Antioxidant Effect of Cynomorii Herba on HepG2 Cells and Diphenyl-picryl-hydrazyl (DPPH) Radical Scavenging Activity (쇄양(鎖陽)의 Diphenyl-picryl-hydrazyl (DPPH) 소거 활성 및 HepG2 세포에 대한 항산화 효과)

  • The purpose of this study was to investigate the anti-oxidant effect of Cynomorium songaricum. The extract of Cynomorii Herba was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, HepG2 cell viability and $H_2O_2$-induced cytotoxicity by a modified MTT assay. DPPH radical scavenging activity was measured after 30 minutes. The extract was tested by 1. 5, 10, 50, 100 and 500 ${\mu}g/ml$ concentrations. HepG2 cell viability by a modified MTT assay was measured in the concentrations of 10, 50, 100, 250, 500 ug/ml for 24 h. The results showed that the extract scavenged DPPH radical up to 52.2% with 50 ug/ml concentration. The extract did not reduced the cell viability and $H_2O_2-induced$ cytotoxicity (69.4%) was blocked by the extract in the concentrations of 50, 100, 250 and 500 ${\mu}g/ml$. In conclusion, the extract of Cynomorii Herba has potent antioxidant activity.

Preliminary antioxidant, antibacterial and cytotoxic activities of Momordica charantia Linn leaf

  • We undertook the present study to evaluate different pharmacological as antioxidant, antibacterial and cytotoxic activities of the crude ethanolic extract of Momordica charantia (Family: Cucurbitaceae) leaves. The antioxidant property of the extract was assessed by 1, 1-diphenyl-2-picryl hydrazyl free radical scavenging assay. The extract showed antioxidant activity where $IC_{50}$ was about ${\sim}500{\mu}g/ml\;and\;IC_{50}$ was about ${\sim}10{\mu}g/ml$ for standard drug ascorbic acid. The extract showed a broad spectrum of antibacterial activity against all the tested gram positive and gram-negative bacteria where Staphylococcus epidermidis, Staphylococcus aureus, Salmonella typhi and Shigella dysenteriae were prominent. And the zones of inhibition were ranging from 8-15 mm for all the tested bacteria. Its cytotoxic property was evaluated by brine shrimp lethality bioassay. The extract showed significant lethality and the $LC_{50}$ value was $20{\mu}g/ml$.

Anti-Inflammatory and Anti-Oxidative Effects of Danggwisusan on Macrophages

  • Background: Danggwisusan is a herbal medicine which is used to treat bruises, static blood, external injuries, and somatalgia in Korean medicine. The objectives of this study were to investigate whether Danggwisusan hot aqueous extract had an inhibitory effect upon inflammatory cytokine production and oxidation. Methods: Cytotoxic activity of Danggwisusan extract was examined by 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The amount of nitric oxide produced was measured using Griess reagent. Prostaglandin E2 production was measured using an enzyme immunoassay. Inflammatory cytokines ($IL-1{\beta}$, IL-6 and $TNF-{\alpha}$) were measured by an enzyme linked immunosorbent assay. The anti-oxidative effect of Danggwisusan was measured by the 1,1-Diphenyl-2-picryl hydrazyl method. The amount of polyphenol and flavonoid contents were measured by Folin and Ciocalteauea phenol reagent and aluminum nitrate. Results: Danggwisusan hot aqueous extracts did not show significant toxicity at 10, 20, 50, and $100{\mu}g/mL$. At a dose of $100{\mu}g/mL$, Danggwisusan hot aqueous extract significantly inhibited nitric oxide and $PGE_2$ production, and significantly reduced $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ production. At a dose of $100{\mu}g/mL$, 1,1-Diphenyl-2-picryl hydrazyl free radical scavenging capability was over 50%. Conclusion: This study showed that Danggwisusan hot aqueous extract may have anti-inflammatory and anti-oxidative effects on macrophages.

Pharmacological profiles of Holarhena antidysenterica (L.) Wall bark

The antioxidant property of the extract was assessed by 1,1-diphenyl-2-picryl hydrazyl free radical scavenging assay. The extract showed antioxidant activity ($IC_{50}$ about ${\sim}08$ ${\mu}g/ml$), which was comparable to standard drug ascorbic acid ($IC_{50}$ about ${\sim}10$ ${\mu}g/ml$). The extract showed a broad spectrum of antibacterial activity against all tested gram positive and gram negative bacteria most prominent against Enterococci, Staphylococcus pyogenase and Shigella sonnie. And the zones of inhibitions were ranging from 10 - 21 mm for all the tested bacteria. Its cytotoxic property was evaluated by brine shrimp lethality bioassay. The extract showed significant lethality and the $LC_{50}$, $LC_{90}$ values were 80 ${\mu}g/ml$, 320 ${\mu}g/ml$ respectively. The results tend to suggest that the extract might possess chemical constituent(s) that are responsible for antioxidant, antibacterial and cytotoxic activities.

Analgesic, cytotoxic and antioxidant activities of Trewia polycarpa bark

  • The crude ethanol extract of the stem bark of Trewia polycarpa (Family: Euphorbiaceae) was subjected to acetic acid induced writhing inhibition, Brine Shrimp lethality bioassay and 1, 1-diphenyl-2-picryl hydrazyl free radical scavenging assay for screening of analgesic, cytotoxic and antioxidant activity respectively. The extract produced significant (P < 0.001) writhing inhibition in acetic acid induced writhing in mice at the dose of 125, 250 and 500 mg/kg body weight respectively, which were comparable to the standard drug diclofenac sodium. The extract showed significant lethality to Brine Shrimp and the $LC_{50}$ value was $8\;{\mu}g/ml$. The extract showed prominent free radical scavenging activity ($IC_{50}$ about ${\sim}10\;{\mu}g/ml$) compare to standard drug ascorbic acid ($IC_{50}about\;{\sim}15\;{\mu}g/ml$). The results tend to suggest that the crude ethanol extract of the bark might possess analgesic, cytotoxic and antioxidant activities or active constituent(s) responsible for the activities.

Effects of Salviae Miltiorrhizae Radix Hot Aqueous Extract on Nitric Oxide and Prostaglandin E2 Production and on 1,1-diphenyl-2-picryl hydrazyl Radical Scavenging in Macrophages

  • Objectives: The objective of this study is to investigate the effects of Salviae Miltiorrhizae Radix hot aqueous extract on nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production and on 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical scavenging in macrophages. Methods: Salviae Miltiorrhizae Radix (300 g) was heated at $100^{\circ}C$ with distilled water (2 L) for 4 hours. The extract was filtered and concentrated to 100 mL by using a rotary evaporator, was frozen at $-80^{\circ}C$, and was then freeze-dried by using a freezing-drying system. The RAW 264.7 macrophage was subcultured by using $10-{\mu}g/mL$ lipopolysaccharide (LPS). In order to evaluate cytotoxicity, we performed 3-(4,5-dimrthylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays and measured the cell viability. The NO production was measured by using Griess assays, and the $PGE_2$ production was measured by using enzyme immunoassays. The antioxidant activity, the 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical scavenging capability, was measured by using the DPPH method. Results: Cell viability with the 1-, 5-, 25-, 125- and $625-{\mu}g/mL$ Salviae Miltiorrhizae Radix hot aqueous extract was not significantly decreased compared to the cell viability without the extract. When 125 and $625{\mu}g/mL$ of Salviae Miltiorrhizae Radix hot aqueous extract were used, nitric oxide (NO) production in LPS-stimulated RAW 264.7 macrophages was significantly inhibited compared to that in the control group. When 25, 125, and $625{\mu}g/mL$ of Salviae Miltiorrhizae Radix hot aqueous extract were used, $PGE_2$ production in LPS-stimulated RAW 264.7 macrophages was significantly inhibited compared to that in the control group. The 125- and $625-{\mu}g/mL$ Salviae Miltiorrhizae Radix hot aqueous extracts had high DPPH free-radical scavenging capabilities in RAW 264.7 macrophages. Conclusion: This study indicates that Salviae Miltiorrhizae Radix hot aqueous extract suppresses NO and $PGE_2$ production and improves DPPH free-radical scavenging capability. Thus, it seems that Salviae Miltiorrhizae Radix hot aqueous extract may have an anti-inflammation effect and antioxidant activity.

Antioxidant Effect of Rubi Fructus on TM4 Sertoli Cells (남성생식세포 Sertoli cell에 미치는 복분자(覆盆子)의 항산화 효과)

  • Objectives : The purpose of this study was to examine the antioxidant effects of the extract of Rubi Fructus on TM4 Sertoli cells. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity and cell viability assays on Sertoli cells. In addition, hydrogen peroxide-induced oxidative stress on Sertoli cells were examined by MTT assay. The antioxidant enzyme of Cu/Zn SOD, Mn SOD, catalase protein expression on Sertoli cells were also measured. Results : The results showed that the extract scavenged DPPH radical dose-dependent manner. The extract showed no cytotoxicity at concentration of 1, 5, 10, 50, $100{\mu}g/ml$. The hydrogen peroxide-induced cytotoxicity of Sertoli cells was protected to 88.3% by the extract at concentration of $100{\mu}g/ml$. Cu/Zn SOD and Mn SOD protein expression were significantly increased on Sertoli cells, but catalase protein expression was not significantly changed. Conclusions : In conclusion, the extract of Rubi Fructus has antioxidant effects on Sertoli cells and protect male reproductive system against oxidative stress.

Antioxidant and Anti-wrinkling Effects of Extracts from Vitex trifolia L. (만형자 추출물의 항산화 및 주름개선 효과)

  • In this study, the antioxidant and anti-wrinkling effects of fraction from Vitex trifolia L. were investigated. Among the fractions, ethyl acetate fraction showed the highest antioxidant activities in 1-1-diphenyl-2-picryl-hydrazyl, 2,2'-azino-bis(3-ethylbenzothiazoine-6-sulfonic acid) radical scavenging and elastase inhibition with 76, 89, and 74%, respectively, at a concentration of $1,000{\mu}g/mL$. This fraction, at the concentration of $25{\mu}g/mL$, inhibited 70% fibroblast cell viability and 86% the matrix metalloprotease (MMP)-1. In addition, the results from Western blot assay showed that this fraction ($25{\mu}g/mL$) expressed the MMP-1 protein level by decreasing 50%. The findings suggest that the ethyl acetate fraction from V. trifolia has great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

Antioxidant Effects of Corni Fructus in GC-1 Cells (산수유(山茱萸)가 남성 생식세포 GC-1의 항산화에 미치는 영향)

  • The purpose of this study is to examine the antioxidant activity in the germ cells of the extract of Corm fructus. The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, GC-1 cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay and lipid perixidation by malondialdehyde (MDA) formation, respectively. The results showed that the extract scavenged DPPH radical with the IC50 being $200{\mu}g/mL$. The extract at concentrations of $10-500{\mu}g/ml$ showed dose-dependent in growth of GC-1 cell. $H_2O_2$-induced cytotoxicity (63.0%) was blocked by the extract (10, 50, 100, 250 and $500{\mu}g/ml$) concentration-dependently. Furthermore, the extract (50, 100 and $250{\mu}g/ml$) also displayed a dose-dependent reduction of MDA formation on $H_2O_2$-induced lipid peroxidation. In conclusion, the extract of Corm fructus has potent antioxidant activity.

UV-induced skin damage improvement effects of solvent fraction from Maekmoondong-tang (맥문동탕 용매 분획물의 UV에 의한 피부손상 개선 효능평가)

  • In this study, we verified the 1-1-diphenyl-2-picryl-hydrazyl radical scavenge, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenge, elastase, tyrosinase inhibitory effect by using the solvent fractions of Maekmoondong-tang hot water extract. As a result, the ethyl acetate fraction (MW-EA) showed the highest inhibitory activity. In cell-based assays, MW-EA treatment confirmed a 34% ($100{\mu}g/mL$) efficacy in reactive oxygen species inhibitory activity, and at the same concentration, MMPs showed more than 50% inhibition and tyrosinase inhibited 25% ($50{\mu}g/mL$). Therefore Maekmoondong-tang is considered high development potential as a material to improve the skin.
