Mucous cell and Epithelial sensory receptor cells in the Sucker of Korean Squid (Sepniella maindroni) (한국산 쇠오징어 (Sepiella muindroni)의 흡반(Sucker)내 점액세포 및 상피성 감각수용세포)

  • 쇠오징어(Sepiella maindroni)흠반의 외측 상피조직에서 3종류의 감각세포(A, B 및 D형 감각세포)와, 1종류의 감각상피세포(C형 감각 상피세포) 그리고 2종류의 점액세포(a 및 b형 점액세포)가 각각 관찰되었다. A 및 B형 감각세포는 분포상태가 다를 뿐 형태는 거의 유사한 세포이고 많은 분비액포를 지닌 해면질 형태의 세포로서 전자 밀도가 낮은 물질을 분비하는 분비성 감각세포로 확인되었다. 0형 세포는 매우 작은 감각세포로서 세포의 자유면에 긴 융모가 밀생되어있었고, 측면원형질막은 불규칙한 수지상 연접을 이루고 있었다. 이 세포는 전자밀도가 매우 높아서 검게 보였다. C형 감각 상피세포도 세포의 자유면에 긴 미세융모가 밀생되어있는 단층 원주상 세포로서. 이 세포 역시 측면원형질막이 불규칙한 수지상 연접을 이루고 있고. 엽접내에는 많은 소포와 약간의 사립제가 관찰되었다. 3형 점액세포는 세포질속에 내강이 팽창된 활면소포체를 소지하고 있으며 전자밀도가 높은 구형의 점액과립을 형성하였고, b형 점액세포도 전자밀도가 중등도인 점액질 과립을 소지하고 있었다.

  • 동물에서 발생되는 질병의 수는 헤아릴 수 없이 많으며, 병인체도 다양하다. 이러한 질병을 효과적으로 관리하기 위하여, 우리나라의 가축전염병예방법(개정법률 제 4,885호; 1995. 1. 5)에는 제1종 전염병 26종과 제 2종 전염병 28종, 총 54종을 법정가축전염병(法定家畜傳染病)으로 지정해 놓고 있다. 최근에 들어, 1996년 3월에 영국에서 소해면형뇌증 (일병 광우병, BSE)이 인체에서의 변형 크로이츠휄트-야곱병 (v-CJD)과 관련하여 세계적인 주목을 받음은 물론 유럽연합 (EU)을 중심으로한 쇠고기의 국제교역에 크게 영향을 미치고 있으며, 지난해(1997) 3월에는 대만에 구제역 (口蹄疫, FAD)이 발생하여, 여러 양돈장에 급속히 퍼져, 대만의 수출주력 산업인 양돈업의 붕괴를 가져왔을 뿐만 아니라 국가적인 경제위기를 초래하였으며, 연간 약 9조원의 경제적인 손실이 예상되고 있고 양돈경기 회복을 위하여는 막대한 자금(약 40조원)과 긴 세월(최소 4-5년)이 필요할것으로 생각되고 있다. 한편, 국제적으로는 아주 전통적인 질병이며 한동안 자취를 감추었던 광견병과 기종저가 개발하여 피해를 입히고 있으며, 아주 최근에 알려진 새로운 원충성 유산증을 일으키는 네오스포라증도 확인되고 있다. 본 편에서는, 최근 국내에서 특히 문제시 되고 있는 풍토병인 기종저(blackleg)에 대하여 그 발생실태와 임상진단, 방역대책 등에 관하여 고찰해 보고자 한다.

  • 임신 10일부터 20일까지 흰쥐 태반형성과정에서 heat shock protein 27(HSP27) 분포를 면역조직화학적으로 조사하였다 임신 10일 태반에 자궁간막 탈락막세포. 영양막세포 및 거대영양막세포가 찰찰되었다. 이후 탈락막세포수는 감소하고 영양막세포는 증가하는데 특히 임신 14일에 현저하며 영양막은 해면영양막과 미로영양막으로 구분되었다 거대영양 막세포수의 변화는 현저하지 않았다. 자궁간막 탈락막에서 HSP27은 임신 10일 및 12일에 탈락막세포의 세포질 또는 핵에 반응이 나타나고 특히 핵에 반응하는 세포수가 많았으며 임신 14일 이후 탈락막 세포수 감소와 함께 반응이 나타나지 않았다 거대영양막세포는 임신 10일부터 일부 세포의 세포질 또는 핵에 반응이 관찰되나 임신 18일부터는 반응이 나타나지 않았다 영양막의 HSP27은 임신 10 및 12일에 일부 세포에서만 세포질 또는 핵에 반응이 관찰되나, 임신 14일부터 반응세포수 증가와 함께 대부분 핵에 반응을 나타내고 반응정도는 임신 16일 및 18일에 가장 현저하였으며, 미로영양막에서 반응세포수가 더 많았다.

Analysis of Seabottom and Habitat Environment Characteristics based on Detailed Bathymetry in the Northern Shore of the East Sea(Gyeongpo Beach, Gangneung) (정밀 해저지형 자료 기반 동해 북부 연안(강릉 경포) 서식지 해저면 환경 특성 연구)

  • In this study, we analyze seabottom conditions and characteristics integrated with topographic data, seafloor mosaic, underwater images and orthophoto(drone) of soft-hard bottom area around the Sib-Ri rock in the northern shore of the East Sea(Gyeongpo Beach, Gangneung). We obtained field survey data around the Sib-Ri rock(about 600 m × 600 m). The Sib-Ri rock is formed by two exposed rocks and surrounding reef. The artificial reef zone made by about 200 ~ 300 structures is shown the western area of the Sib-Ri rock. The underwater rock region is extended from the southwestern area of the exposed the Sib-Ri rock with 9 ~ 11 m depth range. The most broad rocky seabottom area is located in the southwestren area of the Sib-Ri rock with 10 ~ 13 m depth range. The study area were classified into 4 types of seabottom environment based on the analysis of bathymetric data, seafloor mosaics, composition of sediments and images(underwater and drone). The underwater rock zones(Type I) are the most distributed area around the Sib-Ri Rock(about 600 m × 600 m). The soft seabottom area made by sediments layer showed 2 types(Type II: gS(gravelly Sand), Type III: S(Sand)) in the areas between underwater rock zones and western part of the Sib-Ri rock(toward Gyeongpo Beach). The artificial reef zone with a lot of structures is located in the western part of the Sib-Ri rock. Marine algae(about 6 species), Phylum porifera(about 2 species), Phylum echinodermata(about 3 species), Phylum mollusca(about 3 species) and Phylum chordata(about 2 species) are dominant faunal group of underwater image analysis area(about 10 m × 10 m) in the northwestern part of the Sib-Ri rock. The habitat of Phylym mollusca(Lottia dorsuosa, Septifer virgatus) and Phylum arthropoda(Pollicipes mitella, Chthamalus challengeri hoek) appears in the intertidal zone of the Sib-Ri rock. And it is possible to estimate the range and distribution of the habitat based on the integrated study of orthphoto(drone) and bathymetry data. The integrated visualization and mapping techniques using seafloor mosaic images, sediments analysis, underwater images, orthophoto(drone) and topographic data can provide and contribute to figure out the seabottom conditions and characteristics in the shore of the East Sea.

Marine Algal Flora and Grazing Effect of Sea Urchins in the Coastal Waters of Cheju Island (제주연안의 해조상과 성게의 섭식효과)

  • The structure of benthic algal flora and feeding behavior of sea urchins (Anthocidaris crassispina and Hemicentrotus pulcherrimus), were investigated from the eight different locations along the coast of Cheju Island, Korea. Biomass distribution and dominance of the algal communities were studied in relation to the gut contents and population parameters of the sea urchins. As a result, although the largest algal biomass and biological characteristics of sea urchins, such as test diameter, test hight, body weight, gonad weight, etc. This inhdicated that sea urchins would not be dependednt sorely on the abundance of algal foods, and they might feed on other food items. In order to know such a trend with regard to food selectivity, the diets consumed by sea urchins were analyzed, and it was found that they were composed of benthic and drifted algae, various animal components (sponges, bryozoans, crustaceans, and various unidentified capsulated eggs, etc.) and biets turned out to be smaller than might be expected, and it is postulated that sea urchins were not likely to be the major grazing animals for the formation of algal bed and also canopy deterioration, so-called "white incrustation."

High fructose and high fat diet increased bone volume of trabecular and cortical bone in growing female rats (고과당 및 고지방 식이의 섭취가 성장기 동물모델의 골성장과 골성숙에 미치는 영향)

  • Purpose: The objective of this study was to investigate the effects of a high fructose and fat diet on bone growth and maturation in growing female rats. Methods: Three-week-old female SD rats were randomly assigned to four experimental groups; the control group (CON: fed control diet based on AIN-93G, n = 8); the high-fructose diet group (HFrc: fed control diet with 30% fructose, n = 8); the high-fat diet group (Hfat: fed control diet with 45 kcal% fat, n = 8); and the high-fat diet plus high fructose group (HFrc + HFat: fed diets 45 kcal% fat with 30% fructose, n = 8). Each group was assigned their respective diets for the remaining eight weeks. Bone-related parameters (bone mineral density (BMD) and structural parameters, osteocalcin (OC), deoxypyridinoline (DPD)) and morphologic changes of kidney were analyzed at the end of the experiment. Results: Final body weights and weight gain were higher in the HFat and HFrc + HFat groups and showed higher tendency in the HFrc group compared with those of the CON group (p < 0.05); however, no significant difference in caloric intake was observed among the four experimental groups. The serum OC levels of the HFrc and HFrc + HFat groups were lower than those of the CON and HFat groups (p < 0.05). Urinary levels of DPD did not differ among the experimental groups. BV/TV and Tb.N of trabecular bone were higher in the HFrc + HFat group and showed a higher tendency in the HFrc group than those of the CON and HFat groups (p < 0.05). Tb.Pf of trabecular bone were lower in the HFrc + HFat group than those in the CON and HFat groups (p < 0.05). However, no difference in trabecular BMD was observed among the experimental groups. Cortical bone volume was higher in the HFat and HFrc + HFat groups than in the CON and HFrc groups (p < 0.05). No morphology change in kidney was observed among the experimental groups. Conclusion: Our study suggests that 8 weeks of high-fructose and high fat intake could improve the bone quality (Structural parameters) of trabecular and cortical bone of tibia in growing female rats.

Aquatic animal health management in stock enhancement (방류용 수산종묘의 질병 관리에 대한 고찰)

  • Ministry of Agriculture, Fisheries and Food (MAFF) and National Fisheries Research and Development Institute (NFRDI) have inspected the hatchery-reared seeds of 22 marine species and 11 freshwater species for aquatic animal diseases in stock enhancement program in 2009. Results showed that total 12 local selfgovernments have been restocking the sea with cultured juveniles. Gyeongsangnam-do, Jeollanam-do, Jejudo and Chungcheongnam-do have a preference for marine species seeds to freshwater species. On the contrary, freshwater species were released mostly in Gyeonggi-do, Jeollabuk-do and Chungcheongbuk-do. In the marine species group, abalone was the most abundant as 24.5%, and then sea cucumber (15.2%), olive flounder (11.5%), swimming crab (5.6%), black sea bream and rockfish (6.8%), rock bream (5.1%), black rockfish (4.6%) and scorpionfish (4.5%) were followed. Crucian carp was the most abundant as 19.4%, and then eel (17.0%), Korean bullhead (12.3%), melanian snail (12.0%), catfish (8.4%) were followed in the freshwater species group. The total number of inspection cases in this study were 1,080 and disqualification cases were 19 by detection of aquatic animals pathogens such as red sea bream iridovirus (RSIV), koi herpesvirus (KHV) or white spot syndrome virus (WSSV).

Detection of fish pathogens in cultured juveniles for stock enhancement in 2010 (2010년 방류용 수산종묘에 대한 병원체 검출)

  • Aquatic animal raised in hatcheries play an important role in supplying seedling to stock enhancement and seed quality, especially, seed health is the key factor for survival in the field after release and for stocking effectiveness. We have inspected the hatchery-reared seeds of 33 marine species and 12 freshwater species for legally designated diseases in stock enhancement program in 2010. Results showed that abalone was the most abundant as 20.0% in the marine species group and then sea cucumber (15.6%), olive flounder (8.4%), rockfish (6.7%), black sea bream (6.3%) and swimming crab (6.1%) were followed. Crucian carp was the most abundant as 19.4%, and then eel (11.8%), Korean bullhead (10.9%), mandarin fish (10.8%), melanian snail (8.4%), catfish (7.7%) were followed in the freshwater species group. The total number of inspection cases for eight pathogens in this study were 2,105 and disqualification cases were 30 by detection of aquatic animals pathogens such as koi herpesvirus (KHV), red sea bream iridovirus (RSIV), white spot syndrome virus (WSSV) or viral haemorrhagic septicemia virus (VHSV).

Anti-proliferation Effect of Coscinoderma sp. Extract on Human Colon Cancer Cells (Coscinoderma sp.의 대장암세포 증식 억제 효과)

  • Natural products are attractive as the source of new drug development. Especially, numerous unknown marine bioresources are an object of attention because the ocean occupies three fourth of the earth. Survival of marine bioresources in extreme environment may induce the production of biological active compounds. As previous study, we examined over 40 specimens of marine sponges collected from Micronesia and screened their anti-proliferative activities in various cancer cell lines. Among them, we investigated Coscinoderma sp.'s activity and mechanism in human colon carcinoma HCT116 and RKO cells. Furthermore, we also used the p53-knockout of HCT116 cells and the p53 loss of RKO cells for elucidating the role of p53. Coscinoderma sp. inhibited cellular viability independently of the p53 status. Therefore, we compared the expression level of cell death-related proteins by Coscinoderma sp. in HCT16 and in HCT116 p53KO cells. Coscinoderma sp. increased p53 level and NOXA levels and induced apoptosis under the condition of p53 existence. On the other hand, Coscinoderma sp. increased p21 and mTOR levels in HCT116 p53KO cells. These results suggest that Coscinoderma sp. induced anti-proliferation effect through different pathway depending on p53 status.

Inhibitory Effect of RANKL-Induced Osteoclast Differentiation and ROS Generation by Sphaerotylus antarcticus Extract (극지해면동물 Sphaerotylus antarcticus 추출물의 RANKL 유도 파골세포 분화 및 ROS 생성 억제 효과)

  • In order to maintain bone homeostasis, it is necessary to balance bone resorption and remodeling through the differentiation of osteoclasts that absorb old bone and osteoblasts that form new bone. However, bone resorption due to excessive osteoclast differentiation is a major cause of osteoporosis and controlling excessive osteoclast differentiation has been known as a treatment strategy for osteoporosis. Therefore, in this study, the effect of an ethanol extract of Sphaerotylus antarcticus Kirkpatrick, 1907 (SAE), polar-derived sponge with unknown biological activity, on the osteoclast differentiation process of RANKL-induced RAW264.7 cells and the generated ROS was evaluated. In the study results, SAE down-regulated the formation and function of RANKL-induced osteoclasts and osteoclast differentiation specific proteins, genes in a concentration-dependent manner. In addition, it was possible to confirm the result of restoring the lost antioxidant enzyme along with down-regulation of ROS generated by RANKL. Therefore, in this study, we propose the possibility of SAE as a potential regulator of osteoporosis due to excessive osteoclast differentiation and report the biological value of the diversity of marine-derived natural products by identifying the first biological activity against SAE that is not yet known.
