The Study for Practical use of Bioremediation Agent in Oil-Contaminated Area (해상유출유 오염지역에서의 미생물처리제 활용 방안 연구)

  • Recently more than 450 incidents of oil spill a year have occurred in nearshore of Korea, which caused unmeasurelable losses in fisheries and severe damage in marine ecosystem. Two approaches remain paramount in any response to marine oil spill : the enhancement of natural dispersion of the oil by using dispersants, and mechanical recovery using booms and skimmers. A technique currently receiving fresh attention is the enhancement of the natural bioremediation of oil through the application of micro-organisms and/or nutrient. Oil, like many natural substances, will biodegrade over a period of time into simple compounds such as carbon dioxide, water and biomass. Bioremediation is the term used to describe a range of processes which can be used to accelerate natural biodegradation. More specifically biostimulation is the application of nutrients, and bioaugremetation or seeding is the addition of microbes specially selected to degrade oil. Bioremediation is an economically attractive method for the clean-up of oil-contaminated area. Bioremediation has been demonstrated to be an effective oil spill countermeasure for use in cobble, sand beach, salt marsh, and mud flat environment.

Radiocarbon for Studies of Organic Matter Cycling in the Ocean (방사성탄소를 이용한 해양 유기탄소 순환 연구 동향)

  • Radiocarbon is a powerful tool for studies of carbon cycling in the ocean. Development of measurement technology of accelerator mass spectrometry has enabled researchers to measure radiocarbon even in specific compounds. In this paper, a brief introduction on radiocarbon measurement and reporting of radiocarbon data is provided. Researches that used radiocarbon measurements on bulk organic matter, organic compound classes, and specific organic compounds are reviewed. Examples include works to understand the cycling of particulate and dissolved organic matter, biochemical composition of particulate organic matter, post-depositional transport of sedimentary organic matter, selective incorporation of fresh organic matter by benthic organisms, chemoautotrophy by archaea, and sources of halogenated chemical compounds found in marine mammals.

Microbiological Contamination in Domestic and Imported Squid Todarodes pacificus Jeotgal Distributed at On-line Marketplaces: An Investigation (온라인에서 유통 중인 국내산 및 수입산 오징어(Todarodes pacificus) 젓갈의 미생물학적 오염도 조사)

  • Squid jeotgal is a very popular traditional, salted, and fermented seafood in Korea. Due to the development of a distribution structure, a lot of imported squid jeotgal has recently entered Korea. In this study, we analyzed and compared the microbial contamination levels of domestic and imported squid jeotgal. The total viable bacterial count in domestic and imported jeotgal was 5.20 and 5.38 log CFU/g, respectively. The highest contamination level was 6.94 log CFU/g in one of the imported squid jeotgal samples. Coliforms were not detected in domestic jeotgal but were detected at 50% with 2.21 log CFU/g in imported jeotgal. Escherichia coli and Staphylococcus aureus were not detected (ND: <1 log CFU/g) in both domestic and imported jeotgal. All samples of jeotgal were negative for Bacillus cereus and Clostridium perfringens. The microbial contamination levels determined in the current study may be utilized as primary data to execute microbial risk assessments on domestic and (or) imported jeotgal.

A Study on the Development of Marine Detector Using Nano-technology (나노기술과 해양용 센서 개발에 관한 연구)

  • It is generally recognized that monitering of bio-molecules, which are related to the ocean environment, becomes more important. So far, for the detection of the bio-molecules, ocean samples were brought to laboratory to be analyzed using a complicate and expensive measuring system The "ship and dip" method takes a relatively long time to complete a analysis cycle and causes significant errors due to the time difference between the analysis processes. In order to overcome the drawbacks, developments of sensors for the detection of bio-molecules were suggested using nano-technology, such as nano-spintronic device, carbon nano tube device, and nano-semiconductors. The pros and cons of the technology were examined and reinvestigated to overcome the technical problems in the application to real sensors.

Genetic Composition Analysis of Marine-Origin Euryarchaeota by using a COG Algorithm (COG 알고리즘을 통한 해양성 Euryarchaeota의 유전적 조성 분석)

  • To figure out the conserved genes and newly added genes at each phylogenetic level of Archaea, COG (clusters of orthologous groups of proteins) algorithm was applied. The number of conserved genes within 9 species of Archaea was 340 and that of 8 species of Euryarchaeota was 388. Many of conserved 265 COGs, which are specific to Archaea and absent in Bacteria and S. cerevisiae, were concerned with 'information storage and processing' (94 COG, 35.5%) and 'metabolism' (82 COG, 30.9%). COGs related to these functions were assumed as highly conserved and permit peculiar life form to Archaea. It seemed that there was some difference in 'nucleotide transport and metabolism' and there was little difference in 'information storage and processing' between Euryarchaeota and Crenarchaeota. Marine-origin Euryarchaeota showed different conserved COGs with terrestrial Euryarchaeota. Conserved COGs, related to carbohydrate transport and metabolism and others, were different between marine- and terrestrial-origin Euryarchaeota. Hence it was assumed that their physiology might be different. This study may help to understand the origin and conserved genes at each phylogenetic level of marine-origin Euryarchaeota and may help in the mining of useful genes in marine Archaea as Manco et al. (Arch. Biochem. Biophy. 373, 182 (2000)).

Anaerobic dechlorinating enrichment culture on tetrachloroethene (PCE) (PCE 탈염소화를 위한 혐기성배양)

  • Starting at the beginning q the 20th century, increasing amounts of tetrach1oroethene (PCE) and trichloroethene (TCE)were manufactured due to the extensive use of these compounds in industry, in the military, and in private households, mainly as nonflammable solvents. This widespread use, along with careless handling and storage, are among the most serious contaminants of soil, sediment and groundwater. Highly chlorinated ethenes are typically not degraded through oxygenation by aerobic bacteria Since complete reductive dechlorination of PCE and TCE to ethene (ETH) has been observed in anaerobic enrichment culture, anaerobic dehalorespiring bacteria have received increased attention in the last decade. Under anaerobic conditions, these compounds con be reductively dehalogenated to less-chlorinated ethenes or innocuous ethene by microorganism through dehalorespiration. We have been studying anaerobic enrichment culture which used lactate as the electron donor for reductive dechlorination of PCE to ETH the anaerobic mixed microbial culture was enriched from the sediment sample taken from site contaminated with PCE. PCE was consistently and completely converted to ethene. In addition, the accumulation of intermediate products such as 1,2-ds-dichloroethene (cis-DCE) and vinyl chloride (VC) was observed in the anaerobic mixed microbial culture. the established dechlorinating enrichment culture was analyzed by DGGE using primers specific to DefrJ1ococcoides 16S rRNA gene sequences. In conclusion, we established the PCE dechlorinating enrichment culture and confirmed the existence of Dehalococcoides in an enrichment culture.

Risk Analysis and Safety Assessment of Microbiological and Chemical Hazards in Katsuobushi Products Distributed in the Market (시중에서 유통되는 가쓰오부시의 미생물학적·화학적 위해요소분석 및 안전성 평가)

  • For the safety assessment of microbiological and chemical hazards in katsuobushi, fifteen samples of katsuobushi were purchased from supermarkets. The contamination levels of total viable bacteria, coliforms, Escherichia coli, and nine pathogenic bacteria [Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Bacillus cereus, Vibrio parahaemolyticus, Clostridium perfringens, Enterohemorrhagic E. coli (EHEC), Yersinia enterocolitica and Campylobacter jejuni/coli] were quantitatively or qualitatively assessed. Additionally, the heavy metals (total and methyl mercury) content, radioactivity (131 I, 134 Cs+ and 137 Cs) were quantitatively assessed. Microbial and chemical analyses were performed using standard methods in Korean food code. The contamination level of total viable bacteria was 2.70 (1.18-4.42) log CFU/g. Coliforms, E. coli and S. aureus were not detected in any samples. Other eight pathogenic bacteria were negative in all samples. The contamination levels of total and methyl mercury were 0.366 (0.227-0.481) and 0.120 (0.002-0.241) mg/kg, respectively. In addition, radioactivity was not detected in any samples. The results will be helpful in revitalizing domestic use and boosting exports of katsuobushi because the microbiological and chemical safety of katsuobushi has been assured. Furthermore, the results may be used as a basis for performing chemical and microbial risk assessments of katsuobushi.
