Antioxidative and Physiological Activity of Extracts of Angelica dahurica Leaves (구릿대 잎 추출물의 항산화 효과 및 생리활성)

  • This study prepared extracts of Angelica dahurica leaves using reflux water extraction (RW), reflux ethanol extraction (RE) and pressure heating water extraction (PW). The extracts were extraction for levels of polyphenol compounds, antioxidant activities, and inhibitory potencies for xanthine oxidase and tyrosinase. The PW extraction method yielded the highest content of polyphenol compounds (95.23 mg/g). The electron donating abilities (EDAs) of RE and PW extracts were 76.02% and 70.08% respectively. The superoxide dismutase (SOD)-like activities were 13.45 19.00%, when extracts were assayed at 1 mg/mL. The nitrite scavenging ability (pH 1.2) of the PW extract was 54.33% higher than levels shown (44.24%) by the RE and RW extracts. The inhibition of xanthine oxidase by the RW extract was highest (99.71% at 5 mg/mL) while that of the PW extract was over 97% at 500 g/mL. Tyrosinase inhibition was highest in the RE extract (46.25% at 5 mg/mL). All extracts showed dose-dependent inhibitory activities. The results indicated that the PW extract had the highest polyphenol content, the RW and RE extracts had the best nitrite scavenging ability, and the RE extract showed the most pronounced effect on EDA, SOD-like activity and tyrosinase inhibition.

Study on Antioxidant Effects of Fractional Extracts from Ligularia stenocephala Leaves (곤달비(Ligularia stenocephala) 잎 분획물의 항산화활성 평가)

  • Ligularia stenocephala leaves were subjected to extraction by using several solvents with different polarities, which were then investigated for their antioxidant and antimicrobial activities. Extraction yield of ethanol extract of Ligularia stenocephala was 21.36%. The water fraction showed the highest extraction yield of 60.35%, followed by the n-butanol, n-hexane, and ethyl acetate fractions. Total polyphenolic content was the highest (254.00 mg/g GAE) in the ethyl acetate fraction of Ligularia stenocephala extract. The ethyl acetate fraction had $IC_{50}$ values of 0.28 mg/mL for DPPH radical scavenging, and 96.$67{\pm}0.09%$ for ABTS radical scavenging activity. Further, FRAP value was significantly higher in the ethyl acetate fraction. The ethyl acetate fraction showed antimicrobial activities against B. cereus, and the diameter of the zone of inhibition was 10.2 mm at 5 mg/disc. These results suggest that the ethyl acetate fraction of Ligularia stenocephala possesses the antimicrobial activities against B. cereus, and its high antioxidant activity could be applicable to food additives and as a natural cosmetic ingredient.

Antioxidative and Physiological Activities of Fractions from Pleurospermum kamtschaticumin Extracts (누룩치(Pleurospermum kamtschaticumin) 추출물의 항산화 효과 및 생리활성)

  • This study investigated the antioxidant activities, whitening effects, and antimicrobial activities of fractions from Pleurospermum kamtschaticumin extract. Total polyphenolic contents of fractions from Pleurospermum kamtschaticumin extract were 116.44~382.73 mg/g GAE (gallic acid equivalent), with the highest value in the ethyl acetate fraction. Fractions of Pleurospermum kamtschaticumin showed the highest DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity ($IC_{50}$=0.04 mg/mL) and ABTS radical scavenging activity (98.86% at 0.5 mg/mL), which was similar to ascorbic acid. Further, reducing power and FRAP activity were significantly high in the ethyl acetate and n-butanol fractions. The ethyl acetate and n-butanol fractions showed significantly high SOD-like activities (1 mg/mL, 86.93%, and 78.23%, respectively) compared to other fractions. Tyrosinase inhibition activities of the n-butanol fraction and 80% ethanol extract were 43.52% and 41.64%, respectively. Antimicrobial activity against Escherichia coli was only observed in the chloroform fraction (16.7 mm in 0.5 mg/disc). These results suggest that fractions from Pleurospermum kamtschaticumin extract show comparatively high antioxidant activity and thus could be used as a food additive or cosmeceutical ingredient.

Antioxidative Effects of Campanula takesimana Nakai Extract (섬초롱(Campanula takesimana Nakai) 용매 추출물의 항산화 효과)

  • The study analyzed the total polyphenolic contents, DPPH radical scavenging activities, ABTS radical scavenging activities, reducing power levels, SOD-like activities, and tyrosinase inhibition activities of fractions from Campanula takesimana Nakai extract. Total polyphenolic content of the ethyl acetate fraction from Campanula takesimana Nakai extract was the highest. The ethyl acetate fraction of Campanula takesimana Nakai extract showed the lowest $IC_{50}$ value (0.13 mg/mL) as well as greater than 98% ABTS radical scavenging activity, which was similar to ascorbic acid. Further, reducing power was significantly higher in the ethyl acetate fraction. The SOD-like activity of edible plant was 70.56% (n-butanol fraction) and 62.27% (ethyl acetate fraction) at 1 mg/mL. Tyrosinase inhibition activity of Campanula takesimana Nakai extract was reduced to 34.77% in the n-butanol fraction and 30.85% in the ethyl acetate fraction at 5 mg/mL. These results suggest that the ethyl acetate and n-butanol fractions of Campanula takesimana Nakai extract could be applicable as food additives or cosmeceutical ingredients.

Lactic acid Production from Hydrolysate of Pretreated Cellulosic Biomass by Lactobacillus rhamnosus (전처리된 섬유소계 바이오매스로부터 Lactic acid생산)

  • Lactic acid, the most widely occurring hydroxy-carboxylic acid, has traditionally been used as food, cosmetic, pharmaceutical, and chemical industries. Even though it has tremendous potential for large scale production and use in a wide variety of applications, high cost lactic acid materials are primarily problems. Lactic acid can be obtained on either by fermentation or chemical synthesis. In recent years, the fermentation approach has become more successful because of the increasing market demand for naturally produced lactic acid. Generally, lactic acid was produced from pure starch or from glucose. As an alternative, biomass which is the most abundant renewable resources on earth have been considered for conversion to readily utilizable hydrolysate. In this study, we conducted the fermentation method to produce L(+)-lactic acid production from pretreated hydrolysate was investigated by Lactobacillus rhamnosus ATCC 10863. The hydrolysate was obtained from pretreatment process of biomass using Ammonia percolation process (AP) followed by enzymatic hydrolysis. In order to effectively enhance lactic acid conversion and product yield, controlled medium, temperature, glucose concentration was conducted under pure glucose conditions. The optimum conditions of lactic acid production was investigated and compared with those of hydrolysate.

Inhibitory Effect on Tyrosinase, ACE and Xanthine Oxidase, and Nitrite Scavenging Activities of Jubak (Alcohol filter cake) Extracts (주박 추출물의 아질산염 소거와 Tyrosinase, Xanthine Oxidase, ACE 저해 효과)

  • The purpose of this study was to investigate the tyrosinase, xanthine oxidase, angiotensin converting enzyme (ACE) inhibitory effects, and nitrite scavenging abilities of Jubak (alcohol filter cake, AFC) hot water and ethanol extracts. Tyrosinase inhibitory effects of Jubak hot water and ethanol extracts were above 85%. Nitrite scavenging ability was higher at pH 1.2 than at pH 4.2 and 6.0. Nitrite scavenging abilities of water and ethanol extracts were above 90% at pH 1.2. Xanthine oxidase inhibitory effect of Jubak hot water and ethanol extracts showed a lower, and the effect did not different by hot water and ethanol. ACE inhibitory effects of Jubak hot water and ethanol extracts were approximately 43~53% and 36~47%, respectively. Overall, our results indicate that Jubak hot water and ethanol extracts may have tyrosinase and ACE inhibitory effects and nitrite scavenging ability, but no xanthine oxidase inhibitory effect. Therefore, Jubak has potential as a cosmetic raw material. Additionally, Jubak could be used as a food additive.

Evaluation of Antioxidant and Antimicrobial Activities of Solvent Extracts from Coriolus versicolor (구름버섯(Coriolus versicolor)의 용매 추출물에 대한 항균 및 항산화 활성 조사)

  • Antioxidant activities of various solvent extracts from Coriolus versicolor were investigated for their total polyphenol content, total flavonoid content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging activity. C. versicolor extracts were produced by organic solvents such as ethanol, ethyl acetate, and methanol. Total polyphenol and flavonoid contents in the ethanol and ethyl acetate extracts were higher than those in the methanol extract. DPPH radical scavenging activity of methanol extract showed the highest value of 80.3%, which was similar to that of ascorbic acid (85.5%). All extracts showed good (>90.0%) ABTS cation radical scavenging activity. The antimicrobial activities of these extracts from C. versicolor were evaluated against six strains of bacteria using the disc diffusion method. All extracts showed antimicrobial activities against all tested bacteria except Staphylococcus aureus. These results indicate that various extracts from C. versicolor could be useful as natural antioxidant and antimicrobial agents.

Antioxidative Activities and Whitening Effects of Ethyl Acetate Fractions from The Immature Seeds of Abeliophyllum distichum (미선나무 미성숙 종자의 항산화 및 미백 활성)

  • Abeliophyllum distichum Nakai is deciduous shrubs of flowering plant in Oleaceae. It is important plant resources and consists of one species in the world. Also the endemic plant of A. distichum has been protected and designed endangered plant in Korea. For this reason, study on the immature seeds of A. distichum (ADS) hasn't progressed. In the present study, we evaluated the antioxidant activity and inhibitory effects on proteins and mRNA levels were related in the whitening effect in B16F10 cells. ADS was effective for reaction oxygen species (ROS). ROS causes various diseases such as aging, inflammation, cancer, and etc. Antioxidant properties were evaluated DPPH, ABTS radical scavenging activity and Reducing power. Plants were known that contained phenolic compounds were related in antioxidant activity. Phenolic compounds were phytochemicals commonly named natural polyphenols. These are secondary metabolites of plants involved in the defense against different types of stresses. In results, ADS suppressed the expression and transcription of Tyrosinase, TRP-1, TRP-2, and Microphthalmia-associated transcription factor (MITF). Tyrosinase, tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 1 (TRP-2) are known to play an important role in melanin biosynthesis. MITF regulated the expression and transcription of Tyrosinase, TRP-1, and TRP-2. In conclusion, ADS was effective in both antioxidant activities and whitening effects. Also, they were associated with the content of phenolic compounds. We suggested that ADS can be use antioxidants and skin-whitening functional cosmetics material derived from natural plant resources.

Antioxidant, Antimicrobial and Anti-inflammatory Activities of Essential Oil from Erigeron annuus L. Flower (개망초꽃 에센셜 오일의 항산화, 항균 및 항염 활성)

  • This study was designed to examine the in vitro antioxidant, antimicrobial and anti-inflammation effects of essential oils of Erigeron annuus L. Flower. Erigeron annuus L. essential oils were obtained by solvent extraction. Antioxidative ability was evaluated by bioassays using ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid diammonium salt) radical scavenging effect and 2, 2-diphenyl-1-1-picrydrazyl (DPPH) free radical scavenging activity. Erigeron annuus L. essential oil exhibited free radical scavenging activity on ABTS and DPPH 98.6%, 48.3% respectively, at a concentration of $500{\mu}g/ml$. Antimicrobial activity of essential oils of Erigeron annuus L. were tested against Staphylococcus aureus (S. aureus), Propionibacterium acnes (P. acne) and Escherichia coli (E. coli) by paper disc method, MIC and MBC. Erigeron annuus L. essential oil showed excellent antibacterial activities against S. aureus with MIC and MBC values of 0.31 mg/mL. The clear zone, indicating antimicrobial activity against P. acnes, was 14 mm, MIC and MBC values 0.31 mg/mL, 0.63 mg/mL, respectively. For the anti-inflammatory activity in RAW 264.7 cell, the Erigeron annuus L. essential oils inhibited not only NO production but also the expression of pro-inflammatory cytokines such as, TNF-${\alpha}$, IL-6 in a dose-dependent manner. These results suggested that Erigeron annuus L. essential oils has considerable potential as a cosmetic ingredient with antioxidative, antimicrobial and anti-inflammation effects.

Antioxidant activity and anti-inflammatory effects of ethanol extract from Allium schoenoprasum (향부추 에탄올 추출물의 항산화 활성 및 항염증 효과)

  • This study evaluated the antioxidant and anti-inflammatory activity of ethanol extracts using three parts of the chives plant: the bulb, the leaf, and the flower. As a result of DPPH and ABTS radical scavenging ability, the scavenging activity of the flower extract was higher than that of the bulb and leaf. In addition, as a result of FRAP analysis, antioxidant activity increased in all extracts depending on the extract concentration. The total polyphenol content was high in the following order: flower (11.29±0.37 mgGAE/g) > leaf (6.61±0.14 mgGAE/g) > bulb (5.7±0.67 mgGAE/g) extract. The cytotoxicity of the three extracts against rat macrophage RAW264.7 cells and HaCaT cells, both of which are human cutaneous keratinocyte cell lines, was minimal. NO by LPS was generated as a result of examining the anti-inflammatory activity of each extract through the NO colorimetric analysis method and ELISA. TNF-α secretion was decreased to a significant level in the flower ethanol extract. Therefore, these results indicate that there is a high possibility that the ethanol extract of chives, a natural plant resource, can be used as a cosmetic raw material.
