Evaluation of the Antioxidant Potential of Korean Indigenous Plant Extracts by Free Radical Scavenging Activity

  • Since reactive oxygen radicals play an important role in carcinogenesis and other human diseases including neurodegenerative states, antioxidants present in natural products have received considerable attention for alleviation of these disease states. Therefore, in order io identify antioxidants in plant extracts, fifty-seven methanolic extracts derided from indigenous Korean plants were primarily assessed for potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals. As a result, nine plant extracts were found to exhibit the DPPH free radical scavenging activity in the criteria of $IC_{50}<40\;{\mu}g/ml$. In particular, the extracts of Melioma oldhami $(IC_{50}=0.1\;{\mu}g/ml)$, Myrica rubra $(IC_{50}=16.2\;{\mu}g/ml)$, Sympolocos paniculata $(IC_{50}=23.0\;{\mu}g/ml)$, Carpinus laxiflora $(IC_{50}=25.1\;{\mu}g/ml)$, and Cleyera japonica $(IC_{50}=26.2\;{\mu}g/ml)$ showed a potent radical scavenging activity. Further study for the identification of active compounds from these lead extracts might be warranted.

Evaluation of antioxidant property of heat shock protein 90 from duck muscle

  • Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1-pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products.

The Butanol Fraction of Bitter Melon (Momordica charantia) Scavenges Free Radicals and Attenuates Oxidative Stress

  • To investigate radical scavenging effects and protective activities of bitter melon (Momordica charantia) against oxidative stress, in vitro and a cellular system using LLC-$PK_1$ renal epithelial cells were used in this study. The butanol (BuOH) fraction of bitter melon scavenged 63.4% and 87.1% of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals at concentrations of 250 and $500{\mu}g/mL$, respectively. In addition, the BuOH fraction of bitter melon effectively scavenged hydroxyl radicals (${\cdot}OH$). At all concentrations tested, the scavenging activity of the BuOH fraction was more potent than that of the positive control, ascorbic acid. Furthermore, under the LLC-$PK_1$ cellular model, the cells showed a decline in viability and an increase in lipid peroxidation through oxidative stress induced by pyrogallol, a generator of superoxide anion ($O_2{^-}$). However, the BuOH fraction of bitter melon significantly and dose-dependently inhibited cytotoxicity. In addition, 3-morpholinosydnonimine (SIN-1), a generator of peroxynitrite ($ONOO^-$) formed by simultaneous releases of nitric oxide and $O_2{^-}$, caused cytotoxicity in the LLC-$PK_1$ cells while the BuOH fraction of bitter melon ameliorated oxidative damage induced by $ONOO^-$. These results indicate that BuOH fraction of bitter melon has protective activities against oxidative damage induced by free radicals.

전도산(顚倒散)의 항산화 효과에 관한 실험적 연구 (The Experimental Study of Jeondo-san on Antioxidant Effects)

  • Objectives : This study was carried out to investigate the antioxidant effects of Jeondo-san(JDS). Methods : The antioxidant effects of JDS were measured by the scavenging for 2,2-Diphenyl-1-picrylhydrazyl(DPPH) radical, the formation of intracellular glutathione(GSH), the inhibition for reactive oxygen species(ROS). Results : 1. All concentrations of JDS showed antioxidant effect by decreasing the DPPH radicals. 2. All concentrations of JDS did not effect on the formation of intracellular GSH in HaCaT cell. 3. All Concentrations of JDS inhibited the production of ROS in the HaCaT cell stimulated with $H_2O_2$. Conclusion : The present date suggest that JDS has effects on the stage of inflammation.

Antioxidant and Antimicrobial Activity of Zostera marina L. Extract

  • Methanol crude extract of the sea grass Zostera marina L. and organic solvent fractions (n-hexane, chloroform, ethyl acetate, n-butanol, and water) were screened for antioxidant activity (total phenolic contents, DPPH scavenging activity, and reducing power) and antimicrobial activity against three human skin pathogens, two bacteria and a yeast; Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans. Total phenolic contents and 2, 2- diphenyl-1-picrylhydrazyl (DPPH) scavenging activity were highest in the ethyl acetate fraction with 968.50 $\mu$g gallic acid equivalent per milligram of extract, and ca. 95% scavenging activity on the DPPH radicals at 10 mg $ml^{-1}$. In antimicrobial activity tests, MICs (Minimum Inhibitory Concentration) of each Zostera marina extract partitioned ranged from 1mg to 8 mg $ml^{-1}$ (extract/ 10% DMSO) against all three human skin pathogens. The MICs of the ethyl acetate and n-butanol fractions were the same with 1 mg $ml^{-1}$ against S. aureus and C. albicans. The ethyl acetate fraction of Z. marina does protect against free radicals and may be used to inhibit the growth of human skin pathogens.

상황버섯(Phellinus pini)의 라디칼 소거작용 (Radical Scavenging Activities of Phellinus pini)

  • Phellinus pini (CY001) 추출물에서의 페놀류성분의 농도는 P. pini (CY001)분획 g 당 GAEs mg으로 나타내었고, P. pini의 EtOAc 분획 (436.5 mg GAEs/g)이 다른 분획에 비해 가장 높은 함량을 나타내었다. 환원력과 같은 항산화 활성을 screening 하기 위해 몇 가지 생화학적 시험이 수행되었고, 그것은 2, 2-diphenyl-1-picrylhydrazyl(DPPH) 라디칼 소거작용, NBT/XO superoxide system, DCF/AAPH peroxyl radicals 저해능 등이다. 여섯 개의 버섯추출물 분획 중에서 EtOAc 분획이 DPPH, superoxide 라디칼, peroxyl 라디칼 소거작용이 뛰어났고, $IC_{50}$ values는 각각 $11.49\;{\mu}g/ml$, $8.32\;{\mu}g/ml$, and $1.91\;{\mu}g/ml$이었다. P. pini (CY001)의 EtOAc 분획은 중요한 효소적 지질과산화 저해와 cytotoxicity 없이 RAW 264.7 macrophages의 LPS 유도 NO 생성을 효과적으로 감소시켰다. 또한 EtOAc 분획이 HepG2 cell의 타크린 유도 cytotoxicity에서 간 보호 작용을 나타내었다. 이러한 결과를 바탕으로 P. pini (CY001)가 라디칼 소거작용을 하는 성분을 함유한 천연 항산화제로써의 잠재력을 갖고 있는 것으로 사료된다.

Antioxidant and inhibitor of matrix metalloproteinase-l expression from leaves of Zostera marina L.

  • Apigenin-7-O-${\beta}$-D-glucoside, chrysoeriol, and luteolin were isolated from the aqueous ethanolic extract of Zostera marina L. leaves as the scavengers of reactive oxygen species (ROS) with the SC$\_$50/ values of 0.18 mM, 0.68 mM, and 0.18 mM against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 0.04 mM, 0.03 mM, and 0.01 mM against superoxide radicals in the xanthine/xanthine oxidase system, respectively. The luteolin suppressed the expression of matrix metalloproteinase-1 (MMP-1) up to 44% at 4.0 ${\mu}$M. (omitted)

왕느릅나무 수피의 페놀성 화합물 및 항산화 활성 (Phenolic Compounds from Barks of Ulmus macrocarpa and Its Antioxidative Activities)

  • Phytochemical examination of Barks of Ulmus macrocarpa has led to the isolation and characterization of two fla-vanonol, taxifolin $7-O-{\beta}-D-glucopyranoside$ (1), taxifolin $3'-0-7-{\beta}-glucopyranoside$ (2), two flavanone eriodictyol $7-O-{\beta}-D-glucopyranoside$ (3), nalingenin $7-O-{\beta}-D-glucopyranoside$ (4), three flavan 3-ol, (+)-catechin (5), (-)-epicatechin (6), (+)-catechin 7-O-{\beta}-D-glucopyranoside (7) and one proanthocyanidin, procyanidin B-1 (8). Antioxidative activity of these compounds was determined by measuring the radical scavenging effect on 1,1-diphenyl-2- picrylhydrazyl (DPPH) radicals . (+)-Catechin (5) , (-)-epicatechin (6), (+)-catechin $7-O-{\beta}-D-glucopyranoside$ (7) and procyanidin B-1 (8) showed significant antioxidative activity.

Reticulone, a Novel Free Radical Scavenger Produced by Aspergillus sp.

  • Bioassay-guided fractionation of the culture broth of Aspergillus sp. FN070449 (KCTC 26428) using a DPPH (2,2-diphenyl-1-picrylhydrazyl) assay led to the isolation of two compounds: reticulone (1) and reticulol (2). Their chemical structures were elucidated on the basis of UV, IR, NMR, and MS spectroscopic analyses. Compound 1 exhibited more potent free radical scavenging activity on $ABTS^{\cdot+}$ (2,2'-azino-bis [3-ethylbenzthiazoline-6-sulphonic acid]) and DPPH radicals than did butylated hydroxyanisole (BHA) and caffeic acid.

Antioxidant Activities and Tyrosinase Inhibitory Effects of Different Extracts from Pleurotus ostreatus Fruiting Bodies

  • We evaluated the antioxidant activity and tyrosinase inhibitory effects of Pleurotus ostreatus fruiting bodies extracted with acetone, methanol, and hot water. The antioxidant activities were tested against $\beta$-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, and ferrous chelating ability. Furthermore, phenolic acid and flavonoid contents were also analyzed. The methanol extract showed the strongest $\beta$-carotene-linoleic acid inhibition as compared to the other exracts. The acetone extract (8 mg/mL) showed a significantly high reducing power of 1.54 than the other extracts. The acetone extract was more effective than other extracts for scavenging on 1,1-diphenyl-2-picrylhydrazyl radicals. The strongest chelating effect (85.66%) was obtained from the acetone extract at 1.0 mg/mL. The antioxidant activities of the extracts from the P. ostreatus fruiting bodies increased with increasing concentration. A high performance liquid chromatography analysis detected seven phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, naringenin, hesperetin, formononetin, and biochanin-A in an acetonitrile and 0.1 N hydrochloric acid (5 : 1) solvent extract. The total phenolic compound concentration was $188{\mu}g$/g. Tyrosinase inhibition of the acetone, methanol, and hot water P. ostreatus extracts increased with increasing concentration. The results revealed that the methanol extract had good tyrosinase inhibitory ability, whereas the acetone and hot water extracts showed moderate activity at the concentrations tested. The results suggested that P. ostreatus may have potential as a natural antioxidant.
