Development of a New Disinfestation Method Using Methyl Bromide and PH3 in Cut Rose Fumigation (절화장미 훈증처리시 Methyl Bromide와 PH3를 이용한 새로운 살충방법의 개발)

  • Fumigation by combination of methyl bromide/phosphide ($MB/PH_3$) was much more effective in disinfestating the rose insect pests (Aphis gossypii, Tetranychus urticae, and Frankliniella occidentalis) than that by individual MB or $PH_3$. Fumigation for 2 hours in the chamber using ;$MB/PH_3$ at the concentration of $10/3g{\cdot}m^{-3}$ brought about 91.3% and 100% mortality in aphids and thrips, respectively. Fumigation for 3 hrs using ;$MB/PH_3$ at the concentration of 5/3 or $7/3g{\cdot}m^{-3}$ showed over 96% mortalities in mites and thrips as well as no phytotoxicity symptoms, e.g. tip burn or bent neck, in cut roses, while mortalities of mites and thrips were not influenced by increasing $CO_2$ concerntration from 0.03% to 10% or decreasing $O_2$ concentration up to 15% in fumigation chamber.

Evaluation of Phosphine Efficacy on Anoplophora chinensis and A. glabripennis in Export Bonsai (수출분재 주요 해충인 알락하늘소와 유리알락하늘소에 대한 포스핀 훈증 효과 검정)

  • Anoplophora chinensis and A. glabripennis is classified as important quarantine Cerambycidae beetles worldwide. These beetles cause serious damage on natural host plants and export bonsai trees in Korea. We evaluated efficacy of phosphine fumigant for control of two Cerambycidae beetles in bonsai. In the preliminary test to achieve CT (Concentration x time) values of phosphine to Cerambyicdae beetles larvae, we used Monochamus alternatus of larvae as an alternative insect. A 24 h phosphine fumigation with $2g/m^3$ phophine achieved 100% eggs mortality against Anoplophora chinensis and A. glabripennis. Larva of A. chinensis was also completely controlled when treated for 168-h at same dose desiccator. 100% mortality on larval stage of two beetle species were achieved both in dormancy (January) and vegetative period (February) of bonsai when treated with $2g/m^3$ phosphine. However, phosphine fumigation against vegetative period of Acer palmatum, Carpinus turczaninowii and Malus sieboldii bonsai showed slight phytotoxicity (shorten leaf and flower size and delayed flowering).

A New Disinfestation Approach Against Some Greenhouse Pests Using Ethyl Formate Fumigation (훈증제 에틸포메이트를 이용한 몇 가지 시설하우스 해충에 대한 새로운 방제 전략)

  • Ethyl formate (EF) is a rapid kill, environmentally safe, and low mammalian toxicity fumigant, registered to disinfest quarantine insect pests from imported agricultural products. A new concept for controlling insect pests of agricultural crops was tested in a fumigation chamber with EF. Control efficacy of and phyto-toxicity due to EF fumigation were evaluated against four pests (Thrips palmi, Bemisia tabaci, Myzus persicae, and Tetranychus urticae) and on seedlings of four fruit vegetables (FVs; yellow melon, cucumber, tomato, and pepper). Ethyl formate fumigation at a dose of 1.5 g m-3 for 12 h produced >93.3% mortality in T. palmi, B. tabaci, and M. persicae. However, T. urticae was tolerant to fumigation, showing only 20% mortality at 2.0 g m-3. In terms of concentration × time (CT) products, at least 8.9 g·h m-3 CT at 20 ± 1.5℃ was needed to achieve > 90% mortality against the three susceptible insect pests. Fumigation at 1.5 g m-3 for 12 h caused no phyto-toxicity to any of the four FV seedlings. Ethyl formate application, as a new disinfestation method in greenhouses, could be an alternative to reduce the use of conventional insecticides. However, further studies are needed to determine the efficacy of this method at different pest developmental stages and in different greenhouse environments. Additionally, research is needed to elucidate the phyto-toxicity of EF application at different growing stages of a wide variety of crops.

Comparative Effects of Gamma Irradiation and Methyl Bromide Fumigation on Disinfestation and Physicochemical Quality of Acorn (감마선과 Methyl Bromide 처리가 도토리 종실의 해충사멸과 이화학적 품질에 미치는 영향)

  • Comparative effects of gamma irradiation and methyl bromide (MBr) fumigation on disinfestation and some physicochemical attributes of acorn seeds were investigated. Insects in domestic acorns were identified to be Curculio dentipes Roelofs and Dichocrocis punctiferalis Guenee, which were easily disinfestated immediately after commercial MBr fumigation. Irradiation also showed similar effects on mortality of the insects in different post-irradiation periods; at 3rd day with doses of 1-3 kGy and at around 30th day 0.5-1 kGy. MBr fumigation caused the decrease in lightness (Hunter L value), total phenolics and reducing sugars and the increase in yellowness (Hunter b value) and browning intensity for the subjected samples, as compared with the nontreated control and irradiated ones at 0.5-2 kGy. Furthermore, MBr fumigation resulted in decay for the most part of samples during storage at 5-10 $^{\circ}C$ for 6 months in a PVC box packaging with nets. As a result, optimum levels of irradiation is expected to be roe of the alternatives to chemical fumigants.

Sanitary Sterilization of Dried Fishes and Mixed Condiments (주요건조 수산물 및 혼합조미료의 위생적 살균)

  • 건조 수산가공품 및 혼합조미료 각 5종의 위생적 살균 및 저장방법의 개발을 목적으로 현행살균 방법으로 살균효과의 불충분, 제품의 품질저하, 약제성분의 잔유 등 많은 문제점을 내포한 ethylene oxide에 의한훈증처리와 방사선 조사와의 미생물 살균효과, 이화학적 특성 및 관능적 품질에 미치는 영향을 검토한 결과는 다음과 같다. 시료의 미생물 오염은 전 세균이 $10^3~10^7$ CFU/g으로 그중 90%이상이 내열성 세균이었으며, 곰팡이류는 $10^2~10^3$ CFU/g, 대장균도 시료 5종에서 $10^2~10^6$ CFU/g이나 오염되었다. 살균효과 실험에서 대장균군, 곰팡이류는 5~10 kGy 방사선 조사로서 완전 사멸되었으며, 전세균도 오염도가 매우 높았던 혼합조미료 몇종을 제외하고는 제거되었다. 반면, ethylene oxide훈증처리는 전 미생물의 살균이 불충분하였다. 이 화학적 특성 즉, TBA가 TMA-N 함량, 아미노산, 무기질, 색도 등은 적정 선량의 방사선 조사로서는 무처리구 시료와 거의 차이가 없었으나, ethylene oxide 훈증처리구는 심한 품질의 변화를 나타내었다. 또한 관능검사에 의한 전반적 기호성에 있어서도 방사선조사구가 무처리구 및 ethylene oxide 처리구보다 유의적으로 우수함을 나타냈다.

  • This experiment was carried out to investigate control efficacy by cultural practice and fumigation of tacterial wilt caused by Pseudomonas solancearum in resistant tobacco line NC82 at the Korea Ginseng and Tobacco Research Institute, Eumseong Experiment Station in 1982 and 1983. The bacterial wilt of tobacco occurred severely from mid-July to last August in applicable temperature and soil humidity for increasing bacteria. Disease severity appeared low and slowly at fumigation and resistant variety treatment. Incidence of bacterial wilt in tobacco line NC82 was $44.7\~55.8\%$ being compared with susceptible variety, NC2326 and $95\~99\%$ when resistant variety, NC32 was cultivated with soil fumigation treatment. Control efficacy of cultural practices appeared low with $0.8\~20\%$ and was not different from resistant variety and fumigation treatment. Control system against bacterial wilt in flue-cured tobacco was accomplished by control efficacy over $95\%$ when resistant variety(NC82) was cultured after treatment of cultural practices (Tillering after harvest and before transplanting, stalk and root destruction, early transplanting early removal of the mulching film) and soil fumigation(Cylon).

Bactericidal Efficacy of Fumagari OPP®, Fumigant Against Escherichia coli and Salmonella typhimurium (훈증소독제, Fumagari OPP®의 Escherichia coli와 Salmonella typhimurium에 대한 살균효과)

  • This test was performed to evaluate the bactericidal efficacy of Fumagari OPP$^{(R)}$, fumigation disinfectant, containing 20% ortho-phenylphenol against Escherichia coli (E. coli) and Salmonella typhimurium (S. typhimurium). In preliminary tests, both E. coli and S. typhimurium working culture suspension number (N value) was $4.0{\times}10^8$ CFU/mL. And all of the colony numbers on the carriers exposed the fumigant (n1, n2, n3) were higher than 0.5N1 (the number of bacterial test suspentions by pour plate method), 0.5N2 (the number of bacterial test suspentions by filter membrane method) and 0.5N1, respectively. In addition, the mean number of bacteria recovered on the control-carriers (T value) was $3.4{\times}10^6$ CFU/mL. In the bactericidal effect of the fumigant, the reduction number of S. typhimurium and E. coli (d value) was 5.26 and 5.64 logCFU/mL, respectively. According to the French standard for the fumigant, the d value for the effective bactericidal fumigant should be over than 5 logCFU/mL. With the results of this study, Fumagari OPP$^{(R)}$ has an effective bactericidal activity, then the fumigant can be applied to disinfect food materials and kitchen appliances contaminated with pathogenic bacteria.

Microbiological changes and quality characteristics of dried persimmon by chlorine dioxide gas fumigation treatment (이산화염소 가스 훈증 처리에 따른 곶감의 미생물학적 변화 및 품질특성)

  • This study was conducted to investigate the effect of chlorine dioxide fumigation as a substitute for sulfur fumigation which has been used as a method to prevent the quality change of persimmon during storage and distribution process. Dried persimmons were treated with chlorine dioxide gas concentration (0, 15 30, and 45 ppm) and time (0, 15, 30, and 45 min) and microbiological changes, texture properties and color of the treated samples were investigated during storage at room temperature. Total aerobic bacteria, yeast and mold numbers after chlorine dioxide gas fumigation were decreased when compared with the control group. The inhibitory effects of total aerobic bacteria, yeast, and mold were observed during storage. The texture properties and color value of dried persimmons were not affected by chlorine dioxide gas fumigation concentration and time. There was no difference in quality between chlorine dioxide gas fumigation treatment group and control group. These results suggested that chlorine dioxide gas fumigation treatment can be utilized as a processing technique to secure microbiological storage stability of dried persimmons.

Control of Powdery Mildew (Uncinula necator) in Vineyards by Spraying and Vapor-Action Treatments of Triazole Fungicides (Triazole계 살균제의 살포 및 훈증처리에 의한 포도 흰가루병(Uncinula nectator)의 방제)

  • 비가림재배 포도원에서 발생하는 흰가루병에 대하여 triazole계 살균제 flusilazole, myclobutanil, difenoconazole, penconazole, triflumizole, triadimefon의 방제 효과를 검정한 결과는 다음과 같다. 살균제 처리구의 이병과방율과 이병엽율은 무처리구에 비해 현저히 감소하였고, 이들의 방제가는 과방에서 약제에 따라 94.7∼97.9%, 잎에서 85.5%∼90.9%였으며 대조약제인 polyoxin B의 방제가와 유사한 정도를 보였다. 공시살균제의 보호효과는 살균제를 병발생 1주일전에 살포하는 경우, 살포 3주 후에 방제가는 약제에 따라 72.2∼90.5%였으나 4주 후에는 21.2∼41.6%로 감소하여 대조약제 polyoxin B의 52.5%보다 감소시켰으나 myclobutanil의 발아억제 효과는 약제 농도증가에 크게 영향받지 않았다. 살균제에 침지한 cheesecloth를 포도원 선반에 매달아 조사한 공시살균제의 훈증효과는 살포농도의 2배로 처리했을 때 처리점으로부터 반경 30cm이내의 포도송이에서는 약제에 따라 57.2∼71.1%의 방제가를 보였으나 전체 식물체에서는 49.1∼65.8%로서 10일 간격으로 3회 살포한 것 보다 낮은 방제가를 보였다.

  • 건고추는 대부분 세척이나 살균의 과정을 거치지 않고 저장, 유통되므로 해충이나 미생물 오염에 의한 식품위생면이나 상품가치면에서 문제가 되고 있다. 고춧가루의 미생물과 해충 방제를 위하여 ethylene oxide나 propylene oxide 등의 훈증제가 사용되고 있으나 문제점이 많아 점차 사용이 금지되고 있는 실정에 있다. ${\gamma}$-선 조사는 훈증제, 자외선 조사, 초단파 처리등 기존의 방법에 비해 살균 효과는 물론 식품의 품질과 건전성 실험 결과에서도 우수하다고 보고되었다. (중략)
