Susceptibility of Cigarette Smoke Condensate-Exposed Human Bronchial Epithelial Cells to Hypoxia-Reoxygenation (담배연기 농축액 처리에 의해 유도된 사람 기관지 상피세포주의 Hypoxia-Reoxygenation에 대한 민감성)

  • 사람 기관지상피세포주인 BEAS-2B에 담배연기농축액(CSC)을 처리하여 유도된 1198 세포주는 대조군 세포주인 1799에 비해 현저하게 낮은 glutathione 농도와 낮은 glutamate-cysteine ligase(GCL), glutathione peroxidase(GPx), glucose-6-phosphate dehydrogenase(G6PD), catalase 효소활성을 보였다. 두 세포주를 포도당 존재 하에서 4시간 hypoxia 처리 후 reoxygenation 하면서 시간에 따른 세포의 항산화계 활성을 측정한 결과, 1799 세포주에서는 의미 있는 변화가 관찰되지 않은 반면, 1198 세포주에서는 hypoxia 처리에 의해 glutathione의 농도 및 GSH/GSSG 비와 G6PD 활성이 감소되었고, reoxygenation 기에는 GPx, glutathione reductase(GRd), G6PD, superoxide dismutase 활성이 감소되었다. 그러나 reoxygenation 2시간 이후에는 GRd와 G6PD 활성의 회복이 관찰되었으며, 그 결과 GSH/GSSG 비율이 회복되었다. 이 실험 결과는 CSC가 능력을 현저히 저하시킬 수 있음을 보여준다. Glutathione은 hypoxia-reoxygenation에 의한 산화적 스트레스 하에서 항산화제로서의 역할뿐 아니라, 세포 내 GSH/GSSG 비의 변화를 통해 산화적 스트레스에 대한 항산화계의 적응 반응 여부를 결정하는 중요한 인자로 작용할 것으로 보여진다.

Effect of Insecticide Carbofuran and Phenobarbital Sodium and 3-Methylcholanthrene on Activity of Enzyme in Rat (살충제 Carbofuran과 Phenobarbital Sodium 및 3-Methylcholanthrene이 쥐의 효소활성에 미치는 영향)

  • Effect of insecticide carbofuran and phenobarbital sodium(PB) or 3-methylcholanthrene(3-MC), they were orally administered by the chemicals, alone or in combination, on activities of several enzymes in rats were investigated. In in vivo test for the effect of this chemicals on activity of enzyme in rat, activities of acetylcholinesterase(AChE) and butyrylcholinesterase(BuCheE) were inhibited by $20{\sim}70%$ for 48 hrs after the oral administration of carbofuran alone of 3.8mg/kg, whereas those were lowered at the beginning, but recovered to the control level after 24 hrs, in case of the mixed administration of carbofuran+PB or carbofuran+3-MC. The activity of glutathione S-transferase(GST) was inhibited by more than 15 to 35% for an early period of 0.5 to 6 hrs, in the case of the administration of carbofuran alone, whereas that was slightly inhibited at the beginning, recovered almost to the control level after 3 hrs, and raised by mere than 20% above the control after 6 hrs, in case of the mixed administration of carbofuran+PB or carbofuran+3-MC. When carbofuran was administered alorig with PB or 3-MC, the activities of UDP-glucuronosyltransferase(UDPGT) and cytochrome P-450 were more than 2.6 to 2.8 times higher than that in the case of the administration of carbofuran alone for 6 hrs. These results suggest that a simultaneous application of carbofuran and PB or 3-MC is critical for the enhancentment of activity of GST, UDPGT and cytochrome P450 and the protection of rat from carbofuran toxicity.

The Effects of Extraction Conditions on the Antioxidative Effects of Extracts from Campbell Early and Muscat Bailey A Grapevine Leaves (추출조건에 따른 캠벨얼리와 Muscat Bailey A 포도잎 추출물의 항산화 효과)

  • We investigated the physiological activities of extracts from Campbell Early and Muscat Bailey A (MBA) grapevine leaves. Total phenol and flavonoid contents were highest in ethanol extracts from MBA grapevine leaves compared to extracts from Campbell Early grapevine leaves. Specific polyphenols higher in ethanol extracts from MBA grapevine leaves include gallic acid, epicatechin, caffeic acid, naringin, and resveratrol. Resveratrol content from MBA grapevine leaves increased when extracted for more than two hours in ethanol and water. The hydroxyl radical scavenging ability of ethanol extracts was higher than the water extract from both strains of grapevine leaves. DPPH and total antioxidants were highest in ethanol extracts from MBA grapevine leaves among the other extracts. Therefore, these results suggest that ethanol extracts from MBA grapevine leaves are a highly valuable resource for the development of natural functional foods.

Inhibitory Activities of Digestive Enzymes and Antioxidant Activities of Fermented Beverages Using Momordica charantia L. (여주를 첨가한 발효음료의 소화효소 억제와 항산화 활성)

  • This study was conducted to develop Momordica charantia L. juice fermented by four Lactobacillus species such as Lactobacillus paracasei (LPA), Lactobacillus plantarum (LPL), Lactobacillus rhamnosus (LRH), and Lactobacillus reuteri (LRE) as well as to investigate their inhibitory effects against digestive enzymes and antioxidant activities. Fermentation was performed at $37^{\circ}C$ without nutrient supplementation for 72 h. The pH and total lactic acid contents were within the ranges of 3.75~3.96 and 5.21~10.04% in fermented juices, respectively. The type of starter culture and fermentation time induced changes in flavonoid contents more than total phenolic contents. All juices fermented for 48 h strongly inhibited ${\alpha}$-glucosidase activity with the percentage of inhibition ranging of 91.24~95.05%. Antioxidant activities of all juices mostly increased after 48 h of fermentation. Our results suggest that fermented juice possesses inhibitory activity against digestive enzymes and antioxidant activity, and they can be used as health functional beverages.

  • A bacterium with potent activity of pigment production and protease was isolated and identified as being Bacillus sp. CS-17. Cell growth, protease activity and pigment production of the strain reached to its maximum point after 24 hrs, 48 hrs, 72 hrs, respectively. The best pigment producing ability of Bacillus sp. CS-17 was shown on basal medium for pigment production added 1.0% soybean. The high effcient conditions for pigment production was obtained at culture of pH 8.5, $37^{\circ}C$ and 72 hours. Among the tested 5 gram positive strains and 6 gram negative strains, weak antibacterial activity of pigment concentrated extracts was appeared against growth of B. subtilis, P. aeruginosa, S. typhimurium, E. aerogenes, B. cereus, A. hydrophila.

Production of Bioactive Components and Anti-Oxidative Activity of Soybean Grit Fermented with Bacillus subtilis HA according to Fermentation Time (Bacillus subtilis HA를 이용한 soybean grit의 고체발효 기간에 따른 생리활성물질 생산 및 항산화 효과)

  • Soybean grits, fortified with various bioactive components, were produced by solid-state fermentation using Bacillus subtilis HA. ${\alpha}$-Amylase activity gradually increased during fermentation over 5 days. Fibrinolytic and protease activities were highest in the soybean grits fermented for 7 days. The grits fermented for 5 days also showed the highest tyrosine content, indicating a higher peptide content. Peptides of low molecular weight (below 3,000 daltons) and browning pigments increased with increasing fermentation time. The fermented soybean grits showed higher contents of total phenolic compounds, to approximately 18 mg/g. DPPH free radical scavenging effects were higher in the soybean grits fermented for 3 days. Also, ABTS radical scavenging effects were greater in the fermented grits compared to the unfermented grits. Overall, the soybean grits fermented by solid-state fermentation for 5 days showed enhanced production of bioactive compounds and greater antioxidant properties.

Effects of Ozone on Photosynithetic Activity in Chloroplast of Barley (Hordeum vulgare L.) (오존이 보리(Hordeum vulgare L.) 엽록체의 광합성 활성에 미치는 영향)

  • The effects of 0.5 and 1.0 ppm of ozone on the developing chloroplast of barley (Hordeum vulgare L) seedling during greening were investigated by PSI and II activities, chlorophyll fluorescence, and the contents of chlorophyll. Etiolated barley seedling was treated 0.5 and 1.0 ppm ozone for the first 4 h during greening. In 24 h greening experiment, the contents of chlorophyll were decreased by increasing ozone concentration from 0.5 ppm to 1.0 ppm. In 24 h greening experiment, Fo, Fv, Fm and qE were greatly decreased as the concentration of ozone was increased, but those were not considerably decreased in 48 h greening experiment. In another experiment, the developing barley seedling was treated with 0.5 and 1.0 ppm ozone for the last 4 h during greening period, which was 24 h or 48 h. In both experiments the PS II activity was decreased as the concentration of ozone was increased, but not in PS I activity. Fv, Fm and Fv/Fo were also decreased as the concentration of ozone was increased. qP and qR were strikingly decreased as the concentration of ozone was increased in both experiments.iments.

생약재 추출물의 수율 및 항고혈압 활성

  • 본 실험에 사용한 생약재는 금산 약초시장에서 2003년 6월에 구입하여 추출수율을 조사하였다. 그 결과, 생약재의 물 추출물의 동결건조 중량(%)이 30~40%에 달하는 생약재는 가자, 갈근, 맥문동, 산수유가 있고, 20~30%에 달하는 생약재는 감초, 구기자, 당귀, 오미자, 천궁, 황기가 이에 속했으며, 중량(%)이 10~20%에 달하는 생약재는 복분자, 오수유, 애엽, 작약, 진피, 지유, 파고지, 황련이 이에 속했다. 마지막으로 10%이하의 추출 수율을 나타낸 생약재들은 가시오가피, 강황, 계피, 석곡, 소목, 영지, 오약, 육두구, 정향, 죽엽, 필발, 초두구, 호장근이 이에 속했다. 생약재의 70% Ethanol 추출물의 동결건조중량을 살펴본 결과, 동결건조 중량(%)이 30~40%에 달하는 생약재는 가자, 당귀, 산수유가 이에 속하였고, 20~30%에 달하는 생약재는 감초, 맥문동, 구기자, 오미자, 오수유, 천궁, 황기가 속하였으며, 동결건조 중량(%)이 10~20%에 달하는 생약재는 복분자, 육두구, 애엽, 작약, 정향, 지유, 파고지, 호장근, 황련이 이에 속했다. 또한 10% 이하의 수율을 나타내는 생약재는 가시오가피, 강황, 갈근, 계피, 석곡, 소목, 영지, 오약, 익지인, 죽엽, 필발, 초두구가 이에 속하였다. 생약재의 추출 시간 및 온도에 따른 수율을 조사한 결과, 추출 시간이 경과함에 따라서 수율이 점차 증가하는 경향을 보였으며, 가자, 갈근, 황련, 복분자, 지유, 정향, 계피, 소목 순으로 높은 수율을 얻을 수 있었다. 생약재에 여러가지 효소를 이용한 가수분해 결과, 대부분의 생약재에 대해 95$^{\circ}C$에서 2시간동안 추출한 수율과 그다지 큰 차이는 보이지 않았지만 대부분의 생약재가 Termamyl 효소를 사용하여 가수분해하였을 때 가장 높은 수율을 나타내었다. 또한 지유의 경우는 Viscozyme 효소를 처리하였을 경우 가장 좋은 수율을 나타내었다. 또한, 효소의 최적 온도로 조정한 Water bath에서 0, 30, 60, 120, 210분간 효소 반응 시켜, 420nm에서 갈변도를 시간별로 측정해본 결과, Viscozyme 효소를 처리한 생약재에서 시간이 흐름에 따라 흡광도 수치의 변화가 크게 나타났다. 32종의 생약재 물 추출물의 항고혈압 활성을 측정한 결과, 파고지, 소목, 죽엽의 항고혈압 활성이 높은 것으로 나타났으며, 파고지의 효과가 가장 우수한 것으로 나타났다. 10%에탄을 추출물의 경우에는 오미자, 오수유, 소목에서 항고혈압 활성이 높은 것으로 나타났으며, 오미자의 효과가 가장 우수한 것으로 나타났다. 파고지 물 추출물을 칼럼(Sephacryl S-300, High Resolution)으로 분획하여 3개의 Peak를 얻었으며, 2번째 Peak의 항고혈압 활성이 매우 높은 것으로 나타났다.

  • 본 연구의 목적은 비외과적 치주치료시 부가적으로 사용하기 위해 실험적으로 개발한 젤 형태의 테트라사이클린 및 테트라사이클린-구연산 혼합젤의 치근면에 대한 시간에 따른 활성도를 측정하고, 이를 용액 형태의 테트라사이클린 제제 또는 클로르헥시딘 들과 비교하는 것이다. 6명의 환자로 부터 18개의 발치된 치아를 실험대상으로 하였으며, 치아는 발치한 즉시 치석제거술과 치근활택술을 시행한 후 각 각 4개씩 4군으로 나누어 다옴과 같은 처치롤 하였다 ; 1) O.1% 클로르핵시딘 용액에 5분간 침전; 2) 50mg/ml의 테트라사이클린 용액에 5분간 침전; 3) 5% 테트라사이클린 젤에 5 분간 처리 ; 4) 테트라사이클린-구연산 혼합첼로 5 분간 처치; 5) 그리고 2개의 치아는 대조군으로서 멸균된 생리식염수에 5분간 처리하였다. 침전후 치아는 1ml의 tris-buffered saline이 담긴 용기에 옮겨 24시간 간격으로 탈착된 TBS용액올 교체하면서 실온에서 22일간 보관하였다. Porphyromonas gingivalis를 indicator organism으로 하여 microtiter assay를 이용하여 홉광도를 측정 함으로써 제거 된 용액 의 항 미생물 활성올 측정하였다. 1. 50mg/ml 의 테트라사이클린 수용액에 침전되었던 군은 생리식염수로 처리한 군에 비하여 17 일간 클로르헥시딘으로 처리한 군에 비하여는 16일간 항미생물 활성에 있어서 유의성 있는 차이를 보였다. 2. 테트라사이클린 젤과 테트라사이클린-구연산 혼합첼로 처리한 군은 대조군에 비하여 각 각 4일과 3일 까지 활성올 보였다. 3. 0.1% 클로르핵시딘 용액으로 처리한 군은 생리식염수로 처치한 군에 비하여 24시간 밖에 활성을 나타내지 못했다. 4. 전반척으로 테트라사이클린-구연산 혼합첼로 처리한 군에 비하여 테트라사이클린 첼로 처리한 군의 활성이 높았으나 유의성 있는 차이롤 보이지는 않았다.

Characteristics of Activated Carbon Prepared from Waste Citrus Peel by KOH Activation (KOH 활성화법으로 제조한 폐감귤박 활성탄의 특성)

  • An activated carbon was prepared from waste citrus peel produced in large amounts in Jeju Island, Korea, using KOH activation and its characteristics was examined. Under the condition of the KOH ratio between 100 and 300%, activation temperature from 400 to $900^{\circ}C$ and activation time from 0.5 to 1.5 h, the iodine adsorptivity of the activated carbon prepared increased but the yield decreased with respect to the increase of each conditions. The iodine adsorptivity and yield of the activated carbon prepared at the activation time of more than 1.5 h were similar to those of using 1.5 h. In addition, as the KOH ratio increased, the specific surface area and pore volume of the activated carbon increased, but the pore diameter decreased. The activated carbon has an average pore diameter of $20{\sim}25{\AA}$. Also, the activated carbon prepared at 300% KOH and $900^{\circ}C$ for 1.5 h has the highest specific surface area of $1,527m^2/g$ and iodine adsorptivity of 1,246 mg/g.
