The Effects of Kimchi with Monascus purpureus on the Body Weight Gain and Lipid Metabolism in Rats Fed High Fat Diet (홍국 첨가 김치가 고지방식이를 투여한 흰쥐의 체중변화 및 지질대사에 미치는 영향)

  • Obesity is emerging as one of the major risk factors of death in Korea as well as western societies. This study was performed to investigate the effect of kimchi with Monascus purpureus (Hongkuk) on liver function, plasma lipid composition of rats fed high fat diet for four weeks in rats. Rats were divided into the following 6 groups; I group (normal diet), II group (normal diet with $1\%$ kimchi powder using $5.0\%$ Hongkuk), III group (high fat diet), IV group (high fat diet with $1\%$ kimchi powder), V group (hish fat diet with $1\%$ kimchi powder using $2.5\%$ Hongkuk), and VI group (high fat diet with $1\%$ kimchi powder using $5\%$ Hongkuk). Weight gains showed to decrease in group IV, V, VI. Food intake and food efficiency ratio were no significantly different among the groups. Plasma total cholesterol showed to decrease with kimchi using Hongkuk. LDL-cholesterol levels were lower in kimchi using Hongkuk than that of control group. These results demonstrated that the kimchi and kimchi using Monascus purpureus(Hongkuk) decreased weight gains and lowered serum cholesterol levels in rat.

Quality characteristics of sausage added red yeast rice (홍국쌀 첨가 소시지의 품질 특성)

  • This study was carried out to investigate the effects and characteristics of sausages with red yeast rice added, particularly the Monascus purpureus KFRI 1134 strains, and to decide the optimal quantity of red yeast rice used. The additional rate of red yeast rice which has been used as a material for reducing nitrite, a coupler in meat products were 0, 1, 3, and 5% (w/w). In terms of the effects of the red yeast rice on the color, texture properties, and sensory characteristics of the sausages, the optimal temperature at which color stability was shown was below $100^{\circ}C$, and the addition of 1% red yeast rice resulted in a distinctly attractive color and increasingly improved the red color in the UV-visual Spectrophotometer analysis. However, the addition of over 1% (w/w) resulted in an unattractive color. The red colors also had strong heat stability. The texture properties and taste of the sausages were not affected by the red yeast rice.

Content of Rutin and Monacolin K in the Red Buckwheat Fermented with Monascus ruber (홍국균 발효 메밀에서의 rutin과 monacolin K의 함량 변화)

  • Buckwheat contains dietary rutin, a flavonol glycoside, which is able to antagonize hypertension in human and has antioxidant activity. Monacolin K, a secondary metabolite produced by fungi Monascus species, is an effective specific inhibitor of cholesterol biosynthesis. To develop functional health food, dehulled buckwheat was fermented with Manascus ruber to produce Anka, and contents of rutin and monacolin K and Anka were determined. The rutin content in dehulled buckwheat decreased slightly after germination, but increased with the growth of sprout. Growth of fungi had no influence on rutin content in dehulled buckwheat Anka, Monacolin K content in fermented dehulled buckwheat malt was lower than that in dehulled buckwheat Anka. Monacolin K content in dehulled buckwheat Anka decreased with increasing length of sprout in fermented dehulled buckwheat malt, probably due to consumption of nutrients, such as polysaccharides as carbon sources, during sprouting for growth of M. ruber.

Effect of Monascus purpureus-Fermented Korean Red Ginseng Powder on the Serum Lipid Levels and Antioxidative Activity in Rats (홍국발효홍삼 분말 급여에 의한 흰쥐의 혈청 지질 농도 및 항산화 활성에 미치는 영향)

  • Monascus-fermented Korean red ginseng (MFRG), fermented by Monascus purpureus KCCM 12002, may be an ideal candidate for the hepatoprotectic, hypolipidemic and antioxidative activities. Effect of MFRG powder on these parameters in rats was investigated. Body weight gain, food intake, and water consumption were not significantly different among the groups. Total and relative weights of liver were significantly higher in MFRG group than that in other groups. The activities of AST and $\gamma$-GTP were highly lowered in MFRG group compared to control group. Contents of serum total lipid and triglyceride were significantly lowered in silymarin group and were significantly increased in MFRG group compared to control group, but tended to be lowered in RG group. Serum content of total cholesterol tended to be lowered in silymarin, RG, and MFRG groups compared to control group. HDL-cholesterol contents was only significantly increased in MFRG group compared to control group. At the same time, atherogenic index (AI) was also significantly lowered in silymarin, RG and MFRG groups compared to control group, and this effect was more pronounced in MFRG group. Content of thiobarbituric acid reactive substances (TBARS) in the liver was significantly lowered in MFRG group and tended to lowered in silymarin and RG groups compared to control group. The hepatic glutathione concentration was significantly higher in silymarin and MFRG groups. Hepatic morphology in all experimental groups revealed clear-cut hepatic lobules with the uniform pattern of the polyhedral hepatocytes radiating towards the periphery from the central vein. These results suggested that MFRG may have anti-atherogenic index (AI) and antioxidative activity in normal dietary feeding rats.

A Study on Quality Characteristics of Sourdough Breads with Addition of Red Yeast Rice (홍국(Red Yeast Rice)을 이용한 Sourdough Bread의 품질특성 연구)

  • This study was carried out to investigate the effects of red yeast rice addition on the physical characteristics of sourdough starters ($SD1{\sim}4$) and the quality characteristics of sourdough bread (CSB, $SDB1{\sim}3$). Moisture contents of red yeast rice, wheat flour, and rye flour were 6.15, 12.53, and 8.56%; ash contents were 0.15, 0.44, and 1.64%; protein contents were 7.30, 12.57, and 11.18%; crude lipid contents were 0.97, 1.16, and 2.49%, respectively. The pH decreased with increasing red yeast rice addition. Reducing sugar and total sugar increased with increasing red yeast rice addition. Lactic acid bacteria were not detected from 0 day for all samples and was maximum on the 2nd day. The yeast counts of SD1 were not detected from $0{\sim}4th$ day, but $SD2{\sim}4$ increased until the 3rd day. Peak time, peak value, and width of tail of CSB were higher than $SDB1{\sim}3$, and they increased with increasing red yeast rice contents of sourdough. The fermentation expansive power of $SDB1{\sim}3$ was higher than that of CSB. Baking loss and specific volume of SDB1 were higher than other samples and when baking loss of CSB and SDB1${\sim}$3 increased, the specific volume increased. L values decreased with increasing red yeast rice contents of sourdough bread whereas, a and b values increased. Springiness and brittleness of CSB and gumminess of SCB1 were lower than other samples. Springiness, brittleness, and gumminess increased with increasing red yeast rice content of sourdough bread. The sensory evaluation indicated that color, hardness, springiness, sweetness, and sourness increased with increasing red yeast rice content of sourdough bread. Aircell non-uniformity of SDB1 was lower than other samples, whereas off-flavor was higher than other samples.

Study on Monascus Strains and Characteristic for Manufacturing Red Yeast Rice with High Production of Monacolin K (Monacolin K 강화 홍국쌀 생산을 위한 균주 및 특성 연구)

  • Red yeast rice (RYR) is the product of fermented yeast by Monascus strains on rice, and has recently become a popular dietary supplement as a traditional food in Asia. RYR contains monacolin K substances known to inhibit cholesterol synthesis as efficiently as statin drugs. To determine the optimal rice cultivar for manufacturing RYR, 7 rice cultivars (Goami, Goami2, Sangjuchalbyeo, Seolgaeng, Saegyejinmi, Yeonghojinmi and Chilbo) were fermented using two Monascus strains (M. ruber KCTC6122 and KCCM60141 of M. ruber) in this study. The monacolin K content of Sangjuchalbyeo were 47.24 ppm on KCTC6122 cultures and 117.03 ppm on KCCM60141 cultures, respectively. Other cultivars, especially Goami and Goami2, which had less content of monacolin K could not seem to ferment normally because those didn't show red color. These results imply that Sangjuchalbyeo can be optimal rice cultivar as a commercial RYR which is well fermented rice and has high content of monacolin K.

Antibacterial and Anticancer Effects of Kimchi Extracts Prepared with Monascus purpureus Koji Paste (홍국첨가 김치추출물의 항균활성 및 암세포 증식억제 활성)

  • Kimchi was prepared with 2.5% and 5% Monascus purpureus Koji(MPK) paste(20%), fermented at $10^{\circ}C$ for 15 days, and sampled at 3-day intervals during storage. Samples were extracted using 80% ethanol. Ethanol extracts of kimchi prepared with MPK paste (MPK kimchi extracts) and control kimchi extracts at 1mg/mL showed 40% decrease in proliferation of cancer cells, such as AGS, KATOIII, HepG2, and Hela. MPK kimchi extracts showed highest cytotoxic effect against cancer cells compared with control at 2mg/mL. Antibacterial effect of MPK kimchi extracts decreased during fermentation, and was higher than that of control kimchi extracts during fermentation, particularly after six days. The 5% MPK kimchi extracts showed higher antibacterial activity against Escherichia coli O157:H7 and Vibrio parahaemolyticus than other groups. Results indicate kimchi added with M. purpureus Koji paste has stronger antibacterial and anticancer effects in vitro than control kimchi.

The Relations Between Antihypertensive Effect and ${\gamma}-Aminobutyric$ acid, Mycelial Weight and Pigment of Monascus (홍국의 혈압강하효과와 ${\gamma}-aminobutyric$ acid, 균체량 및 색도의 영향)

  • To determine the relations between antihypertensive effect and ${\gamma}-aminobutyric$ acid, mycelial weight and pigment of Monascus, ethanol koji extracts were prepared from Monascus koji and each of three grade was classified based on ${\gamma}-aminobutyric$ acid content, glucosamine content and hue angle value, respectively. Each extract was orally administrated on male spontaneously hypertensive rats and its antihypertensive effect was compared. Most of koji extracts showed antihypertensive activity regardless of their ${\gamma}-aminobutyric$ acid content, glucosamine content or hue angle value. Therefore, hypotensive activity of koji extract was not dependent on above three components.

Physiological characteristics of Extracts from Monascus-Fermented Chinese Yam by Monascus sp. strain (Monascus 속 균주를 이용한 발효 홍국마 추출물의 생리학적 특성)

  • This study was conducted to investigate antioxidative and physiological activities of ethanol extracts concentration from Monascus-Fermented Chinese Yam (MFCY). The ethanol extracts from MFCY were measured to examine pigments, DPPH radical scavenging activity, reducing power and monacolin K contents. As a results show that Monascus sp. MK805, with Dioscorea japonica as the substrate can produce pigments (yellow, orange and red), DPPH radical scavenging activity, reducing power and monacolin K content at 29.6 (yellow), 15.1 (orange), 20.4 (red), 72.8%, 1.74(OD at 700nm), 480.6 mg/kg in EtOH extract, respectively. Therefore, fermented chinese yam (Dioscorea japonica) was estimated to be effective biological activity material.

Screening and Identification of Monascus Strain Producing Monacolin K (Monacolin K 대량생산 Monascus 균주의 탐색 및 동정)

  • We had screened the Monascus strain capable of producing monacolin K dominantly among 29 Monascus strains. Red yeast rice was prepared by culturing each Monascus sp. with 200 g of steamed rice (12$0^{\circ}C$, 20 min) at 3$0^{\circ}C$ for 10 days and drying at 8$0^{\circ}C$ for 20 min. As a result, red yeast rice cultured by M. purpureus ATCC16457, M. purpureu IFO 32316, M. purpureus IFO 32228, M. kaoliang ATCC 46595 and M. kaoliang ATCC 46596 produced lots of red pigment and monacolin K. An unidentified Monascus sp. showed the highest productivityof red pigment and monacolin K among 29 Monascus strains. Its production of red pigment and monacolin K was 1.3∼39 times and 2.4∼8 times higher than other strains, respectively. Although the morphological characteristics of unidentified Monascus strain were a little different from the typical M. purpureus, it was identified as M. purpureus CBS 281.34 from the result of sequencing of ITS (Internal transcribed spacer) and 28S ribosomal RNA (partial).
