Basic Studies on the Development of Diet for the Treatment of Obesity I. The Inhibitory Effect of Alginic Acid as a Dietary Fiber on Obesity (비만치료식 개발을 위한 기초연구 1. 식물섬유로서의 알긴산의 비만억제효과)

  • This work was designed as basic studies on the development of diet for the treatment of obesity. The effect of alginic acid which is the major component of brown algae was investigated by comparing with agar, cellulose, carboxymethylcellulose (CMC), chitin, and lignin as dietary fibers. These dietary fibers ($10\%$) supplemented to basic diet were fed ad libitum to Sprague Dawley rat for 4 weeks, and the inhibitory effects on obesity were evaluated by measuring body weight gain and feed efficiency, the content of glucose and cholesterol in serum, lipase activity in serum, lipid content in liver, adipose tissue around epididymis and ovarium, and Lee index etc. Among the inhibitory effect of these dietary fibers on obesity, lignin was the most effective (p<0.001), followed by Na-alginate (p<0.01) for body weight gain, but lignin was the most effective, followed by CMC, and followed by Na-alginate for feed efficiency (p<0.001). In the inhibitory effects on lipid accumulation in liver and adipose tissue around epididymis and ovarium, lignin and Na-alginate group among these dietary fibers were more effective than others, but there was no significant difference between male and female. The inhibitory effect on obesity evaluated by Lee index was effective in the order of Na-alginate>lignin>CMC>chitin>cellulose (p<0.001). The decreasing effects of lipid content in liver by dietary fibers were found in the order of agar>CMC>cellulose>Na-alginate>chitin>lignin (p<0.001). Glucose content in serum was significantly decreased in cases of CMC, lignin and Na-alginate, whereas a slight difference was found in chitin, but cholesterol content in serum was decreased for all dietary fibers examined except cellulose group. The increasing effect of lipase activity in plasma was found in cases of Na-alginate and chitin, while cellulose, CMC and lignin groups were decreased.

Predicting the Nutritional Value of Seafood Proteins as Measured by Newer In Vitro Model 2. C-PER and DC-PER of Marine Crustacea (수산식품 단백질 품질평가를 위한 새로운 모델 설정 2. 해산 갑각류의 C-PER 및 DC-PER)

  • To confirm the application of a newer in vitro assays to determining the nutritional value of marine crustaceans (mainly shrimps and crabs), which have been considered to be highly nutritive depending on their levels of the essential amino acids and digestibility, their C-PERs and DC-PERs were determined and studied the factors influencing their in vitro results. Four species of seawater shrimps and 2 species of seawater crabs were used in this experiment. The in vitro digestibilities showed $83{\sim}86\%$ for raw shrimps and the trypsin indigestibile substrate content (TIS) was ranged from 1.32 to 3.33 mg/g solid expressed quantitatively as mg of purified soybean trypsin inhibitor. The smaller size of shrimps revealed a greater in vitro digestibility and a lower contents of TIS. It was noted that the in vitro digestibility of raw blue crab meat was around $85\%$ while boiled tenner crab meat showed $86\%$ or above, and the leg meat had the greatest in vitro digestibility in the various parts of crab meats. The poor in vitro digestibilities for shrimp's and crab's meat, compared with that of the other seafoods as noted in previous reports, suggest that the drop in pH, due to the change in their freshness during harvesting and frozen storage, resulted in underestimating their digestibilities using four-enzyme digestion technique. The lysine contents in all samples were higher than that of ANRC casein but they contained a slightly lower sulfur-containing amino acids than those in ANRC casein. But the other EAA, such as valine, tyrosine and phenylalanine, were found to be a half as little as that in casein and played a key-factor in calculation of C-PER or DC-PER. It was observed that the value of C-PER and DC-PER for all samples ranged from 2.1 to 2.4, and the predicted digestibilities showed $90\%$ or above in all samples. It was a different results from the fact that the animal proteins bear a higher values and predicted digestibilities than those of C-PER values. The lack of correlation between C-PER and DC-PER values is attributable to the fact that the lower content of valine, tyrosine and phenylalanine, and drop in pH owing to the changes of freshness in marine crustacea proteins. Therefore, if a newer in vitro digestion technique-which are taken into account the pH drop before digestion, TIS content and released free amino acids and/or peptides-developed, C-PER assays can provide more advantages in assessing the protein nutritional value of marine crustacea than any other in vitro assays.

Removals of 1-Naphthol in Aqueous Solution Using Alginate Gel Beads with Entrapped Birnessites (버네사이트를 고정화한 알긴산 비드(Bir-AB)를 이용한 수용액 중 1-Naphthol의 제거)

  • In this study, alginate beads containing birnessite (Bir-AB), a highly reactive oxidative catalyst for the transformation of phenolic compounds, was prepared and its 1-naphthol (1-NP) removal efficiency was investigated in a batch test. Based on scanning electron microscopy image, it can be inferred that the alginate gel cluster acts as a bridge which bind the birnessite particles together. Kinetic experiment with Bir-AB of different mixing ratios of birnessite to alginate (Bir : AG=0.25 : 1~1 : 1 w/w) indicate that pseudo-first order kinetic constants, $k(hr^{-1})$ for the 1-NP removals increased about 1.5 times when the birnessite mixing ratio was doubled. The removals of 1-NP was found to be dependent on solution pH and the pesudo-first order rate constants were increased from 0.331 $hr^{-1}$ at pH 10 to 0.661 $hr^{-1}$ at pH 4. The analysis of total organic carbon for the reaction solutions showed that a higher removal of dissolved organic carbon was achieved with Bir-AB as compared to birnessite. HPLC chromatographic analysis of the methanol extract after reaction of 1-NP with Bir-AB suggest that the reaction products could be removed through incorporation into the aliginate beads as a bound residue. Mn ions produced from the oxidative transformation of 1-NP by birnessite were also removed by sorption to Bir-AB. The Bir-AB was recovered quantitatively by simple filtration and was reused twice without significant loss of the initial reactivity.

An Inquiry into the Formation and Deformation of the Cretaceous Gyeongsang (Kyongsang) Basin, Southeastern Korea (한반도 동남부 백악기 경상분지의 형성과 변형에 관한 질의)

  • Previously published stratigraphic, sedimentologic, paleontologic, paleomagnetic and geophysical data are reviewed to make an understanding on the tectonic evolution of the Cretaceous Gyeongsang (Kyongsang) basin, southeast Korea. A stratigraphic framework and a tectonic model on the formation and deformation of the Gyeongsang Basin are newly proposed on the basis of integration these data with magmatism and mineralization ages in the basin. A newly proposed stratigraphic framework indicates that strata in the basin can be subdivided into five distinct stratigraphic units that represent pre-rifting, syn-rifting, inversion I, II, and III stages. The Gyeongsang Basin was formed initially as a pre-rifting stage due to north-south extension in the Late Jurassic prior to a syn-riftins stage that resulted from east-west extension during the Early Cretaceous. In the Late Cretaceous, the basin was deformed by three-staged sequential deformation of north-south, northwest-southeast, and east-west compressions. The tectonic history of the basin has been largely controlled by the change of motion of the Izanagi Plate from north to northwest during the Cretaceous. In the early Cretaceous, the Izanagi Plate began to subduct northward beneath the Eurasian Plate and caused the left-lateral strike-slip fault systems in the southern part of the peninsula. The left-lateral wrenching of these fault systems was causally linked to development of pull-apart basins, such as the Gyeongsang Basin in the southeastern part of the peninsula. However, northwestward movement of the Izanagi Plate during the Late Cretaceous probably led to the extensive volcanism as well as sequential deformations in the basin. The stratigraphic and tectonic model, which is newly proposed as a result of this study, may be expected to enhancing the efficiency for exploration and exploitation of useful mineral resources in the basin as well as establishing geologic history in the Cretaceous Gyeongsang Basin. Together with the spatial and temporal correlation of the Cretaceous basins in adjacent areas, this stratigraphic and tectonic model provides a new geologic paradigm to delineate the sophisticated tectonic history of East Asia turing the Cretaceous.

Groundwater-Stream Water Interaction Induced by Water Curtain Cultivation Activity in Sangdae-ri Area of Cheongju, Korea (청주 상대리지역에서 수막재배가 지하수-하천수 상호작용에 미치는 영향)

  • Most of riverside in Korea, in case of application of water curtain cultivation (WCC) technique, has been inveterately suffering from the gradual drawdown of groundwater level and related shortage of water resources during the WCC peak time. We believe that the water resources issue in these riverside areas can be effectively solved when the interaction between groundwater and nearby surface water is well understood. To investigate the connection between stream and ground water, and the influence of stream water on the nearby aquifer, this study examined the water temperature and oxygen and hydrogen stable isotopic compositions. The study area is well-known strawberry field applying the WCC technique in Sangdae-ri, Gadeok-myon, Cheongju City, and the sampling was done from February 2012 through June 2014 for stream and ground water. Some groundwater wells near stream showed big temporal variations in water temperature, and their oxygen and hydrogen stable isotopes showed similar compositions to those of adjacent stream water. This indicates that the influence of stream water is highly reflected in the stable isotopic composition of groundwater. Four cross-sectional lines from stream to hillside were established in the study area to determine the spatial differences in water quality of wells. At the late stage of WCC in February to March, groundwater of wells in line with short cross-sectional length showed the narrow range of isotopic compositions; however, those in the long cross-sectional line showed a wide compositional range. It was shown that the influence of the stream water at the late WCC stage have reached to the distance of 160 to 165 m from stream line, which is equivalent to the whole length and one-third point in each short and long cross-sectional line, respectively. Therefore, the wide compositional range in the long cross-sectional lines was not only due to the influence of stream water, but apparently resulted from the change of relative impact of each groundwater supplying from two or more aquifers. In view of stable isotopic compositions, there seems to be three different aquifers in this study area, which is competing for dominance of water quality in wells at each period of WCC.

Effect of Supplemental Medicinal Plants(Artemisia, Acanthopanax and Garlic) on Growth Performance and Serum Characteristics in Lactating Sows, Suckling and Weanling Pigs (사료내 약용식물(인진쑥, 오가피 및 마늘)의 첨가가 포유돈, 포유자돈 및 이유자돈의 성적 및 혈청특성에 미치는 효과)

  • In Exp. 1, a total of forty eight multiparous sows were used in a 21-d performance assay. All litters were standardized at eleven piglets within 24h of parturition. Sows were fed one of the four experimental diets 1) CON(basal diet; Control), 2) MP1(basal diet added 0.05% of medicinal plant mixtures), 3) MP2 (basal diet added 0.1% of medicinal plant mixtures) and 4) MP3(basal diet added 0.2% of medicinal plant mixtures). Backfat thickness difference from farrowing to weaning was tended to increase in CON treatment compared to those of medicinal plants mixture(MP) treatments. However, there were not significant differences among the treatments(P>0.05). Sow’s ADFI was increased in sows fed MP2 treatment compared to MP3 treatment(P<0.05). Piglet weight gain was tended to increase in MP1 treatment compared to CON treatment(P<0.05). The glucose concentration of MP3 treatment was higher that of CON treatment(P<0.05). Exp. 2, a total of one hundred twenty weaning pigs ($L{\time}Y{\time}D$, 4.70$\pm$0.63kg average initial body weight) were used in a 20-d performance assay. Weanling pigs were fed treatments diets included 1) CON(basal diet; Control), 2) MP0.05(basal diet added 0.05% of medicinal plant mixtures), 3) MP0.1(basal diet added 0.1% of medicinal plant mixtures) and 4) MP0.15(basal diet added 0.15% of medicinal plant mixtures). Through entire experimental period, as MP increased in the diets, there was an increase in ADG(linear, P<0.067), improvement gain/feed(linear, P<0.018) and a decrease in ADFI (linear, P<0.008). DM digestibility was significantly increased with addition of MP(linear, P<0.004; quadratic, P<0.030). In conclusion, sows fed MP2 had showed less body weight loss and increased weight gain for suckling piglet. In weaning pigs, as supplementation MP from 0.05 to 0.1% in diet had improved growth performance and DM digestibility.

The analysis of pesticide residue in leafy vegetables using the modified QuEChERS pre-treatment methods (QuEChERS 시료 처리법을 활용한 엽채류 중 잔류농약분석)

  • In analyzing pesticide residue, LLE (liquid liquid extraction) is generally applied as one of the existing methods, but needed quite a lot of organic solvents and analytical apparatuses for the sample pre-treatment. In addition to its long analysis time and complex analytical processes, it is required to develop a more rapid and efficient method at present. In order to establish an economic and simple pesticide residue analytical method, this study carried out a comparative experiment on the existing analytical method with a new sample pre-treatment method named QuEChERS (quick, easy, cheap, effective, rugged and safe), which extracts and refines pesticide components by directly adding solid powder into the sample. Both the two analytical methods showed favorable values of correlation coefficient ($R^2$ > 0.99) of calibration curves. In terms of the detection limit (identification limit), imidacloprid showed 0.02 mg/kg, while the rest of pesticides showed a level around 0.05 mg/kg. The results of this experiment revealed that the recovery of LLE was 92.8-100.9% and the RSD was below 2.5%. On the other hand, the recovery of QuEChERS was 92.2-101.6% and RSD was below 1.9%. As a result of comparing the amount of pesticide residue by the time between the two analytical methods by using Paired t-Test, there was no significant difference between the two analytical methods as the p-value ranged from 0.3148-0.9890. Considering the results of the two methods, the QuEChERS method had similar recovery, compared to the analytical method using the existing LLE, and the analytical time was shortened by about one fourth of that of the existing method. Moreover, since it excludes the use of harmful organic solvents like dichloromethane during the process of extraction, thus leading to protecting experimenters health and remarkably reducing the amount of disused solvents, it is judged as an echo-friendly and economic analytical method.

Study of the Optimize Radiotherapy Treatment Planning (RTP) Techniques in Patients with Early Breast Cancer; Inter-comparison of 2D and 3D (3DCRT, IMRT) Delivery Techniques (유방암 방사선치료 시 최적의 방사선치료계획기법에 대한 고찰)

  • Purpose: A various find of radiotherapy treatment plans have been made to determine appropriate doses for breasts, chest walls and loco-regional lymphatics in the radiotherapy of breast cancers. The aim of this study was to evaluate the optimum radiotherapy plan technique method by analyzing dose distributions qualitatively and quantitatively. Materials and Methods: To evaluate the optimum breast cancer radiotherapy plan technique, the traditional method(two dimensional method) and computed tomography image are adopted to get breast volume, and they are compared with the three-dimensional conformal radiography (3DCRT) and the intensity modulated radiotherapy (IMRT). For this, the regions of interest (ROI) such as breasts, chest walls, loco-regional lymphatics and lungs were marked on the humanoid phantom, and the computed tomography(Volume, Siemens, USA) was conducted. Using the computed tomography image obtained, radiotherapy treatment plans (XiO 5.2.1, FOCUS, USA) were made and compared with the traditional methods by applying 3DCRT and IMRT. The comparison and analysis were made by analyzing and conducting radiation dose distribution and dose-volume histogram (DVH) based upon radiotherapy techniques (2D, 3DCRT, IMRT) and point doses for the regions of interest. Again, treatment efficiency was evaluated based upon time-labor. Results: It was found that the case of using 3DCRT plan techniques by getting breast volume is more useful than the traditional methods in terms of tumor delineation, beam direction and confirmation of field boundary. Conclusion: It was possible to present the optimum radiotherapy plan techniques through qualitative and quantitative analyses based upon radiotherapy plan techniques in case of breast cancer radiotherapy. However, further studies are required for the problems with patient setup reproducibility arising from the difficulties of planning target volume (PVT) and breast immobilization in case of three-dimensional radiotherapy planning.

Effect of Temperature, Relative Humidity on Germination and Development of Powdery Mildew (Leveillula taurica) on Pepper and Its Inoculation Method (고추 흰가루병균(Leveillula taurica)의 발아특성 및 접종법)

  • Pepper powdery mildew, Leveillula taurica is one of the most important pathogens of pepper in the greenhouses and fields in Korea and is becoming a worldwide disease. These experiments were carried out to investigate the optimal germination, disease development conditions, disease incidence and effective inoculation methods of pepper powdery mildew. The incidence of powdery mildew was investigated throughout the country based on the major pepper growing areas in 2009. The average rate of infected leaves ranged from 15.3% to 81.8% in greenhouses and fields. Powdery mildew incidences were more severe in greenhouses than those of fields. The optimal temperature for conidial germination was $25^{\circ}C$ and showed the highest germination at 6 hs after treatment. The range of temperature for germination was $10^{\circ}C$ to $35^{\circ}C$. Temperature of greater than $30^{\circ}C$ and below $20^{\circ}C$ affected the abnormal germination and germ tube elongation. The optimal relative humidity for germination and germ tube elongation was 85% and germination increased as relative humidity increased. Disease development started 8 days after inoculation and showed the highest disease severity at 15 days in greenhouse and field regardless of varieties. Among three different inoculation methods tested the spore dropping method was better than touching with infected leaves and spraying with spore suspension. However, the method has limitation in mass inoculation due to the amount of time consumed.

The Association of Growth Rate with Body Weight or Shank Length at Birth in Broiler Chickens (육용계의 부하시 체중 혹은 정강이 길이와 성장률과의 상관관계에 관한 연구)

  • The objective of this study was to investigate the association of growth rate with body weight or shank length at birth in broiler chickens. Cobb and Ross breeds were used as experimental stocks and four groups were assigned in each breed; thirteen-male and twelve-female chicks(25 birds) were allocated in each group. The heavier body weight group(HBW) and the lighter body weight group(LBW) were established by birth weight. Whereas, the longer shank length group(LSL) and the shorter shank length group(SSL) were established by shank length at birth. The heavier group chicks in birth weight were significantly(P < 0.05) the longer in shank length at birth. The differences between HBW and LSL or between LBW and SSL in the mean 5-wk body weight of Cobb were not significantly different in each other; however, HBW and LSL were significantly(P < 0.05) heavier than LBW or SSL. Whereas, there were no significant differences in the mean 5-wk body weight of Ross although HBW was heavier by 13.1 g than LBW and LSL was heavier by 68.0 g than SSL. In the mean 5-wk shank length, the LSL of Cobb and Ross was longer by 2.9 mm and 1.3 mm than SSL, respectively; however, the significant(P < 0.05) difference was showed in Cobb only. In both breeds, the differences in the mean 5-wk shank length between HBW and LBW were not great. By the results of correlation analysis, the associations of the overall mean birth weights with the overall mean shank length at birth or the association of the 0~5 wk growth rate with the change of shank length during 0~5 wk showed highly significant(P < 0.01~0.001) in both breeds. The present study suggested that the selection based on birth weight could be at advantage due to easier and more accurate tool in using for the improvement growth rate in broiler chickens because the measurement of shank length takes individually longer and less accurate than the measurement of body weight.
