Comparative karyological analysis of the Korean tree frogs, Hyla japonica and Hyla suweonensis (Anura, Hylidac) (C-banding 방법에 의한 한국산 청개구리 두 종(Hyla japonica와 Hyla suweonensis)의 핵형 비교분석)

  • The comparative karyological analysis of the Korean treefrogs, Hyla japonica and Hyla suweonensis, were performed by C-banding method. Heteromorphic sex chromosomes, female heterogamety, has been identified in the 3rd chromosomes of H. suweonensis H. suweonensis seem to have sex chromosomes which are Zz/ZW type. The Z chromosomes contain large amount of constitutive heterochromatin, but little heterochromatin is located in the W chromosomes. This is in contrast to all previously known amphibian and most other vertebrate's W or Y chromosomes, except Gastrotheca oui'ern and G. walken (Schmid et al., 1988).

  • Cytogenetic characteristics of three Hemibarbus species (H. labeo, H. longirostris and H. mylodon) were analyzed based on erythrocyte measurement, flow cytometric estimation of cellular DNA content, and karyological analysis. Average nuclear volumes for H. labeo, H. longirostris and H. mylodon were 22.5, 21.7 and $26.0\;{\mu}m^3$, respectively. The estimated genome sizes of those three species were not significantly different from one another, being recorded as 2.51, 2.33 and 2.35 pg/cell for H. labeo, H. longirostris and H. mylodon, respectively. Modal chromosome numbers of the three species were the same as 2n = 50. However, their karyotypes and fundamental numbers (FN) were different among species; 16M+16SM+18T/A (FN = 82) for H. labeo, 18M+16SM+16T/A (FN = 84) for H. longirostris and 18M+24SM+8T/A (FN = 92) for H. mylodon.

Karyotypes of Pneumocystis carinii from Korean Rats (한국산 횐쥐 카리니주폐포자충의 핵형)

  • Molecular karyotyping was applied to Pneumocystis carinii (Pc) from two strains of experimental rats, Sprague Dawley(SD) and Fisher(F), in Korea. Field inversion gel electrophoresis and contour clamped homogeneous electric field electrophoresis resolved 15 chromosomal bands from the Pc. The size of the bands was estimated 270kb to 684kb from SD rats, and 273kb to 713 kb from F rats. The bands of 283 kb from SD rats and of 273 kb from F rats stained more brightly suggesting duplicated bands. Total number of chromosomes was at least 16, and total genomic size was estimated 7×106 bp. All of the bands from F rats hybridized to the probe of repeated DNA sequences of Pc and the band of 448 kb size was proved to contain rDNA sequences, but Pc. chromosome bands from SD rats showed no reactions to the probes. The 2 different karyotypes of p. carinii from 2 strains of rats were maintained consistently for 2 years.

G-, C-, and NOR-banding of Korean Native Pig Chromosomes (한국재래돼지의 G-, C-, 및 NOR-banding)

  • Using the G-, C-, and NOR-banding techniques, a karyotyping for Korean Native Pig was performed. Blood samples were collected from 50 male Korean Native Pigs that had been bred at the National Livestock Research Institute and then blood cells were prepared from in vitro cultures followed by karyotyping; G-, C-, and NOR-banding patterns of metaphase chromosomes were analyzed. The karyotype of Korean Native Pig is 38, XX or XY which consists of 5 pairs of submetacentric chromosomes(Group I), 2 pairs of acrocentric chromosomes with short p-arm(Group II), 5 pairs of medium metacentric chromosomes(Group III), 6 pairs of acrocentric chromosomes(Group IV) and metacentric X and Y sex chromosomes. On GTG-banding, the Korean Native Pig exhibited a typical and identical banding pattern in each homologous chromosomes. Overall chromosomal morphology and positions of typical landmarks of the Korean Native Pig were virtually identical to those of Committee for the Standardized Karyotype of the Domestic Pig(CSKDP). However, numbers of G-bands of the Korean Native Pig chromosomes were more than those of CSKDP. In chromosomes 1, 3, 5, 6, 7, 8, 13, 14, 15, 16, 17, 18 and X, the Korean Native Pig exhibited more separated bands as compared with CSKDP. In C-banding patterns, although the quantity of heterochromatin was variable in each chromosome, most of the Korean Native Pig chromosomes had heterochromatic C-bands on centromeres. However, the heterochromatic C-band was constantly observed on the whole Y chromosome. In AgNOR staining, the NORs were located at centromeres on the chromosomes 8 and 10. The number of NORs per metaphase ranged from 2 to 4 giving a mean value of 2.13. The number of NORs were distributed on all chromosome pair 10 but not on chromosome 8. The sizes of NORs were also differed between homologous chromosomes 8. Numbers of NORs of Korean Native Pig were significantly higher than those of Yorkshire. The pattern of pig NORs was polymorphic in breeds, individuals and cells, especially on chromosome 8.

  • Samples of striped field mice, Apodemus agrarius Pallas, from eight localities in s southwestern islands and coasts in Korea were collected and analyzed together with samples from the mainland and Jeju island. In chromosomal analyses no variation was revealed in karyotypes among samples from Korea. In morphometric analyses samples from Wan and Bogil islands formed a large-size g group (A. agrarius chejuensis) with those from Jeju island, whereas samples from Jin and H Hajo islands formed a small-size group (A. agrarius coreae) with those from the mainland including southwestern coasts It is supposed that small-size population of striped field mice in Wan and Bogil islands w were extinct and replaced by large-szie population of striped field mice immigrated from Jeju island by humans.

  • The task for chromosome analysis and diagnosis by experienced cytogenetists are being concerned as repetitive, time consuming job and expensive. For that reason, intelligent agent based on chromosome knowledge base using web has been developed to be able to analyze chromosomes and obtain necessary advises from the knowledge base instead of human experts. That is to say, the knowledge base of IF THEN production rule was implemented to a knowledge domain with normal and abnormal chromosomes, and then the inference results by the knowledge base could enter the inference data into the database. Experimental data were composed of normal chromosomes of 2,736 cases and abnormal chromosomes of 259 cases that have been obtained from GTG-banding metaphase peripheral blood and amniotic fluid samples. The completed intelligent agent for the chromosome knowledge base provides variously morphological information by analysis of normal or abnormal chromosomes also has the advantage of being able to consult with the user on the chromosome analysis and diagnosis.

  • Samples of Siberian chipmunks (Tamias sibiricus borberi ) from Korea were analyzed by multivariate, air-drying , and blot-hybridization methods in order to determine the degree of variations of morphological characters, chromosomal karyotype, and mtDNA restriction fragment patterns, respectively, Moreover, morphometric characters of samples of Siberian chipmunks from Manchuria(T.sibiricus orientalis) were also analyzed in order to clarify the taxonomic status of the subspecies barberi. Siberian chipmunks from six localities in Korea were in morphometric characters more or less similar with one another and Siberian chipmunks from Manchuria were not so distinct enough to from a subgroup. Siberian chipmunks from Four localities in Korea were identical in their karyotypes (2n=38) : samples from three localities in Korea were nore or less similar in their mtDNA fragment patterns. Siberian chipmunks from Korea appeared to be a omogeneous population and as noted by Corbet (1978) subspecies barberi is the synonym of subspecies orientalis. The scientific name of Siberian chipmunks from Korea and Manchuria is T. sibiricus orientalis, although systematic studies with the samples from North korea, Manchuria and China will be necessary for their taxonomic reconsideration.

  • 한국산 가시군부속 (Acanthochitona)의 좀털군부(A. achates),참털군부 (A. circellata), 털군부(A.defilippii)및 애기털군부(A. rubrolineata)등 4종의 정소를 재료로 warm drying mithod를 통해 염색체를 관찰하였다. 염색체의 수는 4종 모두 2n=16, n=8로 확인되었다. 핵형분석결과 형태적으로 좀털군부는 다섯 쌍의 중부염색체, 한쌍의 차중부염색체, 두 쌍의 차단부염색체로;참털군부는 한쌍의 중부염색체, 네 쌍의 차중부염색체, 두 쌍의 차단부염색체, 한 쌍의 말단부염색체로; 털군부는 세 쌍의 중부염색체, 세 쌍의 중부염색체, 한 쌍의 차중부염색체, 한 쌍의 차중부 또는 차단부염색체로 구성되어 있었다. 외부 형태적으로 매우 유사한 애기털군부와 좀털군부의 경우, 특히 4번 염색체에서 현저한 형태적 차이를 보였다. 8번 염색체의 경우, 애기털군부와 좀털군부는 중부염색체인 반면 참털군부와 털군부는 말단부염색체로 나타났다. 동북아시아와 유럽에 분포하는 가시군부류 사이에 나타나는 염색체의 속내 종간 숫적변이를 보면 동북아시아산(2n=16)이 유럽산(2n=16)이 유럽산(2n=24, 18)보다 분화된 것으로 보인다.

  • Isozymes and chromosomes of Bufo bufo and B. kangii were studied by starch gel electrohporesis and bone marrow air-drying method. Twently-three enzymes and nonenzymatic proteins in Bufo species collected in Korea provided a basis for the estimating the proportion of polymorphic loci in the genum. Of 23 loci controlling the proteins examined, 26% and 17$% were polymorphic in B. bufo and B. kangii, respectively. In B. bufo, 4 loci had 3 alleles and no loci had more than 2 alleles in B. kangii. Both species had same karyotypes with 22 chromosomes except one pair of chromosome which had sat-chromosomes in B. bufo karyotypes. The genetic similarity was approximately 0.5.

  • New somatic chromosome numbers and karyotype analyses of 33 medicinal herbs (30 genera, 23 families) in Korea were investigated. The chromosome numbers of 4 taxa, Euryale ferox, Rodgersia podophylla, Cirsium japonicum var. ussurience, Eehinops setifer, showed results that are different from previous reports. Among 33 taxa, 23 taxa were reported for the first time, and karyotype analyses were newly conducted for 2 taxa (Tiarella polyphylla, Crepidiastrum denticulatum) in Korea. In addition, we observed for the first time the new chromosome numbers for 4 taxa distributed evenly over the world (Lindera erythrocarpa, Corylopsis glabrescens var. gotoana, Ardisia crenata, Callicarpa japonica var. luxurians).
