유기노의 항산화 활성 및 멜라닌 생성 억제 효과 (Effect of Artemisia anomala S. Moore on Antioxidant Activity and Melanogenesis)

  • In mammalian melanocytes, melanin synthesis is controlled by tyrosinase, the key enzyme in the pigment synthesis. In this study, to develop a new whitening agent, we have investigated the antioxidant and the inhibitory effect of Artemisia anomala extract on tyrosinase activity and melanigenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of butanol fraction from A. anomala was higher than that of arbutin ($97.5{\pm}0.5%$ at the concentration of 2 mg/ml). The butanol fraction was shown scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radicals in a dose dependent manner. The highest inhibitory activity of melanogenesis was also butanol fraction ($25.0{\pm}3%$ at the concentration of $200\;{\mu}g/ml$). From these results, we suggest that the A. anomala extract might be used to be a potential agent for skin whitening.

Antioxidant Activity of Gamma-Irradiated Asparagus cochinchinensis (Asparagi radix) (Lour.) Merr. Extract and Inhibition Effect on Lipid Oxidation of Emulsion-Type Pork Sausage

  • The objective of this study was to determine the antioxidant activity of gamma-irradiated Asparagus cochinchinensis (Asparagi radix) (Lour.) Merr. Extract (ARE) and its inhibition effect on food lipid oxidation using emulsion-type pork sausage as a model. ARE was prepared from dried Asparagi radix root and ARE solution (1.0 g/mL) was gamma-irradiated with designated doses at 5, 10, and 20 kGy. Antioxidant activity of ARE solution was determined by measuring 1,1-diphenyl-e-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-9-sulphonic acid) (ABTS) radicals. Activities of DPPH and ABTS radicals were decreased, whereas total phenolic contents increased after gamma irradiation with a dose dependence. Addition of gamma-irradiated ARE dose-dependently retarded lipid oxidation of emulsion-type pork sausage during storage at $4^{\circ}C$. These results indicated that gamma-irradiated ARE might have antioxidant activity more than non-irradiated ARE due to increase of the content of polyphenolic compounds by ionizing radiation.

Radical scavenging activity of ethanol extract and solvent partitioned fractions of lotus seeds

  • This study focused on the evaluation of the antioxidative effects of lotus seeds from golden colored flowers. The lotus seeds were extracted with ethanol and then fractionated into 4 fractions, ethyl acetate (EtOAc), n-butanol, methylene chloride, and n-hexane. The comparison of antioxidative activities of the extract and fractions from the lotus seeds was carried out using an in vitro radical scavenging model and the total phenol content was analyzed. Of the tested extracts and fractions, the EtOAc fraction of the lotus seeds showed the strongest 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity with 96.24% at a concentration of $100{\mu}g/mL$. In addition, the hydroxyl radical scavenging activity of the lotus seed EtOAc fraction was also increased in a concentration dependent manner with the concentrations tested ranging from 5 to $100{\mu}g/mL$. Moreover, the EtOAc fraction showed the highest scavenging activity for nitric oxide and superoxide anion radicals. In particular, of all the extracts and fractions, the EtOAc fraction showed highest contents of total phenols. These results indicate that lotus seeds have potential as an antioxidative agent against oxidative stress involving reactive oxygen species. Furthermore, the EtOAc fraction of lotus seeds includes promising oxidative stress-protective compounds.

Inhibitory Activities of Palmatine from Coptis chinensis Against Helicobactor pylori and Gastric Damage

  • Helicobacter pylori (H. pylori) is the most important factor of gastric disease in clinical practice. Moreover, smoking, stress and a poor diet may be additive factors for gastric damage. With these factors, increasing infection of H. pylori triggers gastritis, gastric ulcers and gastric cancer. To develop a new protective agent, we are concerned with plant-derived extract. The extract of Coptis chinensis (C. chinensis) and its constituents were investigated to assess their protective activities against gastric damage. The C. chinensis extract showed a scavenging effect against 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and superoxide radicals, inhibition of H. pylori colonization and antiulcerogenic activities in rat. In particular, palmatine derived from C. chinensis was found to be the novel protective agent. It is better than the C. chinensis extract, berberine, a well-known constituent of C. chinensis. We suggest that palmatine from the root cortex of C. chinensis may be a good candidate for the development of new pharmaceuticals to prevent gastric disease.

Free Radical Scavenging Activity and Inhibition of Linoleic Acid Peroxidation of Commercial Tocotrienol Fraction

  • Tocotrienols (T3) are minor plant constituents found abundantly in rice bran, which provide a significant source of vitamin E in animal feeds. T3 was reported to have an intrinsic hypocholesterolemic effect by inhibiting HMG-Co A reductase. It has similar antioxidative properties as tocopherols in food and biological system due to their similar chemical structures. However, the antioxidant activity and mechanism of T3 to scavenge free radicals and to inhibit the peroxidation of linoleic acid are less understood. The purpose of this study was to investigate the scavenging effect of T3 on free radicals and its inhibition of peroxide formation. Free radical scavenging activity was monitored by the DPPH (1,1-diphenyl-2-picrylhydrazyl) method whereas inhibition of linoleic acid peroxidation was evaluated using the thiocyanate method. Thiobarbituric acid (TBA) test was used to determine malonaldehyde formation from linoleic acid peroxidation. Free radical scavenging activity increased with increasing concentration levels of T3. T3 exhibited 38.2, 78.6, 92.7 and 96.2% radical scavenging activity at concentrations of 2, 8, 32 and 128 ppm, respectively. At 128 ppm, it was highly effective in inhibiting linoleic acid peroxidation. The activity of T3 evaluated by the thiocyanate method showed low absorbance values indicating a high level of antioxidant activity. All treatments showed similar trends in antioxidant activity when evaluated by both the thiocyanate method and TBA test.

Antioxidant Activities of Agrimonia pilosa Ledeb: In Vitro Comparative Activities of Its Different Fractions

  • The antioxidant potentials of the methanol extract and its various fractions from Agrimonia pilosa Ledeb were measured as 1,1-diphenyl-2-picrylhydrazyl (DPPH), and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities as well as the reducing power decreasing NO in vitro. The methanol extract of Agrimonia pilosa Ledeb showed significantly strong scavenging effects on free radicals of DPPH, ABTS, and NO. Hexane fraction (HF), ethyl acetate fraction (EF) and n-butyl alcohol fraction (BF) were prepared by solvent fractionation. By comparison of polyphenol contents among BF and HF, EF with high polyphenol contents showed the highest DPPH, ABTS, and NO scavenging effects with the value of 50.2%, 80.4%, and 65.7%, respectively, at the highest tested dose. HPLC analysis revealed the presence of various phenolic compounds in EF of Agrimonia pilosa Ledeb. These results suggest that EF could be considered as natural antioxidant sources and dietary nutritional supplements to prevent oxidation-related diseases.

돌나물의 항균활성 및 위암예방효과 (Antibacterial Activity and Protective Role against Gastric Cancer by Sedum sarmentosum)

  • 본 연구에서는 돌나물 분획물의 항산화 및 위암예방 효과를 다양한 균들과 위암세포주를 이용하여 검토하였다. Escherichia coli, Staphylococcus aureus 그리고 helicobacter pylori에 대한 돌나물의 n-hexane, methylene chloride (MC), ethyl acetate, n-butanol 분획물의 항균효과를 disc 확산법으로 알아본 결과, MC 분획물이 E. coli에 대해 13 mm의 억제존을 보여 가장 우수한 항균효과를 나타내는 것을 알 수 있었다. $100{\mu}g/mL$ 농도에서 모든 분획물은 1,1-diphenyl-2-picrylhydrazyl (DPPH)와 hydroxyl radicals (${\cdot}OH$)에서 50% 이상의 소거능을 나타냈다. 특히, MC 분획물이 DPPH와 ${\cdot}OH$에서 가장 높은 소거능을 보였다. 또한 MC 분획물은 H. pylori와 위암세포의 성장을 억제하는 것을 확인할 수 있었다. 그러므로 돌나물의 분획물 중 특히 MC 분획물이 radical 소거능, H. pylori에 대한 항균성 및 AGS 인체 위암세포 생존 저해효과가 우수한 활성물질을 함유하고 있을 것으로 생각된다.

자외선이 조사된 섬유아세포에서 말채나무 잎추출물의 항산화작용 (Antioxidative Activity of Extract of Cornus walteri Wanger Leaves in Human Dermal Fibroblast Irradiated by UVB)

  • We investigated antioxidative activity of the ethanol extracts of leaves of Cornus walteri Wanger (CWE) by treated enzyme in human dermal fibroblast (HDFs) irradiated by UVB. We examined the in vitro chemical and cellular antioxidant activities of CWE in HDFs. We employed scavenging assay for the 1,1-diphenyl-2,5-picrylhydrazyl (DPPH) radicals and cellular antioxidative activity of CWE, and we was investigated in $H_2O_2$-treated or UVB-irradiated HDFs. The CWE effectively scavenged DPPH radicals ($IC_{50}$ $7.03{\pm}0.4{\mu}g/mL$) when compared to the scavenging activities of L-ascorbic acid ($IC_{50}$ $4.69{\pm}0.3{\mu}g/mL$). CWE reduced UVB-induced cellular damage in HS68 cells by MTT assay and inhibited intracellular ROS generation in dose-dependent manner. In addition, CWE also attenuated the elevated levels of 8-isoprostane resulting from UVB-mediated oxidative stress. Collectively, these results suggest that CWE could be a new potential candidate as antioxidant against UVB-induced oxidative stress in HDFs.

Antioxidant capacity of crude extract and fractions from Woodfordia fruticosa flower

  • Woodfordia fruticosa Kurz. (Lythraceae), commonly known as Dhayero, is used in the treatment of various ailments in Nepal. In this study, the antioxidant capacity of crude extract and different polarity fractions of Woodfordia fruticosa flowers were assayed for their scavenging abilities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and nitric oxide radicals, competitive ${\beta}$-carotene bleaching, reducing power, metal chelating ability and total phenolic content. Crude extract and polar fractions showed stronger antioxidant capacity and contained very high level of total phenolics. They exhibited strong DPPH radical scavenging, nitric oxide scavenging and reducing power, medium ${\beta}$-carotene bleaching and poor metal chelating capacity. Positive correlation obtained between total phenolic content and antioxidant capacity assays, indicated the contribution of phenolics toards antioxidant capacity.

흑삼의 항산화 활성 (Antioxidant Activity of Black Panax ginseng)

  • This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.
