Studies on the Synthesis of Yellow Coupler and Color Development (황색 발색제의 합성과 발색현상에 관한 연구)

  • In this paper, yellow coupler was prepared by the reaction of ${\alpha}$-pivaloyl- 2-chloro-5-aminoacetaniline hydrogen chloride with 1-hexadecane sulfonyl chloride in the presence of pyridine. The product was identified by using various analytical tools such as melting poin elemental analyzer, IR spectrophotometer, UV-Vis spectrophotometer, mass spectrometer. The reaction of yellow coupler with CD-3(color development agent) was shown yellow color.

크릴과 아스타크쟌틴 첨가량에 따른 붉은쏨뱅이 (Sabastiscus tertius)의 체색변화와 성장

  • 최근 인공종묘생산기술이 개발된 붉은쏨뱅이(Sabastiscus tertius)는 자연산과 양식산의 체색이 붉은색과 검정색으로 많은 차이가 있어 양식산업화에 많은 어려움이 있다. 따라서 상품가치와 경쟁력 향상을 위하여 사료에 카로티노이드 성분을 함유한 크릴새우(25, 50, 75, 100%)와 astaxanthin(5, 10, 15, 20%)를 비율대로 첨가하여 양식산 붉은쏨뱅이의 체색 변화를 살펴보았다. 크릴새우의 경우 50% 첨가한 사료실험구에서의 적색도와 황색도는 각각 6.29와 13.42로 높게 나타나기 시작하였으며 크릴새우 75% 첨가실험구에서는 적색도와 황색도가 각각 8.31과 14.91로 최고치를 보였다. 그리고 크릴 100%에서는 적색도와 황색도가 각각 7.27과 10.33로 75% 실험구에 비하여 감소하는 것으로 나타났다. astaxanthin 실험구들에서는 5%와 10% 실험구에는 체색도가 높게 나타나지 않았으나 15%에서 적색도가 7.86 황색도가 9.32로 가장 높게 나타났다. 그리고 20% 실험구에는 7.18과 6.32로 나타났다. 본 실험결과 붉은쏨뱅이의 체색회복을 위해서는 크릴새우의 경우 최소 50% 이상을 첨가해야만 가능할것으로 판단되며 astaxanthin는 15% 이상 첨가해야 효과가 있는 것으로 판단된다. 그리고 astaxanthin보다는 크릴새우가 더 효과적이였으며 모든 실험구는 대조구(펠렛사료 100%)에 비하여 체색 회복효과가 있는 것으로 나타났다. 또한 자연산에서는 적색도와 황색도는 각각 23.80과 13.44로 적색도가 아주 높게 나타났으나 양식산에서는 적색도와 황색도가 1.09와 1.98로 매우 낮게 나타났다. 그리고 체색의 변화는 모든실험구에서 등지느러미와 꼬리지느리미 그리고 턱과 가슴주변에서부터 점차 배와 등쪽으로 색이 변화하는 것이 관찰되었다. 크릴 75%와 astaxanthin 15%를 공급하였을 때 체색이 변화하는 시간은 크릴의 경우 사료공급 20일 후부터 체색의 변화가 시작하여 30일이 경과 될때에는 등지느러미, 꼬리지느러미 및 턱주변이 붉은색으로 변화하였다. astaxanthin의 경우는 30일부터 등지느러미와 꼬리지느러미 부근에서 체색의 변화가 시작되었으며 40일 이후부터 붉은색으로 변화하였다. 크릴새우와 astaxanthin를 함유하여 실험하였을 때 붉은쏨뱅이의 성장은 크릴 75% 실험구에서 전장 172.3mm, 체중 65.9g으로 가장 좋았다. 그러나 대조구에서는 전장 148.7mm, 체중 49.5g으로 가장 성장이 저조하였으며 astaxanthin 실험구들에서는 전장 152.3~159.8mm, 체중 56.9~59.3g으로 서로 유의적인 차이를 나타내지 않았다.

A Study on Developing Intersection Dilemma Zone Evaluation Model (교차로 딜레마 구간 위험도 평가 모델에 관한 연구)

  • This study focuses on development of a risk level evaluation model for the intersection dilemma zone based on vehicle speed distribution approaching the intersection. The study results show that dilemma zone exists for all approaches selected for the study because the values of amber time have not been properly set, and that re-optimized values significantly reduce the dilemma zone risk level. It is also found that when normal distribution is assumed for vehicle approach speed the risk values similar to those for actual speed distributions can be obtained. When the risk level is found to be high, the amber time must be adjusted to reduce the length of the dilemma zone, or speed calming measures must be introduced so that vehicle speed be maintained under the speed value used when determining the amber time.

SYBR Green I-based Real-time PCR Assay and Melting Curve Analysis for Rapid Detection of Staphylococcus aureus from Raw Milks Samples (Real-time PCR을 이용한 원유시료 유래 황색포도상구균의 신속 검출)

  • The aim of this study was to develop a LightCycler-based real time PCR (LC-PCR) assay and to evaluate its diagnostic use for the detection of Staphylococcus aureus in raw milk samples. Following amplification of 113 bp of coa gene encoding an coagulase precursor specific for Staphylococcus aureus, melting curve and DNA sequencing analysis was performed to verify the specificity of the PCR products. Amplification of 209 bp gene encoding an altered penicillin-binding protein, PBP2a (mecA), melting curve analysis and DNA sequencing analysis was performed to verify methicillin resistance Staphylococcus aureus (MRSA). According to this study, 6 of 647 raw milk samples showed S. aureus positive and 2 of them showed a mecA positive and the detection limit was 10 fg of DNA. And we also isolated Staphylococcus chromogenes a causative agent of exudative epidermitis in pigs and cattle from 3 samples.

Carotenoid Contents of Yellow Sweet Potatoes (황색고구마의 Carotenoid 색소 함량)

  • Twelve varieties of yellow sweet potatoes were measured for carotenoids content to assess their potential as a source of natural food colorant. Benihayato variety had the highest content of carotenoids (20.2 mg/100 g fr wt) but other varieties ranged with $4.6{\sim}16.7\;mg/100\;g$ fr wt. Hunter's +a-values (redness) increased linearly $(R^{2}=0.826)$ with carotenoids content of yellow sweet potatoes. Absorption spectrum of carotenoids extracted from the Benihayato variety was similar to that of standard ${\beta}-carotene$ indicating that carotenoids in yellow sweet potato are mostly composed of ${\beta}-carotene$.

Single-Dose Oral Toxicity of the Gardenia Yellow Pigment in Rate and Rabbits (랫드 및 토끼에 대한 치자 황색색소에 관한 단회 투여 경구 독성 시험)

  • This study was performed to evaluate single-dose toxicity of the gardenia yellow pigment in Sprague-Dawley rats and New-Zealand White rabbits via oral routes. The yellow pigment was administered in rats at does levels of 5,000, 2,500, 1,250, 625, 312.5mg/kg and 9mg/kg. And also yellow pigment was administered in rabbits at does levels of 5,000, 2,500, 1,250mg/kg 0 unit /kg. The rats and rabbits of both sexes were observed daily for 14 days after single oral administration. Yellow pigment treated rats and rabbits did not induce any mortalities and abnormal signs in clinical findings, body weights, gross findings and histopathological finding. Based on these results, it is impossible to estimate LD50 values in rats and rabbits. Therefore, it was concluded that gardenia yellow pigment have no effect on acute toxicity and side effect in rats and rabbit.

Characteristics of Antioxidative and Antimicrobial Activities of Various Cultivars of Sweet Potato (고구마의 품종별 항산화성과 항미생물 특성)

  • Antioxidative and antimicrobial activities of ethanol extracts from 2 colored sweet potatoes such as purple and yellow, 5 pale yellow sweet potatoes, and 8 general cultivars of sweet potato were investigated. Colored sweet potatoes showed the highest antioxidative activity, followed by Saengmi and Yeonmi, which were pale yellow sweet potato cultivars, and Hwangmi, which was a general cultivar of sweet potato. Colored sweet potatoes also showed distinctively lower mold and surface microbial number (cfu) than the pale yellow sweet potatoes and the general cultivars of sweet potato. Although all sweet potatoes tested did not represent any antimicrobial activity against yeast and mold, they exhibited strong antimicrobial activity against Streptococcus faecalis. Especially, purple and yellow sweet potatoes showed antimicrobial activity against wide range of bacteria.

  • Staphylococcus aureus is found on the skin of $78{\sim}100%$ of children and adults with atopic dermatitis (AD) but only on the skin of $2{\sim}25%$ of healthy subjects. It is known that S. aureus and their endotoxins as superantigen have important roles in the exacerbation and prolongation of AD. This study was carried out for the detection of S. aureus in the skin of AD, age, sex, outbreak age of AD, treatment duration, aggravation season, and the relation of ooze and S. aureus. Most patients (84%) with AD show colonization of the skin with S. aureus and there is a correlation between the degree of colonization and the serous exudate. It seems likely that the inhibition of S. aureus is associated with improvement in the skin of AD patients.

  • The mechanical properties of the propellant with yellow iron oxide were slightly increased compared to the propellant with red iron oxide. The propellant with yellow iron oxide used two types of AP. As the ratio of small particles of AP increased, the burning rate increased. The propellant may be applied to the propellant under operating conditions of 17.5 mm/sec or less having a pressure index of 0.5. The burning rate downs in the mixer scale-up. The stress at maximum load of propellant decreased and the strain at maximum load increased in the mixer scale-up. The yellow iron oxide did not affect the adhesive force between the insulation/liner/propellant.

Prevalence and Characterization of Staphylococcus aureus Pathogenic Factors Isolated from Various Foods in Korea (국내 다양한 식품에서 분리한 황색포도상구균의 오염도 및 병원성 인자의 특징)

  • Staphylococcus aureus, a major human pathogen, produces a wide array of toxins, which causes various types of disease symptoms. Prevalence of S. aureus in various foods collected during 2006-2008 in Korea was investigated. S. aureus was isolated from 275 of 5,186 (5.3%) food samples collected from hyper-markets in Korea. Seasonal temperature affected the prevalence of S. aureus in various foods with high isolation rate during the summer. Most of the enterotoxigenic strains produced enterotoxin A only or enterotoxin A in combination with another toxin. A total of 54.5% of the tested strains contained either one or more enterotoxin genes and 3.6% possessed a tst gene. This study offers basic information for securing the stability of food during storage and circulation, and provides an epidemiological tool to study the cause, origin and temporal spread of S. aureus food poisoning.
