Efficiency of Dyes Extraction and Dyeing of Safflower according to pH Condition (pH에 따른 홍화(紅花)의 색소(色素) 추출(抽出)과 염색(染色)의 효율성(效率性)``)

  • Effect of the pH condition was examined on extraction and dyeing of the yellow and red dyes of safflower. Absorption of dye solution extracted from safflower was changed by pH of solvent. In the case of yellow dyes, cotton and ramie weren't dyed regardless of pH of dye bath, but silks were significantly dyed at pH4. Raw silk showed better dyeability than refined silk. In the case of red dyes, cotton and ramie absorbed red dyes selectively, but silks absorbed yellow dyes more significantly than red dyes. Efficiency of extraction and dyeing of yellow dyes in acidic range were find out higher than that in neutral range. Red dyes was effectively extracted at pH11 and dyed at pH6.

  • 적황색토와 플라이애쉬의 혼합비율에 증가에 따라 건조밀도도 증가하며 최적함수비는 감소하는 경향이고 2차 첨가재인 소석회 및 시멘트 첨가에 의한 건조밀도의 증가는 보이나 처리토의 경량화의 범위는 1:03~1:0.5 정도임을 확인할 수 있었다. 프라이애쉬의 혼합비율이 증가해도 프라이애쉬의 고유산화칼슘(CaO)의 함유량이 적으면 유리산화칼슘의 증가도 크지 않으나, 2차 첨가재인 소석회 및 시멘트 첨가량의 증대에 따라 증가한다. 이는 첨가재에 의한 수화반응의 증가로 확인되며, 본 처리토는 Ion 교환작용과 Pozzolan 반응 생성물인 규산칼슘석회수화물(5CaO.6SiO$_2$.5$H_2O$, Tobermorite)과 알민산유산석회수화물(3CaO.Al$_2$O$_3$.3CaSO$_4$.32$H_2O$, Ettringite)가 주된 반응생성물이며 확인된 알민산유산석회수화물(Ettringite)의 회절 X-선 강도는 2차 첨가재의 첨가에 따른 수화반응으로 수화물은 점차 증가하며 상대적으로 일축압축강도도 상응한 강도발현을 하여 고결화 효과에 기여하고 있음을 보여준다.

Effect of Light Quality on Shape and Greening of Selling Mungbean Sprouts (판매용 숙주나물의 형태(形態) 및 녹화(綠化)에 미치는 광질의 효과)

  • Storage conditions of selling mungbean sprouts should affect their marketability. The study was done to examine the effect of light quality (blue, green, yellow, red, far-red) on greening period, shape, fresh and dry weights of mungbean (cv. Zhong Lu 1) sprouts for 5 days to get some information on designing their various polyethylene envelops with different colors. Periods taking to green were 2 to 3.5 days in blue, red and far-red lights but longer than 6 days in green and yellow lights. Yellow lights had the longest total length adding hypocotyl and root, and blue light did the widest hypocotyl diameter while all light treatments showed similar lateral roots for the period. Total fresh and dry weights were the highest in red light but the least in far-red light. Their lengths and weights in green and yellow lights showing the longest periods for their greening declined with increased storage periods but showed no difference between them, meaning that the envelops should be designed mainly by green and yellow colors.

  • Conversion of Gardenia jasminoides yellow pigment into blue-green pigment by 8 bacterial species was examed. Bioconversion pattern can be categorized into three types according to absorption spectra characteristics. The same pattern of the value of ${\Delta}E$ estimated by color differencemeter was also observed. Conversion rate by S. epidermidis was faster than other bacterial species. It took 16 hour for S. epidermidis to convert pigment at $37^{\circ}C$. Gardenia jasminoides yellow pigment and conversion pigment were completely separated by Amberlite XAD column chromatography with $H_2O-MeOH$ solvent system. Storage stability of the conversion pigment was better than Gardenia jasminoides yellow pigment.

Microbiological Hazard Analysis of Foodservice Facilities and Equipment at Kindergarten Foodservice Establishments (유치원 급식시설.설비에 대한 미생물학적 위해분석)

  • This study was conducted to analyze the food safety management practices and to provide data that could be used to improve food safety management at kindergarten foodservice establishments in Daegu and Gyeongbuk province. Microbiological hazard analysis was conducted from May to July, 2008 at the four foodservice establishments. A total (20 items) of foodservice facilities, cooking utensils, and equipment were tested about total plate counts, coliforms, E. coli, Staphylococcus aureus, Salmonella spp., and Listeria monocytogenes. The microbiological detected levels were higher than the critical limits for prepreparing and cooking knives, chopping boards, working tables (both prepreparing and cooking), refrigerator (inside wall), bart, basket, and tray. E. coli, Salmonella spp., and Listeria monocytogens were not detected in any samples. However, Staphylococcus aureus were detected in rubber gloves for cooking and trench in the bottom at one foodservice establishment. In conclusion, these results suggest that a prerequisite programs guideline should be provided to improve the food safety levels at kindergarten foodservice establishments and the foodservice manager must maintain proper food safety technique for foodservice facilities, cooking utensils, and equipment at kindergarten foodservice establishments to prevent cross-contamination and spread of foodborne pathogens.

Molecular Characterization of Clinically Isolated Staphylococcus aureus (인천지역에서 분리된 황색포도상구균의 분자생물학적 특성 분석)

  • Staphylococcus aureus is one of the most significant pathogens and a causative agents of nosocomial infections. The emergence of methicillin resistant S. aureus (MRSA), in particular, has become a major clinical and epidemiological problems worldwide. In this study, we analyzed the toxin genes and investigated molecular epidemiological characteristics of S. aureus isolated from stools of diarrheal patients at the hospitals in Incheon. Of the 609 strains from 2,281 specimens, 173 strains retained enterotoxin; 68 isolates (39.30%), 100 isolates (57.80%) were classified to A and C type, respectively. In the antibiotic susceptibility, all of enterotoxin positive isolates were resistant to oxacillin. Eighty eight strains (50.86%) of 173 MRSA isolate possessed tsst gene, but eta and eth genes were not detected at all. In the detection of MRSA associated genes by PCR method, mecA genes were detected in 167 strains (96.53%). From the result of PFGE analysis, we classified tsst-positive MRSA to 10 types and 24 subtypes. Type A, H and F were the major strains comprised of 57.95% (51 strains), 10.22% (9 strains) and 9.09% (8 strains) respectively.

Hazard Analysis of Staphylococcus aureus in Ready-to-Eat Sandwiches (즉석섭취 샌드위치류의 황색포도상구균에 대한 위해분석)

  • This study investigated the hazard analysis of ready-to-eat sandwiches sold in various establishments. Sandwich samples were collected from convenience stores, discount stores, sandwich chain stores, bakery shops, fast-food chain stores, and food service operations located in Daegu and Gyeongbuk. Out of 174 samples, 18 (10.3%) contained coagulase positive staphylococci with counts ranging from 0.30 to 4.08 log CFU/g. There was significant seasonal difference in Staphylococcus aureus isolation; the average count in summer (3.24 log CFU/g) was 3 times higher than that of winter (1.10 log CFU/g) (P<0.001). According to the microbiological guidelines of PHLS for ready-to-eat foods, 95.4% of the samples were acceptable. As a result of enterotoxin producing experimental data ($35^{\circ}C$, pH 5.8, NaCl 0.5%), enterotoxin was not produced in a sandwich until Staphylococcus aureus increased to a level greater than 4.95 log CFU/g. This microbiological hazard analysis data could be applied to future studies on quantitative risk assessment of ready-to-eat foods.

Characteristics of Staphylococcus aureus Isolated from Patients with Diarrhea. (설사 환자에서 분리된 Staphylococcus aureus의 특성)

  • The major causative bacteria of food poisoning were Salmonella spp. $(35.6\%)$, Staphylococcus aureus $(11.3\%)$ and Vibrio parahaemolyticus $(3.2\%)$ in our country. In this study we attempted isolation of S. aureus from stools of patients with diarrhea. Sixty-four strains $(9.1\%)$ were isolated from 704 the stools of patients with diarrhea. The enterotoxin was detected from 29 isolates $(45.3\%)$: 24 isolates $(37.5\%)$, 3 isolates $(4.7\%)$ and 2 isolates $(3.1\%)$ were A, B and C type, respectively. In the antibiotic susceptibility, 63 isolates $(98.4\%)$ were resistant to penicillin, 60 isolates $(93.8\%)$ to ampicillin, 35 isolates $(54.7\%)$ to erythromycin, 32 isolates $(50.0\%)$ to gentamycin, 22 isolates $(34.4\%)$ to tetracycline and 20 isolates $(31.3\%)$ to oxacillin. All of S. aureus isolates were susceptible to chloramphenicol and vancomycin, 20 isolates $(31.3\%)$ were methicillin-resistance S. aureus (MRSA). MRSA isolation rate was higher in male $(35.7\%)$ than female $(26.3\%)$. With the exception of two isolates which were resistant only to penicillin, sixty-one isolates were multiple antibiotic resistance.

  • 1991년∼2000년의 우리 나라 원인 균별 식중독 발생 상황을 보면 살모넬라균, 황색포도상구균, 그리고 비브리오균에 의한 식중독이 우리 나라 식중독 사고발생의 85∼90%를 차지하는데, 특히 전체 식중독 발생건수에 대해 살모렐라균에 의한 식중독이 차지하는 비율이 1993년에서 97년도 사이에 46.5%, 1999년도와 2000년에는 36.3%로 보고되었다. 이처럼 살모렐라균은 대표적인 식중독균으로서 우리 나라 뿐만 아니라 전세계적으로 세균성 식중독의 주요 원인이 되고 있는데 식육, 달걀 둥 동물성 식품 및 가공품, 어패류 등이 주요 원인식품으로 알려져 있다. Quaility Evaluation of Jellies Prepared with Refined Dietary Fiber from Ascidian (Halocynthia roretzi) Tunic (우렁쉥이 껍질로부터 정제된 섬유소 첨가 젤리의 품질평가)

  • Jellies enhancing physiological functions were prepared with using 5 and 10% dietary fiber isolated from ascidian (Halocynthia roretzi) tunic collected from recycled seafood waste. The texture development of the samples was examined with two-bite compression test and analyzed using five sorts ofrheological parameters with texture profile analysis as follows; hardness, adhesiveness, springiness, cohesiveness and gumminess. All the rheological parameters decreased in the fiber enhanced jellies. However, the springiness and cohesiveness increased slightly in the fiber enhanced jellis. Hunter L' and d' values increased in the fiber enhanced jellies and accordingly the color was light yellow. a' value showed green with the addition of fiber. As a result of sensory evaluations, the color and overall acceptability of 10% fiber enhanced jelly were significantly different at p<0.05. The 10% fiber enhanced jelly was noted as having high sensory scores and peferable acceptability.
