 때문이라기보다는 높은 극성때문으로 추정되었다. 이 약의 C $L_{R}$은 GFR의 문헌치보다 컸으며 C $L_{T}$의 약 2/3을 차지하므로 약물소실에 있어서 신장의 기여도가 크고 신배설 과정에 신분비가 관여하고 있음을 알 수 있었다. 이런 사실은 CAZ나 CTX등의 기존 세파계 항생제들과 유사했으며, 추후 다회투여시와 용량의존적 체내동태에 관해 더 많은 연구가 필요하리라 생각되었다.되었다.
