Effects of Egg White and Ion Exchange Resin Pretreatment on Separation of Egg White Lysozyme (난백 및 이온교환수지의 전처리 조건이 난백 Lysozyme의 추출에 미치는 영향)

  • Pretreatment of egg white and ion exchange resins was attempted to separate lysozyme from egg white efficiently. Apparent viscosity of egg white could be decreased to 3cp by homogenization for 30 minutes at 2, 000rpm and ultrasonication for 45 minutes. The result of testing adsorption capacity of lysozyme was as follows; CM-Sephadex C-25 >Duolite C464>Amberlite C-50>Dowex MSC-1>Amberlite IRC-50>Amberlite IRC-84. Although CM-Sephadex C-25 showed highest adsorption capacity of lysozyme, egg white could not eluted easily. Duolite Cf64 was selected based on relatively high lysozyme adsorption and good egg white eluting property for separation of egg white lysozyme. Na$^{+}$ form of Duolite C-464 was most effective on adsorption of Iysozyme. To separate lysozyme from egg white efficiently rinse buffer and eluting solution were selected 0.1M sodium phosphate buffer at pH 6.5 and 10% ammonium sulfate respectively. After separating lysozyme from egg white, foaming power of egg white was decreased to 85.3%. Color of egg white gel was not changed while hardness of egg white gel was decreased by 30% after separating lysozyme. However, elasticity of egg white gel was increased by 13% in lysozyme-separated egg white.

Functional Properties of Lupinseed Protein Concentrate (루우핀콩 단백질 농축물(LPC)의 식품기능성)

  • The functional properties of lupin seed protein concentrate (LPC) were examined and compared to those of soybean protein isolate (SPI) and Na-caseinate. LPC-50, of which protein level was 50%, was prepared by a two phase solvent (hexane: alcohol: water= 10:7:3) extraction method. LPC-70 was made from LPC-50 by removing the fractions solubilized by carbohydrate decomposing enzymes. The solubilities of LPC-50 and LPC-70 were similar to that of of SPI but slightly higher at pH 4-5, and less susceptible to the added salt. The apparent viscosity of LPC increased exponentially as the concentration increased over 6% level, and the change was similar to that of Na-caseinate. LPC showed strong pseudoplastic non-Newtonean flow behavior, which was similar to that of SPI The emulsifying capasity of LPC-70 was similar to that of SPI when salt was added. The foaming capacity of LPC was comparable to that of SPI. LPC showed high oil and water absorption capacities, which increased as the protein level was elevated. LPC-70 showed the highest oil absorption capacity of all the samples tested.

  • Batch and column tests were performed to investigate the sorption capacity of bottom soils of Kimpo metropolitan waste landfill. Sorption capacity and sorption isotherm from the batch test for inorganic and heavy metal substances and retardation capacity of heavy metals from the column test are obtained. It was recognized from the tests that the samples match well with Freundlich isotherm and marine clay has good sorption capacity for potassium and heavy metals such as lead and cadmium. The soil samples with different geologic origins did not show significant differences in the sorption capacity of tested components.

  • 에탄올아민 (ETA; Ethanolamine)은 에틸렌옥시드를 진한 암모니아수와 함께 가열하여 얻어지는 물질로 흡수성이 있는 무색의 액체 또는 고체이며 탄소, 질소, 산소로 이루어진 매우 안정된 유기화합물이다. 이러한 ETA는 부식방지제, 산성가스 흡수제, 화장품 등 각종 산업에서 매우 유용하게 사용되는 물질이다. 하지만 ETA는 눈, 피부, 호흡기, 폐 등에 접촉하여 호흡기 질환 및 만성 천식을 유발하고 피부에 자극을 일으키므로 ETA를 제거하기 위한 물리화학적 연구가 필요하다. 따라서 본 연구에서는 냉각온도 및 진공펌프압력에 따른 ETA 응축 특성과, 흡착제에 따른 ETA 흡착특성을 조사하였다. 조사결과 ETA는 냉각수의 온도 및 진공펌프압력에 영향을 받았으며 냉각수 온도 및 진공펌프압력이 증가할수록 응축율은 감소하였다. ETA 흡착에서 활성탄의 경우 액상의 ETA와 상호간에 흡착력은 존재하지 않았으며, 기상의 ETA는 흡착되지만, $100^{\circ}C$이상에서 탈착 반응이 일어났다. 제올라이트의 경우 액상 및 기상의 ETA를 모두 흡착하였다.

  • Meniscus, adsorbed layer thickness, capillary pressure and disjoining pressure was deduced in extended meniscus region in cement paste pore by hydrostatic equilibrium. From the results, the relationship between pore size and adsorbed layer thickness could be derived and adsorbed layer thickness represents $0.299{\sim}2.700nm$ according to pore size $1nm{\sim}1{\mu}m$. Especially, disjoining pressure rapidly Increased in less than 10 nm pore size according to adsorbed layer thickness. Therefore, it is interpreted that autogenous shrinkage of cement paste is highly increases in formation of less than 10 nm pore size. Predictions of autogenous shrinkage in cement paste considering driving force for autogenous shrinkage with capillary pressure and disjoining pressure was low in comparison with experiment values between $1{\sim}4$ days and high in later period. These tendency could be thought that pore damage by mercury injection in early age makes shrinkage driving force underestimate and assumption for unsaturated independent pore makes overestimate. These interactions might be needed corrections considering on hydration or pore replacement model.

  • A surfactant is used to assist the effect of cleaning, dispersibility and adhesion during leather manufacturing process. Existing surfactant for that process includes many hydrophilic groups that may cause problem such as stain, bad water resistance and poor durability, etc. It is potential problem to make high-performance property for future leather market. In this study, we have synthesized the fluorinated surfactant of which property decreases surface tension, increases dispersion, cleaning effect and the better chrome absorption by the high bond energy of C-F to complement weakness that the present alkyl derivative surfactants have. Using fluorinated surfactant, we can confirm that dispersion is increased, chrome absorption ratio is augmented with high osmosis and coherence, chrome content in the tanning waste water is reduced, BOD and COD contents are diminished and physical characteristics are improved.

  • A number of industrial zones in South Korea were reported contaminated by heavy metals. Such contamination could cause severe damage to the subsurface environment including soil and groundwater. The treatment of zeolite mixing with soil at the bottom of such industrial zones might prevent, or at least reduce the damage of contamination by adsorption of the heavy metals from the leakage. However, such mixtures should maintain the proper bearing capacity as a foundation fill material from the geotechnical point of view at the same time. To investigate the effect of mixtures of zeolite with local soils for the adsorption of heavy metals (Zn, Pb) and sustainability of bearing capacity, adsorption isotherm tests and direct shear test with compaction tests were performed. Results showed that the mixing zeolite with local soils effectively reduces the spreading of the heavy metal contamination when maintaining its proper geotechnical properties as a fill material of industrial zones.

  • This study was peformed to improve extraction of insoluble proteins and to evaluate funtional properties of abolished proteins by the phytase produced by Asporgillus sp. The optimum pH, temperature, treatment time and unit of the enzyme for extraction of protein were pH 4.0~5.0, $50^{\circ}C$, 8~10 hrs and 120 units. The foaming capacity and foaming stability of sesame meal protein after enzyme treatment were virtually unchanged as compared to control. The emulsion capacity and emulsion stability of sesame meal protein was higher than control. Oil absorption as well as water absorption capacities of sesame meal protein were higher than control.

Adsorption Behavior of Monosubstituted-Halophenols by Amberlite XAD Resins (Amberlite XAD 수지에 대한 일치환 할로 페놀들의 흡착거동에 관한 연구)

  • The adsorption mechanisms of phenols on XAD-2 and XAD-7 resins were studied by using the distribution coefficient(log Kd) measured in the optimum adsorption conditions. It was observed that the Langmuir adsorption isotherm, indicating a molecular size-dependent adsorption, was appropriate for characterizing the adsorption behaviors of phenols on XAD-2 and XAD-7 resins. The adsorption energies of phenols on XAD resins were calculated by Lennard-Jones potential equation. In the calculation of the adsorption energy, the molecular radii and dipole moments of the resins and phenols were calculated by their van der Waals volumes and Debye equation, respectively. The stacking factor (F) were determined from the radio of the equilibrium distance to the stacking distance of molecules. In order to explain the adsorption energy calculated from the stacking factor it was compared with the adsorption enthalpy for each of phenols which was experimentally determined by batch adsorption shake method. It was observed that the adsorption enthalpy of phenolate ions on the XAD resins was likely to be more seriously affected by dispersion interaction than by a dipole or a charge interaction.
