Production of Haploid and Doubled Haploid Plants from Isolated Microspore Culture of Hot Pepper (Capsicum annuum L.) (고추 소포자를 이용한 반수체 및 배가반수체 생산)

  • Haploid/double haploid plants developed from isolated microspores can significantly accelerate plant breeding. Haploid plants can naturally double their chromosomes to create a pure homozygous line of diploid plants. We present a method for producing embryos from isolated microspores of hot peppers (Capsicum annuumL.). We analyzed the polyploidization levels of the regenerated plants. The donor plants produced the optimal stage of microspores following short-term growth under low-intensity light, which resulted in high rates of embryogenesis and cotyledonary embryogenesis. To find an efficient culture method, liquid, doubled-layer, and 2-step cultures were tested. Liquid culture yielded the highest number of embryos, whereas the highest efficiency for cotyledonary embryogenesis was afforded by the doubled-layer culture. When normal cotyledonary embryos were transplanted onto a regeneration medium, they developed into complete plants. From these, 208 plants were tested via flow cytometric analysis, and 35.6% and 72.7% of the chromosomes from the Milyang-jare and LV2319 genotypes, respectively, were found to be spontaneous double haploids. These results are the same as those obtained on analyzing horticultural characteristics, including the size of leaves and the size and shape of fruits. The present study provides information on the practical application of isolated microspore culture of hot peppers, factors that affect embryogenesis, and methods for polyploidy testing.

  • Land shrimp farms can control the growth environment more stably than coastal ones, making them advantageous for high-quality, large-scale production. In order to maintain an optimal shrimp growth environment, various factors such as water circulation, maintaining appropriate water temperature, oxygen supply, and feed supply must be managed. In particular, failure to properly manage water quality can lead to the death of shrimp, making it difficult to have people stationed at the farm 24 hours a day to continuously manage them. In this paper, to solve this problem, we design an integrated monitoring system for land farms that can be operated with minimal manpower. The proposed design plan uses IoT technology to collect real-time images of land farms, pump status, water quality data, and energy usage and transmit them to the server. Through web interfaces and smartphone apps, administrators can check the status of the farm stored on the server anytime, anywhere in real time and take necessary measures. Therefore, it is possible to significantly reduce field work hours without the need for managers to reside in the farm.

A Study on Edutech Types and Utilization of School Libraries in a Non-face-to-face Educational Environment (비대면 교육 환경에서 학교도서관의 에듀테크 유형과 활용에 관한 연구)

  • This study aims to retrospectively review how school libraries utilized digital educational tools in a non-face-to-face environment during the pandemic, and to suggest types of digital educational tools suitable for use in the New Normal. Initially, a list of edutech used in schools during the pandemic was compiled by reviewing previous studies. By grasping the frequency of use of edutech expert teachers and librarian teachers, the list of edutech was readjusted to the type of edutech that can be used in school libraries. The use of edutech by school libraries during the pandemic, as reported in the 'School Library Journal,' was analyzed and integrated the previous list of edutech. As a result, edutech suitable for school libraries was categorized into five main areas: management, communication, productivity, consumption, and evaluation. Additionally, specific edutch examples for each category were presented, based on insights from the 'School Library Journal.'

Field research of cultivation technique for stable production of common mushroom(Agaricus bisporus) (양송이 안정생산을 위한 생산기술 현장연구)

  • Common mushroom production per area has been decreased and are up to less than 50% of the 1980 production. To determine the main reasons for the decrement, we performed this study. Two main reasons, which are mushroom disease and the low compost quality because of mechanized compost making, were assessed. In mechanized mushroom farms, nitrogen concentration in compost was lower than recommended and total compost quantity was about 100-150 $kg/3.3m^2$, which was also lower than usual. Our study revealed that higher nitrogen concentration (about 1.5%) in compost gave better production. Also, use of large amount of compost appeared to increase the mushroom production, although more insects and disease problems were observed. The relationships between the presences of microorganisms and occurrence of diseases were assessed by monitoring the microorganism densities near the mushroom farms. Higher number of microorganisms were observed near the mushroom farm area, compared to control region, Daechon beach. Most contaminating molds were found in the circulating fans, tunnel and culture room floor. The bacterial isolates were collected from the air in mushroom culture room and killed with 0.005% Benzalkonium solution, indicating treatment of Benzalkonium are the effective methods to sanitize the mushroom culture room.

Characteristics of Places to Visit and Hanbok-Trip Class as a Landscape Prosumer - Focused on Gyeongbokgung Palace - (경관 프로슈머로서 한복나들이 향유계층과 방문 장소 특성 연구 - 경복궁을 대상으로 -)

  • This study identifies factors of Hanbok-trippers - a term for people who dress in Hanbok(Korean traditional costume) while going on a trip - who converge on Gyeongbokgung Palace by determining the characteristics of class, places to visit and preferred places. This study interprets the voluntary hobby activities of Hanbok-trippers from a viewpoint of a landscape prosumer and the meaning of the urban landscape. As a result of in-depth interviews, on-site survey, and observation surveys focused on Hanbok-trippers, there were various levels of participants. They are classified into three groups - leading group, entry group, temporary-experience group - according to their cognitions, types of Hanbok use, activities, etc. The leading group and entry group are a voluntary hobbyist class due to the ongoing tendencies of their participation. There are differences in the purpose and factors of visiting Gyeongbokgung Palace as a place for a Hanbok-trip. The leading group visited Gyeongbokgung Palace for cultural activities, regular get-together, public relations, and as a gathering place to go neighboring destinations. In this case, the main factors of the visit are the traditional landscape, convenient transportation, chances for traditional culture exhibitions and events in Gyeongbokgung Palace and its neighborhood. The entry group visits Gyeongbokgung Palace because of its traditional landscape and cultural activities nearby. The traditional landscape and many Hanbok-trippers are main factors of visiting Gyeongbokgung Palace for the Temporary-experience group. This study found that Gyeongbokgung Palace has a new sense of place of 'Introductory course of Hanbok-trip', 'Hanbok Playground' because temporary-experience group visits there to experience a Hanbok-trip for the first time. Hanbok-trippers consume places and landscape in actual places offline, producing a new landscape at the same time, and has the characteristics of a 'landscape prosumer' by producing landscape images online through their own personal or social media. Their colorful and voluntary movements contribute to the dynamism of the urban landscape and can become a new cultural asset for the city. The voluntary hobbyist class can be considered a new type of participants in bottom-up planning such as urban regeneration and place marketing. This study has significance in that it conceptualized the 'landscape prosumer' through the voluntary hobbyist class of Hanbok-trippers with the concept of the 'prosumer' that has been studied only in the consumer studies and marketing fields, and has identified the significance of the urban landscape.

Grazing Rate and Pseudofaeces Production of Native Snail Cipangopaludina chinensis malleata Reeve on Toxic Cyanobacterium Microcystis aeruginosa (한국산 논우렁이의 유해조류 섭식율 및 배설물 생산)

  • Grazing rates (GR) and pseudofaeces production (PFP) of native snail, Chinese mystery snail (Cipangopaludina chinensis malleata Reeve) on natural colonial morphs of Microcystis aeruginosa was measured. C. chinensis was collected from the upstream of the Geum River (Boryeong, Korea), where they co-habituated with Unio douglasiae and Lanceolaria acrorhyncha. The experiments were performed to evaluate the GR and PFP at different conditions such as; incubation time (1, 3, 5, 7, 9 and 11 hr), body size (3 to 6.1 cm, n=28), snail density (0.5, 1, 1.5 and 2.0 ind. $L^{-1}$) and prey concentration (168.3, 336.7, 505.0 and $673.0{\mu}g\;Chl-{\alpha}L^{-1}$). All experiments were triplicated, and conducted in transparent acrylic vessel (3L in volume). Regarding feeding time, a highest GR (0.538L $gAFDW^{-1}h^{-1}$) and PFP $(7.18mgAFDW^{-1})$ appeared at 1hr and 7hr after snail stocking, respectively. Interestingly, the snail, smaller than 4.5cm in body size, showed a wide range of GR ($-4.173{\sim}1.087L\;gAFDW^{-1}h^{-1}$) for the initial period (1 and 4hrs of stocking), compared to those greater than 4.5cm, which showed a stable FR, higher than 0.5L $gAFDW^{-1}h^{-1}$. Upon density effect, the density of 1.5 ind. $L^{-1}$ induced the most effective inhibition on Microcystis biomass with highest PFP. On the prey concentration, highest GR (0.897L $gAFDW^{-1}h^{-1}$) and PFP (3.67 mg $gAFDW^{-1}h^{-1}$) were induced at the level of $168.3{\mu}g\;Chl-{\alpha}L^{-1}$ and $673{\mu}g\;Chl-{\alpha}L^{-1}$, respectively. GR and PFP of this freshwater snail on the cyanobacterial bloom (M. aeruginosa) varied with the feeding conditions, and they were comparatively high for a short period of time less than 7hrs regardless of the stocking condition. Our results suggest that this freshwater snail has a potential to control cyanobacterial bloom when provided with suitable condition.

Studies on the Main Level-Grading Factors for Establishment of LFQC (Liquid Fertilizer Quality Certification) System of Livestock Manure in Korea (가축분뇨 액비품질인증제도 구축을 위한 목표요소에 관한 연구)

  • Establishment of the LFQC (Liquid Fertilizer Quality Certification) system is very urgent issue for recycling livestock manure as renewable resources in Korea faced with environmental problem of manure application to land due to intensive livestock farming. In this study, we investigated relevant laws and regulations on livestock manure fertilizer, certifications of eco-friendly agricultural products, government policies on livestock manure management to establish reasonable direction of Korean LFQC (Liquid Fertilizer Quality Certification) system. As a result from this study, the liquid fertilizers in 'LFQC' system could be classified as three levels according to the usage patterns in field; 1st. Individual Farm Level (IFL), 2nd. Joint Farm Level (JFL), and 3rd. Commercial Level (CML). And finally, we found some characteristics in 'Main Level-Grading Factors' of liquid fertilizer such as fertilizing value, harmfulness, stability, uniformity, economic effect, storage potential, commercial value, functionality. Those items were considered to be the key factors for the establishment of 'LFQC' system. More research on 'Evaluation Standards' for concrete guideline and on the 'Main Level-Grading Factors' be needed to complete Korean LFQC system.

Development of Smart Digital Agriculture Technology for Food Crop Production in Korea-The Path Forward Based on Expert Feedback (식량작물 생산에 대한 스마트디지털 농업기술의 발전 방향 - 전문가 설문조사 연구)

  • Building self-sustainable rural infrastructure and environment through smart digital agriculture technology innovation is one of the major goals of the Korean agricultural administration as a part of the nation's 4th industry revolution. To identify areas for improving and effectively investing in the acceleration of rural development, 207 experts in the areas of crop science and smart digital agriculture technology were interviewed for their opinions and suggestions on 22 questions designed to recognize fundamental agricultural issues to be addressed and solutions to advance technology innovation and rural development. Majority of the participants expected smart digital agriculture technologies to resolve major agricultural issues and help build a better rural environment. To overcome technology gaps and resolve issues more effectively, further investment in training new technology experts and building stronger agricultural technology infrastructure is urgent, and persistent and systematic support from agricultural administration appears to be the key for accelerating the process. While the leading global groups of both public and private sectors have advanced their technologies beyond the field application stage, most of the Korean technologies remain at the early pilot stage. Aging population and lack of labor in rural areas, unknown future climate change, and challenges in sustainable rural development are expected to be resolved by smart digital agriculture technologies. Technological innovations by research institutes should be promptly deployed in the crop production field, and farm training systemically organized by local technology centers can accelerate farming revolution. Standardization of equipment and data systems is another key to the success of digitalization of food crop production and food supply chains nationwide.

Detecting Phenology Using MODIS Vegetation Indices and Forest Type Map in South Korea (MODIS 식생지수와 임상도를 활용한 산림 식물계절 분석)

  • Despite the continuous development of phenology detection studies using satellite imagery, verification through comparison with the field observed data is insufficient. Especially, in the case of Korean forests patching in various forms, it is difficult to estimate the start of season (SOS) by using only satellite images due to resolution difference. To improve the accuracy of vegetation phenology estimation, this study reconstructed the large scaled forest type map (1:5,000) with MODIS pixel resolution and produced time series vegetation phenology curves from Normalized Difference Vegetation Index (NDVI) and Enhanced Vegetation Index (EVI) derived from MODIS images. Based on the field observed data, extraction methods for the vegetation indices and SOS for Korean forests were compared and evaluated. We also analyzed the correlation between the composition ratio of forest types in each pixel and phenology extraction from the vegetation indices. When we compared NDVI and EVI with the field observed SOS data from the Korea National Arboretum, EVI was more accurate for Korean forests, and the first derivative was most suitable for extracting SOS in the phenology curve from the vegetation index. When the eight pixels neighboring the pixels of 7 broadleaved trees with field SOS data (center pixel) were compared to field SOS, the forest types of the best pixels with the highest correlation with the field data were deciduous forest by 67.9%, coniferous forest by 14.3%, and mixed forest by 7.7%, and the mean coefficient of determination ($R^2$) was 0.64. The average national SOS extracted from MODIS EVI were DOY 112.9 in 2014 at the earliest and DOY 129.1 in 2010 at the latest, which is about 0.16 days faster since 2003. In future research, it is necessary to expand the analysis of deciduous and mixed forests' SOS into the extraction of coniferous forest's SOS in order to understand the various climate and geomorphic factors. As such, comprehensive study should be carried out considering the diversity of forest ecosystems in Korea.

Standardization of Identification-number for Processed Food in Food-traceability-system (가공식품에 대한 이력추적관리번호 부여체계의 표준화 방안)

  • Facing a number of global food-related accidents, the concept and system for food traceability have been designed and introduced in many countries to manage the food-safety risks. To connect and harmonize the various food traceability-information in food traceability system according to the food supply chain, the coding system of identification-number for food-traceability has to be standardized. The GTIN (Global Trade Item Number) barcode system which has been globally standardized and implemented, is reviewed with the mandatory food-labeling regulation in expiration date of processed foods. The integration of GTIN-13 bar-code system for food-traceability is a crucial factor to expand its function in the food-related industrial areas. In this literature, the standard coding system of identification-number for food-traceability is proposed with 20 digit coding number which is combined with GTIN-13 bar-code (13 digit), expiration date (6 digit), and additional classification code (1 digit). This proposed standard coding system for identification-number has a several advantages in application for prohibiting the sale of hazard goods, food-recall, and inquiring food traceability-information. And also, this proposed coding system could enhance the food traceability system by communicating and harmonizing the information with the national network such as UNI-PASS and electronic Tax-invoice system. For the global application, the identification-number for food-traceability needs to be cooperated with the upcoming global standards such as GTIN-128 bar-code and GS1 DataBar.
