Antioxidant, anti-inflammatory, and anti-pruritic effects of grape branch extract (포도가지 추출물의 산화방지, 항염증 및 항가려움 효과)

  • This study was designed to investigate the antioxidant activities of the ethanol extracts (GBE) of grape branches (Campbell Early). The total polyphenol and flavonoid content of GBE was $201.42{\pm}4.16$ and $11.85{\pm}0.44mg\;GAE/g$, respectively. The antioxidant activity of GBE was measured using the ABTS and DPPH assays, and the $IC_{50}$ values were $45.60{\pm}0.09$ and $299.13{\pm}0.22$, respectively. GBE inhibited the production of pro-inflammatory mediators (NO, iNOS, $PGE_2$, COX-2, $IL-1{\beta}$, and IL-6) in lipopolysaccharide-stimulated RAW 264.7 macrophages in a dose-dependent manner. Moreover, GBE treatment significantly suppressed the production of $TNF-{\alpha}$ and IL-6 cytokines in phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated HMC-1 human mast cells. Furthermore, the administration of GBE markedly inhibited the scratching behavior induced by the compound 48/80 in ICR mice. These results suggested that GBE has potential as a therapeutic agent against inflammation and itch-related skin diseases.

Anti-inflammatory Effects of Extracts and Their Solvent Fractions of Rice Wine Lees (주박 추출물과 이들의 유기용매 분획물에 의한 항염증 활성)

  • In the current study, we prepared eighty-five different kinds of solvent fractions of rice wine lees and nuruk extracts and investigated their effects on cell viability and nitric oxide (NO) production in mouse RAW 264.7 cells. Among the treated solvent fractions, only three solvent fractions (KSD-E1-3, KSD-E2-3 and KSD-E4-3) significantly decreased NO production in LPS-activated RAW 264.7 cells without affecting cell viability. And, they also reduced the expression of pro-inflammatory genes such as COX-2, TNF-alpha and iNOS. To understand the molecular mechanisms involved in the inhibition of inflammation in (KSD-E4-3)-treated RAW 264.7 cells, we carried out oligo DNA microarray analysis using Agilent Mouse microarray. To confirm microarray data, 6 genes (IL-1F6, iNOS, IL-10, Fabp4, IL-1RN and CSF2) were selected and performed RT-PCR and quantitative real-time PCR analysis with gene specific primers. The results of RT-PCR and real-time PCR agreed with microarray data. Overall, our results suggest that rice wine lees can be a novel resource for the development of foods and drugs which possess anti-inflammatory activity.

Quality Characteristics and Antioxidant Activities of Macaron with Opuntia ficus-indica var. saboten Powder (백년초 분말을 첨가한 마카롱의 품질 특성 및 항산화 활성)

  • The aim of this study was to evaluate the quality characteristics, antioxidant activities, and sensory properties of macaron with Opuntia ficus-indica var. saboten powder (OP). Physicochemical and sensory properties of macaron with different amounts (0%, 2%, 4%, and 6%) of OP were examined. The quality of macarons was evaluated based on spread factor, baking loss rate, color, texture, antioxidant activities, and sensory evaluation. As amount of OP addition increased, spread factor and a value increased, whereas baking loss rate, and L and b values decreased (p<0.05). For texture properties, hardness, springiness, and brittleness were lower as OP content increased while cohesiveness and gumminess were not significantly different (p<0.05). For total polyphenolic contents, DPPH and reducing power significantly increased upon addition of OP at high concentrations (p<0.05). In the sensory evaluation of macarons, scores for color, flavor, taste, texture, and overall acceptance were highest with 4% OP, whereas macaron containing 6% OP showed the lowest scores for taste and texture (p<0.05). These data suggest that addition of 4% OP is the optimal concentration for making macaron.

Purification and characterization of the extracellular alginate lyase from Streptomyces sp. MET 0515 (Streptomces sp. MET 0515의 균체외 Alginate lyase의 정제 및 특성)

  • We isolated a new extracellular alginate lyase-producing microorganism, which displayed alginate-depolymerizing activity in plate assays, from coastal soils in Wando, Jeollanam-do, Korea. This alginate-depolymerizing bacterium belonged to the genus Streptomyces and it was named Streptomyces sp. MET 0515. An extracellular alginate lyase(ALY1) secreted by Streptomyces sp. MET 0515, was purified to homogeneity by a combination of acetone precipitation, anion-exchange chromatography (Q-Sepharose and DEAE-Sepharose) and Sephacryl S-200 HR gel filtration chromatography. Its molecular mass was 26 kDa as determined by SDS-PACE analysis. The enzyme had an optimal temperature of $70^{\circ}C$ for its activity, and was most active at pH 7.5. The thermal and pH stability were $0-50^{\circ}C$, and pH 6.0-9.0, respectively. The enzyme activity was stimulated by 1mM $Mn^{2+}$, and inhibited by 1mM $Fe^{3+}$, 1mM EDTA and 1mM $Zn^{2+}$. Preliminary analysis of substrate specificity showed that this alginate lyase had activity on both poly-alpha 1,4-L-guluronate and poly-beta 1,4-D-mannuronate in the alginate molecule.

Antioxidant Activity of Hot-Water Extract from Yuza (Citrus junos SIEB ex TANAKA) Peel (유자과피 열수 추출물의 항산화 활성)

  • This study was to investigate of characteristics and antioxidant function of yuza from 4 different area. The hot-water extracts of yuza peel extracts from Geoje, Goseong, Goheung and Namhae(Changseon, Seolcheon) were tested for antioxidant activity in different reaction systems. Contents of total phenolic compounds and flavonoids were $122.18{\pm}1.44$ mg/100 g and $114.39{\pm}0.94$ mg/100 g in hot water extracts from Namhae-Seolcheon, respectively. The highest contents of hesperidin and naringin were $55.45{\pm}1.36$ mg/100 g and $28.41{\pm}0.64$ mg/100 g in hot water extracts from Geoje, respectively. Antioxidant activity of yuza peel hot-water extracts were significantely increased as the increament of sample adding concentration ($500{\sim}10,000{\mu}g$/ml). Reducing power was $6{\sim}9$ folds higher in 10,000 ${\mu}g$/ml concentration than 500 ${\mu}g$/ml and it's O.D. value showed $0.68{\pm}0.012{\sim}0.97{\pm}0.021$. ABTs scavenging activity was more than 80% in 10,000 ${\mu}g$/ml concentration samples, except from Goseong ($78.13{\pm}1.30%$). Hydroxyl radical scavenging activity was higher in Namhae-Seolcheon ($31.36{\pm}1.36%$) and Namhae-Changseon ($30.28{\pm}1.60%$) at 10,000 ${\mu}g$/ ml concentration, others activity were below 30%. At 10,000 ${\mu}g$/ml concentration, NO radical scavenging activity and antioxidant activity in ${\beta}$-carotene linoleic acid system were $26.49{\pm}1.77{\sim}40.85{\pm}0.95%$ and $24.40{\pm}1.91{\sim}38.17{\pm}0.56%$, respectively.

Convergence study of oxidative stress from fraction of Xanthium strumarium L. (도꼬마리 추출물의 산화적 스트레스에 대한 융합연구)

  • Xanthium strumarium L. is an annual plant belongs to the family Asteraceae which is is called a 'Cocklebur' that is used for medicinal purposes. Convergent phyto-activity of various extracts of Xanthium strumarium L. (Asteraceae) was examined. We estimated antioxidant activity from ground part and fruit extract of X. strumarium using 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH) and ABTS assay. The extract of X. strumarium was separated each fraction that of ethanol, petroleum ether, and ethyl acetate. It showed potent radical scavenging effect against the DPPH radical and ABTS. The study revealed that X. strumarium could be used as a potential source of natural antioxidant.

  • Eight kinds of poly(oxyethylene, oxyisopropylene) dodecyl ethers were synthesized by adding ethylene oxide and isopropylene oxide with each 5 and 10 moles alternatively on dodecanol. The addition of EO and PO for eight kinds of sodium poly(oxyethylene, oxyisopropylene) dodecyl ether sulfates was identified with HPLC, $^1H$ NMR, hydroxy value, and IR spectrum. In order to verify the sulfation the number of EO and PO molecules was obtained by spectra and Epton method. The yields of products sulfated by chlorosulfonic acid were 90~96%.

A Study of Structural Characteristic Control and Reaction Activity of V/TiO2 for NH3-SCR according to Preparation Method (제조방법에 따른 NH3-SCR용 V/TiO2의 구조적 특성 제어 및 반응활성 영향 연구)

  • In this study, the activity and structural properties of catalysts prepared by mechanochemical method under dry condition were studied. A dry milling was used as a mechanochemical method. The precursors of vanadium were $NH_4VO_3$ and $V_2O_5$. The activity and characterization of the catalysts prepared by dry milling were compared with those prepared by impregnation. In addition, the correlation between the catalytic activity and the structural characteristics was observed through XRD, Raman, and $H_2$-TPR analysis. As a result, the monomeric vanadate species exhibited excellent redox characteristics, which were confirmed to be related to the catalytic activity.

  • This study was investigated experimental condition which is able to evaluate photocatalytic activity of various commercial $TiO_2$. The experiments were performed for three representative substances (ethanol, phenol and methylene blue) and four kinds of commercial $TiO_2$, under the experimental conditions such as pH, reactant concentration, amount of $TiO_2$, reaction time and UV intensity. The optimum experimental conditions to evaluate photocatalytic activity were as follows : for ethanol, the initial concentration 1000 ppm, initial pH 8, $TiO_2$ loadings 0.1 wt%, and reaction time 90 minutes: for phenol, the initial concentration 200 ppm, initial pH 8, $TiO_2$ loadings 1 wt%, and reaction time 60 minutes: for methylene blue, the initial concentration 200 ppm, initial pH 4, $TiO_2$ loadings 0.5 wt%, and reaction time 30 minutes.

Characterization Studies for the Selective Catalytic Oxidation of Ammonia Utilizing Ce/TiO2 Catalyst (Ce/TiO2 촉매를 이용한 암모니아의 선택적 산화반응 특성 연구)

  • In this study, selective catalytic oxidation (SCO) of $NH_3$ using $Ce/TiO_2$ catalyst was examined to control the slipped NH3 from various pollutants. It was found that the catalytic activity increased with increasing the Ce loadings till reaching 10 wt% Ce loading. However, when Ce loaded over 10 wt%, the activity of catalysts rather decreased than that of catalysts, below 10 wt% Ce. Therefore, the composition of $Ce/TiO_2$ catalyst optimized in this study can be applied to industrial fields.
