Effects of Gibberellin Treatment on the Dormancy-breaking and Germination Promotion of Carpinus laxiflora Seeds Stored for 16 Years (지베렐린 처리에 따른 서어나무 16년 저장 종자의 휴면타파 및 발아촉진 효과)

  • This study assessed the vitality of Carpinus laxiflora seeds stored for more than 15 years in order to discover optimal pre-treatment conditions for removingthe causes of seed dormancy and improving revitalization rate. Seeds were collected in October 2000 and stored at -18℃ for 16 years. Experiments to assess the revitalization ability of the seeds were performed under the following conditions: a controlled environment; in warm stratification (30 days at 23℃); in cold stratification (30 days at 4℃ andfor 30, 45, 60, and 120 days); gibberellin (GA3) treatment (24 hours per day in GA3 solutions of 100, 500, 1000, 2000, and 3000 mg·L-1); and both cold stratification and GA3 treatment (30 days at 4℃, then in a GA3 solution of 100, 500, and 1000 mg·L-1 for 2 hours). The average germination percentage(GP) of untreated seeds was 2%, and the average GP of warm-stratification seeds was 10%. Cold-stratification seeds had the highest GP at 81% for the 45-day process, while the 120-day cold-stratification seeds had the lowest GP at 67.3%. The average GP of seeds treated with GA3 ranged from 77% (100 mg·L-1) to 99% (1000 mg·L-1), indicatingsignificant differences between the treatment concentrations. The treatment effect of GA3 was highest at 500 mg·L-1 and 1000 mg·L-1, and lowest at 100 mg·L-1. The average GP of seeds treated with GA3 following cold stratification was 68%, which was lower than the cold stratification-only (73.2%) and GA3-only (88.4%) treatments. A comprehensive comparison of the seed germination characteristics according to the four treatments determined that a GA3 500 mg·L-1 pre-treatment, with the highest average GP, was ideally suited to the revitalization of long-term stored C. laxiflora seeds. Consequently, C. laxiflora stored at -18℃ for 16 years indicated strong vitality and could be regenerated by proper pre-treatment.

Comparison of the Colony Development in the Bumblebees, Bombus ignitus and B. terrestris (호박벌(Bombus ignitus)과 서양뒤영벌(B. terrestris)의 봉세발달 비교)

  • Chilling temperature and colony development in Korean native bumblebee, Bombus ignitus and introduced B. terrestris were firstly investigated. Among four chilling temperatures of -2.5, 0, 2.5 and 5$^{\circ}C$, 2.5$^{\circ}C$ showed the best result at the survival rate after artificial hibernation in both species. The survival rate after chilling was somewhat higher in B. ignitus until three months of cold treatment, but it was higher in B. terrestris at four months of cold treatment. In colony development of queens broken diapause by CO$_2$ treatment, oviposition rate and preoviposition period of B. ignitus were 72.5% and 17.4 days, respectively. These values were 6.1% higher and 7.4 days shorter than those of B. terrestris. The period up to colony foundation, the first male and queen emergence of B. ignitus were 62.8,66.4 and 63.0 days, respectively, and these values were 2.1-29.5 days shorter than those of B. terrestris. But, the duration up to first worker emergence of B. ignitus and B. terrexrris did not differ as 28.6 and 28.4 days, respectively. On the other hand, the rate of colony foundation and progeny-queen production of B. terrestris, which are the important indication in evaluating the quality of colony, were 14.2 and 13.5%, respectively and these values are 3.8 to 5.7 fold higher than those of B. ignitus. Besides, the numbers of progenies of B. terrestris reached 104.2 workers, 317.9 males and 21.1 queens, which corresponded to 1.1-1.8 fold those at B. ignitus. Therefore, above results showed that colony development of B. terrestris is superior to that of B. ignitus.

Effects of Gibberellic Acid and Alternating Temperature on Breaking Seed Dormancy of Panax ginseng C. A. Meyer (개갑된 인삼종자 휴면 조기타파에 미치는 GA3 및 변온처리 효과)

  • Background: Developing new ginseng cultivars is a significant time-consuming process owing to the three years of growth required for ginseng to flower. To shorten the ginseng breeding process, it is necessary to establish rapid progression through each generation. In this study, we examined it was possible to rapidly break ginseng seed dormancy using gibberellic acid ($GA_3$) treatment and alternating temperature. Methods and Results: Seeds were obtained from local variety. Seeds were treated with either $GA_3$ at a concentration of $100mg/{\ell} $, constant temperature ($-2^{\circ}C$ and $2^{\circ}C$), alternating temperature ($2^{\circ}C$ followed by $-2^{\circ}C$, followed by $2^{\circ}C$) or a combination $GA_3$ and temperature treatment. Following experimental treatment, seeds were sown into trays and placed in a greenhouse. Low germination rates were observed in seeds that did not receive $GA_3$ treatment, which were similar following $2^{\circ}C$ and $-2^{\circ}C$ constant temperature treatment. Germination rates increased in proportion to $GA_3$ and more so when combined with alternating temperature treatment. In additon, stem and leaf lengths of the resulting ginseng plants were increased following $GA_3$ treatment, although no synergistic effect was observed with alternating temperature treatment. Conclusions: These results suggest that a combination $GA_3$ and alternating temperature treatment enhances ginseng seed germination, which can contribute to shortening the time required to progress through a single ginseng generation for breeding.

Changes of asparagine content in potato (Solanum tuberosum L.) tuber during storage (저장 중 감자 아스파라진 함량의 변화)

  • In order to research changes of asparagine content according to stage after harvest to storage, it was analysed asparagine content of potato tuber and potato sprout. The results of the asparagine content in potato cultivars after harvest ranked in cv. Superior, cv. Gahwang, cv. Atlantic and cv. Goun in descending order. The content of asparagine in potato was approximately 1300 mg/100g (D.W.) The changes the asparagine content according to storage condition and the difference of asparagine between tuber and sprout can be summarized as follows. In the asparagine content of potato cultivars, Little variation in the content was observed while potato tubers were stored at $4^{\circ}C$. However, after 3 months of storage at $10^{\circ}C$, the content increased by 9.7%, indicating that the content increases as the storage temperature and period increases. The content of asparagine also increased during sprouting of potato tubers. In the sprout, the proximal region contained 21% higher content of asparagines than the distal region, which suggests that the sprout develops with the increasing of the content of asparagines.

Developmental Rate Equations for Predicting Bud Bursting Date of 'Campbell Early' (Vitis labrusca) Grapevines (발육 속도 모델을 이용한 포도 '캠벨얼리'의 발아기 예측)

  • To predict the bud bursting date of 'Campbell Early' grapevines, the bud developmental rate (DVR) models were constructed. The DVRs for bud bursting were calculated from the demanded times at controlled air temperatures. The DVRs were examined on the 'Campbell Early' grapevines incubated in three different temperatures at 4.6, 11.8, and $16.6^{\circ}C$. The DVR increased exponentially or linearly on the air temperature with a slope of about 0.0019. The DVR equations were computed as $DVR=0.0249+0.0020e^{0.1654x}$ or DVR = 0.0019x + 0.0187. These DVR equations offered developmental indices and predicted dates for bud bursting with air temperature data. The DVR equations were validated to the bud bursting data observed in the field. When bud bursting dates were calculated with daily temperature data, the root mean squared error (RMSE) between the observed and the predicted dates was less than 4 days. When those were calculated with hourly temperature data, on the other hand, the RMSE was less than 3 days. These results suggest that the DVR models are useful to predict bud bursting date of 'Campbell Early' grapevines.

Study on the Seed Germination Characteristics of Trapa japonica FEROV. (마름(Trapa japonica FEROV.) 종자의 발아특성에 관한 연구)

  • In this study, germination characteristics and effect of some chemicals on germination of Trapa japonica $F_{EROV}$ seeds were investigated to obtain a fundamental data for controlling them. When the seeds were stored at low temperature for at least 3 months, their dormancy was completely released with a germinability of above 90%. However, those stored under the room or high temperature condition were never germinated. Germination was best at constant temperature of around $25^{\circ}C$ or at alternate temperature of 25/$15^{\circ}C$ (12 hr-day/12 hr-night), and was nat dependent on light. They could germinate though planted at the depth of 15 cm, but their initial growth were: suppressed with increase of planting depth, especially above 10 cm depth. Germinability was lost when the fresh weight was decreased more than 30%, indicating that the seed is sensitive to dehydration. The freezing at $-20^{\circ}C$ for 1 hr didn't affect germination, but 5 hr treatment completely killed them. Their germination was inhibited by 100% at 100 ppm ABA and slightly inhibited above 25 ppm of butachlor, whereas pyrazosulfuron-ethylnt is likely to control T. japonica physically and herbicide treatment to inhibit germination may not be recommendatory. and bensulfuron-methyl didn't inhibit the germination at the concentration of 100 ppm. Taken together, dehydration treatme is likely to control T. japonica physically and herbicide treatment to inhibit germination may not be recommendatory.

Effects of GA3 and Alternating Temperature on Breaking Bud Dormancy of Panax ginseng C. A. Meyer Seedling (인삼 묘삼 휴면 조기타파에 미치는 GA3 및 변온 처리 효과)

  • Background : Considerable time and effort is required to develop new Panax ginseng varieties. Ginseng breeders have been developing techniques to shorten the breeding cycle to resolve this problem. In this study, we investigated the effects of adding $GA_3$ and alternating temperature (AT, $2^{\circ}C{\rightarrow}-2^{\circ}C{\rightarrow}2^{\circ}C$) on breaking bud dormancy in the varieties (Chungsun and Sunun) of ginseng root. Methods and Results : The $GA_3$ soaking treatment and AT were applied to one year old roots, which greatly accelerated the emergence of new buds. In one year old roots, new buds emerged from the 4th day post transplanting and after breaking dormancy with $GA_3$ and AT treatments. The emergence of new buds was completed within two weeks. The rate of bud emergence for Chungsun was 60% - 98% over 15 - 60 days after the AT and $GA_3$ treatments. The emergence rate of Sunun was 46% - 92%. Normal growth of the ginseng seedling was observed in spite of the early breaking of bud dormancy by combined $GA_3$ and AT treatments. Conclusions : $GA_3$ and AT treatments shortened the dormancy period and facilitated the stable emergence of ginseng seedlings. However, some plants suffered deformities and early sprouting owing to the combined $GA_3$ and AT treatments. Early sprouting was free from dormancy after leaf fall from the of aerial part of the plant.

Studies on Growth Characteristics and Yield of Solanum njgrum L. (까마중(Solanum nigrum L.)의 생육특성과 전초수량에 관한 연구)

  • This study was carried out to effect on Growth Characteristics and Yield of Solanum njgrum L. The experimental results are summarized as follows. Germination, growth characteristics and yield of Solanum nigrum Linne collected from southern parts of Korea were investigated. The one fruit weight was the heaviest in Gochang traditional S. nigrum among the collected sites. The number of seeds in one fruit was high in the orders of Gochang traditional sites, Gwangju traditional sites, Suncheon traditional sites and Naju traditional sites. The earliest flowering of Solanum nigrum was observed on 10. August. The plant height, stem diameter, leaf numbers and branch numbers were ranged from 65 cm to 75 cm, 7.1 mm to 8.2 mm, 237 ea to 270 ea and 12 ea to 17 ea, respectively. The fresh weight per plants were ranged from 265 g to 279 g, and yield of biomass were ranged from 1,298 kg to 1,367 kg per 10a. The insect pest numbers of S. nigrum according to the collected sites were showed no significant in this test.

Effect of Soil Temperature on the Emergence - Speed of Rice and Barnyardgrasses under Dry Direct - Seeding Condition (토양온도(土壞溫度)가 벼와 피의 출아속도(出芽速度)에 미치는 영향(影響))

  • Seeds of rice, cv. Ilpoom, and barnyardgrasses(Echinochloa crus-galli, vars. oryzicola, crux-galli, and praticola) were sown for a characterization of their responses to temperature during emergence under a dry direct-seeded condition. A laboratory-made aluminum block apparatus for emergence-temperature control conferred a linear continuous temperature gradient from 10 to $30^{\circ}C$ to the seeds from cooling to heating ends of the apparatus. The lowest temperature for emergence was $12.3^{\circ}C$ for rice cv. Ilpoom, and $11.0^{\circ}C$ for the three varieties of Echinochloa spp.. Percent emergence of rice increased sharply with an increase in temperature by ca. $20^{\circ}C$, then leveled-off, while those of barnyardgrasses increased almost linearly with temperatures up to $30^{\circ}C$. In rice the time required for emergence after seeding was shortened exponentially with increased temperature while those for barnyardgrasses were shortened almost linearly from 11 to $30^{\circ}C$. The temperature-response characteristic of rice in emergence-speed was almost the same among those for the 1st emergence, emergence by 25, 50, 75%, or average emergence time. At $13^{\circ}C$, $346.7^{\circ}C$ days of accummulated temperature(26.67 days) were required for the 1st emergence in rice while 131.7, 136.0, and $138.7^{\circ}C$ days(10.13, 10.46, and 10.67 days) were required for the 1st emergence in E. spp., vars. crus-galli, praticola, and oryzicola, respectively. Greater cold tolerance and increasingly faster emergence of barnyardgrasses than rice below $20^{\circ}C$ seem to render the barnyardgrasses as much more competitive than rice at lower temperatures.
