Production, Characterization, and Variable Region Analysis of Monoclonal Antibodies Specific for Hepatitis B Virus S Antigen (Hepatitis B Virus의 S항원에 특이적인 단세포군 항체 생산, 특성 연구 및 가변지역유전자 분석)

  • Background: Hepatitis B virus (HBV) infection is one of the worldwide public health problem affecting about 300 million people. The envelope protein of HBV consists of three components known as preS1, preS2, and S antigen. According to the recent study, anti-HBs Ab showed effective neutralization ability against HBV from chronic hepatitis B and liver transplant patients, suggesting the possible development of therapeutic antibody. Methods: Spleen cells immunized with S antigen of HBV were fused with myeloma cell line to obtain HBsAg specific monoclonal antibodies. High affinity antibodies against HBsAg (adr, ad and ay type) were selected by competitive ELISA method. Nucleotide sequence of the variable regions of monoclonal antibodies was analyzed by RT-PCR followed by conventional sequencing method. Results: We produced 14 murine monoclonal antibodies which recognize S antigen of HBV. Two of them, A9-11 and C6-9 showed the highest affinity. The sequence analysis of A9-11 revealed that variable regions of the heavy chain and light chains are members of mouse heavy chain I (B) and light chain lambda 1, respectively. Likewise, the sequence analysis of C6-9 revealed that variable regions of the heavy chain and light chains are members of mouse heavy chain II (B) and light chain kappa 1, respectively. Neutralization assay showed that A9-11 and C6-9 effectively neutralize the HBV infection. Conclusion: These results suggest that A9-11 and C6-9 mouse monoclonal antibodies can be used for the development of therapeutic antibody for HBV infection.

  • To clarify characteristics of yolk protein of abalone, yolk protein was purified from the ovarian egg extracts of mature female Haliotis discus hannai by a gel chromatography of sepharose CL-4B. From the results of immuno-electrophoresis and Ouchterlony's diffusion test to male and female sera and ovarian egg extracts using antibodies raised against mature female and male sera and male sera and ovarian egg extrascts, it was identified that the mature female serum had female specific serum protein and its antigenecity shared with ovarian egg extracts. A single type of yolk protein was purified from ovarian egg extracts, and it was composed of two subunits. Their molecular weights were estimated to be approximately 166 KDa and 113 KDa by SDSPAGE. The antiserum against yolk proteins cross-reacted with a mature female specific serum protein and extracts of hepatopancreas of vitellogeing females, but did not reacted with extracts of hepatopancreas of mature male.

  • In this paper, an optimal design method of clonal selection based Fuzzy-Neural Networks (FNN) model for complex and nonlinear systems is presented. The FNNs use the simplified inference as fuzzy inference method and Error Back Propagation Algorithm as learning rule. Also Advanced Clonal Selection (ACS) is proposed to find the parameters such as parameters of membership functions, learning rates and momentum coefficients. The proposed method is based on an Immune Algorithm (IA) using biological Immune System and The performance is improved by control of differentiation rate. Through that procedure, the antibodies are producted variously and the parameter of FNN are optimized by selecting method of antibody with the best affinity against antigens such as object function and limitation condition. To evaluate the performance of the proposed method, we use the time series data for gas furnace and traffic route choice process.

Isolation of Non-toxic Bacillus thuringiensis Strains from the Dead Larvae of Apriona germari and Aphodius apicalis (뽕나무하늘소(Apriona germari) 및 왕똥풍뎅이 (Aphodius apicalis) 사충으로부터 무독성 Bacillus thuringiensis의 분리)

  • Four strains of Bacillus thuringiensis were isolated froin the dead larvae of mulberry longicorn beetle (Apriong germari) and dung beetle (Aphodius apicalis). One nf four B. thuringiensis isolates turned out to be subspecies darinstadiensis but the remains were not identified using 33 B. thuringiensts flgellar ( H ) antibodies. Furthermore. bioassays of spore-parasporal inclusion protein mixture conducted against third instar larvae of A. gerrntrri or A. apicalis, second instar larvae of Bombyx mori, and third instar larvae of Cu1ex pipiens pullens showed that the isolates were non-toxic. To further confirm, four isolates were characterized and analysed by SDS-PAGE and agarose gel electrophoresis. The results revealed that parasporal protein and plasmid DNA patterns of four isolates are different from those of darmstadiensis and 20 known non-toxic B. thuringiensis strains, suggesting that the four isolates are novel non-toxic B. thuringiensis.

  • A 65-year-old man was admitted due to low grade fever, dry coughing, and dyspnea on exertion. The chest radiograph and CT scan showed diffuse ground glass opacities and small nodules in the both lung fields resulting in a diagnosis of severe interstitial pneumonia. Conservative treatment with antibiotics and bronchodilators decreased the symptoms, but the dyspnea and cough reappeared when he returned home. An inspection of his house revealed the presence of fungi under the wallpaper. His symptoms disappeared completely after these were removed. His clinical course raised the suspicion of hypersensitivity pneumonitis and these fungi believed to be the cause of hypersensitivity pneumonitis. The histological findings of a lung specimen by video-assisted thorachoscopy were compatible with hypersensitivity pneumonitis. The fungi were identified as Alternaria.

Anti-inflammatory Effects of Hataedock with Douchi in Atopic Dermatitis-like Skin Lesions in House Dust Mite-Induced NC/Nga Mice (PKC 활성 조절을 통한 두시 하태독법의 항염증작용이 Mite 항원 유도 아토피유사피부염 발병 조절에 미치는 효과)

  • Objectives Hataedock (HTD) is an oral Korean herbal medical oral treatment that removes fetal toxic heat and meconium from new born babies. The purpose of this study is to evaluate whether Hataedock treatment of Duchi extracts has anti-inflammation effects in atopic dermatitis-like skin lesions in House Dust Mite-Induced NC/Nga Mice. Methods The mice were divided into 3 groups (n=10 per group) as follows: the control group (Ctrl group), AD-induced group (AE group), AD-induced with HTD treatment group (DT group). 3-week-old NC/Nga mice were introduced to Hataedock treatment, made of Duchi extract. After 4 weeks, House Dust Mite-Induced application was used six times per week for 3 weeks to induce the first atopic dermatitis, and second AD in 7 weeks after. To examine skin injuries and anti-inflammatory effect, PKC, MMP-9, iNOS immunohistochemistry were used. Results The alleviate effect of the skin damage and angiogenesis was observed in DT group. The damage of stratum corneum, hyperplasia, edema, infiltration of lymphocytes and distribution of capillary were decreased in DT group. Also, the study results suggested that Hataedock treatment made of Duchi extracts in DT group remarkably decreased skin damages by 51% (p < 0.001), as well as PKC by 91%, MMP-9 by 48% (p < 0.001), iNOS by 51% (p < 0.001). Conclusions Based on the study results, we observed that Hataedock treatment of Duchi extracts alleviates AD by diminishing various inflammatory cytokines, initial steps of AD development, in the skin lesions. Potential applications for prevention and treatment of atopic dermatitis are expected.

Expression of Antibody Genes Specific for Human Hepatitis-B Virus in Transgenic Tabacco Plants (형질전환된 담배에서 사람 B형 간염바이러스 항체 유전자의 발현)

  • Chimeric kappa chain and gamma chain cDNA clones (pCKS2 and pCHS2) of a monoclonal antibody specific for pre-S2 surface antigen of human hepatitis-B virus were ligated into Xbal site of plant expression vector pBKS-1. Plasmid DNA containing each of the chimeric gene were then mobilized from E, coli to Agrobacterium tumefaciens strain LBA4404. The chimeric antibody genes were then introduced into tobacco by Ti plasmid-mediated transformation. The putative Transformants were selected on medium containing kamaycin sulfate. Shoots that formed on shoot induction medium were analyzed by Western blot analysis for the expression of kappa-chain or gamma-chain genes. The Western blot analyses clearly showed that the introduced genes were stably expressed in transgenic plants.

  • The present investigation demonstrates HLA-DR and PGP9.5 double positive cells accumulate thymus cortical region in normal baboon thymus and baboon lung. But, these cells disappeared in thymus and lung of bronchopulmonary dysplasia (BPD) animals. 125d GC animal model is more suitable for BPD than 140d GC animal. Anti-bombesin antibody, 2A11 treated baboon recover normal level of HLA-DR positive cells from BPD animal. In addition, thymocytes show responsiveness for bombesin. These observation suggest that blocking BLPs protects a chronic lung injury by BPD and 2A11 is possible agent for passive therapy of BPD.

Reduction in antigenesity of whey protein by alcalase (Alcalase에 의한 유청단백질 가수분해물의 항원성 저감 효과)

  • The aim of this study was to produce enzymatic hydrolysis of ${\alpha}$-LA, ${\beta}$-LG and BSA with alcalase for the possible application of hypoallergenic foods toward cow's milk allergenic infant. The molecular weights of most of the peptides in hydrolysates from ${\alpha}$-LA, ${\beta}$-LG and BSA by alcalase were below 3,000 dalton. Antigenesity of ${\alpha}$-LA, ${\beta}$-LG and BSA hydrolysates to rabbit anti-${\alpha}$-LA antiserum, ${\beta}$-LG antiserum and BSA antiserum were remarkably decreased by more than $10^{-3}$ at 20% inhibitionrate. Antigenesity of polyvalent antigenic peptide in ${\alpha}$-LA, ${\beta}$-LG and BSA hydrolysates to specific rabbit anti-${\alpha}$-LA antiserum, ${\beta}$-LG antiserum and BSA antiserum was determined by PCS test using guina-pig. Hydrolysates of ${\alpha}$-LA, ${\beta}$-LG and BSA with less than 3,000 dalton did not show polyvalent antigenic reaction against rabbit antiserum. Hydrolysates of ${\alpha}$-LA, ${\beta}$-LG and BSA could be a source for the manufacturing of hypoallergenic food.

A research into perceptionality of students of the dental hygiene department on hepatitis B virus (치위생과 학생들의 B형 간염 바이러스에 대한 인지도 조사연구)

  • This study aims to furnish basic data about prevention and infection control of Hepatitis B Virus(HBV) for those who, working in dental offices, are particularly exposed to a high risk of HBV infection. A survey was conducted to 310 students including freshmen, sophomores and juniors enrolled in the dental hygiene department in order to examine their knowledge about infection routes of HBV, clinical history of their family members and their own health. The outcomes of the survey showed following facts; 1. Students were found to lack knowledge about the present conditions of their HBsAg and HBsAb of HBV(PF0.05), conduct of preventive vaccination(PE0.05), completion of 3 required vaccinations(PF0.05) and formation of antibody(PF0.05). 2. Students named "blood"(88.6%) and "infected needles"(82.5%) as most likely infection routes of HBV(PE0.05 and PE0.01). These replies came mostly from sophomores(65.6% and 92.1%), followed by juniors(89.2%, 82.5%) and freshmen(81.1%, 73.0%). Least knowledge about infection routes of HBV was sensed with the reply "infection through breast-feeding of positive mother(27.9%)"(PE0.05). Generally, sophomores seemed to have much knowledge about infection routes, followed by juniors and freshmen in order. 3. As to clinical history of family members, 10 students(3.5%) replied that any of their family members is suffering from HBV now, 8(2.6%) revealed that some of their family members once suffered from it and 10(3.2%) reported cases of death of their family members from liver diseases. 4. Ninety-four point seven percent of respondents believed their health to be better than normal.
