저수지 유입부에 조성한 저류지의 수질개선효과 및 미생물학적 특성

  • 유역의 경사가 비교적 급하고, 소규모 축산농가의 미처리된 축산폐수 및 농경지 배수, 산재된 마을하수 등 비점오염원의 유출이 강우시 집중적으로 이루어지는 중산간 농업용 저수지의 수질개선을 위한 공법으로 부영양화된 충남 아산 마산저수지의 유입부에 조성한 저류지를 이용한 수질개선효과 및 물질분해자로서 미생물학적 특성을 2000년부터 2002년까지 3년간 연구함으로서 향후 유사한 농업용저수지의 수질개선 방안으로 실용화 가능성을 모색하고자 하였다. 그 결과 수질정화효과는 총질소와 총인 등 영양염류의 정화효과가 비교적 우수하게 나타났으며, 유기오염물질로서 BOD, COD, SS 등은 평균 수질정화효과는 (-)의 값을 보여주었으나, 강우기에는 정화효율이 높게 나타났다 또한, 저류지의 물질 침전기능 외 오염물질의 활발한 분해작용을 살펴보고자 세균 분포 특성을 조사한 결과 유입수와 저류지, 저수지 각각의 수체에서 조사된 총세균수 및 총세균수에 대한 진정세균수의 비율이 유입수<저류지<유출수의 순으로 나타나 저류지의 유기물질 분해 활성이 높음을 보여주고 있다. 따라서 저수지 유입부에 조성한 저류지의 수질개선효과는 강우시 입자성 유기물질의 효과적 제거 뿐만아니라 영양염류의 제거효과, 특히 질소의 제거효과가 높은 정화기구로서 설치비용이 적고, 유지관리가 간단한 특성을 살려 유사한 중규모 농업용저수지의 수질개선 기법으로 널리 활용할 가치가 있음으로 사료된다.

Comparative Study of Pathogenicity Following Single or Coinfection with Edwardsiella tarda and Streptococcus iniae in Olive Flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus에서 Edwardsiella tarda와 Streptococcus iniae 단독 또는 혼합감염의 병원성 비교 연구)

  • Olive flounders, Paralichthys olivaceus, were intraperitoneally challenged with Edwardsiella tarda or Streptococcus iniae or both bacteria simultaneously. The pathogenicity was respectively compared with blood chemical using alanine aminotransferase (ALT), aspatate aminotransferase (AST), glucose and total protein, lysozyme activity, bacterial number of kidney and spleen, histopathological change, and cumulative mortality. The tested group of coinfection showed increased cumulative mortality, bacterial number of kidney and spleen, AST and histopathological change, but not in lysozyme activity compared with others. This study provides support for the conclusion that simultaneous infection with Edwardsiella tarda and Streptococcus iniae in a susceptible host results in higher pathogenicity, leading to the increment of bacterial number and the destruction of the internal organs.

  • In order to produce alkaline protease, psychrotrophic Bacterium which have high enzyme activity, was isolated by using enrichment culture from soil samples and identified as genus Bacillus sp. The optimal pH and temperature for the enzyme activity were pH 6 and $40^{\circ}C$. The temperature range of high enzyme activity was $20{\sim}40^{\circ}C$. The optimal initial pH of culture condition for enzyme was pH 6. The most favorable carbon and nitrogen sources for the production of protease by Bacillus sp. WRD-2 were 3% maltose and 4% yeast extract, respectively.

Foliar Application of the Fungicide Pyraclostrobin Reduced Bacterial Spot Disease of Pepper (Fungicide pyraclostrobin의 고추 세균점무늬병 예방효과)

  • Pyraclostrobin is a broad-spectrum fungicide that inhibits mitochondrial respiration. However, it may also induce systemic resistance effective against bacterial and viral diseases. In this study, we evaluated whether pyraclostrobin enhanced resistance against the bacterial spot pathogen, Xanthomonas euvesicatora on pepper (Capsicum annuum). Although pyraclostrobin alone did not suppressed the in vitro growth of X. euvesicatoria, disease severity in pepper was significantly lower by 69% after treatments with pyraclostrobin alone. A combination of pyraclostrobin with streptomycin reduced disease by over 90% that of the control plants. The preventive control of the pyraclostrobin against bacterial spot was required application 1-3 days before pathogen inoculation. Our findings suggest that the fungicide pyraclostrobin can be used with a chemical pesticide to control bacterial leaf spot diseases in pepper.

Analysis of Bacterial Community Structure Using FISH in the Juam Reservoir (FISH법으로 분석한 주암호의 세균군집구조)

  • Bacterial community structure in the Juam Reservoir was analysed using fluorescent in situ hybridization (FISH) technique from April 2005 to January 2006. Total bacterial numbers varied in the range of 1.58 ${\sim}\;2.73{\times}\;10^6\;cells\;mL^{-1}$ proportional to the concentration of chi-a and had the minimal value in January. The ratios of ${\alpha}\;{\cdot}\;{\beta}\;{\cdot}\;{\gamma}$-subclass proteobacteria and Cytophaga-Flavobacterium (CF) group to total bacteria ranged from 45.1% to 77.5%, and the ratios of ${\alpha}\;{\cdot}\;{\beta}\;{\cdot}\;{\gamma}$-subclasses to total bacteria in June and September with the concentration of chi-a measured were lower than those ratios in April and January. It suggests that enriched growth of Microcystis aeruginosa may inhibit the metabolic activlty of ${\alpha}\;{\cdot}\;{\beta}\;{\cdot}\;{\gamma}$-subclass proteobacteria. However, the ratio of CF group bacteria represented no large change depending on algal bloom. In terms of nitrifying bacteria, the numbers of ammonia-oxidizing bacteria ranged from 9.9 ${\times}\;10^4\;to\;25.5\;{\times}10^4\;cells\;mL^{-1}$ with sharp fluctuation whereas those of nitrite-oxidizing bacteria varied in 8.7${\sim}9.8{\times}10^4\;cells\;mL^{-1}$ without noticeable change except the maximal value of $20.3{\times}10^4\;cells\;mL^{-1}$ in January maybe due to the high DO.

Antimicrobial Effect of Plagiorhegama dubium Extracts on Food-borne Pathogen (식중독 유발세균의 증식에 미치는 황련 추출물의 효과)

  • This study was performed to investigate the antimicrobial activities of the Plagiorhegama dubium extracts against food-borne pathogens. The methanol extract was partitioned into petroleum ether, chloroform, ethyl acetate, and methanol portions. The antimicrobial activity of the P. dubium extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of P. dubium showed the highest antimicrobial activity against Staphylococcus aureus and Escherichia coli. The synergistic effect has been found in combined extracts of P. dubium and Hedyotis diffsa as compared to each extracts alone. Finally, the growth inhibition curve was determined using methanol extracts of P. dubium against S. aureus and E. coli. The methanol extract of P. dubium showed strong antimicrobial activity against S. aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate portion from P. dubium retarded the growth of S. aureus more than 36 hours and E. coli up to 60 hours. The methanol extracts of P. dubium has been shown the antimicrobial activity against S. aureus and E. coli.

Optimization of γ-Aminobutyric Acid (GABA) Production Using Immobilized Lactobacillus plantarum K154 in Submerged Culture of Ceriporia lacerata (Ceriporia lacerata 배양액과 고정화 Lactobacillus plantarum K154를 이용한 감마아미노뷰티르산 생산 최적화)

  • The production of GABA was optimized by co-cultivation of immobilized Lactobacillus plantarum K154 (ILK) with Ceriporia lacerata cultures. The mycelial culture of C. lacerata was performed in a defined medium containing 3% glucose, 3% soybean flour, and 0.15% $MgSO_4$ in a submerged condition for 7 days at $25^{\circ}C$, resulting in the production of 29.7 g/L mycelia, 3.1 g/L exopolysaccharides, 2% (w/w) ${\beta}$-glucan, 68.96 unit/mL protease, and 10.37 unit/mL ${\alpha}$-amylase. ILK in C. lacerata culture showed viable cell counts of $3.13{\time}10^9CFU/mL$ for immobilized cells and $1.48{\time}10^8CFU/mL$ for free cells after 1 day. GABA production in the free and immobilized cells was 9.96 mg/mL and 6.30 mg/mL, respectively, after 7 days. A recycling test of ILK in the co-fermentation was consequently performed five times at $30^{\circ}C$ for 15 days, resulting in the highest production of GABA. GABA could also be efficiently overproduced by co-cultivation with the produced polysaccharides, ${\beta}$-glucan, peptides, and probiotics.

Characteristics of Antimicrobial Activities for the Human Pathogenic Microorganism by Extracts from Korean Mushrooms (버섯 추출물이 인체 병원성 균에 미치는 항균활성의 특성)

  • This study was performed to screen antimicrobial activities of 198 extracts from 66 Korean mushrooms against 19 human pathogenic microorganisms using paper disc method. Mushrooms were extracted with petroleum ether 80% ethanol and distilled water in that order Among the extracts with antimicrobial activities, 1 water extract of Amanita virgineoides, 8 ethanolic extracts including Amanita and 1 petroleum ether extrac of Psathyrella hydrophila were highly active against fungi, respectively. In addition to, 24 extracts including Amanita pseudoporphyria, Amanita spissacea, 3 extaracts including Paxillus curtisii were highly active against Gram negative and positive bacteria, respectively.

The Antimicrobial Activity of Bacterial-challenged Black Soldier Fly, Hermetia illucens (세균에 의해 면역이 유도된 동애등에의 항균활성)

  • In the larvae of the black soldier fly, Hermetia illucens, innate immunity mechanisms are activated in response to various pathogens and stimulants, resulting in the expression of antimicrobial peptides (AMPs). To induce the mass production of AMPs, H. illucens fifth instar larvae were immunized with five different kinds of bacteria. We isolated from the hemolymph of the H. illucens larvae after bacterial challenge, and their antimicrobial activities against Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli) were measured using the inhibition zone assay. Among these five different kinds of bacteria, the hemolymph of Bacillus subtilis-challenged H. illucens larvae showed the strongest antimicrobial activity against both Gram-positive bacteria and Gram-negative bacteria. The antimicrobial activity of the hemolymph of $1{\times}10^9cfu/ml$ B. subtilis-challenged H. illucens peaks at 24 hr at 48 hr post-infection and gradually declines with time. Moreover, the immunized hemolymph also showed strong antimicrobial activity against various poultry pathogens such as S. enteritidis, S. typhimurium, and S. pullorum. These results suggest that the expression of AMP genes in B. subtilis-challenged H. illucens is up-regulated by innate immune responses, and that B. subtilis-challenged H. illucens overexpressing AMPs may be useful as a feed additive in livestock diets to reduce the need for antibiotics.

Studies on the Development of Natural Preservatives from Natural Products (전통식품 및 천연물에서 천연보존료 개발에 관한 연구)

  • Certain parts of 190 kinds of medicinal herbs and 171 kinds of original materials of food were extracted by methanol. The extracts were tested their microbial inhibition activities against several food spoilage microorganisms, Micrococcus luteus, Bacillus subtilis, Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Escherichia coli, Saccharomyces cerevisiae, Candida albicans, Penicillium citrinum, Aspergillus flavus and Aspergillus niger. The methanol extracts of Cornus officinalis, Evodia officinalis, Glycyrrhiza glabra, Salvia miltiorrhiza. Schizandrae fructus, Coptidis rhizoma, aroma hop and bitter hop were shown inhibitory effect on certain species of gram(+) bacteria. Aroma hop and bitter hop were shown inhibitory effect on certain species of gram(-) bacteria. The methanol extract of Salvia miltiorrhiza exhibited a strong antibacterial activities. It was purified by solvent fractionation, silicagel column chromatography, prep. TLC, prep. HPLC. The purified active substance was identified as cryptotanshinone by EIMS, $1^H-NMR,\;{13}^C-NMR$ and DEPT. Cryptotanshinone showed a strong antibacterial activity against gram positive bacteria $(MIC\;:\;3.91{\sim}62.50\;{\mu}g/mL)$. Especially, this compound was the most strong activity against Bacillus subtilis $(MIC\;:\;3.91\;{\mu}g/mL)$.
