Comparison of Volatile Terpenoid Content from Carrot Cultured Area and Carrot Portions (당근의 재배지역별 부위별 휘발성 Terpenoids 함량 비교)

  • Using direct headspace sampling (DHS) method, volatile terpenoid content was measured according to carrot cultured areas and various portions of carrot. In comparison of volatile terpenoid content in xylem and phloem of carrot, every volatile terpenoid was distributed highly in the xylem rather than the phloem of carrot. The carrot was divided into three different sections; upper (crown), middle (midsection), and lower part (tip), and the volatile terpenoids were measured. The content of ${\alpha}-pinene,\;{\beta}-pinene,\;{\beta}-myrcene,\;and\;{\gamma}-terpinene$ tended to be distributed highly in the upper part rather than the lower part, whereas the content of terpinolene and total terpenoids tended to be high in the lower part. In comparison of volatile terpenoid content according to the carrot cultured areas, volatile terpenoid content of carrot cultured in Cheju island was higher than Yangsan and Haenam areas. In case of total terpenoids, the carrot clutivars cultured in Cheju island contained $1.6{\sim}2.3\;times$ higher than those of Yangsan, and $1.5{\sim}2.4\;times$ higher than Haenam. In order to investigate the above results, we sampled out soils from the carrot fields and measured physical and chemical characteristics of the soils. The pH did not show much difference in the three areas. However the organic carbon, total nitrogen, cation exchange capacity, and total exchangeable cation in soil from Cheju were $2.4{\sim}3.0\;times$ higher than Yangsan and $3.9{\sim}7.1\;times$ higher than Haenam. The physical characteristics of the soils was investigated. The soil from Cheju island was found to be loam; Yangsan, sandy loam, and Haenam, clay loam.

Condensable Gas Separation using Phenol! Alumina Composite Activated Carbon Hollow Fiber Membranes (페놀수지/알루미나 복합 활성탄소중공사막을 이용한 응축성 기체 분리)

  • Carbon membrane materials have received considerable attention for the gas separation including hydrocarbon mixture of ingredients of the volatile organic compounds(VOCs) because they possess their higher selectivity, permeability, and thermal stability than the polymeric membranes. The use of activated carbon membranes makes it possible to separate continuously the VOCs mixture by the selective adsorption-diffusion mechanism which the condensable components are preferentially adsorbed in to the micropores of the membrane. The activated carbon hollow fiber membranes with uniform adsorptive micropores on the wall of open pores and the surface of the membranes have been fabricated by the carbonization of a thin film of phenolic resin deposited on porous alumina hollow fiber membrane. Oxidation, carbonization, and activation processing variables were controlled under different conditions in order to improve the separation characteristics of the activated carbon membrane. Properties of activated carbon hollow fiber membranes and the characterization of a gas permeation by pyrolysis conditions were studied. As the result, the activated carbon hollow fiber membranes with good separation capabilities by the molecular size mechanism as well as selective adsorption on the pores surface followed by surface diffusion effective in the recovery hydrocarbons have been obtained. Therefore, these activated carbon membranes prepared in this study are shown as promising candidate membrane for separation of VOCs.

Quality Properties of White Lotus Leaf Fermented by Mycelial Paecilomyces japonica (동충하초 균사체로 발효시킨 백련잎차의 품질특성)

  • Quality characteristics of white lotus leaf tea (LLT) fermented with or without mycelial Paecilomyces japonica were investigated. Extraction yield and browning index of hot water extract from non fermented and fermented LLTs were higher than those of ethanol extract (p<0.05). In all LLTs, nutritional components such as total free sugar, free amino acids and minerals of hot water extracts were higher than those of ethanol extracts except for total organic acids (p<0.05). Contents of total free sugar and organic acids were markedly increased through fermentation process of mycelial Paecilomyces japonica. in the same solvent extracts (p<0.05). Contents of most taste components of fermented LLT were increased by mycelial solid fermentation (p<0.05), but total free amino acids of two extracts were decreased in the range of $37.1{\sim}67.2%$ as compared to non-fermented LLT. Fifty-nine volatile compounds were identified by GC and GC-MS, including 11 aldehydes, 14 alcohols, 11 ketones, 11 hydrocarbones and 12 acids. Aldehyde and ketone compounds were more identified in fermented LLT than in non-fermented LLT being abundant alcohol compounds by simultaneous steam distillation and extraction. The most abundant compounds of LLT identified in this study were curcumene followed by 2,6-bis(1,1-dimethylethyl)-4-methyl-phenol and cyclohexen. Main compounds of fermented LLT were 2,6-bis(1,1-dimethylethyl)-4-methyl-phenol, butanoic acid, furfural, benzaldehyde, hexanoic acid and 2(3H)-furanone.

Quality characteristics of distilled spirits by different nuruk-derived yeast (누룩 유래 효모의 종류에 따른 증류주의 품질 특성)

  • This study aimed to select a yeast strain for optimizing the quality of distilled spirits. The brewing and distilling properties of 4 KFRI (Korea Food Research Institute) yeasts (Y88-4, Y98-4, Y172-6, Y192-4) and 2 industry yeasts (C1, C2) were compared. For investigating the possibility of using these strains on an industrial scale, diverse analytical methods were applied to assess parameters associated with distilled spirit quality such as alcohol content, pH, total acidity, and soluble solid content. After 11 days of fermentation, the alcohol strength obtained using six yeast strains reached 13.9-16.4% (v/v), while pH was 3.9-4.0, and total acid was 0.40-0.52%. To compare GC-MSD Volatile flavor components, all the distilled spirit samples were diluted to 20% (v/v) alcohol strength. Seven fusel alcohols, 26 esters, 2 acids, and 3 miscellaneous compounds were detected in the distilled spirits. Y88-4 had the most abundant volatile flavor component and scored the highest overall preference in sensory evaluation. After analyzing the various properties of yeasts, strain Y88-4 was finally selected as the best strain for producing distilled spirits.

Analysis of volatile compounds and metals in essential oil and solvent extracts of Amomi Fructus (사인으로부터 추출한 정유와 용매 추출물의 휘발성 물질 및 금속성분 분석)

  • Amomi Fructus with anti-oxidative activity was chosen and essential oil was obtained by SDE (simultaneous distillation extraction), and 39 constituents were determined by GC-MS (gas chromatography-mass spectrometry). Major components were camphor, borneol acetate, borneol, D-limonene and camphene. Three solvent extracts such as hexanes, diethyl ether and methylene chloride from Amomi Fructus were obtained. These were analyzed by GC-MS and 4 more constituents were identified in addition to 39 components discovered in essential oil. Five major components such as camphor, borneol acetate, borneol, D-limonene and camphene were also detected, however the relative peak percents of those components were different from those of constituents in essential oil. To estimate the kind and the amount of materials evaporated at certain temperature and conditions from essential oil and solvent extracts, dynamic headspace apparatus was used and materials evaporated and trapped at certain conditions were analyzed by GC-MS. Recovery yield of SDE method from Amomi Fructus was measured by using camphor and standard calibration solution of camphor methanol solution and, the yield was 82.0%. Content of Hg was measured by mercury analyzer and contents of Cd, Pb, Cr, Mn, Co, Ni, Cu and Zn in Amomi Fructus, essential oils and solvent extracts were determined by ICP-MS (Inductively coupled plasma-mass spectrometer). Pb, Cd and Hg were measured in the concentration of 0.72 mg/kg, <0.10 mg/kg and 0.0023 mg/kg, respectively and these were below permission level of purity test. Contents of Mn, Cu and Zn in Amomi Fructus were 213 mg/kg, 8.29 mg/kg and 31.0 mg/kg, respectively and which were relatively higher than other metals such as Cr, Co and Ni. Metals such as Mn (0.65 ~ 9.08 mg/kg), Cu (1.16 ~ 4.40 mg/kg) and Zn (1.10 ~ 3.80 mg/kg) in essential oil and solvent extracts were detected. At this point it is not clear that the metals were cross-contaminated in the course of treating Amomi Fructus or metals were contained in Amomi Fructus. The influence evaluation toward biological model study of these metals in essential oil and solvent extracts will be needed.

Characterization of Ethanol Fermentation with Wild Type Yeast Strains (야생 효모 종류에 따른 알코올 발효 특성)

  • The objective of this study was to improve the quality of Korean rice wine with wild type yeast strains isolated from various traditional Korean fermented foods. Herein the fermentation and sensory characterization of wild yeast, for the purposes of brewing Korean rice wine, was investigated. 12 yeast strains were examined for their ethanol and glucose tolerance. In addition, the pH, soluble solids, acidity, amino acidity, ethanol content, organic acids, and volatile compounds were also studied for the alcoholic beverages made with the wild yeasts. Almost all Saccharomyces genera yeasts were showed to have a tolerance at 10% ethanol, but non-Saccharomyces genera yeasts displayed a low tolerance. The alcoholic beverages fermented by non-Saccharomyces yeasts demonstrated higher levels of soluble solids, titratable acidity, amino acids, and lower ethanol content, when compared with the alcoholic beverages fermented by Saccharomyces genera yeasts. The organic acid content, such as malic acid, acetic acid, and succinic acid, was seen to also be higher. The electronic nose was analyzed, and discriminant function analysis (DFA) was used for discriminating wild yeast strains. The DFA plots indicated a significant separation of Saccharomyces genera and non-Saccharomyces yeast strains. For volatile compounds, ethyl acetate from non-Saccharomyces yeasts, and ethanol from Saccharomyces genera yeast, a high area ratio was observed.

Thermodynamic Studies on the Adsorption of 4-Octylphenol on Carboxen by GC/MS Analysis (GC/MS 분석에 의한 4-Octylphenol의 Carboxen 흡착에 대한 열역학적 연구)

  • It is common to analyze volatile organic compound (VOC) or semi-VOC (SVOC) in a sample composed of a complex matrix consisting of multiple components such as bloods through a separation process. Adsorption is a physical phenomenon in which certain components accumulate on the surface of other phases. In order to overcome difficulties in the pretreatment process, an adsorption is frequently used. Solid phase microextraction (SPME) equipment with porous carbon carboxen (CAR) is an example of adsorption application. In this study, the adsorption of 4-octylphenol to carboxen was examined. To do so, the extraction efficiency for such solvents as dichloromethane ($CH_2Cl_2$, DCM), ethylacetate ($CH_3COOC_2H_5$, EA) and diethylether ($C_2H_5OC_2H_5$, $Et_2O$) was studied and also the derivatization reaction for 4-octylphenol with reagents of bistrimethylsilyltrifluoroacetamide (BSTFA), methylchloroformate (MCF) and pentafluorobenzylbromide (PFBBr) was compared. The combination of DCM and BSTFA showed good performance thus they were adopted for this study. Thermodynamic adsorption experiments showed that the adsorption process was endothermic and Freundlich isotherm equation was more suitable than Langmuir isotherm. It was also found that the adsorption followed a pseudo-$2^{nd}$ order kinetic model.

Biochemical analysis and physiological activity of perilla leaves (들깨잎의 품종에 따른 성분분석 및 생리활성물질 탐색)

  • The biochemical components of Namcheondlggae, Miryangdlkkae 25, Boradlggae and Ipdlkkae 1 were measured. The samples were extracted with hot water, 60% acetone or 80% ethanol for screening physiological activity. The crude protein content (4.36%) was found in the Miryangdlkkae 25 and calcium content (497.5 mg%) was found in the Namcheondlggae among the tested 4 perilla loaves. Fructose was 30.86 mg% in the Namcheondlggae and free amino acids at all perilla leaves was detected seventeen. In Boradlggae, glutamic acid and alanine were 25.37 and 11.91 mg%. Totally nine non-volatile organic acids were also detected and the contents of malic acid and glutaric acid were 28.34 and 14.57 mg% in Boradlggae. The Miryangdlkkae 25 had the highest vitamin C amount which was 113.24 mg%. Angiotensin converting enzyme (ACE) inhibition activity of 60% acetone extract of Miryangdlkkae 25 was 39.20% when added as addition of 200 ppm level and xanthine oxidase inhibition activity of 80% ethanol extract of Boradlggae was 46.71%. Electron denoting activity of 60% acetone extract from Namcheohndlggae was the strongest inhibition activity as 98.19% when 200 ppm level of the sample extracts were added.

원료배합을 달리한 무증자 탁주의 성분함량 및 관능평가

  • 우리 전통주의 질적인 향상과 저장성이 우수한 무증자 발효주를 제조하기 위하여 쌀과 옥분의 배합 비율을 쌀 100%, 쌀 80%+옥분 20%, 쌀 60%+옥분 40%, 쌀 40%+옥분60%, 쌀 20%+옥분 80%로 달리하여 무증자로 담금한 발효주와 쌀 100%로 증자하여 발효시킨 주류의 물리화학적인 변화 및 성분함량을 측정하였으며 관능검사를 병행하여 비교 검토하여 다음과 같은 결론을 얻었다. 1) 술덧의 온도변화는 발효 초기 30 에서 시간의 경과에 따라 서서히 감소하여 발효중기 27.5 를 거쳐 발효 종료 시 모두 26.5 였다. 2) 술덧의 주정도 변화는 옥분의 양을 첨가할수록 낮아지는 경향을 보였으며 16∼18.3v/v% 였다. 3) 유기산은 무증자주는 쌀 100%일 때 0.6 g/dl로 증자주 0.49g/dl보다 높았으며 옥분을 첨가한 무증자주에서는 옥분이 첨가될수록 차즘 감소하여 80%일 때 0.34 g/dl 였다. 4) 유리아미노산 함량은 옥분이 40% 첨가되면서부터 leucine의 함량이 쌀 100% 증자주에 비해 2배 이상 증가하였으며 leucine>arginine>lysine의 순이었다. 5) 유리당 함량은 무증자로 옥분을 첨가할수록 증가하여 옥분 80%에서는 9.81mg/dl였다. 6) 휴젤유는 278.6ppm∼441.6ppm으로 쌀100% 무증자에 비해 옥분이 첨가될수록 감소하여 옥분 80%일때 가장 적은 278.6ppm이었다. 7) 휘발성 향기성분중 ester함량은 100% 무증자주에서 총 9종류가 검출되었으나 옥분이 첨가되면서 10∼12 종으로 많아졌다. Ethyl octanoate는 옥분 60%첨가 무증자주에서 19.26%로 많이 나타났으며 ethyl acetate는 옥분이 첨가되면서 증가하였다. 특히 옥분 80% 무증자주는 총 ester류함량은 적었으나 isoamyl acetate, phenylethyl acetate, ethyl octanoate, ethyl acetate의 함량이 비교적 많이 검출되어 독특한 향기를 기대할 수 있었다. 8) GC/O를 사용하여 향기성분을 동정한 결과 무증자 발효주 시험구에서는 옥분을 첨가할수록 sweet 및 warm note가 강하게 나타나는 현상을 보였다. 9) 발효주의 색도를 측정한 결과 밝기에서는 무증자구가, 적색도에서는 옥분 첨가량이 증가할수록 강하였으며 황색은 증자주보다는 무증자주의 옥분 첨가량의 증가에 따라 높아지는 경향을 보였다. 10) 관능검사 결과 쓴맛, 미묘한 맛, 좋은 맛, 떫은 맛, 신맛 그리고 좋은 맛에 대한 전체 적인 평가를 종합하면 쌀 100% 증자발효주가 가장 점수가 낮아 무증자 발효주에 비하여 기호도가 떨어지는 것으로 나타났으며 특히 옥분 첨가량 80%와 60%의 무증자주가 기호도 면에서 유의적으로 가장 높게 평가되었다.

Studies on the Taste Properties in Processing of Accelerated Low Salt-Fermented Anchovy by adding koji (코오지를 이용한 속성 저식염 멸치젓의 맛 특성)

  • To produce low salt fermented anchovy by an accelerated method with Asp. oryzae and Bacillus sp. koji and taste properties after the 60 day fermentation were examined. The main free amino acids of 60 day fermented anchovy paste were valine, isoleucine, proline, alanine, lysine, glutamic acid and aspartic acid. Total amount of free amino acids was the highest in non koji anchovy paste wit 2,624.76mg%. Among the koji added samples, Asp. oryzae koji added on was the highest in the amount o free amino acids. Hypoxanthine accounted for 84.14~95.4% of total nucleotides and their related compounds; Asp. oryzae koji added anchovy paste was the highest in nucleotides other related. Citric acid and lactic acid accounted for 94.9~96.7% of total non-volatile organic acids; Asp. oryzae koji added sample was the highest in non-volatile organic acids with 287.93mg%. The Hunter a and b values gradually increased during the fermentation, but the L value decreased until day 30 or 40 and increased steadily after that. The a and b values were higher in the use of Asp. oryzae koji than in Bccillus sp. koji, but the L value was to the contrary. The Asp. oryzae koji added anchovy paste was good in the aspect of color and taste compared to others. In the aspect of odor, the anchovy paste using the mixture of Asp. oryzae and Bacillus sp. koji was the best. Overall aceptability of sensory evaluation was higher in the mixture of Asp. oryzae and Bacillus sp. koji tan in the others.
