Review on hazardous microcystins originating from harmful cyanobacteria and corresponding eliminating methods (유해 남세균 유래 마이크로시스틴의 위해성과 제거 방안 고찰)

  • Cyanobacterial harmful algal blooms (Cyano-HABs) are an international environmental problem that negatively affects the ecosystem as well as the safety of water resources by discharging cyanotoxins. In particular, the discharge of microcystins (MCs), a highly toxic substance, has been studied most actively, and various water treatment methods have been proposed for this purpose. In this paper, we reviewed adsorption technology, which is recognized as the most feasible, economical, and efficient method among suggested treatment methods for removing MCs. Activated carbons (AC) are widely used adsorbents for MCs removal, and excellent MCs adsorption performance has been reported. Research on alternative adsorption materials for AC such as biochar and biosorbents has been conducted, however, their performance was lower compared to activated carbon. The impacts of adsorbent properties(characteristics of pore surface chemistry) and environmental factors (solution pH, temperature, natural organic matter, and ionic strength) on the MCs adsorption performance were also discussed. In addition, toward effective control of MCs, the possibility of the direct removal of harmful cyanobacteria as well as the removal of dissolved MCs using adsorption strategy was examined. However, to fully utilize the adsorption for the removal of MCs, the application and optimization under actual environmental conditions are still required, thereby meeting the environmental and economic standards. From this study, crucial insights could be provided for the development and selection of effective adsorbent and subsequent adsorption processes for the removal of MCs from water resources.

Acinetobacter Isolates Growing with Carbon Monoxide (일산화탄소를 이용하여 성장하는 acinetobacter의 분리 및 동정)

  • Three strains (JC1, JC2, and HY1) of aerobic carbon monoxide (CO)-utilizing Acinetobacter were isolated from soil through CO-enrichment culture technique. All of them were Gram-negative, nonmotile, and rod-shated but they were changed to spherical form at the end of logarithmic phase. They were resistant to penicillin and able to frow at $42^{\circ}C$. The guanine plus cytosine contents of the DNAs ranged from 43 to 44.5 mol%. Oxidase was not present in all cells. The colonies were smooth and whitish yellow. Heterotrophic growth occurred on several sugars, organic acids, amino acids, and alcohols. The doubling times under and atmosphere of 30% CO and 70% air at $30^{\circ}C$ were 19h, 25h, and 35h, respectively, for JC1, JC2, and HY1, JC1 was studied in more detail. The cells were grown optimally in a mineral medium (pH 6.8) under a gas mixture of 30% CO and 70% air at $30^{\circ}C$. Growth of the cells with CO did not depend on molybdenum. It was able to grow with 100 ppm of CO in air as a sole source of carbon and energy.

Screening of Myxobacteria Inhibiting the Growth of Collectotrichum acutatum Causing Anthracnose on Pepper (고추탄저균 성장 억제 점액세균의 탐색)

  • As an effort to search new bacterial biocontrol agents against pepper anthracnose, we screened myxobacteria, which might inhibit the growth of Colletotrichum acutatum, the agent of that plant disease. When 93 myxobacterial strains including 59 Myxococcus spp. and 34 Corallococcus spp. were tested against C. acutatum ACYSJ001 on agar plates, 10 strains identified as the genus Myxococcus significantly obstructed the growth of C. acutatum, whereas the majority of strains belonging to the genus Corallococcus did not demonstrate any counteractive effect. Such results have indicated that the strains of the genus Myxococcus have a high potential to play roles of biocontrol agents for control of pepper anthracnose. These also have revealed that the strains of the genus Myxococcus could be used as excellent microbial resources for screening novel antifungal substances.

Antibacterial Effects of Sophora flavescens on Streptococcus mutans (고삼의 Streptococcus mutans에 대한 항세균 효과)

  • The purpose of this study is to investigate antibacterial activities of Sophora flavescens Ait. extracts for the prevention of dental caries caused by Streptococcus mutans. The fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. Showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 ${\mu}$g/ml). The glucosyltransferase activity inhibited the formation of glucan by the fraction 5-4-3 and showed 77% of the antiproliferative effect at 100 ${\mu}$g/ml. This showed a significant activity (p<0.05). These results suggest that the fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. may be a valuable material as the prevention of dental caries.

Isolation of Insoluble Phosphate-Solubilizing Bacteria and Optimum Condition for Solubilization (인광석 가용화 세균의 분리 및 가용화 최적조건)

  • 850 strains of phosphate-solubilizing bacteria were isolated from soil of Chung-nam and Daejeon region using 0.5% calcium phosphate containing medium. The HS-2 strain with the highest rock phosphate-solubilizing activity was selected and identified as Azotobacter sp. HS-2 based on the microbiological characteristics. The optimum culture temperature and initial pH of medium for solubilization of rock phosphate were $30^{\circ}C$ and pH 6.0-7.0, respectively. Addition of oxalic acid(0.5M) into the PDB-rock phosphate medium increased 50% solubilization of rock phosphate.

  • 활성 탄의 상층부위에서 가장 많은 수의 세균이 존재하였으며, 중층, 하층 순으로 점차 줄어들었다. 수온이 낮을 때 $\beta$가 우점 하였고, 수온이 올라가는 때는 $\alpha$, ${\gamma}$의 수치가 높아졌으며, C-F는 전체적으로 골고루 분포하였다. HGC는 수온에 따라 약간의 증가가 있었다.

Enhanced Antibacterial Activity of Sodium Hypochlorite under Acidic pH Condition (산성 pH 조건에서 차아염소산나트륨의 항균 활성 향상)

  • Sodium hypochlorite (NaClO) is a disinfectant widely used in hospitals and food industries because of its antimicrobial activity against not only bacteria but also fungi and virus. The antibacterial activity of NaClO lies in the maintenance of a stable hypochlorous acid (HClO) concentration, which is regulated by pH of the solution. HClO can easily penetrate bacterial cell membrane due to its chemical neutrality and the antibacterial activity of NaClO is thought to depend on the concentration of HClO in solution rather than hypochlorite ions (ClO-). In this study, we investigated the antibacterial activity of NaClO according to pH adjustment by means of time kill test and assays of Reactive Oxygen Species (ROS) and adenosine triphosphate (ATP) concentration changes before and after NaClO treatment. We also investigated that the degree of cell wall destruction through field emission scanning electron microscopy (FE-SEM). Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) exposed to 5 ppm NaClO at pH 5 exhibited 99.9% mortality. ROS production at pH 5 was 48% higher than that produced at pH 7. In addition, the ATP concentration in E. coli and S. aureus exposed to pH 5 decreased by 94% and 91%, respectively. As a result of FE-SEM, it was confirmed that the cell wall was destroyed in the bacteria by exposing to pH 5 NaClO. Taken together, our results indicate that the antibacterial activity of 5 ppm NaClO can be improved simply by adjusting the pH.

Antimicrobial Activity of Korean Leek and Its Application to Food System (부추의 항균활성 특성과 식품에의 응용)

  • Antimicrobial activity of Korean leek was effective on Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, E. coli O157:H7, Pseudomonas fluorescens, Pichia membranaefaciens. Extract from raw Korean leek and that from frozen one showed antimicrobial activity; in contrast, extract stored frozen condition or extract from blanched one lost antimicrobial activity. Filtration, centrifugation, and vacuum concentration did not affect antimicrobial activity of the leek, nor did the changes in storage pHs. Addition of ground leek to soy milk, soy curd, and broth for cold noodle resulted in the prolonged shelf life by significantly reducing the number of viable cells.

  • Hemolytic property is a specific feature of bacteria to obtain iron which is essential for its survival in host tissues. Therefore, it is thought to be one of several factors of virulence. The purpose of this study was to investigate the hemolytic properties of Prevotella nigrescens isolated from the teeth diagnosed as pulp necrosis and apical periodontitis under the presence of hemolysin inhibitors such as $NaN_3$ and dithiothreitol. heat, various pH and cultural conditions. The results were as follows; 1. Clinically isolated P. nigrescens strains and standard P. nigrscens ATCC 33563 showed hemolytic activity. 2. P. nigrescens showed higher hemolytic activity against human erythrocytes than sheep or horse erythrocytes. 3. $NaN_3$ and dithiothreitol (DTT) reduced the hemolytic activity of P. nigrescens in a dose dependent manner (p<0.05). 4. Optimal pH for the maximum hemolytic activity of P. nigrescens was 4.0 and the hemolysin was stable under the $50^{\circ}C$, but the hemolytic activity was significantly decreased at $95^{\circ}C$. 5. P. nigrescens cultured in $10\%\;CO_2$ condition showed higher hemolytic activity than the bacteria cultured in the anaerobic condition.

  • Acetylation of triazinylated monosaccharides was carried out. The products were identified by elementary analysis. IR and NMR with Methyl $[T]-{\beta}-L-acetyl-arabopyranoside$ and Methyl $[T]-\alpha-D-acetyl-lyxopyranoside$ respectively. The triazine derivatives of uronic acids had no remarkable biological activities against dish and rice, and also showed not to be effected on antifungal and antibacterial activities.
